1.Effect and Molecular Mechanism of Paeonol on Renal Interstitial Fibrosis in Rats

Objective To study the effect and mechanism of paeonol(PAE)on renal interstitial fibrosis in rats.Methods The rats were randomly divided into sham operation(Sham)group,unilateral ureteral obstruction(UUO)group,PAE low dose(PAE-L)group,PAE medium dose(PAE-M)group,PAE high dose(PAE-H)group and irbesartan(IRB)group.Except for the Sham group,the UUO model was established in other groups.Each group was given a corresponding intervention for two weeks.Serum creatinine(Scr),blood urea nitrogen(BUN),serum 8-hydroxydeoxyguanosine(8-OHdG)levels,renal tissue superoxide dismutase(SOD),glutathione peroxidase(GPX)activities,α-smooth muscle actin(α-SMA),type Ⅰ collagen(Col-Ⅰ),fibronectin(FN),silent information regulator 1(SIRT1),nuclear factor E2-related factor 2(Nrf2)protein expression were detected;observe pathological changes of kidney tissue and calculate collagen volume fraction(CVF).Results Compared with the UUO group,the serum levels of Scr,BUN,and 8-OHdG in each dose group of PAE were decreased,the activities of SOD and GPX in kidney tissue were increased,the positive expressions of α-SMA,Col-Ⅰ and FN in kidney tissue were decreased,and the protein expressions of SIRT1 and Nrf2 were increased.Masson staining showed a decrease of CVF in renal tissue(all P<0.05),and HE staining showed a different degree of improvement in pathological changes such as inflammatory cell infiltration and tubular dilatation in renal tissue;PAE improves renal interstitial fibrosis in rats in a dose-dependent manner(P<0.05),and the effect of large dose PAE on renal interstitial fibrosis in rats was similar to that of IRB.Conclusion PAE can alleviate UUO-induced rat renal interstitial fibrosis and oxidative stress,and improve rat renal function.And this mechanism may be related to the activation of the SIRT1/Nrf2 signaling pathway.
2.Construction of CRISPR/Cas9 knock-in system for tardigrade damage suppressor gene Dsup and its effect on HEK 293T cell proliferation

Objective To construct HEK 293T cells that express tardigrade Dsup protein fused with green fluorescent protein copGFP in order to study the effect of Dsup protein on proliferation of HEK 293T cells.Methods The CRISPR/Cas9 gene knock-in system was constructed.The target gene fragments of Dsup,copGFP,EF1α and puromycin were amplified by PCR and inserted into pAAVS1-SFFV to construct the fusion vector of Dsup and copGFP,which was known as pAAVS1-SFFV-Dsup-copGFP-EF1α-Puro.pAAVS1-SFFV-Dsup-copGFP-EF1 α-Puro and pAAVS1-CRISPR-Cas9 vector were co-transfected into HEK 293T cells before Dsup gene was inserted into the AAVS1 region of HEK 293T cells via homologous recombination.The HEK 293T cells expressing Dsup gene were obtained following puromycin selection,flow cytometry sorting and genome identification.The expression of Dsup at mRNA and protein levels and proliferation-related genes(MCM2,MCM4,PCNA,Ki-67)were examined to investigate the effects of Dsup gene on the proliferation of HEK 293T-Dsup-copGFP cells.Results The pAAVS1-SFFV-Dsup-copGFP-EF1α-Puro recombinant vector was constructed,and the HEK 293T-Dsup-copGFP cells with Dsup gene inserted in the AAVS1 region were obtained,where both Dsup mRNA and protein were expressed.The cell proliferation rate of HEK 293T-Dsup-copGFP was higher than that of HEK 293T-Control-copGFP(P<0.001).Further investigation revealed that the expressions of Ki-67 and MCM4 protein in HEK 293T-Dsup-copGFP were significantly higher than in the control group,indicating that the knock in of Dsup gene might enhance the proliferation ability of human cells by promoting the expression of Ki-67 and MCM4 protein.Conclusion A gene editing vector is constructed,and stable cell line HEK 293T-Dsup-copGFP for Dsup fusion expression with copGFP is established.The expression of Dsup gene in HEK 293T cells can promote cell proliferation,possibly by upregulating the expressions of Ki-67 and MCM4 protein.
3.Development of a fast Monte Carlo dose verification module for helical tomotherapy

