1.ABCB4 gene mutation-associated liver cirrhosis with gallstones: A case report

The disease spectrum of ABCB4 gene mutation involves various diseases such as progressive familial intrahepatic cholestasis type 3 (PFIC3), gallstone disease, intrahepatic cholestasis of pregnancy, portal hypertension, liver cirrhosis, and even primary hepatic and biliary malignancies. A young male patient was admitted to Department of Hepatobiliary Medicine, Eastern Hepatobiliary Surgery Hospital, and was initially diagnosed with liver cirrhosis and gallstones, and he was planned to receive laparoscopic cholecystectomy. Preoperative examination showed abnormal liver function, liver cirrhosis, splenomegaly, and mild esophageal varices, and next-generation sequencing was performed to make a confirmed diagnosis of ABCB4 gene mutation-associated liver cirrhosis with gallstones. The liver function of the patient gradually returned to normal after cholagogic treatment with ursodeoxycholic acid capsules.
2.Simultaneous multi-slice technique applicated in diffusion tensor imaging for evaluating brain glioma

Objective To explore the application value of simultaneous multi-slice(SMS)technique in diffusion tensor imaging(DTI)for evaluating brain glioma.Methods Thirty-four brain glioma patients were prospectively enrolled,and brain conventional DTI and SMS-DTI were acquired.The subjective scores of image quality,signal-to-noise ratio(SNR)and contrast-to-noise ratio(CNR)were compared between SMS-DTI and conventional DTI,so were the numbers of whole brain fiber bundles,tumor relative fractional anisotropy(rFA)and relative mean diffusivity(rMD)obtained based on SMS-DTI and conventional DTI.Results Among 34 patients,there were 23 cases of high-grade glioma and 11 cases of low-grade glioma.No significant difference of subjective scores of image quality,tumor edge clarity nor magnetic susceptibility artifacts was found between SMS-DTI and conventional DTI(all P>0.05).SNR and CNR on SMS-DTI were both lower than those on conventional DTI(both P<0.05).No significant difference of the numbers of whole brain fiber bundles,rFA nor rMD of gliomas with different pathological grades was detected based on SMS-DTI compared with those on conventional DTI(all P>0.05).Conclusion SMS applicated in DTI for evaluating brain gliomas was able to shorten acquisition time under the condition of ensuring image quality and quantitative analysis accuracy.
3.Research progress on nanomedicine for treating chicken coccidiosis

Chicken coccidia specialize in growing and reproducing within the epithelial cells of the chicken intestinal mucosa,posing a threat to the health of the chicken flock.However,traditional chemical drugs are difficult to pass through the cell barrier and can easily lead to the emergence of drug-resistant strains and residual veterinary drugs,posing a huge challenge to the prevention and control of coccidiosis.Therefore,it is necessary to develop new strategies against coccidiosis.Nano-drugs have the advantages of good biocompatibility,easy modification,and high efficiency,impro-ving the penetration of cell membranes while reducing drug toxicity and side effects.They are ex-pected to be applied in the prevention and treatment of coccidiosis.This article introduces various nanomedicines for treating chicken coccidiosis,summarizes their advantages compared to tradition-al formulations,focuses on the application of nanomedicines in the treatment of chicken coccidio-sis,elucidates the existing problems of nanomedicine treatment for chicken coccidiosis,and looks forward to the challenges and future development directions in this field,in order to provide impor-tant references for the development of nanomedicines for treating chicken coccidiosis.

