1.Relationship between lifestyle habits and obesity based on latent category analysis among 12-18 year adolescents

		                        			Objective:
		                        			To analyze the latent category characteristics of lifestyle habits of adolescents aged 12-18 years, and to explore its relationship with obesity.
		                        		
		                        			Methods:
		                        			Data of 3 252 adolescents aged 12-18 years in the established China Nutrition and Health Survey were used for this study. The contents of the survey include basic situation, living habits, and body mass index. Latent category analysis was performed on 11 lifestyle habits on eating and exercise.  Chi square test was used to explore the relationship between the potential categories of lifestyle habits and obesity.
		                        		
		                        			Results:
		                        			All the participants were classified into 3 categories:mixed ( n =1 092, 35.58%), passive ( n =765, 23.52%), and healthy ( n =1 395, 42.90%) groups. Body mass index showed significant differences across three categories ( χ 2=19.66,  P <0.01). In the BMI classification of adolescents, the healthy type has the highest proportion in the light type, overweight type and obesity type(41.61%,47.87%,50.00%).
		                        		
		                        			Conclusion
		                        			The lifestyle habits of adolescents aged 12-18 years show obvious classification characteristics, which have different correlations with obesity. 2.Epidemiological research progress on the relationship between children's dietary patterns and health.

		                        			As food preferences and eating habits form early in life, the development of healthy eating habits in early childhood is a way to prevent diet-related diseases. The dietary pattern approach examines the effect of an overall diet on health outcomes, instead of individual foods or nutrients, thereby presenting a comprehensive evaluation of children's dietary intake. This article reviews the current literature to summarize the main methods for assessing dietary patterns and explore relationships between children's dietary patterns and obesity, puberty onset, cardiovascular diseases, and neurodevelopment. 3.The role of Circular RNA circ_0000253 in diagnosis and prognosis of osteosarcoma

		                        			Objective:To evaluate the value of circ_0000253 as a biomarker for (osteosarcoma, OS) diagnosis and prognosis.Methods:GEO database was used to screen the differentially expressed circRNAs in OS, microRNAs (miRNAs) with binding sites of circRNA were predicted to further construct the ceRNA network, and GO and KEGG analysis were used to screen the target genes. qRT-PCR was used to detect the expression of circ_0000253, miR-578 and TGF β2 in cancer and paracancerous tissue in OS patients, as well as OS cells and normal osteoblasts. The diagnostic value of circ_0000253 for OS and its impact on prognosis were analyzed, and the relationship between circ_0000253 expression level and clinicopathological parameters of patients was discussed. Further the expression of circ_0000253 and miR-578 in cells was intervened, apoptosis was detected by flow cytometry and cell proliferation was detected by MTT assay. Results:The expression of circ_0000253 in OS tissue was higher than that in paracancerous tissue ( t=11.17, P<0.001) . The expression of circ_0000253 was significantly correlated with Enneking stage, histopathology grade and distant metastasis (all P<0.05) . Circ_0000253 can be used as an effective diagnostic index of OS (AUC=0.84, P<0.001) and the prognosis of patients with high expression of circ_0000253 is poor. Circ_0000253 may regulate OS through miR-578/TGF β2 network. Knockdown of circ_0000253 reduced cell proliferation and induced apoptosis, which could be partially saved by miR-578 inhibitor (all P<0 05) . Conclusion:circ_0000253 may serve as a new biomarker for OS diagnosis and prognosis, which has a positive meaning for targeted therapy of OS.
		                        		
		                        		
		                        		
		                        	
4.Establishment of dietary quality assessment system for children is necessary for growth and development research

To examine the diet-growth relationship, it is necessary to carry out a comprehensive and objective evaluation of diet quality in children and to develop effective nutrition intervention programs, so as to improve children s health. 5.Establishment and Assessment of Mice Models of Type 2 Diabetes Mellitus.

