1.The Contemporary Value, Constraining Factors and Breakthrough Approach of Constructing the External Discourse System of Traditional Chinese Medicine

Strengthening the construction of traditional Chinese medicine (TCM) discourse system for international audiences is necessary for enhancing cultural soft power, accelerating the construction of trade power, integrating into the global healthcare system, and realizing international scientific and technological dialogues.TCM not only garners widespread attention from the international community but also faces numerous challenges. The external discourse of TCM has fallen into difficulties, including blocked channels of expression, lack of a discourse subject, contextual segregation of discourse, and a narrative information flow deficit. Based on this, realistic approaches to breaking through dilemmas are proposed from four dimensions, that is, strengthening the construction of international standards for TCM to increase its recognition overseas, improving the development of international talent teams to enhance the visibility of TCM abroad, expanding the construction of the TCM service trade system to deepen its contribution overseas, and enhancing the efficacy of international communication of TCM to strengthen its reputation abroad.
2.Study on the Mechanism of Huanglian Jiedu Decoction in Regulating Intestinal Microbiota Mediated Tryptophan Metabolism and Intervening in Parkinson's Disease

OBJECTIVE To analyze the effect of Huanglian Jiedu Decoction(HLJDD)on the intestinal flora and metabolites of Parkinson's disease(PD)model mice,and explore the mechanism of HLJDD in intervening in PD based on 16S rRNA technology and non-targeted metabolomics technology.METHODS The PD model of mice was induced by subcutaneous injection of MPTP 20 mg·kg-1·d-1 and peritoneal injection of probenecid 200 mg·kg-1·d-1,and the weight and behavior indexes of mice were measured after drug intervention.HPLC-QTRAP-MS/MS technique was used to detect the levels of neurotransmitters in the striatum of mice.The levels of striatal inflammatory factors were detected by ELISA.The changes of intestinal flora in mice were analyzed by 16S rRNA technology.UHPLC-Q-TOF-MS was used to detect endogenous metabolites in mouse striatum,Orthogonal partial least squares discriminant analysis(OPLS-DA)was adopted to screen potential differential metabolites,and MetaboAnalyst 5.0 was introduced to predict metabolic pathways associated with PD.RESULTS HLJDD significantly improved the motor symptoms and neuroinflammation of PD mice(P<0.01),regulated the level of neurotransmitters,and corrected the intestinal microbiota disorder of PD mice,manifes-ted by the increase of intestinal microbial diversity and the restoration of microbiota profile.After treatment with HLJDD,the abun-dance of Prevotella and Akkermansia in PD mice was significantly increased,and the abundance of Clostridium was decreased(P<0.01).The abnormal metabolite levels were restored mainly by regulating the tryptophan metabolic pathway in the feces and striatum of PD model mice.CONCLUSION HLJDD can significantly improve the pathological damage of PD model mice,and the regulation of disordered intestinal flora and tryptophan metabolism pathway may be the potential mechanism of HLJDD to intervene in PD.
3.Exploration and Practice of Pharmaceutical Equipment & Workshop Design by Using the Task-driven Teaching

OBJECTIVE:To provide reference for the improvement of Pharmaceutical Equipment & Workshop Design teach-ing. METHODS:Task-driven teaching was adopted,CIDO engineering educational model was referred to,task was designed from the conception to run with task-based;theoretical and teaching exploration and practice was carried out by cross-learning of individ-uals and groups,applying of pilotscale experiment platform,developing course design and forming evaluation. RESULTS & CON-CLUSIONS:It changed the traditional teaching method that emphasized on class teaching to make students participate in multi-task learning,so that students can better grasp the equipment selection,understanding and application of workshop design in pharmaceu-tical process. The method gives full play to students' initiative and creativity and it is conducive to students’comprehensive ability for adapting the development of pharmaceutical industry.
4.Preparation and characterization of tetrandrine-loaded PLGA nanocomposite particles by premix membrane emulsification coupled with spray-drying method.

For effective inhalable dry-powder drug delivery, tetrandrine-PLGA (polylactic-co-glycolic acid) nanocomposite particles have been developed to overcome the disadvantages of nanoparticles and microparticles. The primary nanoparticles were prepared by using premix membrane emulsification method. To prepare second particles, they were spray dried. The final particles were characterized by scanning electron microscopy (SEM), dry laser particle size analysis, high performance liquid chromatography (HPLC), X-ray diffraction (XRD), differential scanning calorimetry (DSC), infrared analysis (IR) and confocal laser scanning microscope (CLSM). The average size of the primary particles was (337.5 ± 6.2) nm, while that second particles was (3.675 ± 0.16) μm which can be decomposed into primary nanoparticles in water. And the second particles were solid sphere-like with the drug dispersed as armorphous form in them. It is a reference for components delivery to lung in a new form.
5.Intestinal absorption of berberine alone and in combinations by rats single pass intestinal perfusion in situ.

