Objective To investigate the improvement effect and mechanism of Modified Kongsheng Zhenzhong Dan medicated serum on mitochondrial function of PC12 cells injured by oxygen-glucose deprivation/reperfusion(OGD/R)based on the SIRT1/PGC-1α pathway.Methods PC12 cells were used to construct OGD/R cell model in vitro.Cell grouping:normal group(10%FBS),model group(10%FBS),10%drug-containing serum group,5%drug-containing serum group(5%drug-containing serum+5%blank serum),10%blank serum group(control group).CCK-8 method was used to detect the cell survival rate,and the appropriate oxygen glucose deprivation time(2,4,6,8 hours)was screened.MTT assay was used to detect cell activity;mitochondrial stress test(MST)was used to detect the oxygen consumption rate(OCR).Apoptosis was detected by flow cytometry(Annexin V-PE/7-AAD double staining).The protein expression levels of SIRT1 and PGC-1α in PC12 cells were detected by Western Blot.Results Compared with the normal group,the activity of PC12 cells decreased significantly after oxygen-glucose deprivation for 2,4,6 and 8 hours(P＜0.05),and 6 hours of oxygen-glucose deprivation was selected as the time of subsequent experimental modeling.Compared with the normal group,the cell activity of the model group was significantly decreased(P＜0.05).The OCR values of basic respiration value,maximum respiration value,proton leakage,ATP production and standby respiration ability were significantly decreased(P＜0.05).The apoptosis rate was significantly increased(P＜0.05).The protein expression levels of SIRT1 and PGC-1α in cells were significantly decreased(P＜0.05).Compared with the model group,the cell activity of the Modified Kongsheng Zhenzhong Dan 10%and 5%drug-containing serum groups was significantly increased(P＜0.05).The OCR values of basal respiration value,maximum respiration value,proton leakage,ATP production and spare breathing ability were significantly increased(P＜0.05).The apoptosis rate was significantly decreased(P＜0.05).The protein expression levels of SIRT1 and PGC-1α in cells were significantly increased(P＜0.05).Conclusion The Modified Kongsheng Zhenzhong Dan medicated serum can improve mitochondrial dysfunction,inhibit neuronal apoptosis and promote neuronal survival in OGD/R-injured PC12 cells.The mechanism may be related to the activation of SIRT1/PGC-1α signaling pathway.

Objective To investigate the seasonal Aedes population fluctuation and the resistance of Aedes populations to common insecticides in Jiangsu Province in 2020, so as to provide insights into vector-borne infectious diseases control.. Methods One village was randomly sampled from each of Xinbei District of Changzhou City and Zhangjiagang County of Suzhou City in southern Jiangsu Province, Hai’an County of Nantong City and Yandu District of Yancheng City in Central Jiangsu Province, and Suining County of Xuzhou City and Sihong County of Suqian City in northern Jiangsu Province during the period between May and October, 2020. A small ponding container was sampled, and larval Aedes mosquitoes were collected using straws once each in early and late stages of each month. All larvae were bred in laboratory to adults for population identification. In addition, larval breeding were observed in all small ponding containers in and out of 30 households that were randomly sampled from six surveillance sites, and the larval mosquito density was estimated using Breteau index. Larval A. albopictus mosquitoes were sampled around Cuiyuan New Village in Jintan District of Changzhou City, and bred in laboratory to the first offspring generation, and the susceptibility of adult female mosquitoes to deltamethrin, lambda-cyhalothrin, malathion, and propoxur was tested using the filter-paper bioassay recommended by WHO. Results A total of 1 165 larval Aedes mosquitoes were captured from small ponding containers in six surveillance sites of Jiangsu Province in 2020, and all were identified as A. albopictus following eclosion. The largest number of Aedes larvae captured was found in July. A total of 1 152 households were investigated in six surveillance sites, and the mean Breteau indexes were 9.58, 13.20, 13.71, 13.20, 12.18 and 5.58 from May to October, respectively, while a high Aedes transmission risk was seen in Xinbei District of Changzhou City, with a higher Breteau index than in Suining (H = 23.667, P_adjusted = 0.001) and Sihong (H = 22.500, P_adjusted = 0.003) counties. The field-captured A. albopictus from Cuiyuan New Village in Jintan District of Changzhou City remained sensitive to malathion, but was resistant to propoxur, and developed high-level resistance to deltamethrin and lambda-cyhalothrin. Conclusions A. albopictus was present in southern, central and northern Jiangsu Province in 2020, and the larval density peaked in July. A. albopictus captured from Cuiyuan New Village in Jintan District of Changzhou City has developed high-level resistance to pyrethroid pesticides.

