With the advent of precision medicine and personalized treatment, targeted therapies have become pivotal in oncology. Noninvasive molecular imaging, especially immunoPET/SPECT, plays a crucial role in refining cancer diagnostics and treatment monitoring by visualizing biological processes at the molecular level. This review explores the dynamic field of immunoPET/SPECT imaging using Fab and F(ab')₂ fragments, characterized by advantageous pharmacokinetics and swift clearance from the bloodstream, making them suitable for same-day imaging procedures. We examine contemporary strategies for radiolabeling these fragments with PET and SPECT radionuclides and discuss potential advancements and the challenges anticipated in the further development of Fab and F(ab')₂ fragments. Despite the complexities involved in their development, these fragments hold significant promise for advanceing personalized cancer treatment. Keys to this advancement are innovative radiolabeling techniques, site-specific conjugation chemistries, and short-lived radionuclides, all of which are crucial for overcoming existing limitations and enhancing the clinical utility of these imaging agents. As research progresses, Fab and F(ab')₂ fragments are expected to become central to the future of cancer diagnostics and therapeutic monitoring, thereby improving patient management and contributing significantly to the evolution of personalized medicine.

OBJECTIVE: This cross-sectional study assessed the methodological quality of systematic reviews (SRs) of Chinese herbal medicine (CHM) published in Chinese between Jan 2021 and Sep 2022. METHODS: Chinese language CHM SRs were identified through literature searches across 3 international and 4 Chinese databases. Methodological quality was appraised using A MeaSurement Tool to Assess systematic Reviews 2. Logistic regressions were used to explore associations between bibliographical characteristics and quality. RESULTS: Analyses of methodological quality found that among the 213 sampled SRs, 69.5% were of critically low quality, 30.5% were of low quality, and none achieved high or moderate quality. Common shortcomings included the failure to identify the studies excluded from the analysis, failure to disclose funding sources, and limited evaluation of the potential impact of bias on conclusions. Logistic regressions revealed that SRs led by corresponding authors affiliated with universities or academic institutions tended to be of lower quality than SRs led by authors affiliated with hospitals or clinical facilities. CONCLUSION Recent Chinese language CHM SRs exhibited limited methodological quality, making them unlikely to support the development of clinical practice guidelines. Urgent initiatives are needed to enhance training for researchers, peer-reviewers and editors involved in the preparation and publication of SRs. Adoption of Preferred Reporting Items for Systematic Reviews and Meta-Analyses reporting guidelines in Chinese language journals is crucial to improve the relevance of SRs for Chinese medicine development. Addressing deficiencies in methodology and reporting is essential for promoting evidence-based practices and informed clinical decisions in Chinese medicine. [摘 要] 目的：探讨补肾健脾方（BSJP）介导法尼醇X受体（FXR）抑制肝癌细胞AH-130诱导的癌性恶病质（CC）大鼠模型的作用及其机制。方法：腹腔注射AH-130细胞建立肝癌CC大鼠模型，实验分为空白对照组、模型对照组、FXR激动剂（CDCA）组、BSJP组和BSJP + CDCA组。建模后，用CDCA、BSJP连续治疗大鼠16 d，每周称量大鼠体质量。实验结束时开腹，取腹主动脉血、粪便及附睾、腹股沟和肩胛区棕色脂肪质量。采用液相色谱-质谱法（LC-MS）检测大鼠血清和粪便中胆汁酸组分及含量，WB、qPCR法检测大鼠空肠、棕色及白色脂肪组织中FXR、Wnt家族成员10b（Wnt10b）、β-连环蛋白（β-catenin）、解偶联蛋白1（UCP-1）的表达。结果：与空白对照组相比，模型对照组大鼠体质量明显降低（P < 0.01）；与模型对照组相比，CDCA、BSJP、BSJP + CDCA组大鼠体质量均增加（均P < 0.01）。与空白对照组相比，模型对照组附睾、腹股沟和肩胛区域及总棕色脂肪质量均上升（均P < 0.05）；与模型对照组相比，各治疗组大鼠附睾、腹股沟区、棕色脂肪质量均下降（均P < 0.05），而肩胛区棕色脂肪组织质量下降无差异（P > 0.05）；各治疗组总棕色脂肪质量均显著下降（P < 0.05或P < 0.01）。LC-MS分析显示，各组大鼠血清及粪便中胆汁酸组成及含量均发生改变。qPCR、WB法结果证实，与模型组相比，BSJP和BSJP + CDCA可促进大鼠回肠、棕色脂肪及白色脂肪组织中FXR mRNA和蛋白表达升高（均P < 0.05），Wnt10b、β-catenin、UCP-1 mRNA和蛋白表达均下降（均P < 0.05）。结论：BSJP能够抑制肝癌AH-130细胞诱导的大鼠CC，并减轻由此引起的体质量下降和棕色脂肪组织的形成，其机制可能涉及调控FXR并通过抑制棕色脂肪中与Wnt通路相关的Wnt10b、β-catenin和UCP-1的表达来实现。

