1.Application Progress of RNA Fluorescence Aptamers in Biosensing and Imaging

RNA fluorescence aptamers are RNA sequences that can specifically bind to non-toxic,cell permeable,and self-fluorescent target molecules and activate their luminescent properties.These aptamers provide powerful tools for biosensing and imaging researches due to their simple structure,easy synthesis,and easy transfection.This article summarized the characteristics and development history of various RNA fluorescent aptamers,including Malachite Green,Spinach,Broccoli,Mango,Corn,and Pepper family,as well as their corresponding fluorescent groups.The applications of RNA fluorescent aptamers were also reviewed from two aspects:extracellular detection and cell imaging.This review might provide guidance for labeling,detection and interactions of molecules from proof of concept and clinical assessment to practical clinical and biomedical applications.
2.Polysaccharide of Atractylodis Macrocephalae Rhizoma inhibits expression of immune checkpoint PD-L1 by targeting miR-34a in esophageal carcinoma cells.

The immune checkpoint programmed cell death-ligand 1(PD-L1)-mediated immunosuppression is among the important features of tumor. PD-L1, an immunosuppressant, can induce T cell failure by binding to programmed cell death-1(PD-1). Thus, the key to restoring the function of T cells is inhibiting the expression of PD-L1. The Chinese medicinal Atractylodis Macrocephalae Rhizoma(AMR) has the anti-tumor, anti-inflammatory, antioxidant, and hypoglycemic activities, and the polysaccharide in AMR(PAMR) plays a crucial role in immunoregulation, but the influence on the immune checkpoints which are closely related to immunosuppression has not been reported. MicroRNA-34 a(miR-34 a) expression in esophageal carcinoma tissue is significantly lower than that in normal tissue. This study aims to investigate the inhibitory effect of PAMR on esophageal carcinoma cells, and the relationship between its inhibitory effect on PD-L1 expression and miR-34 a, which is expected to clarify the anti-tumor mechanism of PAMR. Firstly, different human esophageal carcinoma cell lines(EC9706, EC-1, TE-1, EC109 cells) were screend out, and expression of PD-L1 was determined. Then, EC109 cells, with high expression of PD-L1, were selected for further experiment. The result showed that PAMR suppressed EC109 cell growth. According to the real-time quantitative PCR(qPCR) and Western blot, it significantly suppressed the mRNA and protein expression of PD-L1, while promoting the expression of tumor suppressor miR-34 a. The confocal microscopy and luci-ferase assay proved that PAMR alleviated the inhibitory effect of PD-L1 while blocked miR-34 a. Additionally, the expression of PD-L1 was controlled by miR-34 a, and the combination of miR-34 a inhibitor with high-dose PAMR reversed the inhibitory effect of PAMR on PD-L1 protein expression. Thus, the PAMR may inhibit PD-L1 by increasing the expression of miR-34 a and regulating its downstream target genes. In conclusion, PAMR inhibits the expression of PD-L1 mainly by inducing miR-34 a.

