1.Mechanism study of IGHG1 on proliferation,apoptosis,and invasion of THP-1 cells in acute myeloid leukemia

		                        			Objective To investigate the expression of immunoglobulin γ-1 heavy chain constant region(IGHG1)in acute myeloid leukemia(AML)THP-1 cells and its influence on cell proliferation,apoptosis,and invasion via its regulation of the transforming growth factor β(TGF-β)/Smad pathway.Methods Bone marrow specimens from nine children with AML and eight children with fractures,human bone marrow stromal HS-5 cells,and human AML THP-1 and HL60 cells were used in the research.Western blot was used to detect IGHG1 protein expression.THP-1 cells were divided into a blank group(without any treatment),si-NC group,si-IGHG1-1 group,si-IGHG1-2 group,si-IGHG1-3 group,TGF-β group,si-IGHG1-1+TGF-β group,and si-IGHG1-1+TGF-β +LY364947 group.Cell proliferation was measured by CCK-8 method.Flow cytometry and Transwell experiment were performed to measure apoptosis and invasion.Western blot was used to detect the protein expression of IGHG1,TGF-β,p-Smad2 and p-Smad3 in each group of cells.Results Compared with the bone marrow of children with fractures,the bone marrow of children with AML(P<0.05)had significantly higher expression levels of IGHG1((0.24±0.03)vs(0.87±0.12)).Compared with HS-5 cells,human AML THP-1 and HL60 cells had significantly increased expression levels of IGHG1((0.89±0.14)(0.75±0.08)vs(0.21±0.02))(P<0.05).Compared with the blank group,the si-IGHG1-1 group of THP-1 cells showed significantly reduced OD450 values(24 h,48 h,72 h),invading cell numbers and protein expression of TGF-β,p-Smad2,p-Smad3 and their apoptosis rate was increased(P<0.05),while the corresponding indexes showed the opposite trend in the TGF-β group(P<0.05).TGF-β reversed the effects of silencing IGHG1 on the proliferation,apoptosis and invasion of THP-1 cells.Compared with the si-IGHG1-1+TGF-β group,the si-IGHG1-1+TGF-β+LY364947 group had significantly decreased TGF-β,p-Smad2,p-Smad3 and protein levels,OD450 values and cell invasion numbers and the apoptosis rate was increased(P<0.05).Conclusions IGHG1 is highly expressed in AML cells.Silencing the IGHG1 gene can inhibit the proliferation and invasion of AML cells and promote the apoptosis of AML cells.This mechanism may be related to the inhibition of the TGF-β/Smad pathway.
		                        		
		                        		
		                        		
		                        	
2.Pregnancy outcomes in patients with adenomyosis with fertility requirements: retrospective analysis of clinical data from real world