[摘 要] 目的：探究中链脂肪酸癸酸对CD8+ T细胞活化的影响，及其对CD8+ T细胞介导的抗肿瘤免疫反应的作用和机制。方法：建立C57BL/6小鼠黑色素瘤B16F10皮下荷瘤模型，随机分为癸酸组（10 mg/kg癸酸灌胃）和对照组（等量溶剂灌胃），观察癸酸对小鼠肿瘤生长以及生存率的影响，采用流式细胞术检测肿瘤微环境中浸润CD8+ T细胞的活化水平。建立B16F10-OVA和OT-I T细胞共培养体系，采用流式细胞术检测癸酸对CD8+ T细胞的肿瘤细胞杀伤能力的影响。采用α-CD8抗体清除B16F10荷瘤小鼠体内CD8+ T细胞，观察对小鼠肿瘤体积的影响。小鼠原代CD8+ T细胞经癸酸处理后，采用WB、ELISA及qPCR、流式细胞术检测T细胞受体（TCR）活化、效应细胞因子产生以及增殖和代谢水平。在B16F10荷瘤小鼠模型中，观察α-PD-1抗体联合癸酸给药对小鼠肿瘤生长以及生存率的影响。结果：在小鼠黑色素瘤荷瘤模型中，与对照组相比，癸酸组小鼠移植瘤体积显著降低且生存率显著提高（均P<0.05），肿瘤浸润CD8+ T细胞IFN-γ和TNF-α的表达水平显著升高（P<0.01）。经癸酸处理的OT-I T细胞对B16F10-OVA细胞的杀伤水平显著升高（P<0.01）。在荷瘤小鼠模型中用α-CD8抗体清除CD8+ T细胞后，癸酸对移植瘤的抑制作用显著降低（P<0.000 1）。小鼠原代CD8+ T细胞经癸酸处理后，TCR活化水平显著升高、细胞因子IL-2和IFN-γ的产生增多、线粒体代谢水平显著上调（均P<0.05）。在黑色素瘤荷瘤小鼠模型中，癸酸与α-PD-1抗体联用，能够显著抑制小鼠移植瘤生长并提高其生存率（均P<0.05）。结论：癸酸能够促进CD8+ T细胞活化、增强其抗肿瘤免疫反应能力。

[摘 要] 目的：探讨解偶联蛋白2（UCP2）抑制剂京尼平（GEN）对人下咽癌FaDu细胞增殖及线粒体功能的影响。方法：使用不同浓度的GEN作用于FaDu细胞24 h，实验分为GEN 0（对照）、50、100、200和400 μmol/L组。采用CCK-8法检测各组细胞增殖能力，DCFH-DA探针及JC-1染色联合流式细胞术检测GEN对FaDu细胞活性氧（ROS）含量及线粒体膜电位的影响，激光共聚焦显微镜观察GEN对FaDu细胞线粒体膜通透性转换孔的影响，可见分光光度法检测细胞中乳酸的含量，WB法检测细胞中UCP2蛋白的表达变化。结果：与对照组相比，GEN可显著抑制FaDu细胞的增殖活力（P<0.05或P<0.01）、细胞中UCP2蛋白的表达（P<0.05），降低线粒体膜电位（P<0.05或P<0.01）、乳酸含量（P<0.000 1），改变细胞线粒体膜孔道通透性，提高细胞中ROS水平（P<0.05或P<0.01）。结论：GEN通过调节细胞中UCP2的表达水平进而影响细胞的氧化还原能力及线粒体功能，从而发挥抑制人下咽癌FaDu细胞增殖并诱导细胞凋亡的作用。

[摘 要] 目的：构建负载二氧化锰（MnO2）纳米颗粒的可得然（Cur）复合水凝胶MnO2@Cur（简称MGel），研究其对黑色素瘤B16-F10细胞的杀伤效果。方法：采用热诱导法制备Cur水凝胶（Gel），物理负载MnO2构建MGel，表征其宏观和微观形貌，检测其机械性能、降解性能以及光热转换性能等理化性能，并研究其联合PTT对小鼠皮肤黑色素瘤B16-F10细胞的光热杀伤效果。结果：MGel具有优异的机械和可降解性能，抗拉伸强度达（127.97±3.60）kPa、抗压缩强度达（151.44±5.23）kPa，28 d降解率约58.17%。MGel负载MnO2纳米片（粒径约180 nm）获得优异的光热转换性能，负载1.0 mg/mL MnO2的MGel在1.0 W/cm2的808 nm NIR光照4 min后到达最高温度50 ℃。细胞毒性实验和Calcein-AM/PI荧光双染色实验表明，MGel联合PTT有效杀伤B16-F10黑色素瘤细胞，NIR光照使得MGel组细胞存活率降低至（4.68±0.66）%（P<0.000 1）。结论：MGel复合水凝胶具备优异的机械性能、可降解性能以及光热转换性能，其联合PTT能有效杀伤肿瘤细胞，可能成为一种有效治疗黑色素瘤的新手段。