		                        			Objective To establish type 2 diabetes mellitus(T2DM)KM mouse models via the combined use of high-calorie diet and multiple administration of low-dose streptozotocin(STZ). Methods Based on the randomized number table,30 KM mice were equally and randomly divided into 2 groups:modeling group and control group. Mice in the modeling group were given foods with high calories for one month and injected with 30 mg/kg STZ via the left lower abdominal cavity for 2-4 consecutive days,while mice in the control group were fed with standard maintenance foods and the same dose of citrate buffer solution. The general conditions including food and water intake and mice weight were recorded. Blood glucose level was measured 1,2,4,5,12,and 21 weeks after STZ injection. When the glucose level became stabilized,the serum insulin and blood lipids [including total cholesterol(TC),triacylglycerol(TG),high-density lipoprotein(HDL) and low-density lipoprotein(LDL)],and hemoglobin a1c (HbA1c)were measured,and oral glucose tolerance test were performed. Results The modeling group had a 100% survival rate. After STZ injection,the body weight of mice in the modeling group reached the peak in the forth week,and later the growth rate decreased,still significantly lower than that of control group mice till the 21week(t=3.160,P=0.006). Their blood glucose level was significantly higher than that of mice before STZ injection and in the control group(all P<0.05);as time went on,it was also rising,and it remained high till the 21week [(26.38±1.34)mmol/L]. In the 4week,the fasting blood glucose of mice in the modeling group was(11.86±3.33)mmol/L,which was significantly higher than that of mice in the control group [(6.37±1.27)mmol/L](t=-3.830,P=0.002). Fasting serum insulin of mice in the modeling group showed no significant difference compared with control group [(5.73±0.24)mU/L vs.(5.48±0.32)mU/L;t=-0.863,P=0.416]. Insulin sensitivity index was 0.0145±0.0039,which was significantly lower than that(0.0267±0.0039)in control group(t=4.414,P=0.003). In the 6week,the blood glucose levels of mice in the modeling group were(15.35±1.82),(26.45±1.07),(25.58±1.46),and(26.15±1.00)mmol/L 0,30,60,and 120 min after oral gavage of D-glucose,which were all significantly higher than those in the control group [(6.88±1.75)(t=-8.203,P=0.000),(17.65±2.94)(t=-6.884,P=0.000),(13.18±2.04)(t=-12.110,P=0.000),and(7.37±3.40)mmol/L(t=-12.969,P=0.000)]. In the 8week,serum TC and TG levels of mice in the modeling group were(3.83±0.06)and(2.20±0.20)mmol/L,which were significantly higher than those in the control group [(3.10±0.10)(t=11.000,P=0.000)and(0.90±0.10)mmol/L(t=10.070,P=0.000)]. HDL level of mice in the modeling group was(2.03±0.06)mmol/L,which was significantly lower than that in the control group [(2.48±0.02)mmol/L;t=11.662,P=0.000]. LDL level was increased but showed no significant difference [(0.34±0.08)mmol/L vs.(0.26±0.02)mmol/L](t=1.680,P=0.168). HbA1c content of mice in the modeling group was(7.30±0.31)%,which was significantly higher than that(4.40±0.32)% in the control group(t=-11.587,P=0.000). 6.Features of human cytomegalovirus UL146 gene in asymptomatic children

		                        			Objective To investigate the polymorphisms of human cytomegalovirus ( HCMV ) UL146 gene in asymptomatic children. Methods Urine samples were collected from 47 asymptomatic chil-dren who were positive for HCMV DNA. PCR was performed to amplify the open reading frame ( ORF) of UL146 gene. Positive bands were sequenced and variations in UL146 gene were analyzed by using bioinfor-matics software. Results Seventeen samples were successfully amplified and sequenced. Variations spread all over the sequence of UL146 gene and the variability in nucleotide and amino acid sequences ranged from 0% to 42. 5% and 0% to 67. 7% respectively. Compared with the Towne strain, there was diversity in sig-nal sequence and C-terminal region. Phylogenetic analysis indicated that UL146 in the 17 asymptomatic chil-dren belonged to four genotypes, which were G1, G8, G9 and G11. Forms of post-translational modification varied greatly among the four genotypes, while the important functional region of ELRCXC chemokine was highly conservative. Secondary structure prediction showed that random-coli conformation was the predomi-nant structure of active proteins. Isoelectric point ( PI) and molecular weight ( MW) were dissimilar among the four genotypes. Conclusion HCMV UL146 gene in asymptomatic children was hypervariable in both nucleotide sequence and amino acid structure. However, the important functional region was highly con-served. The predominant genotypes of UL146 in these children were G1, G8, G9 and G11, and the geno-type distribution in them showed no significant difference with previous findings in children with symptomatic HCMV infection.
		                        		
