Developing and implementing biosafety standards for pathogenic microbiology laboratories is essential to achieving scientific, efficient, and standardized management and operation. This article analyzes the current standardization construction in biosafety in pathogenic microbiology laboratories domestically and internationally. It proposes a framework for the biosafety standard system of pathogenic microbiology laboratories, which mainly includes four parts: basic standards, management standards, technical standards, and industry applications. It provides a reference for the standardization work of pathogenic microbiology laboratories and helps to standardize the biosafety industry in China.

Positive pressure bio-protective equipment is a key protective equipment for high-level biosafety laboratories, which provides comprehensive protection for professionals working in pathogenic microorganisms or other biological contamination environments. This paper summarizes the development status of positive pressure bio-protective equipment at home and abroad, analyzes the existing problems of the current positive pressure bio-protective equipment, and puts forward suggestions, to provide reference for the development and application of positive pressure bio-protective equipment in our country.
4. Pathogenic composition of hand, foot and mouth disease and enterovirus-A71 and coxsackievirus-A16 genetic characteristics analysis in Ningxia, 2016-2017

Objective:
To understand the pathogenic composition of hand, foot and mouth disease (HFMD) in Ningxia Hui Autonomous Region (Ningxia) from 2016 to 2017, and analyze the genetic characteristics of the main pathogens enterovirus (EV)-A71 and coxsackievirus (CV)-A16.
Methods:
Analysis of the result of nucleic acid testing of HFMD in Ningxia from 2016 to 2017 to determine the pathogenic composition of HFMD. The complete VP1 coding region was amplified by RT-PCR and the gene sequence was determined for the enterovirus strains sent to the National HFMD Network Monitoring Laboratory in Ningxia from 2016 to 2017. BLAST analysis confirmed the serotype of the strain, and the phylogenetic tree was constructed respectively by selecting EV-A71 and CV-A16 isolates.
Results:
The leading pathogens of HFMD in Ningxia of 2016 and 2017 were other EV (397, 43.72%) and EV-A71 (918, 56.18%) respectively, and the dominant pathogens in different months may differ. The pathogenic composition causing HFMD in the past two years has changed from CV-A16 and other EV to EV-A71 and other EV. The isolated EV-A71 strains were C4a evolutionary branch and the isolated CV-A16 strains were B1b evolutionary branch.
Conclusions
Compared to 2016, in 2017 EV-A71, CV-A16 and other EV changed dynamically. Dynamic monitoring of EV-A71 in Ningxia is of great significance to guide the strategy of using EV-A71 vaccine, concentrating medical resources to strengthen the treatment and reduce the mortality rate of severe HFMD cases.
5. A study of establishment of non-standard methods in qualification accreditation of virological laboratories

The laboratories of National Institute for Viral Disease Control and Prevention, Chinese Center for Disease Control and Prevention, have developed a series of experimental method . These method have unique advantages over the national standard method and industry standard method . How to make these method become public products through legal procedures to serve disease prevention and control more extensively is an obligatory task for national virological laboratories. This article explores the establishment, verification, validation, and examination of virological non-standard method under laboratory certification and accreditation conditions through empirical research on the " RT-RAA method for detecting EV71/CA16 and other viruses" .
6.Inflammatory mechanism and significance of tumor necrosis factor-α and interleukin-1β in pathogenesis of conjunctivochalasis

