1.Characterisation of the dynamics of MAOA/FOXA1 in the progression of neuroendocrine differentiation in prostate cancer

Objective To explore the dynamic changes in monoamine oxidase A(MAOA)and forkhead box A1(FOXA1)levels during neuroendocrine differentiation(NED)in prostate cancer,providing new strategies for the treatment of neuroendocrine prostate cancer.Methods Cell models and mouse transplantation models of NED were established through long-term sustained induction with enzalutamide(ENZ).Dynamic expression of MAOA and FOXA1 in NED was detected by Western Blot and Real-time PCR.GEO database data were selected to analyze the dynamic trends in MAOA and FOXA1 levels in multiple NED models.We constructed a mouse transplantation model of human prostate cancer cell lines and analyzed the dynamic expression of MAOA and FOXA1 in the in vivo NED model by immunohistochemistry.MAOA expression was disrupted with lentiviral transfection,and the impact on FOXA1 was detected.Results Both MAOA and FOXA1 concentrations showed dynamic characteristics,increasing and then decreasing during the NED process.Knockdown of MAOA in prostate cancer cells led to decreased expression of FOXA1.This MAOA may play different roles at different stages of NED by acting through FOXA1.Conclusions Both MAOA and FOXA1 levels showed increasing,then decreasing,trends during NED.The expression of MAOA affected the level of FOXA1,and MAOA/FOXA1 may play a dynamic regulatory role in the NED process.
2.Correlation analysis of the expression of LACTB and GST-π in focal tissues of endometrialcarcinoma with their survival and estrogen and progesterone receptors

Objective:To investigate the expression of LACTB and GST-π in endometrial carcinoma and their correlation with estrogen and progesterone receptors.Methods:A total of 165 patients with endometrial cancer admitted to our hospital from Jan. 2015 to Oct. 2020 were selected to observe the expression of LACTB, GST-π, estrogen and progesterone receptors in patients with different tumor stages, degrees of differentiation, muscular infiltration, tissue type, tumor diameter and whether there was lymph node metastasis. The correlation between the expression of LACTB and GST-π and the expression of female and progesterone receptors and the survival of patients with different expressions of LACTB and GST-π were analyzed.Results:The LATCB positive rate decreased in patients with tumor stage III to IV, high differentiation, myometrial infiltration ≥1/2, tissue type I, tumor diameter <2 cm and no lymph node metastasis ( P<0.05), and the GST-π positive rate was not statistically significant ( P>0.05). The positive rate of GST-π in patients with chemotherapy resistance was higher than that in patients with chemotherapy sensitivity, and the difference was statistically significant ( P<0.05). The positive rate of estrogen receptor decreased in patients with tumor stage III to IV, high differentiation, tissue type II, no lymph node metastasis, and chemotherapy resistance, and the difference was statistically significant ( P<0.05). The positive rate of progesterone receptor decreased in patients with tumor stage III to IV, low differentiation, tissue type I and no lymph node metastasis, and the difference was statistically significant ( P<0.05). In this study, 110 patients with LACTB positive expression were detected, with an average LACTB positive staining intensity (25.92±4.77) %, and 99 patients with GST-π positive expression were detected, with an average GST-π positive staining intensity (27.46±4.83). A total of 50 patients with LACTB positive and GST-π negative had an average survival time of (41.48±5.52) months, a total of 39 patients with LACTB negative and GST-π positive had an average survival time of (21.25±3.15) months, and a total of 60 patients with LACTB positive and GST-π positive had an average survival time of (21.25±3.15) months. The mean survival time of 16 patients with both LACTB negative and GST-π negative was 29.31±3.77 months. The mean survival time was 31.54±4.61 months. Pearson correlation test showed that the staining intensity of LACTB positive staining was positively correlated with the survival time of patients. The positive staining intensity of GST-π was negatively correlated with survival (correlation coefficient =0.392, -0.284, P<0.01). Pearson correlation analysis showed that LACTB was positively correlated with estrogen and progesterone receptors. GST-π was negatively correlated with estrogen receptor ( P<0.05) . Conclusions:The expression of LACTB in patients with endometrial cancer is associated with the disease of the patients. The survival of patients with high expression is longer. The expression of GST-π is associated with the chemotherapy resistance of patients, both of which can be used as indicators to evaluate the prognosis of patients. Moreover, the expressions of LACTB and GST-π are correlated with the expression of female and progesterone receptors, which may be regulated by the expression of female and progesterone receptors.
3.Screening for UV-C irradiation-enhanced transcription factors that regulate the metabolism of phenolic compounds in tomato fruit.