Objective To develop a GPU-based Monte Carlo dose calculation module for helical tomotherapy(TOMO),and integrate it into the commercial software ArcherQA to achieve fast and accurate dose verification in clinic.Methods The TOMO treatment head was modeled using TOPAS to obtain phase space files,and a fast weight tuning algorithm was used to simulate particle transport in multi-leaf collimator for improving computational efficiency,and finally,GPU-based Monte Carlo algorithms in ArcherQA were used to simulate particle transport in patients.To verify the model accuracy,the ArcherQA calculated results in water tank were compared with measured data for different open fields.In addition,multiple comparisons among ArcherQA results,TPS results and ArcCHECK results were conducted on 15 clinical cases(5 cases in the head and neck,5 cases in the chest and abdomen,and 5 cases in the whole body).Results In the water tank tests for 40 cm×5.0 cm,40 cm×2.5 cm and 40 cm× 1.0 cm radiation fields,the average global relative errors of the percentage depth dose,transverse dose distribution,and longitudinal dose distribution calculated by ArcherQA with the corresponding measured values were 0.72%,0.66%,and 0.54%,respectively.Over 98%of the voxels had a global relative error of less than 1%.As for 15 clinical cases,in 2%/2 mm criteria,the mean Gamma passing rate was 98.1%between ArcherQA and TPS,99.1%between TPS and ArcCHECK,and 99.4%between ArcherQA and ArcCHECK.The uncertainty of the simulation maintained less than 1%,and the average time taken for calculation based on patient CT vs ArcCHECK phantom was 87 s vs 64 s.Conclusion ArcherQA can be used for independent dose validation for TOMO plans for it can provide fast and accurate dose calculations.
4.QDPR improves UUO⁃induced renal interstitial fibrosis in rats by regulating Beclin1

Objective:
To investigate the effect of QDPR on renal interstitial fibrosis and Beclin1 expression of unilateral ureteral obstruction ( UUO) rat model.
Methods:
The UUO model was constructed by unilateral ureteral ligation. The fibrosis progression , QDPR and Beclin1 expression of UUO model were evaluated by Masson , COL Ⅰ staining and Western blot techniques. UUO model which was over expression QDPR and its control were constructed by retrograde ureteral infusion of corresponding lentivirus. Infection efficiency and QDPR expression level were further evaluated. The QDPR overexpression on fibrosis progression , and Beclin1 was further analyzed.
Results:
Compared with the sham operation group , the collagen fibers level in the UUO model group significantly increased (P <0. 05) , the QDPR expression level was significantly reduced (P < 0. 05) , and the Beclin1 significantly increased (P< 0. 05) . Retrograde injection of lentivirus through the ureter could successfully infect the kidney and increase the expression level of QDPR in the kidney. The overexpression of QDPR in kidney could significantly decrease collagen fibers (P < 0. 05) and Beclin1 (P < 0. 05) expression.
Conclusion
Overexpression of QDPR inhibits the expression level of Beclin1 and progression of fibrosis , suggesting that overexpression of QDPR may relieve renal interstitial fibrosis by inhibiting Beclin1expression.
5.Research progress on total skin irradiation using helical tomotherapy

Cutaneous T-cell lymphomas are a relatively rare group of mature T-cell lymphomas mainly manifesting in the skin, and its common subtype is mycosis fungoides. Total skin electron irradiation is one of the important conventional treatment methods, but there are many disadvantages, such as uneven dose distribution, poor position repetition, and long treatment time, which affect the clinical efficacy and patient prognosis. With the emergence and gradual popularization of helical tomotherapy in recent years, more and more medical institutions are gradually expanding their applications in total skin irradiation due to their ability to treat ultra-long targets and achieve dose-sculpted distribution, aiming to further explore its good or bad, and confirm whether it can replace the traditional total skin electron irradiation. In this article, research progress on total skin irradiation using helical tomotherapy was reviewed, the development of treatment technology, clinical efficacy and current concerns and controversies were illustrated.
6.Gender stereotype of early adolescents and its influencing factors

Objective:
To explore the gender stereotype in early adolescence and the potential influencing factors.
Methods:
Stratified cluster sampling method was adopted to recruit students of grade 6-8 in three middle schools of Shanghai during November to December, 2017. Students were surveyed anonymously using Computer Assisted Self-Interview approach regarding geder stereotype and associated factors.
Results:
Average gender stereotype score was (3.32±0.75), with boys(3.43±0.75)higher than the girls(3.20±0.72)(t=6.37, P<0.05). The "agreement" proportion was highest among gender stereotypes items which reflect personality characteristics such as "it’s important for boy to be strong and determined" "girls should be like a lady" and "girls were expected to be submissive"(60.99%,50.79%,43.95%), and the "agreement" proportion among boys increased with age, while decreased in girls. Boys who had sisters or recent bullying behaviors showed more gender stereotype[β(95%CI)=0.19(0.05-0.33), 0.39(0.12-0.67)]. Girls whose parents allowed to go to opposite sex homes alone had less gender stereotype, however, the gender stereotype became stronger when girls had more female peers[β(95%CI)=-0.22(-0.41--0.03),0.04(0.00-0.07)].
Conclusion
The young adolescents, especially boys, show certain level of gender stereotype. The normal heterosexual friendships and safe campus environments would help promote young adolescents to establish equal gender beliefs.
7.Heterosexual romantic expectation of early adolescents and its influencing factors