BACKGROUND:
LY01005 (Goserelin acetate sustained-release microsphere injection) is a modified gonadotropin-releasing hormone (GnRH) agonist injected monthly. This phase III trial study aimed to evaluated the efficacy and safety of LY01005 in Chinese patients with prostate cancer.
METHODS:
We conducted a randomized controlled, open-label, non-inferiority trial across 49 sites in China. This study included 290 patients with prostate cancer who received either LY01005 or goserelin implants every 28 days for three injections. The primary efficacy endpoints were the percentage of patients with testosterone suppression ≤50 ng/dL at day 29 and the cumulative probability of testosterone ≤50 ng/dL from day 29 to 85. Non-inferiority was prespecified at a margin of -10%. Secondary endpoints included significant castration (≤20 ng/dL), testosterone surge within 72 h following repeated dosing, and changes in luteinizing hormone, follicle-stimulating hormone, and prostate specific antigen levels.
RESULTS:
On day 29, in the LY01005 and goserelin implant groups, testosterone concentrations fell below medical-castration levels in 99.3% (142/143) and 100% (140/140) of patients, respectively, with a difference of -0.7% (95% confidence interval [CI], -3.9% to 2.0%) between the two groups. The cumulative probabilities of maintaining castration from days 29 to 85 were 99.3% and 97.8%, respectively, with a between-group difference of 1.5% (95% CI, -1.3% to 4.4%). Both results met the criterion for non-inferiority. Secondary endpoints were similar between groups. Both treatments were well-tolerated. LY01005 was associated with fewer injection-site reactions than the goserelin implant (0% vs . 1.4% [2/145]).
CONCLUSION:
LY01005 is as effective as goserelin implants in reducing testosterone to castration levels, with a similar safety profile.

Anti-seizure medications (ASMs) are the main therapy for epilepsy.There are many kinds of ASMs with complex mechanism of action, so it is difficult for pharmacists to examine prescriptions.This paper put forward some suggestions on the indications, dosage forms/routes of administration, appropriateness of usage and dosage, combined medication and drug interaction, long-term prescription review, individual differences in pathophysiology of children, and drug selection when complicated with common epilepsy, for the reference of doctors and pharmacists.
6.Prognostic effects of type 2 diabetes mellitus on patients with different TNM stage of hepatocellular carcinoma after operation

Objective:To investigate the effects of type 2 diabetes mellitus (T2DM) on the clinicopathological characteristics and prognosis of patients with hepatocellular carcinoma (HCC) after curative hepatectomy.Methods:From November 2009 to June 2011, the clinicopathological and survival data of HCC patients who underwent radical resection at Eastern Hepatobiliary Surgery Hospital of Naval Medical University were retrospectively analyzed. The patients were divided into T2DM group and non-T2DM group. Clinicopathological characteristics and prognosis of two groups were compared. Chi square test or Fisher exact test were used for statistical analysis. Kaplan-Meier test was used for univariate survival analysis, and Cox proportional hazard regression model was used for multivariate survival analysis.Results:A total of 1 691 patients with HCC were enrolled in this study. Among them, 142 patients (8.4%) were complicated with T2DM. The proportion of patients with onset age≥65 years old, the proportion of men, the proportion of patients with hypertension and γ-glutamyltranspeptidase >61 U/L in the T2DM group were higher than those in non-T2DM group (24.6%, 35/142 vs. 10.4%, 161/1 549; 92.3%, 131/142 vs. 85.7%, 1 327/1 549; 43.7%, 62/142 vs. 12.3%, 191/1 549; 58.5%, 83/142 vs. 49.4%, 765/1 549), while the proportion of patients with chronic hepatitis virus infection and α-fetoprotein >20 μg/L in the T2DM group were both lower than those in non-T2DM group (76.1%, 108/142 vs. 92.0%, 1 425/1 549; 47.2%, 67/142 vs. 59.9%, 928/1 549), and the differences were all statistically significant ( χ2=25.79, 4.75, 100.36, 4.28, 39.01 and 8.66, all P<0.05). The results of univariate survival analysis showed that there was no significant difference in the postoperative overall survival (OS) rate between T2DM group and non-T2DM group ( χ2=3.02, P=0.082). The results of further subgroup analysis showed that among HCC patients with TNM stage Ⅰ, there was statistically significant difference in the OS rate between T2DM group and non-T2DM group ( χ2=4.53, P=0.033). The OS rates at 1 year, 3 years and 5 years after curative resection of HCC patients in T2DM group were lower than those of patients in non-T2DM group (96.0%, 48/50 vs. 97.6%, 558/572; 78.0%, 39/50 vs. 88.6%, 507/572; 68.0%, 34/50 vs. 79.5%, 455/572). The results of multivariate Cox regression survival analysis demonstrated that T2DM was an independent risk factor for postoperative survival in patients with TNM stage Ⅰ HCC (odds ratio=1.663, 95% confidence interval 1.045 to 2.648, P=0.032). Conclusions:The effect of T2DM on prognosis of patients after radical resection of HCC is associated with TNM stage, and its effect may be limited on the early stage of HCC.
7.Influence of long non-coding RNA-CRNDE in stem cell properties and their mechanism in glioma

