1.Network toxicology and its application in studying exogenous chemical toxicity

		                        			With the continuous development of society, a large number of new chemicals are continuously emerging, which presents a challenge to current risk assessment and safety management of chemicals. Traditional toxicology research methods have certain limitations in quickly, efficiently, and accurately assessing the toxicity of many chemicals, and cannot meet the actual needs. In response to this challenge, computational toxicology that use mathematical and computer models to achieve the prediction of chemical toxicity has emerged. In the meantime, as researchers increasingly pay attention to understanding the interaction mechanisms between exogenous chemical substances and the body from the system level, and multiomics technologies develop rapidly such as genomics, transcriptomics, proteomics, and metabolomics, huge amounts of data have been generated, providing rich information resources for studying the interactions between chemical substances and biological molecules. System toxicology and network toxicology have also developed accordingly. Of these, network toxicology can integrate these multiomics data to construct biomolecular networks, and then quickly predict the key toxicological targets and pathways of chemicals at the molecular level. This paper outlined the concept and development of network toxicology, summarized the main methods and supporting tools of network toxicology research, expounded the application status of network toxicology in studying potential toxicity of exogenous chemicals such as agricultural chemicals, environmental pollutants, industrial chemicals, and foodborne chemicals, and analyzed the development prospects and limitations of network toxicology research. 2.Research Progress of Genes Related to Congenital Lipodystrophy

Congenital lipodystrophy (CL) is a group of genetic disorders characterized by an extreme deficiency of fat tissue in the body. The core defect of this disease is adipose metabolism disorder and is associated with a variety of metabolic problems, such as insulin resistance, diabetes, hypertriglyceridemia, and hepatic pathological changes. Over the past few decades, the rapid development of genomics and molecular biology has significantly advanced the identification and functional study of genes related to CL. This article aims to review the recent progress in the identification and function of CL-related genes, deepen the understanding of its pathological mechanisms, and provide references for future treatment strategies.
3.Establishment and identification of a Trappc11 inducible knockout mouse model

		                        			Objective To establish an inducible knockout mouse model of trafficking protein particle complex subunit 11 (Trappc11). Methods and results LoxP sites were introduced on both sides of exon 3-5 of Trappc11,and then the CRISPR/Cas9 technique was used to establish F0 C57BL/6J mice. The positive F0 generation mice were identified by polymerase chain reaction amplification and sequencing. After that,F0 positive mice were mated with C57BL/6J wild type mice to obtain F1 Trappc11flox/+mice. And then,Trappc11flox/+mice were mated with UBC-CreERT2 mice,and finally Trappc11 inducible systemic knockout mouse model was obtained after 2 generations. Conclusion The Trappc11 inducible knockout mouse model is established using CRISPR/Cas9 and Cre-loxP,providing an important tool for revealing the pathophysiological role of Trappc11 in multi-organ system diseases.
		                        		
		                        		
		                        		
		                        	
4.Modified Qianghuo Shengshitang Combined with Ozone Injection and Radiofrequency Thermocoagulation Target Puncture in Treatment of Cervical Spondylotic Radiculopathy with Wind-cold Blockage Type

