2.Ultrasound Diagnosis and Misdiagnosis Analysis of Accessory Cavitated Uterine Malformation

To explore the ultrasound characteristics of accessory cavitated uterine malformation (ACUM) and the causes of misdiagnosis, in order to better understand the disease and improve the diagnostic ability of radiologists. We retrospectively collected clinical, pathological and imaging data of ACUM patients who were diagnosed after surgery at Peking Union Medical College Hospital from December2013 to December 2023. Besides, we analyzed ultrasound features of ACUM and the cause of misdiagnosis. A total of 11 patients were included, with a mean age of diagnosis of (27.1±7.4)years. There was a mean interval of (6.1±5.5)years between menarche and onset of dysmenorrhea. Preoperative ultrasound showed that ACUM appeared as solitary lesions located under the insertion of the round ligament, with a mean maximum diameter of (3.0±1.0)cm. Lesions were round or oval (90.9%, 10/11) and not connected to the uterine cavity (100%, 11/11) with clear boundaries (72.7%, 8/11), presenting as regular thick-walled cystic-solid structures. Cystic areas were observed within the mass (100%, 11/11) with a ground-glass-like appearance (90.9%, 10/11) and hyperechoic endometrial lining (90.9%, 10/11). The surrounding of the mass showed hypoechoic or isoechoic areas resembling the muscular layer and circular or semi-circular vascularity was detected around the mass. No adenomyosis or other uterine lesions were found. Analysis of misdiagnosis: Among the 11 ACUM cases, 6 were correctly diagnosed by the initial ultrasound examination, and 5 cases by MRI. Two patients were misdiagnosed as residual horn uterus by MRI, despite ultrasound suggesting ACUM with visible bilateral uterine horns. ACUM exhibits distinct ultrasound characteristics. When young women present with progressive dysmenorrhea and ultrasound reveals a regular cystic-solid mass with thick wall located under the round ligament insertion, ACUM should be considered.
3.Research advances on in-situ cell electrospinning and its application in wound repair

Currently, there are limited strategies for convenient and rapid wound repair in clinical practice. In recent years, in-situ cell electrospinning (IS-CE) technology, developed from in-situ electrospinning (IS-E) technology, has emerged. IS-CE technology involves encapsulating living cells within micro-nanofibers to construct living fibrous tissue scaffolds in situ, making some progress in wound repair applications. However, this technology still faces limitations such as low cell survival rate and poor fiber stability. This article provides a comprehensive review on the current status of both IS-E and IS-CE technologies, as well as the application of IS-CE technology in wound repair. In addition, the advantages, limitations, and improvement methods of IS-CE technology applied in wound treatment are emphatically discussed, aiming to provide insights for its application in tissue engineering and wound repair.
4.Expression levels of gastrointestinal hormones in the progression of liver fibrosis in patients with nonalcoholic fatty liver disease

Objective To investigate the changes in gastrointestinal hormones during the progression of liver fibrosis in patients with nonalcoholic fatty liver disease (NAFLD), and to provide a basis for digestive function impairment. Methods A prospective analysis was performed for 326 patients with NAFLD who attended the outpatient service and were hospitalized and treated in Ruikang Hospital Affiliated to Guangxi University of Chinese Medicine from October 2018 to June 2020, and FibroTouch was used to measure liver stiffness measurement (LSM). According to the presence or absence of liver fibrosis, they were divided into non-liver fibrosis group (group A, 161 patients with LSM < 7.3 kPa) and liver fibrosis group (group B, 165 patients with LSM ≥7.3 kPa). According to the fibrosis degree, the patients were further divided into F0-1 group (LSM < 7.3 kPa), F2 group (7.3 kPa ≤LSM < 9.7 kPa), F2-3 group (9.7 kPa ≤LSM < 12.4 kPa), F3-4 group (12.4 kPa ≤LSM < 17.5 kPa), and F4 group (LSM ≥17.5 kPa). Related data were collected, including age, sex, liver function parameters, and gastrointestinal hormones. The independent samples t -test and the one-way analysis of variance were used for comparison of normally distributed continuous data between groups, and the nonparametric Mann-Whitney U test and the Kruskal-Wallis H test were used for comparison of non-normally distributed continuous data between groups. A Spearman correlation analysis was used to investigate the correlation between LSM and liver function parameters. Results Comparison of liver function and gastrointestinal hormones showed that there were significant differences between groups A and B in alanine aminotransferase (ALT) ( Z =-3.778, P < 0.001), aspartate aminotransferase (AST) ( Z =-3.320, P =0.001), gamma-glutamyl transpeptidase (GGT) ( Z =-3.040, P =0.002), cholecystokinin (CCK) ( t =-2.944, P =0.003), and lipopolysaccharide (LPS) ( Z =-2.317, P =0.020). There were significant differences in ALT ( χ 2 =23.113, P < 0.001), AST ( χ 2 =23.415, P < 0.001), ALP ( χ 2 =15.962, P =0.003), GGT ( χ 2 =20.172, P < 0.001), and CCK ( F =2.687, P =0.031) between the F0-1 group with 161 patients, the F2 group with 89 patients, the F2-3 group with 46 patients, the F3-4 group with 16 patients, and the F4 group with 14 patients. LSM was positively correlated with direct bilirubin, ALT, AST, alkaline phosphatase, and GGT ( r =0.128, 0.266, 0.225, 0.137, and 0.213, all P < 0.05). Conclusion Liver fibrosis progression in NAFLD can affect gallbladder contraction function and gastrointestinal function, and measurement of the serum levels of CCK and LPS has an important clinical value in the early diagnosis and treatment of digestive diseases related to gallbladder contraction function and gastrointe stinal function in NAFLD patients with liver fibrosis.
5.Association between liver fibrosis progression and endothelin-1/nitric oxide in patients with nonalcoholic fatty liver disease

