Spinal cord injury is a traumatic disease, commonly seen in falling injuries, traffic accidents, heavy injuries, etc, which could cause motor, sensory and autonomic dysfunction below the level of spinal cord injury.Myelin-associated inhibitors play a role in promoting the collapse of growth cones and inhibiting axonal regeneration in the injured spinal cord microenvironment, which is the main reason for the difficult repair of spinal cord injury.Myelin-associated inhibitors(MAIs),such as neurite outgrowth inhibitor(Nogo),oligodendrocyte-myelin glycoprotein(OMgp)and myelin-associated glycoprotein(MAG),along with their receptor proteins, such as Nogo-A/Nogo-66 receptor 1(NgR1),paired immunoglobulin-like receptor B(PirB),sphingosine-1-phosphate receptor 2(S1PR2),are the important regulatory factors in the spinal cord microenvironment. They can inhibit therepair process of spinal cord injury by affecting the signaling pathway of neuron axon growth.Although the mechanism of spinal cord injury repair is still unclear, the regulation of myelin-related inhibitory factor proteins and downstream signaling pathways remain an important therapeutic approach for spinal cord injury. In this paper, the role of MAI proteins and their receptors in spinal cord injury repair in recent years were reviewed to provide a new target for spinal cord injury repair and provide more ideas for clinical treatment after spinal cord injury.

Objective To establish an acute exposure model of extreme plateau hypobaric hypoxia environment and explore transcriptomic changes related to learning and memory impairment in rats. Methods Healthy male SD rats aged 6-weeks,200-250 g,were selected and divided into control group and plateau group. The control group was treated with normal pressure and oxygen(19 rats),and the plateau group was placed in a hypobaric hypoxia chamber(19 rats)at a simulated altitude of 8000 meters and treated for 72 hours.Behavioral changes were detected with 16 animals from each group using contextual fear conditioning and Morris water maze(8 rats each).Three hippocampal tissues were extracted from each group for transcriptomic sequencing, and the molecular mechanism of learning and memory impairment induced by extreme plateau environment was analyzed by Gene Ontology(GO),Kyoto Encyclopedia of Genes and Genomes(KEGG)and gene set enrichment analysis(GSEA)enrichment. Results The behavioral result showed that compared with the control group, the fear memory and spatial learning memory abilities of rats in plateau group were decreased.GO and KEGG analyses showed that the extreme altitude environment reshaped the hippocampal microenvironment and affected the intercellular signal transmission, while GSEA analysis showed that the extreme altitude environment up-regulated the gene set related to the plasma membrane and extracellular matrix. Conclusion The extreme plateau environment at an altitude of 8000 meters could affect the microenvironment of rat hippocampus, destroy intercellular connections and impair intercellular communication and then induce learning and memory impairment.

Aim To investigate the protective effect of berberine(BBR)on mice with unilateral ureteral obstr-uction(UUO)and explore its mechanism. Methods C57BL/6 mice were randomly divided into the sham group,UUO group, and BBR treatment groups(50,100 and 200 mg·kg-1),with eight mice in each group.Except the sham group, the other groups were subjected to left ureteral ligation to establish the UUO model.Af-ter modeling, the mice in the sham and UUO groups were fed normal saline, and the mice in the BBR treat-ment groups were fed(50,100,200)mg·kg-1 BBR by gavage for 14 days, respectively.Biochemical analy-zer was employed to detect the levels of serum creati-nine(Scr)and blood urea nitrogen(BUN).HE,Mas-son,TUNEL and immunohistochemical staining were used to observe the pathological changes of renal tis-sue.ELISA was employed to detect the expression of pro-inflammatory cytokines in renal tissue homogenate.Western blot was used to detect the protein levels of NF-κB p65,TGF-β1 and CTGF in mouse kidney.Re-sults Compared with the UUO group, the levels of Scr and BUN in the BBR group were significantly reduced.Renal injury and interstitial fibrosis were alleviated. The expression of pro-inflammatory cytokines decreased in kidney. The expression of NF-κB p65,TGF-β1 and CTGF decreased. All results showed some degree of dose dependence. Conclusion Berberine has a sig-nificant protective effect on unilateral ureteral obstruc-tion mice, and the mechanism may be that BBR has the potential to inhibit NF-κB p65/TGF-β1/CTGF signa-ling pathway, thus reducing renal inflammation and fi-brosis.