From 1978 to 2019, the cause of neurosurgery in China has developed rapidly, and the surgical treatment of pituitary adenoma has achieved good results.. This article focuses on the surgical treatment of pituitary adenomas from imaging techniques, surgical techniques and anesthesia techniques, and looks forward to the future.

A simple colorimetric method for detection of isocarbophos was developed based on the enhanced peroxidase-like activity of hemin. Hemin could catalyze the oxidation of peroxidase substrate 3,3′,5,5′-tetramethylbenzidine (TMB) by H2O2, which made TMB to lose one electron and caused reaction solution color changing from initial transparent to blue-green. Adding isocarbophos improved hemin affinity to substrates, which further enhanced peroxidase-like activity of hemin and made TMB to lose two electrons. The color of TMB solution was further changed from blue-green to yellow, and the degree of color change was proportional to the concentration of isocarbophos. Under the optimal conditions, the present analytical method for isocarbophos detection had a dynamic range from 2 μg/L to 100 μg/L with a detection limit of 1.2 μg/L (3σ). The selectivity assay demonstrated that other organophosphorus pesticides exhibited negligible interferences for isocarbophos detection. The application of the proposed method in the practical samples showed that the mean recovery of isocarbophos was in the range of 94.4％-113.0％, thus it could be widely applied to rapidly and sensitively detect isocarbophos in the agricultural products.

Objective To describe the lived experience of mothers of children with hydrocephalus. Methods A semi-structured interview was conducted with 14 mothers of children with hydrocephalus. Data were analyzed based on Colaizzi phenomenotogical research method. Results Through the analysis, sorting and refining the theme, the mother's care experience could be divided into three themes: psychological emotional, stress response, social support requirements. Conclusions Nursing staff should have a thorough understanding of the experience to provide medical care and emotional support, improve the ability of the parents care,which could help caregiver adapt to their roles, maintain and promote the health of children patients and their families .

Objective To observe the characteristics of liver-kidney yin deficiency syndrome in late stage of spontaneously hypertension rats (SHRs). Methods Ten 12-month-old healthy male SHRs were put into syndrome observer group (SHR group), while ten male WKY rats with the same age were set as control group (WKY group). Syndrome differentiation method on rats was employed to transform the four-diagnostic information into general behavior indexes with the same significance. The afternoon anal temperature, tongue color, holding power, ability of learning and memory, serum estradiol and testosterone level were observed, compared and analyzed. Results Compared with the WKY group, rats in the SHR group had higher afternoon anal temperature, redder tongues, poorer strength and claw holding power and their ability of learning and memory was retarded; the level of serum estradiol was lower;the ratio of estradiol to testosterone was higher. All of these were largely in line with deficiency of live-kidney yin syndrome. Conclusion SHRs in late stage show characteristics of liver-kidney yin deficiency syndrome.

Objective:To investigate mechanism of imatinib mesylate induced apoptosis of primary CD3+T cells.Methods:The CD3+T cells were stimulated by 0-100 nmol/L imatinib for 24 h,cell apoptosis was detected by flow cytometry;Caspase-3,Caspase-8, A20 and NF-κB expression levels were detected by Real-time quantitative PCR and Western blot.Results: IM significantly increased apoptosis of T cell;Caspase-3 and A20 gene expression levels were upregulated and NF-κB expression level was downregulated both in gene and protein levels.Conclusion:IM increased apoptosis of T cell by upregulating A20 expression.