		                        			ObjectiveTo observe the clinical efficacy of Qianghuo Shengshitang combined with ozone injection and radiofrequency thermocoagulation target puncture in the treatment of cervical spondylotic radiculopathy (wind-cold blockage type). MethodSixty-eight patients with cervical spondylotic radiculopathy who were treated in Henan Province Hospital of Traditional Chinese Medicine from August 2020 to May 2021 were included and grouped into a control group and a research group by random number table, with 34 patients in each group. The control group was administrated with placebo granules of Qianghuo Shengshitang, and treated with ozone injection and radiofrequency thermocoagulation target puncture at the same time. The research group was given modified Qianghuo Shengshitang combined with ozone injection and radiofrequency thermocoagulation target puncture. Before and after treatment, patients in two groups were tested for the levels of tumor necrosis factor-α (TNF-α), procalcitonin (PCT), interleukin-2 (IL-2), interleukin-1β (IL-1β), 6-ketoprostaglandin E1α (6-keto-PGE1α), plasma substance P (SP), β-endorphin (β-EP), lipid peroxide (LPO), malondialdehyde (MDA), and superoxide dismutase (SOD). The pain visual scale (VAS), cervical dysfunction index (NDI), and clinical evaluation scale for cervical spondylosis (CASCS) were given to patients to evaluate the clinical efficacy. ResultCompared with those before treatment, the levels of IL-1β, PCT, and TNF-α in two groups were decreased, and the level of IL-2 was increased (P<0.05). The improvement of IL-1β, PCT, TNF-α, and IL-2 was more obvious in the research group as compared with the control group (P<0.05). After treatment, the levels of SOD in two groups were increased, while the levels of LPO and MDA were decreased (P<0.05). After treatment, the improvement of SOD, LPO, and MDA was more obvious in the research group as compared with the control group (P<0.05). Compared with those before treatment, the levels of SP and 6-keto-PGE1α in two groups were decreased after treatment, and the level of β-EP was increased (P<0.05). The improvement of -keto-PGE1α and β-EP was more obvious in the research group as compared with the control group after treatment (P<0.05). Compared with those before treatment, the VAS and NDI scores of the research group were decreased, and the CASCS scores were increased (P<0.05). After treatment, the improvement of VAS, NDI, and CASCS of the research group was more obvious as compared with the control group (P<0.05). After treatment, the total effective rate of patients in the research group was higher than that in the control group (Z=2.018, P<0.05). ConclusionModified Qianghuo Shengshitang combined with ozone injection and radiofrequency thermocoagulation target puncture in the treatment of patients with cervical spondylotic radiculopathy (wind-cold blockage type) can inhibit inflammation, remove oxygen free radicals, improve the level of pain mediators, relieve pain, and improve cervical spine function and clinical efficacy. 
		                        		
		                        		
		                        		
		                        	
5.miR-184 promotes compensatory lung growth via TIMP-2/MMP-14 in pneumonectomy model

		                        			AIM:To explore the effect of microRNA-184(miR-184)on compensatory lung growth(CLG)af-ter lobectomy in multiple primary lung cancer(MPLC)and its mechanism.METHODS:(1)Lung tissue samples(n= 16)from MPLC patients and patients with good recovery after lobectomy(CLG)were collected,and the expression of miR-184 was measured by RT-qPCR.(2)Human alveolar epithelial cells were divided into NC-mimic group,miR-184 mimic group,OE-NC group,tissue inhibitor of metalloproteinase-2(TIMP-2)overexpression(OE-TIMP-2)group,and miR-184 mimic+OE-TIMP-2 group according to the transfection(n=3).The expression of miR-184,TIMP-2 mRNA and matrix metalloproteinase-14(MMP-14)mRNA was measured by RT-qPCR,and the protein expression of TIMP-2 and MMP-14 was determined by Western blot.The proliferation of the cells was measured by CCK-8 and colony formation assays.(3)C57BL/6J mice were divided into pneumonectomy(PNX)group and PNX+miR-184 mimic group(n=5).The flexiVent system was used to measure the vital capacity and lung compliance of the mice.Lung volume was measured by water dis-placement method,and lung tissue changes were observed by HE staining.RESULTS:The expression of miR-184 was significantly higher in the patients with better recovery after lobectomy(P<0.01).Overexpression of miR-184 promoted the proliferation of human alveolar epithelial cells and the recovery of lung function in mice after PNX.In terms of mecha-nism,miR-184 showed targeted binding with TIMP-2,and overexpression of miR-184 promoted the expression of MMP-14 by inhibiting TIMP-2,thereby promoting the proliferation of human alveolar epithelial cells and the recovery of mouse lung function after PNX.CONCLUSION:miR-184 promotes CLG after PNX through the TIMP-2/MMP-14 axis.
		                        		
		                        		
		                        		
		                        	
6.Analysis of influencing factors of death of epidemic Japanese encephalitis cases in Longnan City, Gansu Province from 2014 to 2018