6.Analysis of Traditional Chinese Medicine Syndromes of 46 Cases of COVID-19

Objective:To analyze the traditional Chinese medicine(TCM) syndromes of 46 patients with coronavirus disease-2019 (COVID-19) by cluster analysis method. Method:The 46 COVID-19 patients with TCM symptoms, signs, tongue and pulse were analyzed by frequency analysis and clustering analysis, and the TCM syndrome types of COVID-19 patients were classified and summarized by combining clinical expertise, and the evolution law was analyzed by combining the median days of the syndrome collection. Result:The major signs and symptoms of the 46 patients with COVID-19: fever (frequency of 39, accounted for 84.8%), poor appetite (frequency of 44, 95.7%), fatigue (frequency of 42, 91.3%), wheezing breath (frequency of 35, 76.1%), dry mouth (frequency of 21, 45.7%), lung condition short (frequency of 20, 43.5%), defecate pond (frequency of 15, 32.6%), red tongue (frequency 30, accounting for 65.2%), yellowish moss (frequency 27, accounting for 58.7%), and slimy pulse (frequency 30, accounting for 62.5%). TCM syndromes were divided into seven types, i.e. dampness and heat syndrome (13 cases), dampness and toxin syndrome (10 cases), heat stasis syndrome (7 cases), dampness and heat syndrome (5 cases), cold dampness internal invasion syndrome (4 cases), dampness poison blood stasis syndrome (2 cases).According to the time of onset, there were cold and dampness syndrome (median days 6.5 days), dampness was heavier than heat syndrome (10 days), dampness and heat syndrome (10 days), dampness and heat syndrome (11.5 days), heat stagnation of liver and gallbladder syndrome (13 days), dampness and blood stasis syndrome (15 days), and Yin injury syndrome (16 days). Conclusion:Among the 46 cases of COVID -19, damp-heat syndrome was the most common, followed by heat stagnation of liver and gallbladder syndrome, Yin injury and gas consumption syndrome, cold and damp internal invasion syndrome, and dampness and blood stasis syndrome. The pathogenesis of the disease evolved into heat and poison, and later appeared to consume Qi Yin performance.
7.Calculation on Medical Insurance Fund Based on the Integration of Basic Medical Insurance for Urban and Rural Residents in the Inner Mongolia Autonomous Region

Objective:To calculate the medical compensation fee,management fee and risk reserve in the medical insurance fund of Inner Mongolia,calculate the total financing amount of the medical insurance fund under the different compensation scheme in 2011-2015 and the amount of fund-raising fund of the per capita health insurance fund,and study the financing feasibility of urban and rural areas in Inner Mongolia in the integration process of Basic Medical Insurance for urban and rural residents.Methods:The medical insurance fund under different compensation schemes in Inner Mongolia was calculated by using the residual qualified model,the correlation degree qualified model,the variance ratio qualified model and the small error probability qualified model.Results:The average health insurance fund under the five compensation schemes in 2011-2015 was between 521.43 yuan and 2012.27 yuan.The five compensation schemes were to be raised between 128.85 yuan and 49.73 billion yuan respectively.Based on different kinds of compensation program,the financing amount of basic medical insurance fund for urban and rural residents in Inner Mongolia increased by years.The average annual growth rate was 20%,the average annual growth rate of per capita health insurance fund financing was 10%.Conclusion:The amount of fund raising increased quickly by years with high financing pressure.This was a difficult task for the advancement of the integration of basic medical insurance for urban and rural residents in Inner Mongolia.Meanwhile,it provided a scientific and effective financing plan of insurance fund for promoting the integration of basic medical insurance in Inner Mongolia.
8.Effect of FSD-C10 on modulation of inflammatory microenvironment in an Alzheimer disease double transgenic mouse model

AIM:To explore the therapeutic effect of a novel Rho kinase inhibitor FSD-C10 onβ-amyloid pro-tein precursor (APP)/presenilin-1 (PS1) double transgenic mice.METHODS: The transgenic mice overexpressing hu-man APP with the Swedish mutation (695) and human PS1 with ΔE9 mutation at the age of 8 months were used in this study.The mice were randomly divided into model group and FSD-C10 intervention group, and wild-type mice at the same age served as normal controls .The mice in FSD-C10 intervention group were treated with FSD-C10 (25 mg· kg-1 · d-1 ) for 2 months by intraperitoneal injection .The mice in model group and the wild-type mice were injected with saline in the similar manner.Morris water maze (MWM) test was applied to examine the capacity of learning and memory .The Aβ1-42 deposition, Tau protein phosphorylation , and the expression of β-site APP-cleaving enzyme ( BACE) as well as inflammato-ry molecules, such as TLR-4 and NF-Κb, and M1/M2 microglial markers, such as Inos and Arg-1, were determined by the methods of immunohistochemistry and Western blot .RESULTS: Compared with model group , FSD-C10 significantly improved the learning and memory abilities of APP/PS1 double transgenic mice , accompanied by reduced Aβ1-42 deposi-tion, Tau protein phosphorylation and BACE expression in the hippocampus .The intervention of FSD-C10 decreased the protein levels of TLR-4 and p-NF-Κb, reduced the expression of Inos and increased the expression of Arg-1 in the brain tissues.CONCLUSION:The novel Rho kinase inhibitor FSD-C10 improves the capacity of spatial learning and memory in APP/PS1 double transgenic mice , which may be related to the inhibition of TLRs/NF-Κb signaling pathway , the reduction of the secretion of inflammatory molecules and the polarization of anti-inflammatory M2 microglia, thus improving the in-flammatory microenvironment of the brain in APP/PS1 double transgenic mice .
9.Prediction of microRNA-296-5p target genes and its application in lung development.