Background Conjunctivochalasis (CCh) is a common age-related ocular surface diseases.Researches showed that the inflammatory factors are upregulated in conjunctiva and tear of CCh patients,inferring inflammation participates in the pathogenesis of CCh,however,its mechanism is unclear now.Studies also showed that the expression of matrix metalloproteinases (MMPs) in CCh conjunctiva is elevated.However,the association between inflammatory factors and MMPs is unknown.Objective This research was to observe the effects of tumor necrosisfactor-α (TNF-α) and interleukin-1 β (IL-1 β) on the expression of MMP-1,MMP-3,tumor necrosis factor-stimulatedgene-6(TSG-6) and pentraxin-3 (PTX3) in cultured CCh fibroblasts.Methods Conjunctival fibroblasts wereisolated and cultured from CCh-derived and cataract-derived human conjunctiva explants.The cells were treated using20 ng/ml PBS,TNF-α or IL-1β for 4 hours,respectively.The expression levels of MMP-1,MMP-3,TSG-6 and PTX3genes and proteins were detected by quantitative real time PCR and Western blot assay,respectively.Results Thesecond-generation cells showed a long and fusiform shape with ovoid nucleus and radial agreement,and the cellprocessors connected each other.The differences of the relative expression levels of MMP-1,MMP-3,TSG-6 and PTX3mRNA in the cells were significantly different between CCh group and cataract group after being treated by PBS,TNF-αand IL-1 β (MMP-1 mRNA:Fgroup =2.611,P =0.116;Fi =161.564,P =0.000;MMP-3 mRNA:Fgroup =5.201,P =0.029;Fintervene =211.021,P =0.000;TSG-6 mRNA:Fgroup =47.209,P =0.000;Fintervene =119.340,P =0.000;PTX3 mRNA:Fgroup =40.512,P =0.000;Fintervene =93.935,P =0.000).Compared with the cataract group with PBS treatment,the expression levels of MMP-1,MMP-3,TSG-6 and PTX3 mRNA were significantly elevated in the CCh group with PBS treatment or the cataract group with TNF-α and IL-1 β treatment (all at P<0.05).Compared with the CCh group with PBS treatment,the expression levels of MMP-1,MMP-3,TSG-6 and PTX3 mRNA were significantly elevated in the CCh group with TNF-α and IL-1β treatment (all at P < 0.05).The differences of the relative expression levels of MMP-1,MMP-3,TSG-6 and PTX3 proteins in the cells were significantly different between CCh group and cataract group after being treated by PBS,TNF-α and IL-1 β (MMP-1:Fgroup =84.702,P =0.000;Fint =48.900,P =0.000;MMP-3:Fgroup =112.818,P =0.000;Fint =194.980,P =0.000;TSG-6:Fgroup =56.867,P =0.000;Fint =70.356,P =0.000;PTX3:Fgroup =1.488,P =0.231;Fint =89.872,P =0.000),and the expression changes of MMP-1,MMP-3,TSG-6 and PTX3 proteins were coincident with the genes among the groups with various treatments.Conclusions The expressions of MMP-1,MMP-3,TSG-6 and PTX3 in conjunctival fibroblasts are upregulated in CCh eyes.The interaction of TNF-α and IL-1 β with MMPs is probably involved with the pathogenesis and development MMPs probably.
7.Application effect of systematic nursing intervention mode on teenager patients with internet addiction ;disorder

Objective To investigate the influence of systematic nursing intervention on family cohesion, adaptability, internet addiction rehabilitation degree and psychological status of patients with internet addiction disorder ( IAD ) . Methods IAD patients were randomly divided into control group ( n=50 ) and experimental group ( n=50) according to random number table. Patients in the control group were intervened by conventional nursing treatment. On the base of that, patients in the experimental group were intervened by systematic nursing intervention mode, such as inspirational education, comment classroom of nursing, systematic health education, regular follow-up, etc. Changes in family cohesion, adaptability, internet addiction rehabilitation degree and psychological status of the IAD patients were evaluated before and three months after the intervention.Results After the intervention, of patients in the experimental group, scores of ideal family cohesion improved from ( 67. 54 ± 4. 58 ) points to ( 74. 63 ± 3. 20 ) points; scores of actual family cohesion improved from (53.14±4.29) to (76.29±3.93); scores of ideal adaptability improved from (46.43±7.98) to (57.50±6.05);scores of actual adaptability improved from (44.90±5.78) to (50.78±5.43);scores of internet addiction rehabilitation degree declined from (83.37±3.46) to (56.01±5.02);various indexes of psychological health status had all improved significantly than before the treatment ( P<0. 05 ) . Indexes of patients in the control group showed no significant improvement (P>0.05).Conclusions Systematic nursing intervention mode can obviously improve physiological and psychological conditions of IAD patients, and increase the rate of internet addiction rehabilitation.
8.Influence of positive psychological intervention on occupational stress and mental health in operating room nurses

