1.Formulation Optimization of the Volatile Oil Microcapsules of Cinnamomi Ramulus and Angelicae Sinensis Radix and Their Pharmacokinetics in Vivo

OBJECTIVE To optimize the preparation technology of the volatile oil microcapsules of Cinnamomi Ramulus and Angelicae Sinensis Radix, characterize the microcapsules prepared after the formulation optimization and study pharmacokinetics characteristics in rats. METHODS Spray drying was used to prepare the volatile oil microcapsules of Cinnamomi Ramulus and Angelicae Sinensis Radix. The preparation process was optimized by Box-Behnken response surface method with the drug loading and encapsulation efficiency as indexes. The microcapsules were characterized by Fourier transform infrared spectroscopy(FT-IR) and scanning electron microscopy(SEM). Blood samples were collected after gastric administration at a dose of 100 mg·kg-1. HPLC method was adopted in the plasma concentration determination, and pharmacokinetics behavior in vivo was also compared. RESULTS The optimal formulation: core material-capsules material ratio was 1∶1.7, capsules material concentration was 10.25%, sodium starch octenyl succinate∶maltodextrin was 3.8∶1, high pressure homogenization pressure was 20 MPa and the inlet temperature of spray drying was 185 ℃. Under the optimized conditions, the drug loading was (18.94±1.09)% and the encapsulation rate was (96.03±2.91)%, respectively. The results of FT-IR and SEM showed that the essential oils had been successfully coated in the capsule wall material. The microcapsules were basically spherical in shape, with concave surface but no obvious cracks. The main pharmacokinetic parameters such as tmax, t1/2, CL, AUC0-t, k10,k12, and k21 of microcapsules had significant difference of the volatile oil(P<0.05 or P<0.01). Microcapsules effectively prolonged the circulation time of volatile oil in the body, promoted the absorption of drugs in the body and the oral bioavailability was enhanced to 2.62 times. CONCLUSION The model established by the Box-Behnken design-response surface method can be used to optimize the formulation of volatile oil microcapsules of Cinnamomi Ramulus and Angelicae Sinensis Radix with great prediction effect. The microcapsules prepared by the optimized process has good drug loading properties and the bioavailability of volatile oil is increased.
2.Framework and core contents of World Health Organization and United Nations Educational, Scientific and Cultural Organization health-promoting schools

Objective To systematically research the policies and core contents of World Health Organization (WHO) and United Nations Educational, Scientific and Cultural Organization (UNESCO) documents on policies, guidelines and standards for health-promoting schools and health services. Methods These policy documents included Making Every School a Health-promoting School: Implementation Guidelines, WHO Guidelines on School Health Services, and Making Every School a Health Promoting School Global Standards and Indicators, which construct a conceptual and policies framework for health-promoting schools. In perspective of health service system, this study systematically explored the policies, guidelines and standards of WHO and UNESCO on health-promoting schools and health services, as well as the role of health-promoting school component systems in promoting the health of children and how physical activity can be an important area of health-promoting schools. Results The Health Promoting Schools Initiative is an important area of research for the WHO health services. Making Every School a Health-promoting School: Implementation Guidelines primarily serves students aged five to 19, teachers and other staff in schools. WHO Guidelines on School Health Services cover a variety of activity types including health promotion, health education, preventive interventions, clinical assessment and health services management. Making Every School a Health Promoting School Global Standards and Indicators covers eight global criteria and thirteen specific areas in the areas of policy resources, curriculum and environment, and community engagement. Conclusion WHO and UNESCO have proposed a series of policies related to building health-promoting schools, which focus on child health and advocate the promotion of child and adolescent health, public health, education, social and economic development through schools to achieve the goal of safeguarding the health rights of children and adolescents and meeting health needs. Following the WHO Guidelines on School Health Services and Making Every School a Health Promoting School Global Standards and Indicators, it proposed to incorporate educational and physical education tools into the school health service system.
3.Preparation of quercetin-human serum albumin-nanoparticles and its inhibitory effects against NASH-induced liver fibrosis in vivo and in vitro