Objective:
To explore the heterosexual romantic expectation in early adolescence(10-14 years)and its influencing factors, and to provide a reference for instructing children of early puberty to establish a positive interpersonal relationship(including heterosexual relationship) and improving their healthy development.
Methods:
Stratified cluster sampling method was adopted to recruit students of grade 6-8 in three middle schools of Shanghai during November to December, 2017. Students were surveyed anonymously using Computer Assisted Self-Interview approach via the tablets. The collected data included the information on demographic, families, peers, use of the media, community cohesion and heterosexual romantic expectation. The ttest, Chi-square test, ANOVA and multiple linear regression were used to explore the relationships between potential factors and heterosexual romantic expectation.
Results:
The mean score of heterosexual romantic expectation was 2.55. Score of the boys was higher than that of girls (2.62 vs. 2.47, t=2.65, P<0.05). There was no statistically significant difference between the scores of the ages before 12 years old (10-11: 2.34, 12: 2.28), but a significant increase of the scores along the age after 12 years old(13: 2.69, 14: 3.05). Multiple linear regression suggested that respondents with older age, poorer caregiver-child relationship, higher proportion of friends of the opposite sex, more friends that thought having boyfriends/girlfriends was important, more TV/movie use and lower community cohesion had higher score of heterosexual romantic expectation of early adolescents(P<0.05).
Conclusion
The early adolescents had relative conservative views on the heterosexual romantic relationships of adolescents of their own age while the boys were more permissive than girls. The heterosexual romantic expectation improved significantly after 12 years and may be influenced by factors of family, peers, media and community.
8. Application of low temperature thermoplastic plate combined with special abdominal band in fixing abdominal pedicled flap for repairing 17 patients with deep electric burn wounds in hands

If the abdominal pedicled flaps are not well fixed after repair of deep electric burn wounds in hands, many problems such as poor blood supply may occur. In order to solve the above problems, we designed and manufactured the individualized low temperature thermoplastic plate combined with special abdominal band to fix abdominal pedicled flaps for repairing of 17 patients (12 males and 5 females, aged 2-35 years) with deep electric burn wounds in hands from February 2016 to August 2018, and achieved the desired results. The shoulder joint, elbow joint, and wrist joint were fixed by low temperature thermoplastic plate according to the 1/2 circumference of the patient′s side chest and upper arm, and the braking of wrist joint and elbow joint was strengthened by special abdominal band. Application of the combined method of fixing abdominal pedicled flaps in repairing deep electric burn wounds in hands has high success rate of flap transplantation. It is simple to make and practical, and worthy of clinical promotion.
9. Clinical efficacy observation on combination of narrow-spectrum medium-wave ultraviolet, red light, and low power He-Ne laser in treating post-burn eczema

Objective:
To investigate the clinical effects of combined utilization of narrow-spectrum medium-wave ultraviolet, red light, and low power He-Ne laser on treatment of post-burn eczema.
Methods:
From July 2014 to July 2017, 80 patients with post-burn eczema who met the study inclusion criteria were treated in our burn rehabilitation center, and their clinical data were analyzed retrospectively. Patients were divided into ultraviolet treatment group, red light treatment group, laser treatment group, and combination treatment group according to the utilized treatment methods, with 20 cases in each group. Patients in ultraviolet treatment group were treated with narrow-spectrum medium-wave ultraviolet once every other day for 10 minutes each time. Patients in red light treatment group and laser treatment group were treated with red-light and low power He-Ne laser respectively once a day for 10 minutes each time. Patients in combination treatment group were treated with combination of the above three methods without sequence or interval time, and the treatment time and interval time were the same as the previously described. All patients were treated for four weeks since the time of admission. The itching degree, surface area of the affected body, degree of keratosis, and degree of cracking of target tissue were evaluated using Eczema Area and Severity Index (EASI) scoring method after each treatment. Eczema improvement rate was calculated according to the total score of EASI to determine the curative effect. The therapeutic effective time was recorded. The therapeutic effective rate was calculated according to the total scores of EASI before treatment for the first time and 4 weeks after treatment. Besides, the adverse reactions of patients were also observed and recorded. 10.Oral health status of patients undergoing hemodialysis: a Meta-analysis.

OBJECTIVEThis study aimed to systematically evaluate the oral health status of patients suffering from chronic renal failure and undergoing hemodialysis (HD).
METHODSElectronic databases, namely, China National Knowledge Infrastructure, Wanfang Data base, PubMed, Web of Science, and Cochrane Library, were searched until May 2016 for cross-sectional and case-control studies that investigated the oral health status of patients suffering from chronic renal failure and undergoing HD. Two reviewers independently screened the trials, extracted the data, assessed the quality, and conducted a Meta-analysis with RevMan 5.3 in accordance with the inclusion and exclusion criteria.
RESULTSSixteen studies satisfied the eligibility criteria. Meta-analysis revealed that plaque index (MD=0.62, 95%CI: 0.51-0.72), calculus index (MD=1.09, 95%CI: 0.56-1.63), probing depth (MD=0.63, 95%CI: 0.29-0.98), and attachment loss (MD=0.63, 95%CI: 0.56-0.69) were significantly higher in the patients undergoing HD than in the healthy control group (P<0.01). No significant differences in decayed missing filled teeth (MD=1.12, 95%CI: -1.08-3.32) were observed between the patients undergoing HD and the healthy control group (P=0.32).
CONCLUSIONSThe prevalence of caries in patients undergoing HD is similar to that in the healthy control group. However, these patients are at a higher risk of acquiring periodontal diseases than the healthy control group. Thus, the oral health status of these patients should be further evaluated in clinical treatments.