Objective To research the influence of positive psychological intervention on occupational stress and mental health in operating room nurses. Methods Totals of 50 operating room nurses working in our hospital from July 2013 to January 2014 were selected. According to the principles of randomized control, they were divided into the experimental group and the control group, with 25 cases in each. The experimental group underwent positive psychological intervention, while the control group got no psychological intervention. The occupational stress and psychological conditions of the two groups were compared and analyzed before and after the intervention. Results Before the intervention, there were no significant differences in occupational stress and mental health between the two groups (P>0. 05). After the intervention, scores of EE, DP, PA and SCL-90 were (16. 7 ± 5. 1), (5. 5 ± 2. 8), (40. 2 ± 8. 9) and (136. 7 ± 40. 4) points in the experimental group, which were significantly better than those of the control group (t=5. 864 3, 2. 397 7, 5. 170 7, 2. 654 5, respectively;P<0. 05). Conclusions Continual positive psychological intervention can significantly ease the pressure on the operating room nurses and improve their mental health, and it is worthy of promoting.
9.Impact of ischemic stroke on the intestinal barrier function in dogs

Objective To explore the impact of ischemic stroke on intestinal barrier changes in dogs.Methods Totally 20 mongrel dogs were divided into 2 groups by random number table with 10 in each.Double silicone cylinders measuring 1.1 mm in diameter and 8 mm in length were placed into their internal carotid arteries in all dogs of group A.Group B served as a control group and received sham operation.Light microscopy was performed for morphological measurement of intestinal epithelial cell.Immunohistochemistry was used to analysis the changes of protein zonula occludens-1(ZO-1)localizing at tight junction of intestinal epithelial cells.Results Ischemic stroke was confirmed by cranial CT scanning in all dogs of group A.Compared with the test results in group B,the occludin and Zo-1 protein levels in group A were significantly lower than those in group B(occludin:0.20 ±0.01 vs 0.22 ±0.01,P =0.007; ZO-1:0.20 ±0.01 vs 0.22 ±0.02,P =0.008).The apoptotic index in group A was significantly higher than in group B(29.04 ± 3.79 vs 6.44 ± 1.24,P =0.002).There was a positive correlation between occludin and ZO-1(R =0.71,P =0.02),and the apoptotic index was negatively correlated with levels of occludin,ZO-1(R =-0.91,P =0.00; R =-0.77,P =0.01).Light microscopy showed that the dogs in group A had intestinal mucousal injuries while no obvious change was detected in group B.Conclusions Dogs with ischemic stroke tend to develop intestinal barrier dysfunction,during which the destruction of tight junction plays a key role.The up-regulated apoptosis of intestinal epithelial cell constitutes one of the cellular bases of intestine injury.
10.Evaluation System for Risk Factors of Nosocomial Infection in ICU

OBJECTIVE To evaluate the risk factors of nosocomial infection(NI),then establish the evaluation system for risk factors of NI in ICU.METHODS A total of 1897 ICU patients were enrolled in our study.The risk factors of NI were classified and quantified,then investigated the relativity of the risk factors and NI.RESULTS NI correlated with APACHEⅡscore,serum prealbumin level,blood products uese,incision of trachea,mechanical ventilation,central vein cannula and enteral nutrition.CONCLUSIONS The risk factors of NI evaluatied in ICU can to be used to lay the foundation for the model of NI early warning.