OBJECTIVETo predict the target genes of rno-microRNA-296-5p (miR-296) using bioinformatics software and databases, and to provide a theoretical basis for further studies of biological effects of miR-296 in fetal lung development.
METHODSPubMed and Google were used to search for all reported literature on miR-296. The miRBase database was used to determine the sequence and evolutionary conservatism of miR-296. The TargetScans database was used to predict the target genes of miR-296. The DAVID Bioinformatics Resources 6.8 database was used for the functional enrichment analysis of the target genes. The KEGG database was used to analyze the signaling pathways of target genes.
RESULTSmiR-296 was reported to play important roles in many biological processes and have a high degree of sequence conservation among species. The target genes of miR-296 were involved in biological processes, cell components, and molecular function. Those target genes were significantly enriched in the mitogen-activated protein kinase signaling pathway, Wnt signaling pathway, and transforming growth factor-β signaling pathway (p<0.05).
CONCLUSIONSThe bioinformatics analysis of the target genes of miR-296 provides a basis for studying biological effects and mechanism of action of miR-296 in lung development.

10.Study on Different Compatibility of Shuangshen Pingfei Decoction Influence on Decocting Rate of Mangiferin in Anemarrhena as-phodeloides

OBJECTIVE To explore the influence of different compatibility of Shuangshen Pingfei decoction on the decocting rate of mangiferin in Anemarrhena asphodeloides ,and ingredients to research influence on the mechanism of the solution rate. METHODS To disassemble formulas on the basis of the prescription of traditional Chinese medicine containing categories of ingredients.With decocting rate of mangiferin in Anemarrhena asphodeloides Bge as indexes,analyse different compatibility influence on mangiferin decocting rate.Based on these results of disassembled formulas,the representative alkaloid 1-Deoxyno-jirimycinand amino acid in different compatibility are used to research the influence of compositions of white mulberry on the decocting rate of mangiferin in Anemarrhena asphodeloides Bge and discuss the influence of the decocting rate mechanism with Anion and cation exchange resin processing of samples.RESULTS Research findings shows that the decocting rates of man-giferin in Anemarrhena asphodeloides Bge decoction,prescription combined decoction,Anemarrhena asphodeloides Bge-sapo-nins from Chinese herbal compound,Anemarrhena asphodeloides Bge-phenolic acid from Chinese herbal compound are 97.33%,47.03%,94.08%,57.53%,34.31%,respectively;Further study shows that Morus alba L.combined with Anemarrhena asphodeloides Bge have the most obvious effect on extraction yield of mangiferin.Anemarrhena asphodeloides Bge decoting with total alkaloids of Morus alba L,DNJ,glycine,β-alanine,respectively.The decocting rate of mangiferin in each group was 1 6.64%,28.13%,40.75% and 48.56%,respectively.CONCLUSION Our results suggest that the decoc-ting rates of mangiferin are mainly influenced by Morus alba L.We also conjectured that alkaloids and amino acids of Morus alba L.form complexes with mangiferin.