Objective:To investigate the influence of long non-coding RNA-CRNDE in stem cell properties and their mechanism in glioma.Methods:The CD133 +U251 stem cells (U251s) were isolated from human glioma cell line U251 by immunomagnetic beads. Cells were divided into U251s group, U251s-CRNDE group, U251 group, and U251-CRNDE group; CRNDE shRNA lentiviral vectors were transfected into cells in the U251s-CRNDE group and U251-CRNDE group to down-regulate the CRNDE expression. The CRNDE mRNA expression was detected by real-time fluorescent quantitative PCR (RT-qPCR); CCK-8 assay was used to detect the cell viability; Transwell assay was used to detect the cell invasion; wound healing method was used to detect the cell migration; plate cloning assay was employed to detect the clone formation; flow cytometry was used to detect the cell cycles; Western blotting was used to detect the expressions of A2B5, CD133, nestin, Sox2, Oct-4 and Nanog, as well as phosphatidylinositol kinase (PI3K)-protein kinase B (AKT)-mammalian target of rapamycin (mTOR) signal pathway key proteins (PI3K, AKT, phosphorylated-AKT, and mTOR). Results:The CRNDE mRNA expression in U251s group was significantly higher than that in U251 group ( P<0.05). As compared with U251s group, U251s-CRNDE group had significantly decreased cell viability, cell invasion and cell migration, statistically smaller number of cell colony, significantly decreased proportion of cells in G2/M phase, significantly increased proportion of cells in G1/G0 phase ( P<0.05); and the same trend was noted between U251 group and U251-CRNDE group. As compared with U251s group, U251s-CRNDE group had significantly decreased expressions of CD133, nestin, Sox2, Oct-4, PI3K, AKT, phosphorylated-AKT, and mTOR ( P<0.05). Conclusion:The CRNDE expression is increased in glioma stem cells, and down-regulation of CRNDE expression can inhibit the differentiation, metabolism and proliferation of glioma stem cells; and this effect is related to the regulation of PI3K-AKT-mTOR signaling pathway.
8.Prediction of Olsen formula in postoperative refractive power of patients with high myopia complicated with cataract

AIM: To evaluate the predictability of Olsen formula for postoperative refractive power in patients with high myopia complicated with cataract.
METHODS: This retrospective study reviewed 65 patients(101 eyes)with high myopia and who had phacoemulsification combined with intraocular lens implantation from October 10th 2016 to August 20th 2019 in our hospital. They were divided into three groups according to the axis length: group A(26mm RESULTS: The postoperative 3mo MAE in the three groups calculated with Olsen formula was 0.15(0.04, 0.22), 0.19(0.14, 0.27), 0.26(0.115, 0.455)D respectively(P>0.05). For patients with the same axial length, the postoperative 3mo MAE of Olsen formula was the smallest. The Bland-Altman method was used to analyze the consistency between the postoperative theoretical diopter and the postoperative 3mo actual diopter. The results showed that the postoperative 3mo actual diopter was the closest to the postoperative theoretical diopter of the Olsen formula, while the postoperative theoretical diopter of the SRK/T formula was the worst. CONCLUSION: For patients with high myopia and cataract, the accuracy and stability of Olsen formula is better than that of SRK/T and Haigis formula, and Olsen formula is least affected by eye axis length.
9.Change and significance of coagulation function and von Willebrand factor antigen level in HELLP syndrome