		                        		
		                        		
		                        	
7.Detection of the plasma miR-92a expression in systemic lupus erythematosus and its clinic significance

		                        			Objective To analyze miR-92a expression and its clinical significance in the plasma of systemic lupus erythematosus (SLE) patients.Methods Plasma samples from 44 SLE patients,16 rheumatoid arthritis (RA) patients and 20 healthy controls were collected.The small RNAs in these plasma samples were isolated and reversely transcribed.Using cel-miR-39 as the external reference,the levels of miR-92a expression were detected by real-time polymerase chain reaction (PCR) method.MiR-92a and cel-miR-39 were analyzed by real-time fluorescence quantitative PCR and agarose gel electrophoresis.The sensitivity and specificity of miR-92a as SLE were analyzed by receiver-operating characteristic (ROC) curve.The correlation between the levels of miR-92a expression and the clinic pathological features of SLE and biological significance of miR-92a expression in SLE were further analyzed by Pearson or Chi-square test.Results Our data indicated that the plasma levels of miR-92a expression was 49.20 (5.33,95.17) in SLE patients,411.30 (320.84,504.69) in healthy controls,and 25.59(11.20,30.54) in RA patients.The difference was significant (x2=40.77,P<0.01).The area under the ROC curve (AUC) was 0.958 for discriminating between SLE patients and normal subjects and 1.00 for discriminating between RA patients and healthy controls.The levels of miR92a expression cutoff values were set the as 198.59 for healthy control and 85.35 for RA patients,the diagnostic sensitivity and specificity were 93.2%,90%,and 100%,100%,res-pectively.The analysis of the correlation between miR-92a expression and the clinic pathological features of SLE had shown that the levels of plasma miR-92a expressions were much lower in SLE patients with down-regulated complement C3,and up-regulated urea nitrogen,creatinine,LDH,ATH (all P<0 05).Conclusion Down-regulated miR-92a expression in plasma of SLE may be involved in the SLE disease occurrence or development and could be used as a novel potential diagnostic biomarker for SLE.
		                        		
		                        		
		                        		
		                        	
8.Constructing a recombinant adenovirus carrying human cytomegalovirus UL138 gene and evaluating the effects of UL138 gene on the functions of monocytes

		                        			Objective To construct a recombinant adenovirus carrying UL138 gene, which was re-lated to the latent infection of human cytomegalovirus, and to investigate the effects of UL138 gene on the functions of THP-1 mononuclear cells. Methods The recombinant adenovirus expressing the UL138 gene was packaged. The titer of the recombinant adenovirus was determined by calculating 50% tissue culture in-fective dose ( TCID50 ) . THP-1 mononuclear cells were infected with the recombinant adenovirus at different multiplicity of infection (MOI) and the optimal MOI was determined (100 PFU/cell) by observing the ex-pression of green fluorescent protein ( GFP ) . Changes in the expression of proinflammatory cytokines by THP-1 mononuclear cells that was induced by overexpressed UL138 were analyzed by quantitative PCR. The expression of chemokines and their receptors were measured by quantitative PCR array. Results The re-combinant adenovirus carrying the UL138 gene was successfully constructed with a titer of 1×1011 PFU/ml. The rate of THP-1 mononuclear cells that was infected with the recombinant adenovirus was 60% at the MOI of 1 ∶ 100. Results of the RT-PCR analysis and Western blot assay further confirmed that the recombinant adenovirus could infect THP-1 mononuclear cells successfully and the expression of UL138 protein increased gradually over time. The overexpressed UL138 in THP-1 mononuclear cells significantly inhibited the expres-sion of IL-18, IL-1β, IL-6, IL-8 and TNF-α as indicated by the results of quantitative PCR. Results ob-tained from the quantitative PCR array analysis showed that most of the chemokines and their receptors were downregulated in the transfected THP-1 mononuclear cells except for the chemokines of CCL17, CCL21, CCL2 and XCL2 and the receptors of CCR2, CXCR1, CXCR2, CXCR4 and CX3CR1 which were upregulat-ed. Conclusion We successfully constructed the recombinant adenovirus carrying UL138 gene which could be used to infect THP-1 mononuclear cells. 9.Berberine hydrochloride combined with sodium ozagrel in the treatment of acute cerebral infarction and its effect on glucose and lipid metabolism

