1.Effect of TBC1D5 on hepatocellular carcinoma progression via JAK/STAT pathway

Objective To investigate the role of Tre2-Bub2-Cdc16 1 domain family member5(TBC1D5)in the de-velopment of hepatocellular carcinoma(HCC).Methods Western blot(WB),Immunohistochemistry(IHC)and quantitative real-time PCR(qPCR)were used to verify the difference in TBC1D5 expression in clinical sam-ples.The HCC cell lines MHCC97H and Hep3B were chosen to construct the knockdown model.The effects on cell proliferation were detected by cell proliferation assay,colony formation assay and EdU assay.Wound assay and Transwell assay were used to detect cell migration and invasion.Flow cytometry was used to detect the changes of cell cycle and H2O2-induced apoptosis of HCC cells.Finally,the effects of knockdown and overexpression of TBC1D5 on JAK/STAT pathway were detected by WB.Results The results of WB,IHC and qPCR showed that the expression of TBC1D5 in HCC tissues was higher at the protein level(P<0.000 1)and mRNA level(P<0.01)than that in corresponding adjacent tissues.Compared with the control group,the proliferation level of HCC cells with TBC1D5 knockdown was decreased(P<0.05),the formation of plate colony number decreased(P<0.001),and the proportion of EdU-positive cells decreased(P<0.001).The results of scratch assay and Tran-swell assay showed that the migration(P<0.01)and invasion ability(P<0.01)of HCC cells after TBC1D5 knockdown were significantly lower than those in the control group.After TBC1D5 knockdown,the cell cycle of HCC cells was slowed down(P<0.05)and the ability to resist apoptosis was reduced(P<0.01)than those in the control group.Compare with the control group,knockdown of TBC1D5 decreased the phosphorylation level of JAK and STAT proteins and inhibit the JAK/STAT pathway.Conclusion TBC1D5 is highly expressed in HCC.After knocking down TBC1D5,the proliferation,migration and invasion ability,cell cycle rate and anti-apoptosis ability of HCC cells decreased and may affect HCC progression through the JAK/STAT pathway.
2.Research progress in roles of olfactory stimulation in improving cognitive dysfunction

Cognitive dysfunction,predominantly a consequence of neurodegenerative disorders,is a burgeoning health issue.Odor molecules have the unique ability to circumvent the blood-brain barrier through the olfactory pathway,exerting a direct influence on cognitive-related brain regions including the entorhinal cortex,hippocampus,and amygdala,without transiting through the thalamus.These olfactory molecules,mostly sourced from natural plant essential oils,are character-ized by significant volatility,minimal adverse reactions,and abundant availability.Hence,research into and applications of olfactory stimulation in improving cognitive dysfunction have raised widespread concerns.This article reviews the rela-tionship between the olfactory pathway and cognitive brain regions,and explores the mechanism by which natural plant es-sential oils alleviate cognitive dysfunction through olfactory stimulation in order to provide a reference for improving cogni-tive function via olfactory stimulation.
3.Effects of rosemary essential oil inhalation on memory function of sleep-deprived mice and underlying mechanisms

Objective To study the influence of rosemary essential oil inhalation on the memory of mice experiencing sleep deprivation and to delineate the possible mechanisms involved.Methods C57BL/6J mice were randomly divided into four experimental groups in this study:a control group(Con),a control group with rosemary essential oil inhalation(Con+REO),a sleep deprivation group(SD)and a sleep deprivation group with rosemary essential oil inhalation(SD+REO).A 72-hour sleep deprivation model was induced using the multiple platform water environment method,with the Con+REO and SD+REO groups exposed to rosemary essential oil inhalation.Cognitive function was evaluated through Y-maze and novel object recognition tests.The hippocampal tissue was analyzed for superoxide dismutase(SOD)activity and the concentrations of malondialdehyde(MDA)and glutathione(GSH).ELISA was used to determine the levels of norepinephrine(NE),dopamine(DA),and serotonin 5-hydroxytryptamine(5-HT)in the hippocampus.The expression levels of postsynaptic density 95(PSD95)and brain-derived neurotrophic factor(BDNF)in the hippocampus were determined using immunoblotting techniques.Results Compared with the Con and Con+REO groups,the SD group demonstrated a significant reduction in the spontaneous alternation percentage in the Y-maze as well as the novel object recognition index.Additionally,there was a pronounced decrease in hippocampal SOD activity and GSH content,a substantial elevation in MDA levels,and a decrease in the levels of DA,NE,and 5-HT.The expressions of PSD95 and BDNF proteins also decreased.In comparison with the SD group,the SD+REO group exhibited a significant increase in the spontaneous alternation percentage in the Y-maze and the novel object recognition index.There was also a marked increase in hippocampal SOD activity and GSH content,a reduction in MDA levels and elevated levels of NE and DA.Moreover,the expressions of PSD95 and BDNF proteins were upregulated.Conclusion The inhalation of rosemary essential oil enhances the memory of sleep-deprived mice,and the underlying mechanism may involve the mitigation of oxidative stress within the hippocampal tissue,the modulation of neurotransmitter levels,and the facilitation of synaptic plasticity.
4.Design and Implementation of Medical Equipment Rational Deployment Information System Based on ASP.NET.

Based on ASP.NET framework, The Intelligent Estimated System for Rational Deployment of Medical Equipment (MERDIS) is designed and developed with SQL Server 2012 database and C# language. The system is used to realize the rational deployment suggestions and evaluation of medical equipment in hospitals. The system input the data of hospital medical equipment and clinical pathway into the database, and then feedback the deployment information to users which are calculated by big data information, so as to achieve the purpose of giving rational deployment of hospital medical equipment.

5.Numerical simulation and optimization research of needle parameters in vial washing machine.

According to the working principle of vertical ultrasonic vial washing machine, receiving respective force of small water droplets on the inside wall of vials and the minimum air velocity of blowing off water droplets can be obtained based on the analysis of water-droplet-related parameters. The inside wall model of 7 mL vial created by GAMBIT was divided into fine grids. Then the Realizable k-epsilon Two Equation Turbulence Model was adopted and the flow field of vial by FLUENT software was simulated when air was flushing inside the wall. In that case, the optimal position, inner diameter and the corresponding minimum air velocity of needle can be acquired to meet the needs of vial washing machine applied to 7 mL vial.

6.Study of the Effects of Improved Wright-Giemsa Staining

Objective To study the effects of improved Wright-Giemsa staining. Methods 40 semen samples by both traditional and improved Wright-Giemsa staining.The morphological characteristics stained by two methods was ob-served. Results The defective rates of the middle and tail parts of sperm were 7.02%± 2.4%and 11.02%± 2.03%respec-tively in improved staining group;while those were 5.48%±2.8%and 8.05%±2.56%in traditional staining group. There was statistical significance between two staining methods (P < 0.05). There was no significant difference of normal sperm and sperm head between these two staining methods;however, improved staining method showed a much clearer nucleus and ac-rosome staining than that of traditional methods. Improved Wright-Giemsa staining is better for sperm, vaginal discharge and Trichomonas vaginalis. More importantly, axostyle and flagella were clearly stained with the improved method. Conclusion Improved Wright-Giemsa staining is simple and can be used in clinical diagnosis.