Objective:To investigate the changes and clinical significance of blood coagulation function and von Willebrand factor antigen (vWF:Ag) in patients with HELLP syndrome (hemolysis, elevated liver function, low platelet count).Methods:The clotting data of patients with severe preeclampsia and HELLP syndrome (observation group) admitted to the department of critical care medicine of the Fifth Center Hospital in Tianjin from May 2015 to December 2019 were retrospectively analyzed, and normal late pregnancy women with the same period were enrolled as the control group. The coagulation indexes such as prothrombin time (PT), activated partial thrombin time (APTT), antithrombin (AT), fibrinogen (Fib), D-dimer and plasma vWF:Ag level were compared between the two groups, and among patients with HELLP syndrome with different disease degree.Results:① Sixty-five patients with HELLP syndrome and 65 normal pregnant women with third trimester were included. Both groups were women of childbearing age, and there were no significant difference in the baseline data. ② The levels of Fib, D-dimer in both groups increased, but they were significantly higher in the observation group than those in the control group [Fib (g/L): 4.94 (4.76, 5.85) vs. 3.58 (2.97, 4.14), D-dimer (mg/L): 3.34 (2.55, 4.32) vs. 1.72 (1.29, 2.08), both P < 0.05], the AT was obviously reduced [62.00 (49.00, 73.00)% vs. 97.50 (90.75, 107.00)%, P < 0.01], and both PT and APTT were in the normal reference range in the two groups. In addition, the plasma vWF:Ag level in the observation group was significantly higher than that in the control group [516.50 (467.20, 563.00)% vs. 246.45 (189.95, 274.10)%, P < 0.01]. ③ According to thrombocytopenia, among the 65 patients with HELLP syndrome, 26 cases were mild [platelet count (PLT) > 100×10 9/L], 22 cases were moderate [PLT (50-100)×10 9/L], and 17 cases were severe (PLT < 50×10 9/L). With the aggravation of the disease, the D-dimer, Fib, vWF:Ag levels in the mild, moderate, severe patients significantly increased, while the AT level significantly decreased, and there was statistically significant difference between the two groups [D-dimer (mg/L): 2.63 (2.60, 2.73), 3.15 (2.55, 3.73), 3.84 (3.52, 4.23); Fib (g/L): 4.23 (4.06, 4.47), 4.72 (4.34, 5.04), 5.43 (5.14, 5.76); vWF:Ag: 465.20 (437.20, 495.40)%, 500.10 (472.40, 534.50)%, 543.50 (521.30, 563.00)%; AT: 67.50 (61.60, 78.00)%, 63.70 (53.30, 70.40)%, 54.40 (44.00, 61.20)%; all P < 0.05]. Conclusion:Patients with HELLP syndrome may show hypercoagulability and excessive expression of peripheral blood vWF:Ag level, which can induce platelet aggregation, leading to thrombocytopenia and thrombotic microangiopathy, and the clinicians should pay attention to that.
10.Annexin A1 involved in the regulation of inflammation and cell signaling pathways

With the deepening of research, proteomics has developed into a science covering the study of all the structural and functional characteristics of proteins and the dynamic change rules. The essence of various biological activities is revealed from the perspectives of the biological structure, functional activity and corresponding regulatory mechanism of proteins by proteomics. Among them, phospholipid-binding protein is one of the hotspots of proteomics, especially annexin A1, which is widely present in various tissues and cells of the body. It has the capability of binding to phospholipid membranes reversibly in a calcium ion dependent manner. In order to provide possible research ideas for researchers, who are interested in this protein, the biological effects of annexin A1, such as inflammatory regulation, cell signal transduction, cell proliferation, differentiation and apoptosis are described in this paper.