Objective To investigate the effect of a nurse-training program of theory combined with practice training methods on pressure ulcer .Methods Forty-two nurse trainees were selected from different departments where the pressure sore was most likely to occur .A two-month training program was conducted based on multimedia, skill training, on-site training, patient rounds, typical case studies, mannequin, and problem analysis .Theoretical examinations were conducted both before and after the program and the skill tests were conducted by the way of on-site intervention .Results Among the forty-two nurses , the mean scores of theoretical examination before and after training were (88.52 ±2.31) and (97.16 ±3.20), respectively, and the difference was significant (t=-14.193,P<0.01).The mean scores of skill test before and after training were (86.19 ±4.68) and (93.35 ±2.41), respectively, and the difference was significant (t=-8.814, P<0.01).The forty-two nurse trainees all passed the tests .Conclusions The training program which combined with theory and practice skills can improve the knowledge and skills of the nurses .It can be served as a model for future training on pressure ulcer treatment by other general hospitals .

Objective To explore the effectiveness of traumatic brain model on the rehabilitation hemiplegic patients .Methods From June 2012 to May 2013 , 120 cases with craniocerebral injury complicated with hemiplegia were selected .60 patients with conventional treatment were in the control group . After established the traumatic brain model , 60 cases were selected for the observation group .Within 24 hours admitted in the hospital and 5 weeks after admission, the Glasgow coma scale (GCS), modified Rankin scale (MRS) and the Barthel index (BI) were surveyed.Two groups were given the routine neurosurgery therapy . The control group was given the conventional care , while the observation group was used the traumatic brain model for the rehabilitation guidance .Results The SCS, MRS and BI scores between groups before and after treatment had significant differences .After the treatment , the scores of SCS , MRS and BI in the observation group were (8.25 ±5.01), (3.49 ±2.13), (45.84 ±15.48), which were significantly higher than (10.21 ± 4.11), (3.00 ±1.24), (54.72 ±17.74) in the control group (t=2.343,2.074,2.922,respectively;P<0.05).The proportion of BI >65, self care were 65.0% and 70.0% in the observation group, which were significantly higher than 40.0% and 41.7% in the control group (χ2 =7.519, 9.766, respectively;P <0.01).Conclusions The traumatic brain model emphasizes the partnership between neuro-surgeons and rehabilitation specialists .Meanwhile, the nurses will give the professional guidance on rehabilitation .All these can guarantee a timely rehabilitation treatment for the patients after their admission to the hospital , which can also provide a solid foundation for further rehabilitation .It avoids the occurrences of adverse events to the uttermost and paves the way for the rehabilitation treatment in the next phase after the patients in a stable condition , which can reduce the time of hospital stays , improve the rehabilitation prognosis and help the patients with the functional recovery to the greatest extent .

OBJECTIVETo study the influence of aconite root, a Chinese medicinal herb with hot property, on energy metabolism and gene expression spectrum, and to analyze the possible mechanism of it effect.

METHODThirty two SPF Wistar rats were randomly divided into aconite root group and control group. Decoction of aconite root and NS were intragastrically administrated with the concentration of 10 mL x kg(-1) respectively once a day for 20 days. Temperature, energy intake (EI), digestive energy (DE) and metabolic energy (ME) were measured. The activity of ATPase and succinate dehydrogenase (SDH) in liver was detected by colorimetry. The gene expression of liver was detected with Illumina's rat ref-12 gene array. The differential expression genes were selected, annotated and classified based on gene ontology (GO). Real-time quantitative reverse-transcriptase PCR (Q-RT-PCR) was used to test the accuracy of the array results.

RESULTCompared with the control group, the toe temperature (TT) on the 10th and 20th day after the administration,the EI/BM( body mass), DE/BM, ME/BM and the activity of Na+ - K+ - ATPase, Ca2+ - Mg2+ - ATPase and SDH of liver in the aconite root group increased significantly (P<0.05). There were 592 differential expression genes in aconite root group compared with the control group. Based on Go analysis, the most significant genes was related to metabolic process (lgP = - 15.5897).

CONCLUSIONAconite root could improve the energy metabolism in rats, by influencing the metabolic process of sugar, lipid and amino acid, which may be the main molecular mechanism of warming yang and dispelling cold for the treatment of the cold syndrome according to Chinese medicine theory.