Solanum lycopersicum phenylalanine ammonia-lyase 5 (SlPAL5) gene regulates the metabolism of phenolic compounds. The study of transcription factors that regulate the expression of SlPAL5 gene is of great significance to elucidate the regulatory mechanism underlying the biosynthesis of phenolic compounds in tomato fruit induced by UV-C irradiation. Here, yeast one-hybrid library of tomato fruit was constructed, and the yeast one-hybrid technology was used to screen the transcription factors that regulate the expression of SlPAL5, the key gene related to the synthesis of phenolic compounds in tomato fruit. As a result, a transcription factor, SlERF7, was obtained and sequenced, followed by the blast homology analysis. Further experiments confirmed that SlERF7 interacted with the promoter of SlPAL5 gene. In addition, UV-C irradiation significantly increased the expression level of SlERF7. These results indicate that SlERF7, which is regulated by UV-C irradiation, might be involved in regulating the transcription of SlPAL5, which provided foundations for further studying the regulation mechanism of the biosynthesis of phenolic compounds in tomato fruit induced by UV-C irradiation.

4.Application of G arm X-ray machine in minimally invasive treatment of thoracolumbar vertebral osteoporotic vertebral compression fractures

Objective To compare the clinical effects of G-arm X-ray machine and C-arm X-ray machine in percutaneous kyphoplasty (PKP) for osteoporotic vertebral compression fracture (OVCF) of thoracolumbar spine.Methods The clinical data of ninety-five patients with thoracolumbar OVCF treated with PKP from May 2016 to August 2017 in Yanan University Affiliated Hospital were retrospectively analyzed.They were divided into two groups according to the different guiding fluoroscopy methods used during the operation.Forty-six cases in G arm group completed PKP under the guidance of G arm X-ray machine,Forty-nine cases in C arm group completed PKP under the guidance of C arm X-ray machine.The operation time,fluoroscopy times,cement leakage cases,the height of injured vertebral leading edge,Cobb angle of kyphosis,visual analogue score and Oswestry dysfunction index were recorded before and after operation,and the related indexes were analyzed and compared.Results Two groups of patients were successfully completed surgery,no complications of vascular and nerve injury.The operation time and fluoroscopy times in G arm group were less than those in C arm group (operation time:(29.6±4.5) min vs.(42.5±5.3) min,and fluoroscopy times in G arm group:(9.1±2.0) vs.(16.9±3.2));the difference was statistically significant (t =-12.747,12.870,P< 0.01).Postoperative height of injured vertebral leading edge(G arm group (22.3±5.3) mm),C arm group (22.4±5.1) mm),kyphosis Cobb angle (G arm group (9.2±3.8)°,C arm group (9.3±3.7) o),visual analogue score (G arm group (2.1±0.7)points,C arm group (2.2±0.9) points),Oswestry dysfunction index (G arm group (21.3±8.5) points,C arm group(21.5 ± 8.3)points),compared with preoperative(the hight of injured vertebral leading edge of G arm group (18.2 ±5.3) mm,C arm group (18.4±5.2) mm,Cobb angle of injured vertebra G arm group (15.7±4.4) °,C arm group (15.9±4.3) °,visual analogue score of G arm group (7.8± 1.2) points,C arm group (7.7± 1.1) points,Oswestry dysfunction index score of G arm group(41.2±8.3)points,C arm group (41.5±8.2) points),the difference was statistically significant (t =-3.709,-3.844,-7.582,-8.144,27.827,27.088,11.360,11.999,P<0.01),but there was no significant difference between the two groups (P >0.05).Conclusion Using G-arm X-ray machine to assist PKP in the treatment of thoracolumbar spine OVCF can effectively shorten the operation time,reduce the intraoperative fluoroscopy time,and the clinical effect is satisfactory.
5.Preliminary application of 3D printing navigation template in the screw insertion of the suprapubic fracture

Objective To introduce the method of using hollow pull screw insertion for the suprapubic fracture with 3D printing navigation template and to evaluate its clinical effect.Methods From March 2016 to August 2017,eighteen cases of suprapubic ramus fractures treated with hollow lag screw fixation in the Department of Orthopedics of Yanan University Affiliated Hospital were retrospectively collected was collected.Before operation,the thin slice CT scan of pelvis was used to get raw data,and the mimics17.0 software was used to reconstruction the model,to design and make the personalized navigation template assisting the suprapubic fractures surgery.The navigation template was printed out pre-operatively,and the nail was inserted via the assisted by the template intra-operatively.After the operation,the fracture reduction was evaluated by Matta imaging scores and the pelvis function was evaluated by Majeed scores.Results All the operations were successfully completed.The Matta scores were excellent in 11 cases,good in 6 cases,and fair in 1 case.The Majeed scores were excellent in 13 cases,good in 3 cases,and fair in 2 cases.Conclusion 3D Printing navigation template assisted the placement of the hollow screw on the suprapubic fracture could provide personalized treatment,reduce the difficulty of operation and satisfy the clinical effect.
6.Application of near-infrared fluorescent dyes in tumor diagnosis

