1.miR-429-3p mediates memory decline by targeting MKP-1 to reduce surface GluA1-containing AMPA receptors in a mouse model of Alzheimer's disease.

Alzheimer's disease (AD) is a leading cause of dementia in the elderly. Mitogen-activated protein kinase phosphatase 1 (MKP-1) plays a neuroprotective role in AD. However, the molecular mechanisms underlying the effects of MKP-1 on AD have not been extensively studied. MicroRNAs (miRNAs) regulate gene expression at the post-transcriptional level, thereby repressing mRNA translation. Here, we reported that the microRNA-429-3p (miR-429-3p) was significantly increased in the brain of APP23/PS45 AD model mice and N2AAPP AD model cells. We further found that miR-429-3p could downregulate MKP-1 expression by directly binding to its 3'-untranslated region (3' UTR). Inhibition of miR-429-3p by its antagomir (A-miR-429) restored the expression of MKP-1 to a control level and consequently reduced the amyloidogenic processing of APP and Aβ accumulation. More importantly, intranasal administration of A-miR-429 successfully ameliorated the deficits of hippocampal CA1 long-term potentiation and spatial learning and memory in AD model mice by suppressing extracellular signal-regulated kinase (ERK1/2)-mediated GluA1 hyperphosphorylation at Ser831 site, thereby increasing the surface expression of GluA1-containing α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors (AMPARs). Together, these results demonstrate that inhibiting miR-429-3p to upregulate MKP-1 effectively improves cognitive and synaptic functions in AD model mice, suggesting that miR-429/MKP-1 pathway may be a novel therapeutic target for AD treatment.
2.Efficacy of fractional CO 2 laser and 1064-nm, Q-switched Nd∶YAG laser on xanthelasma palpebrarum

Objective:To determine the clinical efficacy and safety of a fractional CO 2 laser and a 1 064 nm, Q-switched Nd∶YAG laser therapy in the treatment of xanthelasma palpebrarum. Methods:From October 2020 to October 2021, 30 patients (5 males and 25 females) with bilateralxanthelasma palpebrarum of the eyelid were enrolled in the Department of Dermatology, the First Affiliated Hospital of Xiamen University. The age ranged from 38 to 67 (51±7) years. One side was randomly treated with fractional CO 2 laser as the fractional group, and the other side was treated with Q-switch 1 064 nm Nd∶YAG laser as the Q-switch group. The treatment was given every 28 days for 4 times. Before treatment and 1 month after the last treatment, the general pictures were taken to compare the clinical effect. Skin ultrasound was used to measure the difference of tumor thickness before and after treatment. The incidence of adverse reactions, such as local scar, hyperpigmentation or hypopigmentation after inflammation, were recorded. Results:Under general photos, there was statistically significant difference in efficacy scores between the two groups before and after treatment ( Z=-3.082, P<0.05). By comparison of tumor thickness under skin ultrasound, the difference between the two groups before and after treatment was statistically significant ( t=21.60, P<0.05; t=17.29, P<0.05); there was no statistically significant difference between the two groups before treatment ( t=0.46, P=0.650), but there was statistically significant difference between the two groups after treatment ( t=8.41, P<0.001). No serious adverse reactions occurred in both groups. Conclusions:Fractional CO 2 laser or Q-switch 1 064 nm Nd∶YAG laser can safely and effectively improve xanthelasma palpebrarum, in which the effect of fractional CO 2 laser is much better.
3.Protein O-linked mannose β1, 2-N-acetylglucosaminyltransferase 1 gene is involved in the pathogenic process of Alzheimer's disease model mice