		                        			Objective To investigate the effects of sodium ozagrel and berberine hydrochloride in the treatment of acute cerebral infarction and its effect on glucose and lipid metabolism.Methods According to the digital table,158 patients with acute cerebral infarction were randomly divided into the two groups:the control group was treated with the sodium ozagrel group and the observation group was treated combined with sodium ozagrel and berberine hydrochloride.After 2 weeks,Chinese stroke clinical neural function defect score scale and the National Institutes of Health Stroke Scale (NIHSS) was used to evaluate neurological function.The therapeutic effectiveness was assessed by ADL after treatment 1,3,6 months.The changes of glucose and lipid levels were detected by turbidimetric method.Results The total effective ratein the observation group was obviously higher than that of the control group (87.01% vs 69.14%) (x2 =7.316,P <0.01).Of the above two ways of nerve function score indicateed that the observation group was significantly better than that of the control group (t =3.207,1.274,all P < 0.01).ADL score of all patients was significantly better than that of before treatment with drug in the two groups after 1 month,3 months and 6 months (t =7.266 and t =8.850,t =1.429 and t =2.816,t =9.218 and t =6.739,P <0.01).Compared with the control group,ADL score of the observation group was significantly improved after 3 months and 6 months (t =1.163,0.932,all P <0.01).The significant difference was found between the control group and the observation group after 3 months and 6 months (t =7.861,5.005,all P > 0.05).After the treatment,the fasting blood glucose level was reduced significantly in the observation group (t =0.859,P < 0.01),total cholesteroland high-density lipoprotein,triglyceride,low density lipoprotein level were significantly decreased (t =0.257,0.114,0.378,all P < 0.01),but there was no significant difference in HDL-C level (t =1.960,P > 0.05).Conclusion The therapeutic effect of berberine hydrochloride combined with sodium ozagrel is better in the treatment of acute cerebral infarction,and the level of glucose and lipid metabolism was significantly decreased.
		                        		
		                        		
		                        		
		                        	
10.Effects ofYixintai Granules on Expression of C-Myc mRNA and Its Protein in Myocardial Tissues of Rabbits with Chronic Heart Failure

		                        			ObjectiveTo explore the effects ofYixintaiGranules on expression of C-Myc mRNA and its protein in myocardial tissues of rabbits with chronic heart failure (CHF).Methods CHF rabbit models were established by ear marginal vein injection of adriamycin. Successfully modeled rabbits were divided into the model group, the Losartan Potassium group, the high-, medium-, and low doseYixintaiGranules groups. Besides, a normal control group was set up. Administration groups were given relevant medicine for gavage, equal volume of physiological saline was administered to rabbits of the model group and the normal control group by gastrogavage. The intervention lasted for 4 weeks, once per day. Echocardiographic indexes and mRNA and protein expression levels of C-Myc in myocardial tissue were detected after 4 weeks of medication.Results Compared with the normal group, the LVEF, LVFS, and E/A of the model group decreased significantly (P<0.01), but mRNA and protein expression levels of C-Myc in myocardial tissues increased significantly (P<0.01). Compared with the model group, the LVEF, LVFS, and E/A of YG groups and Losartan Potassium group increased significantly (P<0.01), but mRNA and protein expression levels of C-Myc in myocardial tissues decreased significantly (P<0.05,P<0.01).ConclusionYixintaiGranules can effectively inhibit the expression of mRNA and protein expression of C-Myc, and improve cardiac function.
