2.All-trans retinoic acid suppresses proliferation of breast cancer cells through DOK1/PPARγpathway

AIM:To investigate the potential effect of all-trans retinoic acid (ATRA) on the proliferation of human breast cancer MCF-7 cells and its underlying mechanism.METHODS:Human breast cancer MCF-7 cells were in-cubated in DMEM supplemented with 10%fetal bovine serum and 1%penicillin/streptomycin.Western blot was performed to detect the protein expression and its phosphorylation.MTT assay and bromodeoxyuridine ( BrdU) incorporation, and TUNEL staining were carried out to determine the cell proliferation and apoptosis, respectively.Lentivirus carrying shRNA sequences targeting the gene of docking protein 1 ( DOK1 ) was used to silence DOK1 expression.The activity of peroxi-some proliferator-activated receptor gamma ( PPARγ) was measured using a PPARγtranscription factor assay kit.RE-SULTS:ATRA treatment suppressed the proliferation and promoted the apoptosis of MCF-7 cells.ATRA was also found to induce DOK1 expression in a time-dependent manner.Silence of DOK1 mitigated anti-cancer effect of ATRA evidenced by recovered the cell proliferation and reduced the cell apoptosis.In addition, DOK1 knockdown inhibited PPARγexpression and activity.Furthermore, inhibition of PPARγwith its specific inhibitor GW9662 attenuated impacts of ATRA on the cell proliferation and apoptosis.CONCLUSION: ATRA suppresses MCF-7 cell survival through inhibiting cell proliferation and promoting cell apoptosis, which is mediated by DOK1-activated PPARγ.
3.Ubiquitin-proteasome system regulates adipogenic differentiation by stabi-lizing HSP90-dependent PPARγ

AIM: To investigate the role of ubiquitin-proteasome system ( UPS) in adipocyte differentiation. METHODS:Differentiation of 3T3-L1 preadipocytes into adipocytes was induced by treatment with insulin, 3-isobutyl-1-methylxanthine and dexamethasone.Western blot and immunoprecipitation were performed to detect the protein abundances and association, respectively.Oil red O staining was used to determine the intracellular lipid of 3T3-L1 adipocytes.The levels of mRNA were measured by reverse transcription polymerase chain reaction ( RT-PCR) .RESULTS:UPS inhibitor bortezomib (BZM) suppressed the differentiation of 3T3-L1 pre-adipocytes, evidenced by reduced intracellular content of triglyceride, and decreased mRNA expression of adipogenic marker proteins such as adiponectin and adipocyte protein 2.In contrast, administration of sildenafil (SDN), an activator of protein kinase G which was also found to activate UPS, promo-ted adipocyte differentiation.In addition, BZM treatment decreased the expression of heat shock protein 90 (HSP90) and peroxisome proliferator-activated receptor gamma ( PPARγ) in the soluble fraction and reduced association of HSP90 and PPARγ.Furthermore, HSP90-specific N-terminal inhibitor geldanamic mitigated SDN-induced increase in PPARγlevel and 3T3-L1 cell differentiation.CONCLUSION:UPS modulates HSP90-dependent PPARγstability, thus leading to pro-motion of adipocyte differentiation.
4.Correlation between burden of main caregivers of patients undergoing enterostomy and social supports

Objective To investigate the correlation between burdens of main caregivers of patients undergoing enterostomy and social supports.Method Fifty eight caregivers of patients with enterostomy involved the investigation by using questionnaires of Zarit Caregiver Burden Interview and Social Support Rating Scale.Results The mean score of burdens was(21.10±14.17).The total score of social supports was(43.69±8.61),higher than the national norm(34.56±3.73)(P<0.05).The total score of burdens, personal burdens and responsibility burdens were all negatively correlated with social support(P<0.01 for all).Conclusion The higher social support,the lower care burdens.The doctors and nurses should help the patients and their families to establish more accesses to the social supports so that the caregivers relieved and stress and reduce the burdens.

Based on the study of four diagnostic methods in Treatise on Exogenous Febrile Diseases,this dissertation analysed the cause of differentiation of syndrome unavailable,and indicated that:differentiation of syndrome unavailable resulted from multi-reasons.The patients who had symptoms and signs but all the modern physics and chemistry index were normal,the data get from traditional four diagnostic methods were useful to discriminate and estimate diseases.Modern physics and chemistry index can help in improving exactitude,but we cannot ignore traditional diagnostic methods.

目的探讨中西医结合在脑外伤反应治疗中的作用。方法将120例脑外伤反应患者分成两组:治疗组(80例)在综合措施治疗的基础上,使用理气开窍汤;对照组(40例)按综合措施治疗。观察治疗20d内患者头痛、头昏、耳鸣、恶心呕吐、失眠症状改善情况。结果临床症状有效率比较,两组有显著性差异(P<0.05)。结论理气开窍汤配以高压氧及营养神经类药物,可明显提高疗效。
7.Effects of nerve growth factor on N-methyl-D-asparate receptor 1 after spinal cord injury in rats.