Optical molecular imaging is more and more widely used in the field of biomedical sciences due to its advantageous properties such as non-invasive, real-time and high resolution. As a kind of important optical molecular imaging probe,the near-infrared fluorescent(NIRF)dyes exhibit less tissue absorption and strong tissue penetration,and has been gradually applied to the early diagnosis of tumors. Researchers have developed a number of NIRF dyes with high potential for clinical application by conjugating tumor-targeting ligands,nano-modifications and multimodal NIRF imaging, and have significantly improved the specificity and sensitivity of these NIRF dyes in tumor diagnosis. In this paper we provide a review on the application of NIRF dyes in tumor diagnosis.
7.Application of FTS and NIRF conjugated compounds in imaging and therapy of mouse tumor models

Objective To study the tumor targeting ability and application of farnesylthiosalicylic Acid (FTS) and heptamethine carbocyanine fluorescent dye-mediated near-infrared imagine in living animals, and confirm the inhibitory effect of this compound on growth of tumor cells.Methods Human breast cancer cell line MCF-7, glioma cell line U251 and prostate cancer cell line PC3 were cultured to logarithmic growth phase, and different concentrations of FTS and FTS-IR783 were added, respectively.We observed the inhibitory effect of those two compounds on the growth of tumor cells.Under fluorescence microscopy, specific accumulation of FTS-IR783 in these tumor cells was observed.The tumor cells (1×106) were transplanted subcutaneously into nude mice.These mice were subjected to intraperitoneal injection of FTS-IR783 (10 nmol/mouse) two weeks later.In the in vivo imaging, near infrared fluorescence signal and tumor volume were measured and their correlation was analyzed.Results Compared with FTS, FTS-IR783 significantly inhibited the growth of MCF-7, U251 and PC3 cells in vitro.FTS-IR783 was specifically uptaken by these three kinds of tumor cells, showing strong near infrared fluorescence in cell agglomerates.After subcutaneous injection of FTS-IR783, the correlation between fluorescence intensity and tumor volume was 0.987, 0.998 and 0.971, respectively.Conclusions The compound of FTS conjugated with near infrared fluorescent dye IR-783 can specifically recognize tumor cells, in both in vitro and in vivo imaging.At the same time, the compound can significantly inhibit the growth of tumor cells, and may be expected to become a new potential targeted drug.
8.Therapeutic Observation of Superficial Needling for Temporomandibular Joint Disorder

Objective To observe the clinical efficacy of superficial needling in treating temporomandibular joint disorder. Method Sixty patients with temporomandibular joint disorder were randomized into a treatment group and a control group, 30 in each group. The treatment group was intervened by superficial needling, and the patients were asked to repeatedly open and close mouth after the treatment; the control group was intervened by regular acupuncture. The treatment was given once a day, 10 d as a treatment course. The clinical efficacies were compared after 2 treatment courses.Result After the first treatment course, the total effective rate was 89.7% in the treatment group versus 86.2% in the control group; after the second course, the total effective rate was 96.6% in the treatment group versus 93.1% in the control group. There was a significant difference in comparing the therapeutic efficacy between the two groups after the second course (P<0.05). The relapse rate was 5.0% in the treatment group versus 33.3% in the control group, and the difference was statistically significant (P<0.05).Conclusion Superficial needling is an effective method in treating temporomandibular joint disorder.

10.Purification and activity determination of native and recombinant HBHA protein

Objective To purify native and recombinant heparin-binding hemagglutinin(HBHA)protein,and investigate the activity of HBHA polyclonal antibody against aggregation of Bacillus CalmetteGuerin(BCG)induced by HBHA.Methods After growing BCG to the stationary phase in the 7H9 liquid medium,the native HBHA protein(nHBHA)was obtained by CL-6B column chromatography.At the same time,the HBHA gene fragment was cloned and expressed by transforming Escherichia coli BL-21.Then the polyclonal antibody against rHBHA was prepared by immunizing rabbit.Different comcentration of the HBHA protein was added to the BCG liquid medium,and the aggregation of the BCG was observed.Then,add the HBHA protein that incubated with anti-HBHA antibodies to the BCG culture medium and observe the aggregation of BCG.Results The purity of native HBHA was 99% and the concentration was 1.016 mg/ml.The expressed product contained 36% of total somtic protein.After purified,the purity of the recombinant HBHA protein was 97.1% and the concentration was 10.98 mg/ml.Both the rHBHA and nHBHA could induce the aggregation of BCG.When then concentration of nHBHA is 0.2μg/ml,BCG could be induced to aggregate,while the rHBHA concentration is 2μg/ml could induce the aggregation.Both aggregations could be suppressed by the polyclonal antibody against rHBHA.Conclusions The native and recombinant HBHA are successfully obtained.It is proved that the rHBHA could induce the aggregation of BCG similar as nHBHA,and polyclonal antibody against rHBHA could also suppress the activity of nHBHA.It suggested that rHBHA could be further used in clinical diagnosis and vaccination.
