1.Effect of Polydatin on Growth,Apoptosis, and ROS/p38 MAPK Signaling Pathway of Myeloma Cells

		                        			ObjectiveTo analyze the effects of polydatin on myeloma cell growth,apoptosis, and reactive oxygen species(ROS)/p38 mitogen-activated protein kinase(MAPK) signaling pathway. MethodHuman multiple myeloma (MM) cell line U266 cells were cultured in vitro,and the effects of polydatin at 0,20,40,80,160,200 mg·L-1 on the growth of U266 cells were detected by cell counting kit-8(CCK-8)assay. The half-maximal inhibitory concentration(IC50)was calculated. U266 cells in the logarithmic growth phase were randomly divided into a control group, low- and high-dose polydatin (80 and 160 mg·L-1) groups, and a bortezomib (75 nmol·L-1) group. After treatment with corresponding drugs,the cell viability of each group was determined by CCK-8 assay. The apoptosis rate of each group was measured by flow cytometry. The levels of inflammatory factors, such as tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β), and ROS in each group were measured by enzyme-linked immunosorbent assay (ELISA). The protein expression levels of apoptosis-related factors, including cysteine aspartate-specific protease-9(Caspase-9),B-cell lymphoma-2-associated X protein(Bax),p38 MAPK,and phosphorylated (p)-p38 MAPK in each group were detected by Western blot. ResultCompared with the results in the control group, polydatin of different concentrations could inhibit the growth of U266 cells (P<0.05),and the effect was potentiated with the increase in the concentration,with IC50 of 156.54 mg·L-1. Compared with the control group,the groups with drug treatment showed blunted cell viability (P<0.05) and increased apoptosis rate,TNF-α,IL-1β,and ROS levels, protein expression levels of Caspase-9, Bax,and p-p38 MAPK/p38 MAPK (P<0.05). Compared with the low-dose polydatin group, the high-dose polydatin group and the bortezomib group showed improved indicators mentioned above (P<0.05), and there was no significant difference between the high-dose polydatin group and the bortezomib group. ConclusionPolydatin can activate the ROS/p38 MAPK signaling pathway,promote the expression of inflammatory factors,inhibit the growth of U266 cells,and promote their apoptosis. 
		                        		
		                        		
		                        		
		                        	
2.Clinical effect of EGFR-TKIs combined with platinum-containing chemotherapy and whole brain radiotherapy in the treatment of non-small cell lung cancer with brain metastasis

		                        			Objective To explore the clinical effect of epidermal growth factor receptor-tyrosine kinase inhibitor (EGFR-TKIs) gefitinib combined with platinum-containing chemotherapy and whole brain radiotherapy in the treatment of non-small cell lung cancer (NSCLC) with brain metastasis. Methods One hundred and fifty-six patients of NSCLC with brain metastasis were divided into observation group (82 cases) and control group (74 cases) according to the therapeutic regimens. On basis of whole brain radiotherapy, the patients in control group received platinum-containing chemotherapy (paclitaxel+cis-platinum+pemetrexed), while the patients in observation group received gefitinib. The serum levels of carcino-embryonic antigen (CEA), cytokeratin 19 fragment antigen (CYFRA21-1) and EGFR, short-term effects, progression-free survival (PFS) and incidences of adverse reactions between two groups were evaluated. A subgroup analysis was carried on the 41 cases of patients with EGFR mutant type. The short-term effects and PFS between two groups were compared. Results The levels of CYFRA21-1 and EGFR in observation group were obviously lower than those in control group (P<0.01). The ORR and DCR in observation group was obviously higher than that in control group (81.71% vs. 36.49%, 93.90% vs. 82.43%) (P < 0.05 or < 0.01). The median PFS in observation group and control group was 12 months and 6months. There were statistic differences between two groups (P < 0.05). Subgroup analysis showed that the ORR and DCR in observation group was obviously higher than that in control group (72.0%vs. 25.0%, 96.0%vs. 31.25%)(P<0.01). The median PFS in observation group and control group was 13 months and 5 months. There were statistic differences between two groups (P<0.05). There was no statistic difference in incidences of adverse reactions between two groups (P>0.05). Conclusions For patients with NSCLC with brain metastasis, compared with the therapeutic regimen of whole brain radiotherapy + platinum-containing chemotherapy, whole brain radiotherapy + platinum-containing chemotherapy+EGFR-TKIs (gefitinib) could more effectively reduce the levels of EGFR and CYFRA21-1, improve short-term effects, prolong PFS and do not increase the incidence of adverse reactions.
		                        		
		                        		
		                        		
		                        	
3.Endovenous laser obliteration versus traditional surgical ligation for the treatment of varicosis of lower extremity: a comparative study

		                        			Objective To compare the effectiveness and the influence on life quality of endovenous laser obliteration with those of traditional surgical ligation in treating varicosis of lower extremity.Methods By using numerical random grouping method,40 patients with varicosis of lower extremity were divided into traditional surgical ligation group (high ligation plus stripping,traditional group,n=20) and endovenous laser obliteration group (laser group,n=20).The curative effect and complications were compared between the two groups.According to the brief health survey (SF-36) scale,the influences of the two treatments on the life quality were evaluated.Results During the follow-up period,the differences in the curative effect and complications between two groups were not significant (P>0.05).The life quality assessed by SF-36 scores in the laser group was better than that in the traditional group,the difference between the two groups was statistically significant (P<0.05).Conclusion For the treatment of varicosis of lower extremity,endovenous laser obliteration is safe and effective,it carries less influence on the patient's life quality when compared with traditional surgical ligation therapy.
		                        		
		                        		
		                        		
		                        	
4.Preparation and evaluation of pilocarpine nitrate in situ gel for ophthalmic use

		                        			Objective To develop and charaterize a series of Poloxamer-and Carbopol-based in situ gel for ophthalmic use as to enhance the ability of drug to retain in eyes and delay drug release.Methods The gel was prepared using Poloxamer 407/188 and Carbopol 974P as gelling agent and viscosity enhancer,respectively.Rheological characteristics were evaluated and behaviour of drug release in vitro was investigated by modified Franz diffusion cells.Results The rheological study indicated that the gel was physically entangled polymer solutions at 20 ℃ and then converted into a network structure with secondary bonds at 35 ℃.The gel released the drug molecules slowly in 4 h.The best fit model was Higuchi matrix model(r=0.992 3).Formulations consisting of Poloxamer 188 and Carbopol 974P were proved to be able to decrease the drug release speed efficiently.The impact on elastic modulus G" of the gel caused by those two were different.Conclusion An in situ gel with suitable sol-gel transition temperature and satisfactory release pattern could be achieved by adjusting the ratio of Poloxamer 407 to Poloxamer 188.The developed formulations have the ability to prolong the ocular residence time,which suggests it may be a new durg delivery system with bright future.
