1.Study on screening for 274 suspected cases of corona virus disease 2019

		                        			Objective:To evaluate the diagnostic efficacies of computed tomography (CT), clinical manifestations and 2019 novel coronavirus (2019-nCoV) nucleic acid test results for the screening and diagnosis of novel coronavirus pneumonia.Methods:The clinical data of suspected cases with corona virus disease 2019 (COVID-19) visited fever clinic or stayed in emergency room of the First Affiliated Hospital of Nanchang University from January 23 to February 9, 2020 were collected. Totally 274 cases who met the inclusion criteria were enrolled. Four screening schemes including chest CT screening, scoring, chest CT and scoring series screening, chest CT and scoring parallel screening were designed. The statistical analysis was performed by using chi-square test. The sensitivities, specificities and the areas under the receiver operator characteristic curve of the four screening schemes were calculated, and the diagnostic efficacies were evaluated.Results:Among the 274 cases, 93.80%(257/274) presented with typical clinical symptoms, 47.81%(131/274) had epidemiological history, and the blood routine examination results of 45.26%(124/274) cases met the positive criteria of the scoring , and chest CT results of 43.07%(118/274) cases met the positive criteria of chest CT screening. The 2019-nCoV nucleic acid test positive rate of cases with epidemiological history was 30.53%(40/131), which was higher than that of cases without epidemiological history (9.09%(13/143)), and that of cases with typical imaging performance on chest CT was 40.68%(48/118), which was higher than cases without typical imaging performance (3.21%(5/156)). The differences between the above groups were both statistically significant ( χ2=20.150、60.468, respectively, both P<0.01). The positive rates of viral nucleic acid detection in cases with positive findings of chest CT screening, scoring, series screening, and parallel screening were 40.68%(48/118), 23.74%(47/198), 44.68%(42/94) and 23.87%(53/222), respectively, while those in cases with negative findings of the four screening schemes were 3.20%(5/156), 7.89%(6/76), 6.11%(11/180) and 0(0/52), respectively. The positive rates of the four screening schemes were all significantly different from that of viral nucleic acid detection ( κ=0.402, 0.100, 0.431, 0.106, all P<0.01). The chest CT screening method had a sensitivity of 90.57%and a specificity of 68.33%, and an area under the operating characteristic curve of the subject was 0.794, which had higher diagnostic value than those of the other three screening schemes. Conclusions:For the screening and diagnosis of COVID-19 cases, epidemiological history, positive 2019-nCoV nucleic acid test with ≥2 typical clinical manifestations have highly diagnostic value. On the basis of the preliminary screening of chest CT examination, flexible analysis of the diagnostic results could improve the diagnostic value of each detection method.
		                        		
		                        		
		                        		
		                        	
2.A pilot study on the biodistribution pattern of 68Ga-DOTA-TATE in normal organs of adults

		                        			Objective To retrospectively study the biodistribution of 68Ga-DOTA-TATE as a SSTR imaging agent in human subjects.Methods A total of 106 patients with suspected disease were enrolled in this study.All patients were histologically proven for having either a single tumor <2 cm or without evidence of tumor during follow-up.Patients underwent PET/CT whole-body scan 17-100 min after intravenous injection of 55.2-220.0 MBq 68Ga-DOTA-TATE.ROI was drawn for measuring SUV of tracer-avid pathologies.One-way analysis of variance and two-sample t test were used for statistical analysis.Results High 68Ga-DOTA-TATE avidity was found in pituitary,with SUVmax of 4.00± 1.21.Tracer was excreted mainly through urinary system resulting in highest uptake in the urinary tracts.The SUVmax of kidney cortex was 19.01 ± 5.45.Mediastinal blood pool and liver SUVmax were 0.93±0.33 and 7.69±2.26,respectively.Mild uptake of 68Ga-DOTA-TATE was found in the brain,cerebellum,lung and muscle,all lower than that of mediastinal blood pool.Moderate accumulation of 68 Ga-DOTA-TATE (close to or slightly higher than liver) was found in adrenal gland and spleen,with SUVmax 7.61 ± 3.42 and 8.63± 2.31,respectively.Other organs (pituitary,salivary gland,thyroid,pancreas,small intestine,colon,uterus,prostate and bone) showed tracer uptake in the range between those of mediastinal blood pool and liver.68Ga-DOTA-TATE distribution in pancreas was not uniform.Nine patients had focal accumulation in the uncinate process of pancreas with highest SUVm~ up to 8.48.However,the SUVmax and SUV in the rest of pancreas (head,neck,body and tail) showed insignificant difference (F values:0.703,0.563,both P>0.05).68Ga-DOTA-TATE uptake in each organ reached equilibrium quickly after injection but with slight increase over time.The changes in SUV,however,showed insignificant difference among organs,including different parts of pancreas (t values:from -0.09 to 1.75,from-1.70 to-0.42,respectively,all P>0.05).Conclusions The biodistribution of 68Ga-DOTA-TATE reaches equilibrium shortly after intravenous administration and is stably maintained.The biodistribution activities are organ-specific,and characteristic to that of SSTR concentration.
		                        		