To investigate the intervention effect and mechanism of Zhenwu Decoction on diabetic nephropathy(DN) mice of spleen-kidney Yang deficiency syndrome based on the Rho-associated coiled-coil kinase(ROCK)/IκB kinase(IKK)/nuclear factor-κB(NF-κB) pathway. Ninety-five 7-week-old db/db male mice and 25 7-week-old db/m male mice were fed adaptively for one week. The DN model of spleen-kidney Yang deficiency syndrome was induced by Dahuang Decoction combined with hydrocortisone by gavage, and then the model was evaluated. After modeling, they were randomly divided into a model group, high-dose, medium-dose, and low-dose Zhenwu Decoction groups(33.8, 16.9, and 8.45 g·kg~(-1)·d~(-1)), and an irbesartan group(25 mg·kg~(-1)·d~(-1)), with at least 15 animals in each group. The intervention lasted for eight weeks. After the intervention, body weight and food intake were measured. Serum crea-tinine(Scr), blood urea nitrogen(BUN), fasting blood glucose(FBG), urinary albumin(uALb), and urine creatinine(Ucr) were determined. The uALb/Ucr ratio(ACR) and 24 h urinary protein(UTP) were calculated. Renal pathological morphology was evaluated by HE staining and Masson staining. The levels of key molecular proteins in the ROCK/IKK/NF-κB pathway were detected by Western blot. Enzyme-linked immunosorbent assay(ELISA) was used to detect interleukin-1β(IL-1β), interleukin-6(IL-6), interleukin-8(IL-8), interleukin-10(IL-10), and tumor necrosis factor-α(TNF-α). Compared with the blank group, the model group showed increased content of BUN, uALb, and SCr, increased values of 24 h UTP and ACR, decreased content of Ucr(P<0.05), enlarged glomeruli, thickened basement membrane, mesangial matrix proliferation, inflammatory cell infiltration, and collagen fiber deposition. The protein expression of ROCK1, ROCK2, IKK, NF-κB, phosphorylated IKK(p-IKK), phosphorylated NF-κB(p-NF-κB), and phosphorylated inhibitor of NF-κB(p-IκB) increased(P<0.05), while the protein expression of inhibitor of NF-κB(IκB) decreased(P<0.05). The levels of inflammatory factors IL-1β, IL-6, IL-8, and TNF-α increased(P<0.05), while the level of IL-10 decreased(P<0.05). Compared with the model group, the groups with drug treatment showed decreased levels of BUN, uALb, SCr, 24 h UTP, and ACR, increased level of Ucr(P<0.05), and improved renal pathological status to varying degrees. The high-and medium-dose Zhenwu Decoction groups and the irbesartan group showed reduced protein expression of ROCK1, ROCK2, IKK, NF-κB, p-IKK, p-NF-κB, and p-IκB in the kidneys(P<0.05), increased protein expression of IκB(P<0.05), decreased levels of serum inflammatory factors IL-1β, IL-6, IL-8, and TNF-α(P<0.05), and increased level of IL-10(P<0.05). Zhenwu Decoction can significantly improve renal function and renal pathological damage in DN mice of spleen-kidney Yang deficiency syndrome, and its specific mechanism may be related to the inhibition of inflammatory response by down-regulating the expression of key molecules in the ROCK/IKK/NF-κB pathway in the kidney.

OBJECTIVE:
To analyze the factors related to pregnancy of endometriosis and whether Chinese herbal medicines (CHMs) can improve pregnancy outcomes in patients with endometriosis in long-term management.
METHODS:
This multicenter cohort study retrospectively analyzed the clinical data of endometriosis patients with fertility needs from January 2019 to November 2019. A total of 252 patients with endometriosis from 5 level-III Grade A hospitals in Beijing were included in this study. Univariate and multivariate logistic regression analysis were performed for the relevant factors. The propensity score matching (PSM) function of SPSS software was used to match the CHMs group with the non-CHMs group. The pregnancy rate and live birth rate were analyzed.
RESULTS:
The results of univariate analysis showed that age, disease course, presence of infertility, presence of adenomyosis, time after surgery or use of gonadotropin-releasing hormone agonist (GnRH-a), use of CHMs and follow-up time were influencing factors of pregnancy in endometriosis patients (P<0.05). The results of multivariate analysis showed that age, presence of adenomyosis, time after surgery or use of GnRH-a, use of CHMs and follow-up time were independent factors affecting pregnancy in endometriosis patients, among which, age ⩾35 years old, presence of adenomyosis and follow-up time >6 months were independent risk factors (OR=0.445, 0.348, 0.140, respectively, P<0.05), time after surgery or use of GnRH-a ⩽6 months and use of CHMs were independent protective factors (OR=3.839, 3.842, respectively, P<0.05). After PSM, 99 pairs of two groups were matched successfully. The pregnancy rate of the CHMs group was higher than that of the non-CHMs group [55.56% (55/99) vs. 36.36% (36/99), P<0.05]. The live birth rate of the CHMs group was higher than that of the non-CHMs group [49.49% (49/99) vs. 35.35% (35/99), P<0.05].
CONCLUSION
CHMs can effectively improve clinical pregnancy rate and live birth rate of patients with endometriosis in the chronic disease management.

Chinese Journal of Biochemistry and Molecular Biology 2023;39(11):1650-1658
Biochemistry is one of the basic disciplines of life sciences‚ and plays an important role in the undergraduate teaching of biology. However‚ the biochemistry course system is huge‚ and the knowledge is numerous. How to improve students' interest in learning‚ improve teaching effect‚ and promote course construction are important questions faced by many biochemistry teachers. Here the authors put forward the " three-dimensional teaching concept based on FACT" ‚ and take the biochemistry course construction of the School of Life Sciences‚ Sun Yat-sen University‚ as an example to analyze the connotation and practice of this teaching concept. " Three-dimension" is inspired by the " seemingly identical but actually different" concept of biological chiral molecules‚ which not only reflects the need for universities to carry out course construction based on their own actual situation and seek truth from facts with distinct characteristics‚ but also emphasizes the construction of a multi-dimensional course construction system. The authors will introduce the concept of " three-dimension" in “form” ‚ “angle” ‚ “content” and “team” in an all-round way. Moreover‚ the authors also conduct in-depth analysis from aspects such as teaching methods‚ second classroom‚ knowledge point logic‚ ideological and political education for courses‚ and teaching team construction‚ which has certain reference significance for peer teachers to carry out biochemistry course construction.