[摘 要] 目的：探讨山柰酚诱导人非小细胞肺癌（NSCLC）NCI-H1650细胞发生自噬及其机制。方法：常规培养NCI-H1650细胞，用不同浓度山柰酚处理细胞，用CCK-8法、MTT法以及EdU法检测山柰酚对NCI-H1650细胞增殖能力的影响，用自噬双标记腺病毒mCherry-EGFR-LC3感染实验检测山柰酚对NCI-H1650细胞发生自噬的影响，用WB法检测山柰酚处理后NCI-H1650细胞中自噬相关蛋白及Met/PI3K/Akt/mTOR信号通路相关蛋白的表达，用qPCR法检测山柰酚处理后NCI-H1650细胞中Met mRNA的表达。采用荧光素酶标记A549-luc细胞建立裸鼠移植瘤模型后用山柰酚进行处理，用活体动物成像技术观察移植瘤生长情况，用WB法检测移植瘤组织中自噬相关蛋白以及Met/PI3K/Akt/mTOR信号通路相关蛋白的表达。结果：山柰酚能显著抑制NCI-H1650细胞的增殖能力（P<0.05），山柰酚处理后NCI-H1650细胞内的自噬小体数量明显增加（P<0.05）、自噬标志蛋白LC3B和beclin1表达均明显上调（均P<0.05）、P62表达明显下调（P<0.05），山柰酚可明显抑制NCI-H1650细胞中Met mRNA和蛋白的表达（均P<0.05）、抑制p-PI3K p85、PI3K p85、p-Akt和p-mTOR蛋白表达（均P<0.05）。山柰酚抑制A549细胞裸鼠移植瘤的生长（P<0.05）和影响其体内自噬、Met/PI3K/Akt/mTOR通路相关蛋白的表达（均P<0.05）。结论：山柰酚通过影响Met/PI3K/Akt/mTOR通路诱导NSCLC NCI-H1650细胞发生自噬，进而抑制其增殖能力。

[摘 要] 目的：探讨DJ-1基因过表达对人胃癌MGC803细胞增殖、迁移、侵袭与上皮间质转化（EMT）的影响及其机制。方法：利用基因转染技术构建DJ-1基因过表达MGC803细胞，实验分为MGC803、空载体和DJ-1过表达组。采用MTT、平板克隆形成、细胞划痕和Transwell实验分别检测DJ-1过表达对MGC803细胞增殖、克隆形成、迁移与侵袭的影响；qPCR和WB法检测DJ-1过表达对各组细胞DJ-1、PTEN、Akt、p-Akt、Snail、vimentin、E-cadherin、MMP-9与TIMP-3表达的影响，相差显微镜下观察MGC803细胞形态学的变化。裸鼠荷瘤实验检测DJ-1过表达对MGC803细胞移植瘤体内生长的影响。结果：成功构建DJ-1基因稳定过表达的MGC803细胞。与MGC803组和空载体组比较，DJ-1过表达组细胞的增殖能力与克隆形成数均显著增加（均P<0.05），细胞迁移距离明显增加、划痕距离明显缩短（均P<0.05），迁移与侵袭细胞数显著增多（均P<0.05），DJ-1 mRNA与蛋白表达明显上调、PTEN mRNA与蛋白表达下调（均P<0.05），Akt总蛋白各组比较无明显差异（均P>0.05），p-Akt蛋白表达明显上调（P<0.05），Snail、vimentin与MMP-9表达上调、E-cadherin与TIMP-3表达下调（均P<0.05）。相差显微镜下见长梭形细胞数目增多，圆形与椭圆形细胞减少，异型性更为明显。荷瘤裸鼠体内实验结果表明，与MGC803组相比较，DJ-1过表达组MGC803细胞移植瘤生长速度明显加快、移植瘤质量显著增加（均P<0.05）。结论：DJ-1过表达可通过PTEN/Akt通路在体内外抑制MGC803细胞的增殖、迁移、侵袭与EMT。

Radical gastrectomy with D2 lymphadenectomy has been widely performed as the standard surgery for patients with gastric cancer in major medical centers in China and abroad. However, the exact extent of lymph node dissection is still controversial. In the latest version of the Japanese Gastric Cancer Treatment Guidelines, No. 14v lymph nodes (along the root of the superior mesenteric vein) are again defined as loco-regional lymph nodes, and it is clarified that distal gastric cancer presenting with infra-pyloric regional lymph node (No.6) metastasis is recommended for D2+ superior mesenteric vein (No. 14v) lymph node dissection. To explore the relevance and clinical significance of No.6 and No.14v lymphadenectomy in radical gastric cancer surgery, a review of the national and international literature revealed that No.6 lymph node metastasis was associated with No.14v lymph node metastasis, that No.6 lymph node status was a valid predictor of No.14v lymph node negative status and false negative rate, and that for gastric cancer patients with No. 14v lymph node negative and No.6 lymph node positive, the dissection of No.14v lymph node may also have some significance. The addition of No. 14v lymph node dissection in radical gastrectomy is safe, but it is more important to distinguish the patients who can benefit from it. Professor Liang Han of Tianjin Medical University Cancer Hospital is currently leading a multicenter, large-sample, prospective clinical trial (NCT02272894) in China, which is expected to provide higher level evidence for the clinical significance of lymph node dissection in No.14v.