		                        		
		                        		
		                        	
3.Characteristics of neuroendocrine neoplasm metastasis in somatostatin receptor PET/CT imaging: a retrospective analysis

		                        			Objective To investigate the characteristics of NEN metastasis with SSTR PET/CT and to correlate the results with FDG-PET and pathology.Methods From November 2011 to August 2016,a total of 43 patients with NEN (18 males,25 females;age range:26-74 years) were recruited into this retrospective study;they underwent 68Ga-DOTA-TATE PET/CT (TATE-PET) imaging 40-60 min after 44.4-229.4 MBq 68Ga-DOTA-TATE administration.Metastases in 31 patients were confirmed by histopathology and in 12 patients by follow-up and other imaging modalities.Twenty-eight of 43 patients finished routine FDG-PET in a week after TATE-PET.PET/CT results were considered positive when the metastatic lesions were tracer-avid.ROI was drawn over each lesion for size and SUV measurement.x2 test and Pearson correlation analysis were used for statistical analysis.Results (1) Of 43 patients,TATE-PET detection rates of metastasis in the liver,lymph nodes,bones.and lungs were 85.7%(30/35),12/13,7/7,2/3,respectively,with their corresponding SUVmax of 18.1(11.3-23.3),10.8(5.4-15.6),7.7(4.2-9.9) and 1.8(1.3-2.3),respectively.Statistical correlation between size and SUVmax was found in metastatic bone lesions(r=0.233,P<0.05).(2) In 31 patients with histopathologically proven metastasis,TATE-PET detected 23/24 (95.8%) G2,1/2 G1 and 5/5 G3 metastases.G1 metastases were only found in the liver.(3)Among 28 patients underwent both TATE-PET and FDG-PET,there was no significant difference between the detecting rate of metastasis:89.2% (25/28) vs 71.4% (20/28);x2 =2.389,P>0.05.Compared with FDG-PET,TATE-PET was superior in demonstrating metastasis in the liver and bones (70.0% (14/20) vs 65.0% (13/20),3/3 vs 2/3),equal in detecting lung metastasis (both 2/3) but inferior in demonstrating metastasis in lymph nodes (9/10vs 10/10).Conclusions The capability of TATE-PET in revealing NEN metastasis varies depending on lesion localization and histologic grade.TATE-PET and FDG-PET are complementary to each other in detection of NEN metastasis,but without obvious relationship to histologic grade.
		                        		
		                        		
		                        		
		                        	
4.Study on the ELISA Double-antibody Sandwich Method and Pharmacokinetics of Recombinant Human Calcineurin B Subunit in Rat Plasma

		                        			OBJECTIVE:To establish a method for determining recombinant human calmodulin B subunit(rhCNB)in rat plas-ma,and study its pharmacokinetics characteristics. METHODS:ELISA double-antibody sandwich method was adopted. 1 μg/ml rhCNB monoclonal antibody mAb was wrapped,added to the to-be-test sample,rhCNB polyclonal antibody pAb(dilution ratio of 1∶5 000)and HRP-labeled conjugate of anti-IgG(dilution ratio of 1∶10 000)were added. Using tetramethylbenzidine for develop-ing,microplate reader was conducted in wavelength of 450 nm to determine the absorbance value(OD value)and plasma concen-tration of 6 rats after 2,15,30,60,120,240,480,720 min of iv 2.5 mg/kg rhCNB,and the pharmacokinetic parameters were calculated by BAPP 3.0 software. RESULTS:The linear range of rhCNB were 0.195-12.5 ng/ml(r2=0.995 0),lower limit of quan-titation was 0.195 ng/ml,accuracy were 97.300%-103.622%(RSD<7.5%,n=6);RSDs of within-batch,inter-batch,freezing and thawing 3 times were no higher than 8.5%(n=6,18,15). rhCNB pharmacokinetics characteristics in rat fitted to two-com-partment model,AUC0-720 min was 173.038 mg·min/L and t1/2 was 94.62 min. 5.Study on Formulation Optimization of Nicorandil Sustained-release Matrix Tablet and Drug Release in vitro