Objective To investigate whether the progression of liver fibrosis affects endothelial function in patients with nonalcoholic fatty liver disease (NAFLD), and to early identify the warning of cardiovascular diseases caused by endothelial dysfunction by liver fibrosis progression. Methods A total of 280 patients who attended the outpatient service or were hospitalized in Department of Liver Disease, Ruikang Hospital Affiliated to Guangxi University of Chinese Medicine, from April 2019 to October 2020 were enrolled, and they were diagnosed with fatty liver disease by ultrasound and met the diagnostic criteria for NAFLD. General information and related serological markers were collected and recorded. FibroTouch technique was performed for the NAFLD patients diagnosed by ultrasound to record their fat attenuation parameter (FAP) and liver stiffness measurement (LSM), and according to LSM, the patients were divided into non-progressive fibrosis group (239 patients with LSM < 11 kPa) and progressive fibrosis group (41 patients with LSM ≥11 kPa) to analyze the association between liver fibrosis progression and endothelin-1 (ET-1)/nitric oxide (NO) in NAFLD. The t -test or the Mann-Whitney U test was used for comparison of continuous data between two groups, and the Spearman method was used for correlation analysis. Results There were no significant differences between the non-progressive fibrosis group and the progressive fibrosis group in the expression levels of ET-1( Z =-0.190, P =0.849) and NO( Z =-1.509, P =0.131), and there were significant differences between the two groups in body mass index (BMI), alanine aminotransferase (ALT), aspartate aminotransferase (AST), gamma-glutamyl transpeptidase (GGT), triglyceride (TG), and high-density lipoprotein cholesterol (HDL-C) ( Z =-3.977, -4.162, -3.471, -3.201, -3.202, and -3.311, all P < 0.05). The Spearman analysis showed that LSM was not correlated with ET-1, NO, and NO/ET-1 ( r s =-0.046, 0.086, and 0.104, all P > 0.05). Further analysis of the correlation of ET-1 and NO with each index showed that ET-1 was not correlated with age, NO, ALT, AST, GGT, total cholesterol, TG, HDL-C, low-density lipoprotein cholesterol (LDL-C), FAP, and BMI ( r s =-0.017, 0.054, -0.067, -0.016, -0.031, 0.004, 0.051, -0.084, -0.030, 0.080, and 0.044, all P > 0.05), and NO was not correlated with age, ET-1, ALT, AST, GGT, total cholesterol, TG, HDL-C, LDL-C, FAP, and BMI ( r s =0.004, 0.054, 0.011, 0.052, 0.004, -0.051, -0.052, -0.012, -0.076, -0.013, and -0.021, all P > 0.05). Conclusion This study shows that liver fibrosis progression in NAFLD has no impact on ET-1 and NO, suggesting that fibrosis progression may have no influence on endothelial function.
6.Research advances on the mechanism of fractional laser in treating scars