The aim of the study is to investigate the effects of concentration, intestinal segments, pH, inhibitors of proteins (P-gp), Na(+)-dependent glucose transporter (SGLT1) on the intestinal absorption of berberine, and to compare intestinal absorption of berberine in combinations. With phenol red as the indicator, in situ single pass intestinal perfusion (SPIP) model was used and intestinal absorption of pure berberine at concentrations of 36.70, 46.17 and 92.33 microg x mL(-1), simulated system of HLJDT (mixture of berberine, baicalin and geniposide), HLJDT with the concentration of berberine 92.33 microg x mL(-1) in perfusion solution of different intestinal segments (duodenum, jejunum, ileum, and colon) were determined by HPLC in combination with diode array detection (DAD). The results indicated that Ka values ofberberine at different concentrations had little significant difference among that obtained after perfusing via duodenum, jejunum, ileum and colon indicating that the absorption of berberine was mainly the passive diffusion. It was also suggested that SGLT1 and P-gp might exert some effects on the absorption of berberine. Ka and Peff values of berberine in a mixture of pure compounds and HLJDT for different intestine segments of rat showed an increasing tendency and was significantly different (P < 0.05) indicating that berberine in a mixture of pure compounds and HLJDT was assimilated better in small intestine. These results indicate that the intestinal absorption of berberine may be affected by compatibility of compounds. Additionally, berberine has wide absorption window and better absorption in colon.

7.Interaction of nourishing and tonifying blood effects of the combination of Angelicae sinensis Radix and Astragali Radix studied by response surface method.

The combination of Angelicae sinensis Radix (Danggui, DG) and Astragali Radix (Huangqi, HQ) is a popular herb pair commonly used in clinic for the treatment of blood deficiency syndrome in China. The aim of this paper is to study the interaction of DG and HQ nourishing and tonifying blood effects by response surface method. The blood deficiency mice were induced by injecting N-acetylphenylhydrazine (sc) and cyclophosphamide (ip). The blood deficiency mice were administrated intragastrically with DG-HQ extracts (0:1, 1: 5, 2:5, 2:3, 1:1, 3:2, 5:2, 5:1, 1:0). The changes of the peripheral blood indexes and organ indexes were observed. The indexes were integrated by comprehensive index method; the interactions of DG and HQ were analyzed by the response surface diagram established with Matlab software. The results showed that DG and HQ at most of their combination ratios had synergic effect. Within the range of 1:5 - 5:1, all of the extracts of DG-HQ showed synergic effect, and among which, high-doses had better effects than low-doses. The highest value (-1) of the synergic effect was showed when DG was 10 - 40 g at the same time of HQ as 90 -180 g, and DG was 50 - 100 g at the same time of HQ as 20 - 100 g. DG-HQ at all combination dosages within Chinese Pharmacopeia (DG: 6 - 12 g, HQ: 9 - 30 g) had certain synergic effect, and Danggui Buxue Decoction (DG: 6 g, HQ: 30 g) also was at this range. The results provided scientific basis to the clinical application of DG and HQ. And the response surface method was firstly applied to quantitatively evaluate the bio-activity change of herb combination, which provided a novel way for modern basic research on the interaction of herbs.
8.Comparision of adsorption-desorption dynamics of huanglian jiedu decoction using different macroporous resins combinations.

OBJECTIVETo establish the techniques for the purification of Huanglian Jiedu decoction by macroporous resins combinations, and to lay the foundation of improvement of dosage forms in clinical application and realization of industrialized production about Huanglian Jiedu decoction.
METHODFour kinds of macroporous resins (X-5, S-8, HPD100, AB-8) were chosen and combined in six different forms. Adsorption and desorption experiments were carried out and contents of six index constituents, gardenoside, baicalin, palmatine, berberine, baicalein and wogonin, in different groups were compared before and after purification.
RESULTAdsorption rates of six index constituents in Huanglian Jiedu decoction of different combinations of macroporous resins groups were all nearly 100%, but desorption rates had remarkable differences.
CONCLUSIONThrough comprehensive comparison, macroporous resins combination 6 resulted in high contents of index suitable relatively. It is suitable for purification of Huanglian Jiedu decoction.

9.Analysis of morphic traits between physical and chemical properties and chemical constituents based on different combinations of Shaofu Zhuyu decoction.

The formulae decoctions are commonly used in clinic and have characteristics of simple preparation and rapid effect. It was considered as a complex physical and chemical system. In this article, the physical and chemical properties and chemical components and their relationships were investigated to explore the changes of characteristics resulted from different combinations of drugs of Shaofu Zhuyu decoction. The method of partial leasts-quares regression was adopted to evaluate the correlation between physical and chemical parameters and chemical components. These researches may provide a new way to investigate the state of bioactive components, the action mechanisms, and property of treating diseases of traditional Chinese medicine decoction.

10.Preliminary study to investigate dynamic extract process of Huanglian Jiedu Tang and the mechanism of subsidence produce.

OBJECTIVETo study the phenomenon of subsidence emergence in the process of Huanglian Jiedu Tang decoction extraction, and the mechanism of subsidence emergence.
METHODUV was applied to determine the concentration of total alkaloids and total flavones; Simultaneous determination of berberine, jatrorrhizine, palmatine, baicalin and geniposide were carried out by HPLC; The solid holdup and precipitation rate were calculated by the formula. The relativity among these parameters was analyzed by the SPSS software program.
RESULTThe contains of berberine, jatrorrhizine, palmatine in total alkaloids showed a good correlation with total alkaloids. The correlation between baicalin and total flavones was lower than that between geniposide and total flavones. Compared to precipitation rate, solided hold up shows a larger relevance with index component
CONCLUSIONWith the change of time, the total alkaloids represented by berberine alkaloids and baicalin at a certain concentration can be regarded as the equilibrium point, or one generated by the critical point of precipitation, the reaction can generate "sediments"; because "precipitation objects" generated, reducing the concentration of the above-mentioned components, destroy the" balance". The relevant components of herbal medicine increased dissolution rate, at the same time might partly dissolved sediment, reaching a new equilibrium state.