OBJECTIVE To prepare quercetin-human serum albumin-nanoparticles (Que-HSA-NPs),and to evaluate the in vivo and in vitro inhibitory effects of Que-HSA-NPs on hepatic fibrosis of non-alcoholic steatohepatitis (NASH). METHODS Que-HSA-NPs were prepared by desolvation-chemical cross-linking method ,their appearance characteristics were observed ,and their particle size ,polydispersity index (PDI),Zeta potential and drug loading were detected. Quercetin (Que)and Que-HSA-NPs were applied to murine HSC-T 6 cells. The effects of them on survival rate of HSC-T 6,mRNA expression of transforming growth factor β(TGF-β),Type Ⅰ collagen α1(COL1A1)and α-smooth muscle actin (α-SMA)were compared. Que and Que-HSA-NPs were applied to mice fed with low methionine and choline deficient high-fat diet. The serum levels of liver injury indexes ,liver pathological characteristics ,mRNA expressions of TGF-β,COL1A1 and α-SMA,protein expression of α-SMA in liver tissue were determined to evaluate the improvement effects of them on hepatic fibrosis of NASH in mice. RESULTS The prepared Que-HSA-NPs was spherical ,the particle size was (172.9±2.2)nm,the PDI was 0.233,the Zeta potential was -29.2 mV,and the drug loading was 2.99%. Que and Que-HSA-NPs were nontoxic to HSC-T 6 at concentrations of 0-250 μg/mL. Both of them could significantly decrease mRNA expressions of TGF-β,COL1A1 and α-SMA,especially Que-HSA-NPs (P<0.05). They also could significantly decrease the serum levels of liver inju ry index ,relieve liver injury and down-regulate mRNA expressions of TGF-β,COL1A1 and α-SMA and protein expression of α-SMA, especially Que-HSA-NPs (P<0.05). CONCLUSIONS Que- HSA-NPs is successfully prepared ,and confirm that its anti- NASH hepatic fibrosis effect is better than that of Que .
4.Study on effects and mechanism of interleukin 16 on pathogenesis of atherosclerosis

Objective To analyze the expression of interleukin 16 (IL-16) in atherosclerosis (AS) patients, and to study the roles of IL-16in the pathogenesis of AS.Methods Thirty AS patients in Affiliated Hospital of Jining Medical College from August 2015to August 2016were randomly selected as the case group and twenty-nine healthy subjects were selected as the healthy control group.Peripheral blood of the subjects were collected.IL-16levels were determined with enzyme-linked immunosorbent assay (ELISA).Reverse transcriptase-polymerase chain reaction was applied to analyze IL-16mRNA level.IL-16expression in the atherosclerotic plaque samples was detected with immunohistochemical analysis.IL-16expression in aortic atherosclerotic plaque of AS patients and atherosclerotic ApoE-/-mice were analyzed by immunohistochemical staining.The aortic plaque changes of AS mice injected intraperitoneally with recombinant IL-16were detected.Results Both IL-16protein levels and IL-16mRNA levels were higher in case group than those of healthy control group, the difference was statistically significant (P<0.05).The IL-16mRNA was highly expressed in the atherosclerotic plaque.The aortic plaque area of the mice underwent IL-16intraperitoneal injection were decreased while the plaque stability increased.Conclusion IL-16levels elevated in both AS patients and AS mice, which suggested that IL-16might play aprotective role against AS.
5.Clinical comparative study of breast-conserving surgery and improved surgical in breast cancer patients

