A ratiometric fluorescent probe([Eu(Hpfdap)3(CTMR-Phen-N)])was designed and synthesized for detection of singlet oxygen(1O2)with dimethyl anthracyl-containing β-diketone compound 1,1,1,2-pentafluoro-5-(9',10'-dimethyl-2'-anthracene)-3,5-pentanedione(Hpfdap)as recognition ligand and 5-amino-1,10-phenanthroline conjugated with Rhodamine as auxiliary ligand(CTMR-Phen-N).The results of spectral analysis showed that the europium(Ⅲ)complex had almost no fluorescence due to the presence of anthracene ring in the probe. After reaction with 1O2,the anthracene ring was oxidized to endoperoxide, and the fluorescence of the europium(Ⅲ)complex at 614 nm was restored, followed by the increase of quantum yield from 1.60%to 25.16%.The fluorescence intensity of Rhodamine at 575 nm remained unchanged before and after the reaction with 1O2.Therefore,the ratio of fluorescence intensity at 614 nm and 575 nm could be used to measure 1O2 by the developed rationmetric fluorescence method. The probe showed good selectivity for 1O2,and the detection limit(S/N=3)was as low as 3 μmol/L.The fluorescence imaging results demonstrated that the probe had low cytotoxicity and good membrane permeability, and could be applied to fluorescence imaging of 1O2 in Hela cells.

This article discussed the significance of latent pathogen theory in the understanding of pathogenesis and prevention and treatment of epidemic diseases such as novel coronavirus infection(COVID-19).Latent-qi is the cause of epidemic febrile disease, and it is also the yang-heat qi of human body. The stagnant yang-heat qi hidden in the body can induce the latent heat constitution. The latent heat constitution is particularly closely related to the development and progression of epidemic febrile disease, and is susceptible to the warm pathogen. The significance of the latent pathogen theory on the prevention and treatment of epidemic diseases such as COVID-19 with traditional Chinese medicine is as follows:abnormal climate is an important early warning and is the external condition for the development of epidemic diseases;latent pathogen is easily assimilated into the newly-attacked pathogenic factors for the assimilation of similar nature, and then injuries taiyin-spleen,which causes spleen deficiency and dampness accumulation and then results in latent dampness;preventing and treating epidemic diseases based on the latent pathogen theory is helpful for identifying the key syndromes, formulating the core regimen indicated for the pathogenesis, and cutting off the progression of the disease.Based on the latent pathogen theory,dampness-removing therapy for latent dampness is important for the prevention and early treatment of COVID-19,and our research group has developed a Chinese herbal formula of Wuzhi Fangguan Prescription(composed of Fici Simplicissimae Radix,Coicis Semen,Poria,Herba Polygoni Chinensis,Atractylodis Rhizoma,Pogostemonis Herba, and Glycyrrhizae Radix et Rhizoma)to prevent COVID-19 early by using a variety of dampness-removing methods such as drying dampness, resolving dampness, draining dampness, and invigorating spleen. The prescription is indicated for the prevention and early treatment of COVID-19.The mechanism of dampness-removing therapy is mainly related to the improvement of the disordered immune function and restoration of the body's resistance to diseases.

Clinical and Molecular Hepatology 2024;30(3):468-486
Background/Aims:
Chronic hepatitis C (CHC) patients who failed antiviral therapy are at increased risk for hepatocellular carcinoma (HCC). This study assessed the potential role of metformin and statins, medications for diabetes mellitus (DM) and hyperlipidemia (HLP), in reducing HCC risk among these patients.
Methods:
We included CHC patients from the T-COACH study who failed antiviral therapy. We tracked the onset of HCC 1.5 years post-therapy by linking to Taiwan’s cancer registry data from 2003 to 2019. We accounted for death and liver transplantation as competing risks and employed Gray’s cumulative incidence and Cox subdistribution hazards models to analyze HCC development.
Results:
Out of 2,779 patients, 480 (17.3%) developed HCC post-therapy. DM patients not using metformin had a 51% increased risk of HCC compared to non-DM patients, while HLP patients on statins had a 50% reduced risk compared to those without HLP. The 5-year HCC incidence was significantly higher for metformin non-users (16.5%) versus non-DM patients (11.3%; adjusted sub-distribution hazard ratio [aSHR]=1.51; P=0.007) and metformin users (3.1%; aSHR=1.59; P=0.022). Statin use in HLP patients correlated with a lower HCC risk (3.8%) compared to non-HLP patients (12.5%; aSHR=0.50; P<0.001). Notably, the increased HCC risk associated with non-use of metformin was primarily seen in non-cirrhotic patients, whereas statins decreased HCC risk in both cirrhotic and non-cirrhotic patients.
Conclusions
Metformin and statins may have a chemopreventive effect against HCC in CHC patients who failed antiviral therapy. These results support the need for personalized preventive strategies in managing HCC risk.

The purpose of this study was to evaluate the diagnostic performance of multiparametric ultrasound (mpUS; grayscale US, color Doppler US, strain elastography, and contrast-enhanced US) in the assessment of testicular lesions with negative tumoral markers. MpUS imaging data, patient age, serum tumor markers, scrotal pain, cryptorchidism, and related clinical information were retrospectively collected for patients who underwent mpUS examination between January 2013 and December 2019. Histologic results or follow-up examinations were used as the reference standard. In total, 83 lesions from 79 patients were included in the analysis. Fifty-six patients were finally diagnosed with benign tumors, and 23 patients were ultimately diagnosed with malignant tumors. Chi-square tests or Fisher's exact tests were used to assess the difference between the two groups. Stepwise multivariate logistic regression analysis showed that lesion diameter (odds ratio [OR] = 1.072, P = 0.005), vascularization on color Doppler US (OR = 4.066, P = 0.001), and hyperenhancement during the early phase (OR = 6.465, P = 0.047) were significant independent risk factors for malignancy; however, when compared with neoplastic lesions, pain (OR = 0.136, P < 0.001), absence of vascularization on color Doppler US (OR = 1.680, P = 0.042), and nonenhancement during the late phase (OR = 3.461, P = 0.031) were strongly associated with nonneoplastic lesions. MpUS features are useful for differentiating testicular lesions with negative tumoral markers and improving the preoperative diagnosis, which may avoid inappropriate radical orchiectomy.