Objective To investigate the role of protein O-linked mannose β1,2-N-acetylglucosaminyltransferase 1 (POMGnT1) in the pathogenic process of Alzheimer's disease (AD).Methods The expression of POMGnT1 gene was examined in AD model cells (N2a/amyloid-precursor protein (APP) 695swe,n=3) and N2a/wt cells (n=3) using real-time PCR and Western blotting.This expression was also examined in AD model mice (APP/PS1,n=3) and wild-type mice (n=3) using immunofluorescence staining.Amyloid β-protein (Aβ) were examined in POMGnT1-gene-knockout mice (n=3) and wild-type mice (n=3) using immunochemistRy.Results The expression of POMGnT1 gene decreased in mRNA and protein levels in N2a/APP695swe cells compared to N2a/wt cells (mRNA:0.80±0.02 vs 1.00,t=10.52,P<0.01;protein:0.50±0.02 vs 1.31±0.04,t=18.64,P<0.01).Immunofluorescence results showed the reduced expression of POMGnT1 protein in neurons of APP/PS1 mice.Immunochemistry results showed more Aβ deposits in POMGnT1-gene-knockout mice (2 months old:0.358±0.014 vs 0.048±0.001,t=22.58,P<0.01;1 year old:0.266±0.004 vs 0.229±0.003,t=7.771,P=0.002).Conclusion These findings suggest POMGnT1 gene may play an important role in the pathogenic process of AD.
4.Survey of the readiness for hospital discharge and its influencing factors among chronic renal failure patients undergoing arteriovenous fistula surgery

0bjective To investigate the status of readiness for hospital discharge of chronic renal failure patients undergoing arteriovenous fistula surgery and analyze its influencing factors. Methods A total of 229 diabetic patients who were discharged from the department of endocrinology were surveyed by the general data questionnaire, Readiness for Hospital Discharge Scale (RHDS), Quality of Discharge Teaching Scale (QDTS) and Social Support Rating S1cale (SSRS). Results The total score of RHDS was (157.20±19.39) points, and the items were equally (7.15±0.88) points. Education level, inhabiting pattern, discharge guidance skills and subjective support entered the readiness for hospital discharge regression equation of chronic renal failure patients undergoing arteriovenous, accounting for a total of 33.1% of all the variation. Conclusions The readiness for hospital discharge of chronic renal failure patients undergoing arteriovenous fistula surgery is intermediate level. Education level, inhabiting pattern, discharge guidance skills and subjective support could exert important influence on readiness for hospital discharge of chronic renal failure patients undergoing arteriovenous fistula surgery.
5.Application of enteral nutrition support via naso-jejunal tube in esophageal carcinoma patients treated with ;radiotheraphy

Objective To investigate the effect of enteral nutrition support via naso-jejunal tube in esopha-geal carcinoma patients treated with radiotheraphy.Methods 36 esophageal carcinoma patients were randomly assigned into enteral nutrition(EN)group,while 38 patients assigned to control group.All patients underwent defini-tion IMRT combined with weekly concurrent chemotherapy of paclitaxel-nedaplatin.The naso -jejunal tubes were bedside inserted by hand in EN group.Enteral nutrition support began the day after the tube insertion.The control group took food orally.Nutrition was assessed through body weight,BMI,lymphocyte,albumin,pre -albumin and hemoglobin.Treatment induced complications were recorded.Results The degree of nutritional reduction was lower in EN group and significantly different with the control group.The EN group underwent (4.5 ±1 .1 )cycles concurrent chemotherapy,the control group underwent (3.1 ±2.3)cycles concurrent chemotherapy(t=6.21,P=0.027).The hematotoxicity induced by chemoradiotherapy(CRT)was statistically severe in the control group(χ2 =24.64,P<0.01),while radiation esophagitis was similar between the two groups.Conclusion EN support via naso -jejunal tube in esophageal carcinoma patients treated with radiotheraphy may improve the nutritional status,alleviate CRT induced hematotoxicity,increase tolerance of CRT.
6.Relationship Between Prevalence of Atrial Fibrillation, Ischemia Stoke and CHA2DS2-VASc Score in Elderly Patients