Objective To comparative analysze of the applications value of breast-conserving surgery and improved surgical in breast cancer patients.Methods One hundred and sixty-eight female breast cancer patients from February 2005 to October 2010 in the People's Hospital of Rizhaoour hospital were selected.The patients were divided into breast-conserving surgery group (n =80) and improved surgical group (n =88).The patients in two groups were followed-up and investigated for three years.Operative time,blood loss,postoperative drainage,postoperative drainage time and hospital stay,incidence of complications,local recurrence,distant metastasis and survival after 3 year,satisfaction ratio of breast cosmetology,psychological anxiety scores and quality of life scores after 3 years of the patients in two groups were compared and analyzed.Results The complication rate of breast-conserving surgery group was 12.50% (10/80),lower than the improved surgical group,and the difference was significant statistically (29.55% (26/88),x2 =7.23,P<0.05).The operative time ((72.68±5.28) min),blood loss ((39.21 ± 8.59) ml),postoperative drainage ((162.25 ± 28.56) ml),postoperative drainage time((8.16±2.58) d) and the length of hospital stay((11.43±1.98) d) of the patients in breast-conserving surgery group were lower than those of the improved surgical group,and the differences of two groups were significant statistically ((92.48 ± 6.01) min,(68.26 ± 10.23) ml,(343.29 ± 62.18) ml,(16.59±4.96) d,(18.47±2.94) d;t =22.72,19.99,24.61,13.99,18.35;P<0.05).The satisfaction rate after 3 years of breast-conserving surgery group was 92.50%(74/80),higher than that of the improved surgical group,and the difference was significant statistically (81.82% (72/88),x2 =4.20,P< 0.05).The psychological anxiety scores after 2 and 3 years of breast-conserving surgery group were (40.02 ± 2.63) points and (34.26 ± 3.59) points,lower than those of improved surgical group((42.58±2.37) points and (39.85±2.10) points),the quality of life scores after 3 years of breast-conserving surgery group was (67.58± 11.26)points,higher than that of improved surgical group ((50.85 ± 10.42) points),and the differences were significant statistically (t =6.60,12.16,9.96;P < 0.05).Conclusion Breast-conserving surgery could reduce the incidence of complications and psychological anxiety situations,improve clinically relevant indicators and satisfaction ratio of breast cosmetology and quality of life for the breast cancer patients.
6.Morphology and distribution of EpCAMhigh/CD44+ colorectal cancer stem cells

BACKGROUND: Cancer stem cells have bean a hot topic in life science research. EpCAMhigh/CD44+ colorectal cancer stem cells are beneficial to observe morphological characteristics and distribution of cancer stem cells.OBJECTIVE: To investigate the quantity, location, distribution and hematoxylin-easin staining morphologic features of colorectal cancer stem cells (Co-CSCs).DESIGN, "rIME AND SETrlNG: The observational study was performed at the Department of Pathology, Lanzhou General Hospital of Lanzhou Military Area Command of Chinese PLA from March to August 2008.MATERIALS: A total of 67 colorectal cancer paraffin embedding samples were collected from 200512007 archive at the Department of Pathology, Lanzhou General Hospital of Lanzhou Military Area Command of Chinese PLA, comprising 27 males and 40 females, aged 29 90 years.METHODS: EpCAM and CD44 was used to label cancer stem cells in paraffin embedding colorectal cancer samples. The locations of EpCAM and CD44 double positive cells were detected by SP(streptavidin HRP) immunohistochemical staining and double immunohistochemical staining. The morphologic features of them were investigated on hematoxylin-easin staining at the same position.MAIN OUTCOME MEASURES: Number, location, distribution of EpCAM and CD44-posltive cells, and morphology following hematoxylin-eosin staining.RESULTS: The number of double-positive cells accounted for 0.1% to 30.0% of all tumor cells, and the cells were scattered or distributed focally along the basement of glandular-like structure. The cells with scarcely cytoplasm were cube or oval, and its nucleus was oval or high cylindrical, deep stained and homogeneous; The quantity of double-positive cells were negatively correlated with the differentiation of colorectal cancer.CONCLUSION: Cancer stem cells are the causes of tumor development, metastasis and recurrence and drug resistance. To observe the number, location, distribution and morphology of Co-CSCs and to analyze the relationship between Co-CSCs and pathological parameters will provide guidance for the diagnosis, staging, prognostic evaluation and clinical prognosis of cotorectal carcinoma.
7.Relationship between thrombospondin-1 gene GI678A polymorphism and acute coronary syndrome