		                        			Objective:To retrospectively analyze the pregnancy outcomes of patients with adenomyosis requiring fertility in a single center under real world condition.Methods:From June 2015 to May 2020, 231 cases of pregnancy complicated with adenomyosis diagnosed by ultrasound with fertility requirements were treated in the Women′s and Children′s Hospital Affiliated to Qingdao University with complete clinical data. And they were divided into three groups according to the treatment of adenomyosis before pregnancy: expectation group, drug group and operation group. The relevant data before pregnancy of the three groups were analyzed, and the pregnancy outcomes of the patients were summarized. According to whether the early pregnancy was treated with medication, the patients who were naturally conceived without symptoms of threatened abortion were divided into observation group and fetus protection group, and the pregnancy outcomes of the two groups were compared.Results:(1) Compared with the expectation group, the ages of patients in the drug group and the operation group were larger [(31.5±1.8) vs (34.1±3.7) vs (36.9±3.6) years old], and the difference was statistically significant ( P<0.05). Only 9 patients (11.5%, 9/78) had clinical symptoms in the expectation group, while the patients in the drug group and the operation group had a higher proportion of dysmenorrhea and increased menstrual volume. The uterine volume of the drug group and the operation group were larger than that of the expectation group [(151±46) vs (166±27) vs (97±18) cm 3], the difference was statistically significant ( P<0.05). 78.6% (33/42) of the operation group were focal adenomyosis. The proportion of natural pregnancy in the expectation group was 97.4% (76/78), and in vitro fertilization and embryo transfer was mainly used in the drug group and the operation group. (2) The abortion rates of the three groups were 48.7% (26/111), 4/17, 67.5% (27/78) respectively. Compared with the drug group and the operation group, the preterm birth rate was lower [55.9% (33/111) vs 11/17 vs 12.5% (5/78)] and the natural delivery rate was higher [44.1% (26/111) vs 4/17 vs 67.5% (27/78)] in the expectation group. (3) There were 89 cases of spontaneous pregnancy without threatened abortion symptoms, including 31 cases in the observation group and 58 cases in the fetus protection group. Compared with the observation group, the abortion rate of patients in the fetus protection group was lower [41.9% (13/31) vs 34.5% (20/58)], and the difference was statistically significant ( P<0.05). Conclusions:Patients with adenomyosis who have fertility requirements should be comprehensively evaluated and individualized treatment plans should be given. Pregnancy patients with adenomyosis have a high rate of miscarriage, and they should be included in the management of high-risk pregnant women. 4.Efficacy of probiotics on chemotherapy-induced intestinal mucositis in rats: a meta-analysis

		                        			Objective To evaluate the efficacy of probiotics on chemotherapy-induced intestinal mucositis in rats.Methods The databases including PubMed,EMbase,Cochrane,CBM,CNKI,WanFang and VIP were retrieved from their establishment to April 2016.The related randomized controlled trials(RCT) on the effects of probiotics for treating chemotherapy-induced intestinal mocositis in rats were included.The relevant literatures were screened according to the inclusion and exclusion criteria,then the data were extracted and analyzed.Results Total 6 RCT were included.Compared with the control group,the intestinal secretion and absorption function in the probiotics group was strengtnened[SMD=1.73,95 %CI(0.79,2.68),P<0.01],jejunal anti-oxidant capacity was increased [SMD=-2.12,95%CI(-3.56,-0.67),P<0.01],however low dose probiotics (<1 × 109 cfu/d)had no protective effect on small intestine[SMDjejunum =-0.06,95%CI(-0.51,0.40),SMDileum =0.02,95% CI(-0.71,0.75);P >0.05],while high dose probiotics(≥ 1 × 109 cfu/d) could reduce the intestinal pathological damage[SMDjejunum =-0.64,95 % CI (-1.20,-0.09),SMDileum=-0.85,95% CI(-1.59,-0.12);P<0.05].Conclusion High dose probiotics can reduce chemotherapy-induced intestinal mucositis in rats.Because of less included literatures and the influence of publication bias,the effect of probiotics on chemotherapy induced mucositis could be overestimated.
		                        		
		                        		
		                        		
		                        	
5. Determination of dimethyl sulfate in the air of workplace by GC-MS

		                        			To establish a method to detect the concentrations of dimethyl sulfate in the air of workplace by GC-MS.
		                        		
		                        			Methods:
		                        			DMS in the air of workplace adsorpted by Silicone tube, then desorped by acetone, add 1.0 ml of acetone, shake 1 min, placed after 30 min, the desorption solution qualitative and quantitative determination by gas chromatography-mass.
		                        		
		                        			Results:
		                        			The calibration curves were liner in the range of 0.1-200.0 μg/ml. The within-run and between-run precisions were 2.6%-4.7% and 4.0%-9.0% respectively. The method detection limit is 0.1 μg/ml, the minimum detectable concentration is 0.02 mg/m3 (in terms of sampling 4.5 L) . Add 1 ml of desorption liquid, place 30 min, the average desorption efficiency of more than 90%.
		                        		
		                        			Conclusion
		                        			This method has simple pretreatment, short analysis period, and optimized linear rage and limit of detection, and is suitable for the determination of DMS in workplace air. 
		                        		