Scars are the result of abnormal repair of skin tissue trauma. Recently, fractional laser is more and more widely used in the treatment of scars, but its mechanism is not clear. Studies have shown that fractional laser could produce multiple microthermal zones in target skin, induce wound repair responses, affect the function of epidermal and dermal cells, induce changes in blood vessels and collagens, and change the expression of heat shock proteins, microRNA, matrix metalloproteinases, cytokines such as transforming growth factor β, basic fibroblast growth factor, and facilitate drug delivery, thus achieving the effect of treating scars. This article reviews the mechanism of fractional laser in treating scars from three aspects, including the tissue and cell mechanism, molecular mechanism, and drug delivery.
7.Effect of FOXK2 overexpression on malignant biological behaviors of ovarian cancer SK-OV-3 cells

[Abstract] Objective: To investigate the effects of forkhead box transcription factor (FOXK2) overexpression on the proliferation, migration, invasion and adhesion of human ovarian cancer SK-OV-3 cells and its related molecular mechanism. Methods: The open reading frame (ORF) of FOXK2 was cloned into lentivirus expression vector, which was then enveloped in HEK293T cells and transfected into human ovarian cancerSK-OV-3cells.TheoverexpressionefficiencywasdetectedbyqPCRandWesternblotting.Theproliferation, migration, invasion and adhesion of SK-OV-3 cells were detected by CCK-8, Scratch-healing, Transwell and Cell adhesion assays respectively, and the expressions of epithelial-mesenchymal transition (EMT) markers were detected by qPCR. Results: The FOXK2 overexpression vector was constructed successfully and packaged into lentivirus, which was then transfected into SK-OV-3 cells. After transfection, the expression of FOXK2 was significantly increased (P<0.01); the proliferation, migration and invasion of SK-OV-3 cells were significantly reduced while the adhesion ability was significantly increased (P<0.05 or P<0.01); and the expression levels of E-cadherin and β-catenin were significantly increased while that of vimentin and fibronection were significantly decreased (all P<0.01). Conclusion: Overexpression of FOXK2 in SK-OV-3 cells leads to a significant decrease in proliferation, migration and invasion but increase in adhesion. The molecular mechanism may be related to the reversion of the EMT process in tumor cells, suggesting that FOXK2 may be a potential target for the diagnosis and treatment of ovarian cancer.
8.A study on the mechanism of mi-362 targeting six1 inhibiting proliferation and migration of cervical cancer cells

Objective To investigate the mechanism of Mi-362 targeting Six1 inhibiting proliferation and migration of cer-vical cancer cells. Methods The expression levels of microRNA-362 in cervical cancer tissues and adjacent tissues were measured in 142 patients with cervical cancer in our hospital. At the same time,Hela cancer cell group,microRNA-362 mimics group and mi-croRNA-362 inhibitor group were set up to determine the viability of cancer cells,the number of monoclonal formation of cancer cells,the apoptotic rate of cancer cells,cell cycle,the number of perforations,and the levels of microRNA-362 and Six1 in cervical cancer fluid of Hela. Results The expression level of miR-362 in cervical cancer tissue was lower than that in adjacent tissue(P<0. 05). The higher the infiltration of lymphatic vessel space,pathological stage,TNM stage,lymph node metastasis and depth of infil-tration,the lower the expression rate of miR-362(P<0. 05). The OD value and survival rate in the miR-362 mimics group were lower than those in the Hela cancer cells group(P<0. 05),while the OD value and survival rate in the mir-362 inhibitor group were higher than those in the Hela cancer cells group and the miR-362 mimics group(P<0. 05). The number of clones formed in the miR-362 mimics group was lower than that in the Hela cancer cell group(P<0. 05),and the number of clones formed in the miR-362 inhibitor group was higher than that in the Hela cancer cell group and the miR-362 mimimics group(P<0. 05). The apoptotic rate of miR-362 mimics group was higher than that of Hela cancer cell group(P<0. 05),and that of miR-362 inhibitor group was lower than that of Hela cancer cell group and miR-362 mimics group(P<0. 05). The G1 phase in the miR-362 mimics group was higher than that in the Hela cancer cell group(P<0. 05),and the G1 phase in the miR-362 inhibitor group was lower than that in the Hela cancer cell group and the miR-362 mimimics group(P<0. 05). The number of cell membrane penetration in the miR-362 mimics group was lower than that in the Hela cancer group(P < 0. 05),and that in the miR-362 inhibitor group was higher than that in the Hela cancer group and the miR-362 mimimics group(P<0. 05). The expression level of miR-362 in the miR-362 mimics group was higher than that in the Hela cancer cell group(P<0. 05),and the expression level of miR-362 in the miR-362 inhibitor group was lower than that in the Hela cancer cell group and the miR-362 mimics group(P<0. 05). The expression level of Six1 in the miR-362 mimics group was lower than that in the Hela cancer cell group(P<0. 05),and that in the miR-362 inhibitor group was higher than that in the Hela cancer cell group and the miR-362 mimics group(P<0. 05). Conclusion miR-362 plays an important inhibition in the occurrence and development of cervical cancer,and its mechanism is related to the inhibition of prolifer-ation,migration and invasion of cancer cells by microRNA-362 through negative regulation of Six1.
9.Improvement Effects of Water Extract of Glycyrrhiza uralensis on Acute Hepatic Injury Caused by Triptolide and Its Effects on the Levels of IL- 10 and TNF-α in Rats