Objective To investigate the possible association between the thrombospandin-1(TSP-1) gene GI678A (Ala523Thr)polymorphism and acute coronary syndrome (ACS) in a Chinese Han population.Method he ease cohort studied was compsed of 412 hospitalized patients with ACS recruited from four participating hospitals between November 2003 and May 2006.The diagnosis of ACS was based on the criteria of AHA/ACC set in 2002.The eontrul group was consisted of 319 age- and sex-matched subjects from partiei pating hospitals,and they were free from coronary artery disease judged by history,clinical examination,electrocardiography,exercise test and angiography.The TSP-1 GI678A polymorphism was determined by polymerase ehain reaction and restriction fragment length polymurphism analysis(PCR-RFLP).Results The prevalence OfAA genotype of the G1678A polymorphismin patients with ACS was significantly higher than that in the control subjects (49.5%% vs.40.4%,P=0.015).The frequencies of GA and GG genotypes were not significantly different between patients with ACS and controls (CA:39.3% vs.46.1%,P=0.070;CA;11.2% vs.13.5%,P=0.340).The frequencies of A allele in the ACS group and control group were 69.2% and 63.5%,respectively (P=0.022).Furthermore,multiple logistic regression analysis showed that the AA genotype was a significant risk factor for ACS (OR=1.52;95% CI:1.11~2.08;P=0.010).Conclusions The present findings suggest that the AA genotype in TSP-1 gene GI678A polymorphism may be associated with a risk factor for ACS in the Hart nationality of China.The AA genotype may be a genetic marker of the liability to the inheritance of AC,S.
8.Research progress in colorectal cancer stem cells

BACKGROUND: Colorectal cancer is a common malignant tumor in digestive tract. Theory of tumor stem cells (TSCs) offers a new approach to understand the development mechanism of colorectal cancer. OBJECTIVE: To review the progress in the existence, origin and identification of colorectal cancer stem cells (Co-CSCs). RETRIEVAL STRATEGY: Pubmed was undertaken to identify relevant articles published from January 1990 to July 2007 with the of "colorectal cancer,stem cell,tumor stem cells" in English.At the same time, Vip database was undertaken to identify the relevant articles published between January 1990 and July 2007 with the key words of "colorectal cancer, stem cell, tumor stem cells" in Chinese. The data were selected primarily, and then quotations of each article were checked. Inclusive criterion: the articles related to the advances of the existence, origin and identification of Co-CSCs were selected. Exclusive criterion: the articles with repetitive research were excluded. Totally 63 relevant articles were selected and 33 of them met the inclusive criterion. The 30 excluded articles were of old or repetitive content. LITERATURE EVALUATION: Of the 33 inclusive articles, 28 were on the existence and origin of Co-CSCs, 4 were about the isolation and identification of Co-CSCs, and 1 was about the problems and prospect. DATA SYNTHESIS: TSCs is a rare population of cells in tumor tissues with infinitely proliferative and self-renewal potential that drive the formation and growth of tumors. It may become a new target to eradicate malignant tumor. TSCs have been discovered in leukemia and some solid tumors. Intestine stem cells are poorly differentiated. Its phenotype is difficult to identify. There are many reports on the quantity, location, origin, function and relation with intestinal disease. Co-CSCs have been isolated and identified as EpCAMhigh/CD44+ cells recently, which accelerate to understand the mechanism of colorectal cancer. CONCLUSION: The discovery of Co-CSCs is one of the advances in researching of malignant tumor. TSCs may become a new target to eradicate malignant tumor, and brings hope to cure it.
9.Application and Research of Membrane Separation Technology in Banlangen Keli Production

Objective To investigate the refining and concentration of the Banlangen aqueous extractives using membrane separation technology. Methods The membrane separation process for refining and concentration Banlangen aqueous extractives was optimized by the quality of products and membrane operation parameters, and was tested through medium-scale trials. Results The selected G2+GM+RO2 membrane process was suitable. The inherent quality of the products obtained by membrane separation process was better than that obtained by traditional process. Conclusion G2+GM+RO2 membrane process is suitable to refine and concentrate Banlangen aqueous extractives, and it has technical conditions for industrial scale trials.
10.Distribution and Pharmacokinetics of Lung Targeting Etoposide- bovine Serum Albumin- microspheres in Mice

OBJECTIVE: To prepare etoposide- bovine serum albumin- microspheres (VP- BSA- MS)and to study the distribution and pharmacokinetics of VP- BSA- MS in mice. METHODS: The drug concentrations in various tissues were determined by high- performance liquid chromatograph (HPLC). RESULTS: The VP- BSA- MS was injected into mice and (47.88± 2.56 )% of the total dosage was detected in lung tissue 15min after administration, the pharmacokinetical equation was C=149.0 897e- 1.7 780t+ 3.9 627e- 0.0 398t — 153.0 524e- 3.5 054t. CONCLUSION: The VP- BSA- MS showed remakable targeting action to the lung and the pharmacokinetic regularity could be discribed as two- compartment model