		                        			Objective:To understand the influencing factors of death of epidemic Japanese encephalitis (EJE) cases in Longnan City of Gansu Province.Methods:In the EJE Monitoring Information Report Management System of the Chinese Disease Prevention and Control Information System, data on EJE cases with onset from 2014 to 2018 and current address in Longnan City were derived. An "Individual Questionnaire of Epidemic Japanese Encephalitis in Longnan City" was designed, retrospective study was conducted on enrolled cases, their information on demographic data, consultation, onset, clinical classification, and chronic underlying diseases were collected, characteristics of EJE cases and death-related factors were analyzed.Results:From 2014 to 2018, a total of 260 EJE cases were reported in Longnan City, and 259 cases completed the questionnaire. Among them, 70 cases (27.0%) were aged ≥60 years old, 67 cases (25.9%) were severe and extremely severe, and 55 cases (21.2%) had chronic underlying diseases. Among 259 EJE cases, 46 cases died, with a fatality rate of 17.8%. After multivariate unconditional logistic regression analysis, age ≥60 years old [odds ratio ( OR)=2.667, 95% confidence interval ( CI): 1.140-6.237], severe and extremely severe ( OR = 2.762, 61.820, 95% CI: 1.053-7.091, 5.149-742.239), and chronic underlying diseases ( OR = 2.489, 95% CI: 1.038-5.964) were risk factors for death in EJE cases. Conclusions:The influencing factors of death of EJE cases in Longnan City are age, clinical classification and chronic underlying diseases. Therefore, we should focus on patients over 60 years old, clinically classified as severe or extremely severe, and suffering from chronic underlying diseases, and strengthen the immunization of EJE vaccine for key populations.
		                        		
		                        		
		                        		
		                        	
7.Application of self fixing mesh with " Y-shaped" placement in laparoscopic transabdominal preperitoneal repair of inguinal hernia of female inguinal hernia

		                        			Objective:To investigate the effect of self fixing mesh with " Y-shaped" placement in laparoscopic transabdominal preperitoneal repair of female inguinal hernia.Methods:From February 2019 to September 2020, a total of 40 female patients who underwent TAPP (Keyhole method) in Shaanxi Provincial People′s Hospital were researched.Random number table method was used.According to different patch placement methods, the patients were divided into the self fixing mesh with " Y-shaped" placement group (21 cases) and with normal placement group (19 cases). The operative time, placement time, length of stay, Visual Analogue Scale (VAS), postoperative complication were compared between two groups.Results:All 40 patients completed TAPP, and all patients were followed up successfully, with a median follow-up time of 12.5 months.The operation time and patch placement time were (54.9±9.0) min and (10.8±2.5) min in the Y-shaped placement method group and (62.8±11.2) min and (20.3±3.5) min in the conventional placement method group, respectively; the differences between the two groups were statistically significant ( t=2.490, P=0.017; t=.925, P<0.001). There was no statistically significant difference between the two groups in length of stay, VAS score 1 day after surgery and postoperative complications(all P>0.05). Conclusion:In laparoscopic female inguinal hernia repair (keyhole method), the " Y" shape placement method can effectively shorten the time of self fixing patch placement, thus shorten the operation time, and does not increase the risk of complications.

Early detection, diagnosis, treatment response and prognosis can significantly improve patients' quality of life. The development of non-invasive liquid biopsy represents a major innovation in the field of precision medicine, opening up a new era for precision medicine and individualized treatment of cancer. Circulating biomarkers have shown a great potential utility in the diagnosis, treatment response, and prognosis of urinary tumors. 10.Analysis of prognosis of pulmonary adenocarcinoma with a micropapillary pattern and its influencing factors

		                        			Objective To analyze the prognostic influencing factors of pulmonary adenocarcinoma with a micropapillary pattern (MPPAC).Methods A total of 109 MPPAC patients who received surgical operation in Shandong Cancer Hospital Affiliated to Shandong University from August 2012 to August 2015 were retrospectively analyzed.The median survival time and the survival rate of 1-,3-and 5-year were calculated by using Kaplan-Meier method,log-rank test was used for single-factor analysis and Cox regression analysis was used for multiple-factor analysis.Results The median overall survival time of 109 MPPAC patients was 55.0 months (3-67 months).The overall survival rate of 1-,3-and 5-year was 89.0%,61.5%,48.6%,respectively.Single factor analysis showed that the gender (x2 =7.208,P =0.007),the tumor size (x2 =24.083,P < 0.01),lymph node metastasis (x2 =23.068,P < 0.01),vascular tumor thrombosis (x2 =16.411,P < 0.01),visceral pleural infiltration (x2 =18.438,P < 0.01) and multiple tumors (x2 =28.563,P <0.01) were associated with the overall survival of MPPAC patients.Multiple factor analysis showed that the tumor size (RR =1.629,95% CI 1.145-2.317,P =0.007),lymph node metastasis (RR =1.680,95% CI 1.161-2.430,P =0.006) and vascular tumor thrombosis (RR =2.867,95% CI 1.286-6.392,P =0.010) were the independent prognosis factors for MPPAC patients.Conclusion The MPPAC patients have a poor prognosis.The tumor size,lymph node metastasis and vascular tumor thrombosis could influence the prognosis of MPPAC patients.