OBJECTIVE: To observe the improvement effects of water extract of Glycyrrhiza uralensis on acute hepatic injury caused by triptolide and its effects on the levels of IL-10 and TNF-α in rats. METHODS: A total of 60 healthy male SD rats were randomized into blank control group, model group, positive control group (magnesium isoglycyrrhizinate) and G. uralensis water extract low-dose, middle-dose and high-dose groups. Blank control group and model group were given isochoric normal saline intragastrically; positive control group was given Magnesium isoglycyrrhizinate liquid intragastrically (13.5 mg/kg); G. uralensis water extract low-dose, middle-dose and high-dose groups were given G. uralensis water extract intragastrically 1 mL (120, 240, 480 mg/kg, by the amount of extract), qd, for consecutive 7 d. At 8th day of administration, model group, positive control group and G. uralensis water extract groups were given Triptolide liquid intragastrically (0.6 mg/kg) to establish acute liver injury model. HE staining was used to detect the pathological changes of liver tissue in rats. The activities of ALT and AST in serum were determined by enzyme coupling reaction. The protein expression of IL-10 and TNF-α in liver tissue were analyzed by Western blotting. RESULTS: Compared with blank control group, obvious pathological changes of liver tissue in rats were observed in model group; serum activities of ALT and AST, the protein expression level of TNF-α in liver tissue were increased significantly, while the protein expression level of IL-10 was decreased significantly (P<0.01). Compared with model group, pathological changes of liver tissue were relieved significantly. Except for the activity of AST in serum of rats in G. uralensis water extract low-dose group, the activities of ALT and AST in serum, the protein expression level of TNF-α in liver tissue were decreased significantly in G. uralensis groups, while the protein expression level of IL-10 in liver tissue was increased significantly (P<0.05 or P<0.01). CONCLUSIONS: G. uralensis water extract can relieve triptolide-induced acute liver injury in rats, the mechanism of which may be associated with expression down-regulation of pro-inflammatory factor TNF-α and expression up-regulation of anti-inflammatory factor IL-10.
10.Effects of Different Polar Parts of 5 Kinds of Alpinia on Function of Sympathetic-adrenal System in Gastric Ulcer Model Rats with Cold Syndrome

