2.Efficency and safety of various combined therapies in treating postoperative refractory recurrent liver cancer

Objective:The study aimed to study the efficacy and safety of combined dual therapy using anti-programmed death (PD)-1 and tyrosine kinase inhibitor (TKI) with combined triple therapy using anti-PD-1, TKI and locoregional intervention triple therapy in patients with postoperative refractory recurrent liver cancer.Methods:Patients with postoperative refractory recurrent liver cancer who had undergone either anti-PD-1 and TKI dual therapy or anti-PD-1, TKI and locoregional intervention triple therapy between July 2016 and March 2019 at the First Medical Center, Chinese PLA General Hospital were retrospectively studied. Tumor responses were assessed by the modified response evaluation criteria in solid tumors and overall survival and progression free survival were compared. Adverse events were evaluated according to the National Cancer Institute Common Terminology Criteria for Adverse Events.Results:Of 63 patients who were included in this study, there were 25 patients in the dual therapy group (16 males and 9 females, aged 54.3±8.8 years) and 38 patients in the triple therapy group (31 males and 7 females, aged 55.5±8.4 years). The 1-year survival rate of the triple therapy group was significantly higher than the dual therapy group (94.5%vs 54.9%) ( P<0.01). The disease control rate was 64.0% (16/25) in the dual therapy group and 84.2% (32/38) in the triple therapy group, and the difference was not significant ( P>0.05). The incidence of treatment-related adverse events in the triple therapy group and the dual therapy group were 78.9% (30/38) and 80% (20/25), respectively. There was no treatment-related death in the 2 groups. Conclusions:Anti-PD-1 and TKI dual therapy and anti-PD-1, TKI and locoregional intervention triple therapy were effective and tolerable treatments for postoperative refractory recurrent liver cancer. The latter treatment had a significantly better clinical benefit on survival outcomes.
3.Effects of Acorustatarinowii Schott on chronic restraint stress mice and its mechanisms

Objective To evaluate the effects of Acorustatarinowii Schott(ATS)on chronic restraint stress mice and its possible mechanisms. Methods Chronic stressed mice underwent a repeated 8h/d constraint in a specific behavior blocker ,however ,only CSA mice received once gavages of 4.5 g/(kg · d)ATS decoction for con-secutive 28 days. Morris water maze(MWM)task was conducted for evaluating learning and memory of the mice. ELISA was used to examine the levels of plasma corticosteroid. Results ATS dramatically ameliorated cognitive impairments and decreased serum corticosteroid level in the stressed mice(P<0.01). Conclusion ATS improves cognitive deficits provoked by chronic stress in mice ,which may attribute to decreasing plasma corticosteroid levels.
4.Self care of patients with lung cancer with peripherally inserted central catheter

Objective To investigate the optimal self care in lung cancer patients with peripherally inserted central catheter (PICC) in ladder-type nursing training,network education,continuing nursing care clinics,and other forms of nursing mode care.Methods Based on the baselien data of patients in the control group and observation group,PICC self management ability,PICC quantitative data,PICC-related infections,thrombosis,and other complications were compared,and the impact of continued nursing platform on lung cancer patients with indwelling PICC was analyzed.Results In the observation group self management ability and health behavior is higher than that of the control group (P<0.001).PICC catheter -related complications no difference with the control group (P>0.05),Patient satisfaction of the observation group than the control group,the difference was statistically significant (92.18% vs.79.00%,P=0.015).Conclusion The self care based on the continued nursing mode established by PICC speciality nurses can improve the self-management level of patients with PICC,reduce PICC-related complications,and increase the patient's satisfaction.
5.The application of PDCA cycle in health education management in emergency department

Objective To explore the application of PDCA cycle in health education in e-mergency department.Methods A total of 50 patients were randomly selected from our hospital from April to June 2013 before applying PDCA cycle management as traditional health education group,another 50 patients were randomly selected from our hospital from July to September 2013 after applying PDCA cycle management as PDCA health education group,health education aware-ness and patient satisfaction rate of the two groups were compared.Results Health education awareness and patient satisfaction rate in PDCA health education group were better than traditional health education group,and the difference was statistically significant(P <0.05).Conclusion PDCA cycle management method can meet the needs of emergency patients’health education,and effectively improve patients’awareness of emergency health education and satisfaction rate.