OBJECTIVETo explore the effects of the nerve growth factor (NGF) on N-methyl-D-asparate receptor 1 (NMDAR 1) after spinal cord injury.
METHODSSpinal cord injury of Wistar rats was performed with Allen's method by a 10 gx2.5 cm impact on the posterior T8 spinal cord. NGF was given to the rats of the treatment group via subarachnoid space tube at once, 2, 4, 8, 12 and 24 hours after spinal cord injury, respectively. The expression of NMDAR1 mRNA in spinal cord was detected by in situ hybridization.
RESULTSRare expression sequence of NMDAR1 mRNA was found in rat spinal cord of the normal group. A strong expression sequence of NMDAR1 mRNA was found in rat spinal cord of the normal saline group. The expression of NMDAR1 mRNA in the NGF group was significantly decreased as compared with that in the normal saline group (P=0.01).
CONCLUSIONSNGF can relieve damage of injured spinal cord by prohibiting the expression of NMDAR1 mRNA.

8.Effects of nerve growth factor on neuronal nitric oxide production after spinal cord injury in rats.

OBJECTIVETo explore the protective effects of nerve growth factor (NGF) on injured spinal cord.
METHODSThe spinal cord injury (SCI) model of Wistar rats was established by a 10 gx2.5 cm impact force on the T(8) spinal cord. NGF (60 microg/20 microl) was given to the rats of the treatment group immediately and at 2, 4, 8, 12, 24 hours after SCI. The level of neuronal constitutive nitric oxide synthase (ncNOS) and the expression of ncNOS mRNA in the spinal cord were detected by the immunohistochemistry assay and in situ hybridization method.
RESULTSAbnormal expression of ncNOS was detected in the spinal ventral horn motorneuron in injured rats. The levels of ncNOS protein in the NGF group were significantly lower than those in the normal saline group (P<0.05 ). The ncNOS mRNA expression was found in the spinal ventral horn motorneuron in injured rats and the expression in the NGF group was significantly decreased compared with that in the normal saline group (P<0.01).
CONCLUSIONSNGF can protect the injured tissue of the spinal cord by prohibiting abnormal expression of nitric oxide synthase and the neurotoxicity of nitric oxide.

9.Effect of nerve growth factor on neuronal apoptosis after spinal cord injury in rats.

OBJECTIVETo explore the molecular mechanism of the protective effect of nerve growth factor (NGF) on injured spinal cord.
METHODSThe posterior T(8) (the 8th thoracic segment) spinal cords of 60 Wistar rats were injured by impacts caused by objects (weighing 10 g) falling from a height of 2.5 cm with Allen's way. Solution with nerve growth factors (NGF) was given to 30 rats (the NGF group) through a microtubule inserted into the subarachnoid cavity immediately, and at 2, 4, 8, 12 and 24 hours after spinal cord injury (SCI) respectively. Normal saline (NS) with same volume was given to the other 30 rats (the NS group) with the same method. And 5 normal rats were taken as the normal controls. The expression of bcl-2 and bax proteins in spinal cord was detected with immunohistochemistry. The apoptotic neurons in spinal cord were measured with terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end-labeling of DNA fragments (TUNEL) staining.
RESULTSThe positive expression of bcl-2 protein was strong in the normal controls, but decreased in the NS group, and increased significantly in the NGF group as compared with that of the NS group (P<0.01). The positive expression of bax protein was also strong in the normal controls, but increased in the NS group, and decreased significantly in the NGF group as compared with that of the NS group (P<0.01). Apoptotic neurons were found in the NS group, and they decreased significantly in the NGF group as compared with that of the NS group (P<0.01).
CONCLUSIONSNGF can protect the injured nerve tissues through stimulating the expression of bcl-2 protein, inhibiting the expression of bax protein and inhibiting the neuronal apoptosis after SCI.

10.Application of MR Urography in the Diagnosis of Iatrogenic Ureteral Injury

Objective To evaluate the diagnostic value of magnetic resonance urography(MRU)in patients with iatrogenic ureteral injuries.Methods MRU in 12 patients with iatrogenic ureteral injuries caused by gynecological and rectal operation were performed.Results MRU examinations were successfully done in all cases at once.The accurate rate in localized and qualitative diagnosis was all 100%.The injures of ureter in 12 cases were all located at lower segment of the ureter,of them,8 patients were purely obstruction,4 cases were obstruction with urinal leakage,all cases were accompanied by dilations of ureter and hydronephrosis.Conclusion MRU is helpful for the diagnosis of iatrogenic ureteral injury.