OBJECTIVE: To investigate the effects of different polar parts of 5 kinds of Alpinia on the function of sympathetic-adrenal system in gastric ulcer model rats with cold syndrome, such as the root of Alpinia officinarum, the rhizome and fruit of Alpinia galangal, the seed of Alpinia katsumadai, and the fruit of Alpinia oxyphylla. METHODS: SD rats were given Anemarrhena asphodeloides decoction at 4 ℃ and Glacial acetic acid solution intragastrically to induce gastric ulcer model with cold syndrome; the model rats were randomly divided into model group, Fuzi lizhong pills group (positive control, 9.0 g/kg), cimetidine group (positive control, 0.003 3 g/kg), low-dose and high-dose groups of petroleum ether, ethyl acetate, n-butanol and water extraction parts from 5 medicinal materials (hereinafter referred to as “gaoshidi” “gaoshigao” “dashidi” “dashigao” “hongshidi” “hongshigao” “caoshidi” “caoshigao” “yishidi” “yishigao” “gaoyidi” “gaoyigao” “dayidi” “dayigao” “hongyidi” “hongyigao” “caoyidi” “caoyigao” “yiyidi” “yiyigao” “gaozhengdi” “gaozhenggao” “dazhengdi” “dazhenggao” “hongzhengdi” “hongzhenggao” “caozhengdi” “caozhenggao” “yizhengdi” “yizhenggao” “gaoshuidi” “gaoshuigao” “dashuidi” “dashuigao” “hongshuidi” “hongshuigao” “caoshuidi” “caoshuigao” “yishuidi” “yishuigao”, 0.064/0.256, 0.032/ 0.128, 0.008/0.032, 0.075/0.3, 0.1/0.4, 0.064/0.256, 0.108/0.432, 0.16/0.64, 0.064/0.25, 0.125/0.5, 0.056/0.224, 0.108/0.432, 0.08/0.32, 0.2/0.8, 0.3/1.2, 0.14/0.56, 0.032/0.128, 0.028/0.112, 0.05/0.2, 0.087/0.348 g/kg, by mass of extraction parts), with 10 rats in each group; and the blank group (normal temperature water) was set up. After last medication, urine contents of 17-OHCS, CAs substances (A, NE, DA) and serum contents of ACTH and D-β-H were determined by ELISA. RESULTS: Compared with blank group, the contents of 17-OHCS, A, NE and DA in urine, the contents of ACTH and D-β-H in serum were decreased significantly in model group (P<0.01). Compared with model group, the contents of 17-OHCS (Fuzi lizhong pills group, cimetidine group, gaoshidi, gaoshigao, dashigao, hongshigao, caoshigao and yishigao groups, ethyl acetate part groups of 5 medicinal materials, dazhenggao, hongzhenggao, caozhenggao and yizhenggao groups, gaoshuigao, dashuigao and hongshuigao groups), A (Fuzi lizhong pills group, cimetidine group, gaoshigao, dashigao, hongshidi, hongshigao, caoshidi, caoshigao and yishigao groups, ethyl acetate part groups of 5 medicinal materials, gaozhenggao, dazhenggao, hongzhengdi, hongzhenggao, caozhenggao and yizhenggao groups, hongshuigao group), NE (Fuzi lizhong pills group, cimetidine group, petroleum ether part, ethyl acetate part and n-butanol part groups of 5 medicinal materials, gaoshuigao, dashuigao and hongshuigao groups), DA [Fuzi lizhong pills group, cimetidine group, petroleum ether part (except for gaoshidi group) and ethyl acetate part groups of 5 medicinal materials, gaozhenggao, dazhenggao, hongzhenggao, caozhenggao and yizhenggao groups, hongshuigao group] in urine, serum contents of ACTH [Fuzi lizhong pills group, petroleum ether part (except for gaoshidi, dashidi and caoshidi groups) and ethyl acetate part (except for dayidi and hongyidi groups) groups of 5 medicinal materials, hongzhenggao group, dashuigao group], and D-β-H [Fuzi lizhong pills group, gaoshigao, dashigao, hongshidi, hongshigao and yishigao groups, ethyl acetate part groups of 5 medicinal materials (except for dayidi, hongyidi and yiyidi groups), dazhenggao and hongzhenggao groups, hongshuigao group] were increased significantly (P<0.05 or P<0.01). The contents of 17-OHCS and D-β-H in dashigao group, the contents of ACTH and D-β-H in caoshigao group and the contents of D-β-H in gaoshigao and yishigao groups were significantly lower than hongshigao group. The contents of DA in gaoyigao, caoyigao and yiyigao groups as well as the contents of D-β-H in gaoyigao, hongyigao, caoyigao and yiyigao groups were significantly lower than dayigao group; the contents of DA in gaoyigao and caoyigao groups were significantly lower than hongyigao group, the contents of ACTH in gaoyigao, dayigao, caoyigao and yiyigao groups were significantly higher than hongyigao group. The contents of 17-OHCS, DA and ACTH in gaozhenggao, dazhenggao, caozhenggao and yizhenggao groups, the contents of A in dazhenggao, caozhenggao and yizhenggao groups as well as the contents of D-β-H in gaozhenggao, caozhenggao and yizhenggao groups were significantly lower than hongzhenggao group. The contents of 17-OHCS and D-β-H in caoshuigao and yishuigao groups were significantly lower than dashuigao group. The contents of 17-OHCS, DA and D-β-H in caoshuigao and yishuigao groups as well as the contents of DA and D-β-H in gaoshuigao groups were significantly lower than hongshuigao group (P<0.05 or P<0.01). CONCLUSIONS: Different polar parts of 5 kinds of Alpinia can improve gastric ulcer model rats with cold syndrome to different extents; among them, the fruit of A. galangal is the best, followed by the root of A. officinarum. Above medicinal materials can regulate the function of sympathetic-adrenal system by increasing the contents of 17-OHCS, CAs and D-β-H.