OBJECTIVETo examine apoptosis of SMMC-7721 hepatocarcinoma cells induced by total flavonoids of Oxytropis falcata (TFOF) and its preliminary mechanism.
METHODSMMC-7721 cells were treated for 24 h with TFOF in different concentrations. Inhibition on proliferation of SMMC-7721 cells was assessed by MTT assay. The morphology of treated SMMC-7721 cells was observed by optical microscope. Effect of TFOF on the nuclear morphology of cells was analyzed using Hoechst 33258 staining by fluorescence microscope. Annexin V-FITC/PI staining and flow cytometric measurement were used for investigating the effect of TFOF on induction of apoptosis in SMMC-7721 cells and cell cycle analysis.
RESULTThe results of MTT assay showed that TFOF could induce cytotoxicity in SMMC-7721 cells in a dose-dependent manner. Hoechst 33258 staining analysis indicated that TFOF caused typical characteristics of apoptotic programmed cell death, such as cell shrinkage, apoptotic body formation etc. Flow cytometric analysis demonstrated that TFOF caused a dose-dependent apoptosis of SMMC-7721 cells and arrested cell cycle in G1 phase.
CONCLUSIONIt suggested that TFOF inhibit proliferation of SMMC-7721 cells by inducing apoptosis of the cells and arresting cell cycle in G1 phase.

9.Influences of Oxytropis falcata on proliferation of SMMC-7721 hepatocarcinoma cells and expression of MMP-2.

OBJECTIVETo investigate the influences of extracts of Oxytropis falcata on proliferation of SMMC-7721 cells and expression of MMP-2 (matrix metalloproteinase-2).
METHODSMMC-7721 cells were treated for 24 h with five fractions obtained from 0. falcata in different concentrations. Inhibition on proliferation of the SMMC-7721 cells was assessed by MT method. Secretion of MMP-2 was measured by ELISA in the supernatant of SMMC-7721 cells treated with fractions of essential oil and total flavonoids for 24 h. Transcription of mRNA of MMP-2 was detected by Real-time PCR.
RESULTIn MT assay, essential oil and total flavonoids showed potential antiproliferative activity on SMMC-7721 in a concentration dependent manner. Data of ELISA showed that fraction of essential oil suppressed secretion of MMP-2 significantly. Results of Real-time PCR indicated that both essential oil and total flavonoids restrained expression of mRNA of MMP-2.
CONCLUSIONIt suggested that essential oil and total flavonoids of O. falcata inhibit proliferation of SMMC-7721 cells through down-regulating secretion and expression of MMP-2 in cells. However, further experiments are necessary to carry out to investigate the potential mechanism.

10.Determination of flavones in different origin and parts of Bupleurum smithii var. parvifoliaum by UPLC-PDA.

An ultra performance liquid chromatography method has been developed for determination of flavones in different origin and different parts from Bupleurum smithii var. parvifoliaum. The separation was performed at 30 degrees C on an Acquity UPLC BEH C18 column eluted with methanol and 0.1% phosphoric acid solution as mobile phases in gradient elution. The detection wavelength was set at 256 nm and the flow rate was 0.4 mL x min(-1). There flavones of rutin, quercetin and isorhamnetin were separated well, the linear calibration curves were obtained over the ranges of 0.106-1.06 microg for rutin (r = 0.999 8, n = 6), 0.004 87-0.048 7 microg for quercetin (r = 0. 999 7, n = 6), 0.022 0-0.220 microg for isorhamnetin (r = 0.999 8, n = 6). The mean recoveries of the three compounds were 99.3%, 97.8%, 98.9% with RSD of 2.4%, 3.0%, 3.2%. The result showed that the method is convenient and feasible for determination of the flavone content in B. smithii var. parvifoliaum.