		                        		
		                        		
		                        	
6.Determination of total Bromine in urine by inductively coupled plasma mass spectrometry (ICP-MS).

OBJECTIVETo establish a method to determine total bromine in urine.
METHODDiluted urine samples were directly introduced into ICP-MS then quantized by standard curve.
RESULTTotal bromine in urine was linear within 1.0~50 mg/L with r > 0.999, When spiked at a concentration of 0.020 mg/L, 0.050 mg/L, 0.150 mg/L, the recovery was 95%~98%, intra-assay precision was 1.4% 3.2%, inter-assay precision was 3.4% to 5.0%. Urine could store in -20 °C refrigerator 3 months without any bromine loss.
CONCLUSIONUsing ICP-MS to determine the urinary total bromine, the method is fast, accurate, wide linear range of features, could meet with the requirement of Part 5 of occupational health standards guide: Method determination of chemical substances in biological materials (GBZ/T 210.5-2008), a strong competitive advantage in a wide range of survey, suitable for promotion.

8.Metabolism study of asperosaponin VI by using zebrafish.

		                        			Model organism zebrafish was used to study metabolism of asperosaponin VI from Dipsacus asper Wall. ex Henry for the first time. Metabolic components of asperosaponin VI after exposing to zebrafish for 24 h were identified by high performance liquid chromatography--electrospray mass spectrometry (HPLC-ESI-MS), the separation was performed with a Zorbax C18 column using a binary gradient elution of 0.05% formic acetonitrile--0.05% formic acid water. The quasi-molecular ions of compounds in both negative and positive mode were observed for molecule mass information, and the potential structures were identified by attentive study on the deglycosylation metabolites and one hydroxylation metabolite of asperosaponin VI. The results were highly in consistent with metabolism of asperosaponin VI in rat. It can be concluded that zebrafish model can wonderfully imitate current metabolic model with advantages of small amount of lower cost, far less amount compound, higher efficiency and more simple, and can reflect integrated metabolism results of in vivo method. Zebrafish metabolic model may become a novel organism model for quick predication on metabolism of even mircoamount compound, which can enrich the available models greatly.
		                        		
		                        		
		                        		
		                        	
9.Construction and identification of the human liver-specific miR-122 expression vector.

		                        			miR-122 is the most abundant miRNA in adult human liver. To study the functions of miR-122 in liver disease, we amplified the precursor of human miR-122 gene by polymerase chain reaction (PCR) from the HepG2 genomic DNA, and then constructed miR-122 expression vector pLMP-miR-122. pLMP-miR-122 could overexpress mature miR-122 when human normal liver cells L-O2 and the hepatoma cells HepG2 were transfected with it. When HepG2 cells were co-transfected with HBV1.3 and pLMP-miR-122, we found that miR-122 could down-regulate the expression of HBs and HBe antigen. These results showed that the human liver specific miR-122 expression vector was constructed successfully, and it could regulate the replication and the expression of HBV genes. The plasmid pLMP-miR-122 will facilitate further studies of the functions of miR-122 in the development of liver virus infection diseases and HCC.

10.Observation on the effect of serf-care education and nursing package usage on non-throat patients

		                        			Objective To discuss the effect of self- care education at the early time of postoperation and the use of nursing packages during the later period on non- throat patients. Methods 40 patients having un-dergone total laryngectomy were divided into the control group and the experimental group according to the surgery date with 20 cases in each group. The control group received conventional nursing, while the experi-mental group was given the system intervention by specialized nurses. The intervention content included self-care education at the early time of postoperation and the use of nursing packages during the later period. The questionnaire survey was carried out 15 days and 3 months after operation.The coping style,self-care ability and survival quality were compared between the two groups. Results The comparison of coping style showed re-markable difference in the two groups. The comparison of self- care ability and level of health knowledge showed significant difference 15 days after operation except two other items,but the four items demonstrated significant difference 3 months after operation. Difference also existed in the total scorn of survival quality. Conclusions Early system serf- care education after operation and the use of nursing package during the later period can impel the patients to cope positively, enhance self-care ability and survival quality.