		                        			OBJECTIVE:To optimize the formulation optimization of Nicorandil sustained-release matrix tablet,and evaluate its drug release properties in vitro. METHODS:Based on single factor test,powder direct compression method was used,using nicorandil cumulative release rate (Q) in 1,4,8,12 h as evaluation indexes,central composite design-response surface method was adopted to optimize the amount of hydroxypropyl methylcellulose(HPMC)and ethyl cellulose(EC);Q values within 12 h in different pH (1.0,5.0,6.8,7.4) media were compared. RESULTS:The optimized formulation (every tablet) was nicorandil 10 mg,HPMC 150 mg,EC 90 mg,microcrystalline cellulose 80 mg,lactose 60 mg,magnesium stearate 2%. Q1 h,Q4 h,Q8 h and Q12 h of the obtained formulation were 23.6%,51.3%,83.7% and 96.9%,respectively;deviation from the predicted values were 2.1%,1.6%,1.0%,0.2%. Q values were similar in pH 1.0-7.4 at different time points. CONCLUSIONS:The obtained Nicor-andil sustained-release matrix tablet by optimal formulation shows sustained-release effect,and the change of pH 1.0-7.4 has no in-terference in the release characteristics of main drug.
		                        		
		                        		
		                        		
		                        	
6.Application of tissue spontaneous fluorescence in draw focal cerebral ischemia reperfusion injury of rats

		                        			Aim To discuss the application of tissue spontane-ous fluorescence for draw focal cerebral ischemia reperfusion in-jury in rats based on specific fluorescence detection technology. Methods The change of spontaneous fluorescence WAS COM-PARED between the brains of normal rats and those of rats with draw focal cerebral ischemia-reperfusion injury and an quantita-tive analysis was then made. Result The results showed that spontaneous fluorescence of brain tissue for focal cerebral ische-mia reperfusion injury changed significantly. Spontaneous fluo-rescence signal of injury considerably enhanced. The fluores-cence signal which was quantified by IVIS had significant statisti-cal significance compared with normal brain tissue, P <0. 05. Conclusion Our research shows that spontaneous fluorescence of brain tissue enhances obviously after focal cerebral ischemia-reperfusion injury. 7.HS-4, a highly potent inhibitor of cell proliferation of human cancer cell

		                        			Objective:To investigate the antitumor activity of the compound HS-4 and the action mechanism.Methods:MTT method was used to testin vitroantitumor activity of the compound HS-4. Orthotopic xenotransplantation tumor model of liver cancer was established in nude mice, and,in vivoantitumor activity of compound HS-4 was tested with a small animal in-vivo imaging system. Sequencing of small RNA library and RNA library was performed in HS-4 treated tumor cell group and control group to investigate the anti-cancer mechanism of HS-4 at level of functional genomics, using high-throughput sequencing technology. Results:HS-4 was found to have relatively highin-vitro antitumor activity against liver cancer cells, gastric cancer cells, renal cancer cells, lung cancer cells, breast cancer cells and colon cancer cells. The IC50 values against SMMC-7721 and Bel-7402 of liver cancer cells were 0.14 and 0.13 nmol/L respectively, while the IC50 values against MGC-803 and SGC-7901 of gastric cancer cells were 0.19 and 0.21 nmol/L, respectively. It was demonstrated that HS- 4 possessed a better therapeutic effect in liver cancer.Conclusions: A new reliable orthotopic xenotransplantation tumor model of liver cancer in nude mice is established. 8.Occupational value investigation and analysis of pediatric nurses in level two hospitals

		                        			Objective To investigate the occupational value status of pediatric nurses in level two hospitals, so as to provide evidence for nursing human resource management. Methods From March to April 2013, convenient sampling method was used to select 150 pediatric nurses in 4 level two hospitals who were investigated by occupational value scale. Results The total score of occupational value among pediatric nurses was (4. 38 ± 0. 46), in which care dimension acquired the highest score (4. 46 ± 0. 46) while behavior dimension obtained the lowest score (4. 21 ± 0. 64). Multiple regression analysis presented that occupational value total score of pediatric nurses was impacted by education background and working years. Conclusions Occupational value score of pediatric nurses in level two hospitals is above average level, so nursing managers can manage nursing human resources scientifically according to the recognition characteristics of occupational value in pediatric nurses.

		                        			OBJECTIVE:To get to know medical staff's recognition of ADR monitoring in our city.METHODS:The concept,reporting consciousness,range and reasons for ADR reporting,reporting processes,professional organization of ADR and law consciousness of ADR etc.were investigated in400medical staff members through questionnaires on“Medical Staff's Recognition Degree to the Monitoring of ADR”.RESULTS:The medical staff in our city were basically clear about the range and the reasons for ADR reporting,but they lacked adequate recognition to the concept and the consciousness of ADR report?ing,they were not clear about the reporting processes and the organization of ADR and they lack the law consciousness of ADR reporting.CONCLUSION:It is necessary to carry out training on professional knowledge,laws and regulations of ADR among the medical staff so as to enhance their recognition to ADR monitoring and to make ADR monitoring as a self-conscious behavior.