Objective: To explore the relationship between prevalence of atrial fibrillation (AF), iskhemia stoke and CHA2DS2-VASc score in patients≥65 years in order to provide prevention and treatment basis in clinical practice. Methods: A total of 5016 patients admitted in our hospital from 2013-10 to 2015-10 were enrolled. The patients were divided into 2 groups: AF group, n=437 and Non-AF patients, n=4579; according to age, the patients were further assigned into 4 subgroups as <65 years subgroup, (65-74) years subgroup, (75-84) years subgroup and ≥85 years subgroup. The risk factors for AF occurrence were retrospectively studied. Results: Compared with the Non-AF group, the patients in AF group had the elder age and more male gender, both P<0.001; more patients combining with hypertension, coronary artery disease (CAD), diabetes, sick sinus syndrome and rheumatic heart disease, all P<0.001. Age, male gender, CAD, sick sinus syndrome and rheumatic heart disease were the independent risk factors for AF occurrence. Compared with Non-AF group, AF group showed the higher prevalence rate of ischemic stroke and the elder onset age, both P<0.01. For non-valvular AF, the ratio of patients with CHA2DS2-VASc score≥2 was higher than those with CHA2DS2-VASc score<2 and the rate of anticoagulant therapy was decreasing by age increasing, all P<0.001. Conclusion: Age, male gender, CAD, sick sinus syndrome and rheumatic heart disease were independently related to AF occurrence. Non-valvular AF patients had the higher risk for ischemic stroke than non-AF patients, anticoagulation therapy should be conducted at the early stage.
7.Diterpenoid Alkaloids in the Roots of Aconitum Iochanicum

Objective: To study the chemical constituents in the roots of Aconitum iochanicum Ulbr.Methods: The air-dried roots of A.iochanicum were powdered and extracted by methanol with a percolation method.After removing the solvent under reduced pressure, the crude extract was dissolved in1.5% HCl solution, and then extracted by ecetic ether.The acidic solution was basified to pH 9.0 by NaOH (5%) and extracted with ethyl acetate to obtain crude alkaloidal extract after the removal of ethyl acetate.The compounds were isolated and purified by column chromatography and identified based on spectral analysis (1H-NMR, 13C-NMR and MS).Results: Totally 18 compounds were isolated from A.iochanicum and characterized as 14-O-acetylsachaconitine (1), franchetine (2), crassicaudine (3), indaconoitine (4), 14-benzoyl chasmanine (5), 14-O-acetyltalatisamine (6), talatisamine (7), chasmannine (8), crassicauline A (9), bikhaconine (10), 13,15-dideoxyaconitine (11), crassicautine (12), kongboensine (13), liljestrandisine (14), ludaconitine (15), 8-deacetyl-yunaconitine (16), yunaconitine (17) and ouvrardiantine (18).Conclusion: It''s the first time to study the chemical constituents of A.iochanicum, and 18 diterpenoid alkaloids are isolated.

Methods:The endogenous fungui fermentation product was extracted by methanol followed by recycling methanol to obtain the extract, and the ex-tract was extracted by ethyl acetate to obtain the crude extract. The compounds were isolated and purified by column chromatography and identified based on the spectral analysis ( MS, 1 H-NMR and 13 C-NMR) . Results:Totally 12 compounds were isolated from A. so-lani and characterized as (22E, 24R)-Ergosta-4, 6, 8 (14), 22-tetraen-3-one (1), 1,3-Diolein (2), 5-(3', 3'-Dimethylallyloxy)-3-methoxy-4-methylphthalide (3), pseudomonas aeruginosa H2-sesquiterpene lactone (4), ergosterol peroxide (5), 7-dehydroxyl-zinni-ol (6), ergosterol-7, 22-diene-3β, 5,6-triol (7), 9-octadecenoic acid -2',3'-dihydroxy propyl ester (8), 8-O-Methyltalatisamine (9), Chasmanine (10), yunaconitine (11) and crassicauline A (12). Conclusion:Totally 12 compounds are isolated from the endo-phytic fungus Alternaria solani, and four of them are isolated for the first time.

10.Application of modified Seldinger technology in PICC intubation for chemotherapy

Objective To explore the effect of modified Seldinger technology in PICC intubation for chemotherapy . Methods Forty patients with chemotherapy during January to December 2014 were assigned as the control group, where the traditional PICC intubation was used, and another forty patients during January to December 2014 as the observation group, where modified Seldinger technology was used. The two groups were compared in terms of post-intubation comfort, puncture success rate, bleeding rate and complications. Results The post-intubation comfort degree of the observational group was significantly higher than that of the control group (P<0.05). The total puncture success rate and one-time success rate were both statistically significantly higher than those of the control group, and the rates of obstruction and infection were significantly lower than those of the control one (P<0.05). Conclusion Modified Seldinger technology used for intubation for chemotherapy can help improve comfort degree and safety and reduce the rate of complications.
