1.Research Progress of Genes Related to Congenital Lipodystrophy

Congenital lipodystrophy (CL) is a group of genetic disorders characterized by an extreme deficiency of fat tissue in the body. The core defect of this disease is adipose metabolism disorder and is associated with a variety of metabolic problems, such as insulin resistance, diabetes, hypertriglyceridemia, and hepatic pathological changes. Over the past few decades, the rapid development of genomics and molecular biology has significantly advanced the identification and functional study of genes related to CL. This article aims to review the recent progress in the identification and function of CL-related genes, deepen the understanding of its pathological mechanisms, and provide references for future treatment strategies.
2.Mechanical thrombectomy via carotid artery salvage puncture for acute anterior circulation large vessel occlusive stroke: a clinical analysis of 8 cases

Objective:To investigate the efficacy and feasibility of mechanical thrombectomy (MT) via carotid artery salvage puncture in patients with acute anterior circulation large vessel occlusive stroke. Methods:A retrospective analysis was performed. Eight patients with acute anterior circulation large vessel occlusive stroke underwent MT via carotid artery salvage puncture in Departments of Neurology, First People's Hospital of Xianyang, Xi'an Ninth Hospital, Xi'an Daxing Hospital and XD Group Hospital from June 2021 to September 2023 were enrolled. Occlusion location, time from onset to femoral artery puncture, causes of carotid artery salvage puncture, time from femoral artery puncture to carotid artery salvage puncture, time from successful carotid artery puncture to vascular recanalization, modified thrombolysis in cerebral infarction (mTICI) score immediately after MT, and scores of National Institutes of Health Stroke Scale (NIHSS) 24 h after MT and modified Rankin Scale (mRS) 90 d after MT were analyzed. Results:Of the 8 patients, 7 had M1 segment occlusion and 1 patient had M2 segment occlusion. Direct thrombectomy was not possible resulting from type III aortic arch in 3 patients, aortic arch replacement in 1 patient, right common carotid artery twisting angle in 1 patient, left common carotid artery twisting angle in 2 patients, and bilateral femoral artery occlusion in 1 patient. All 8 patients had successful carotid artery puncture under local anesthesia, including 7 with mTICI 3 and 1 patient with mTICI 2b. Average time from successful carotid artery puncture to vascular recanalization was about 35 min, ranged 10-90 min. All patients had decreased NIHSS score 24 h after MT compared with before MT, and no complications such as hematoma or airway compression occurred in carotid artery puncture site. Six patients had mRS scores of 0-2 and 2 had scores of 6 (death cause: pulmonary infection) 90 d after surgery.Conclusion:For patients with patients with acute anterior circulation large vessel occlusive stroke, MT via carotid artery salvage puncture is a safe and feasible method in cases of difficulty in establishing thrombectomy route via femoral artery such as type III aortic arch, common carotid artery twisting angle, abdominal aorta occlusion.
3.Establishment and identification of a Trappc11 inducible knockout mouse model

Objective To establish an inducible knockout mouse model of trafficking protein particle complex subunit 11 (Trappc11). Methods and results LoxP sites were introduced on both sides of exon 3-5 of Trappc11,and then the CRISPR/Cas9 technique was used to establish F0 C57BL/6J mice. The positive F0 generation mice were identified by polymerase chain reaction amplification and sequencing. After that,F0 positive mice were mated with C57BL/6J wild type mice to obtain F1 Trappc11flox/+mice. And then,Trappc11flox/+mice were mated with UBC-CreERT2 mice,and finally Trappc11 inducible systemic knockout mouse model was obtained after 2 generations. Conclusion The Trappc11 inducible knockout mouse model is established using CRISPR/Cas9 and Cre-loxP,providing an important tool for revealing the pathophysiological role of Trappc11 in multi-organ system diseases.
4.Modified Qianghuo Shengshitang Combined with Ozone Injection and Radiofrequency Thermocoagulation Target Puncture in Treatment of Cervical Spondylotic Radiculopathy with Wind-cold Blockage Type

ObjectiveTo observe the clinical efficacy of Qianghuo Shengshitang combined with ozone injection and radiofrequency thermocoagulation target puncture in the treatment of cervical spondylotic radiculopathy (wind-cold blockage type). MethodSixty-eight patients with cervical spondylotic radiculopathy who were treated in Henan Province Hospital of Traditional Chinese Medicine from August 2020 to May 2021 were included and grouped into a control group and a research group by random number table, with 34 patients in each group. The control group was administrated with placebo granules of Qianghuo Shengshitang, and treated with ozone injection and radiofrequency thermocoagulation target puncture at the same time. The research group was given modified Qianghuo Shengshitang combined with ozone injection and radiofrequency thermocoagulation target puncture. Before and after treatment, patients in two groups were tested for the levels of tumor necrosis factor-α (TNF-α), procalcitonin (PCT), interleukin-2 (IL-2), interleukin-1β (IL-1β), 6-ketoprostaglandin E1α (6-keto-PGE1α), plasma substance P (SP), β-endorphin (β-EP), lipid peroxide (LPO), malondialdehyde (MDA), and superoxide dismutase (SOD). The pain visual scale (VAS), cervical dysfunction index (NDI), and clinical evaluation scale for cervical spondylosis (CASCS) were given to patients to evaluate the clinical efficacy. ResultCompared with those before treatment, the levels of IL-1β, PCT, and TNF-α in two groups were decreased, and the level of IL-2 was increased (P<0.05). The improvement of IL-1β, PCT, TNF-α, and IL-2 was more obvious in the research group as compared with the control group (P<0.05). After treatment, the levels of SOD in two groups were increased, while the levels of LPO and MDA were decreased (P<0.05). After treatment, the improvement of SOD, LPO, and MDA was more obvious in the research group as compared with the control group (P<0.05). Compared with those before treatment, the levels of SP and 6-keto-PGE1α in two groups were decreased after treatment, and the level of β-EP was increased (P<0.05). The improvement of -keto-PGE1α and β-EP was more obvious in the research group as compared with the control group after treatment (P<0.05). Compared with those before treatment, the VAS and NDI scores of the research group were decreased, and the CASCS scores were increased (P<0.05). After treatment, the improvement of VAS, NDI, and CASCS of the research group was more obvious as compared with the control group (P<0.05). After treatment, the total effective rate of patients in the research group was higher than that in the control group (Z=2.018, P<0.05). ConclusionModified Qianghuo Shengshitang combined with ozone injection and radiofrequency thermocoagulation target puncture in the treatment of patients with cervical spondylotic radiculopathy (wind-cold blockage type) can inhibit inflammation, remove oxygen free radicals, improve the level of pain mediators, relieve pain, and improve cervical spine function and clinical efficacy.
5.miR-184 promotes compensatory lung growth via TIMP-2/MMP-14 in pneumonectomy model

AIM:To explore the effect of microRNA-184(miR-184)on compensatory lung growth(CLG)af-ter lobectomy in multiple primary lung cancer(MPLC)and its mechanism.METHODS:(1)Lung tissue samples(n= 16)from MPLC patients and patients with good recovery after lobectomy(CLG)were collected,and the expression of miR-184 was measured by RT-qPCR.(2)Human alveolar epithelial cells were divided into NC-mimic group,miR-184 mimic group,OE-NC group,tissue inhibitor of metalloproteinase-2(TIMP-2)overexpression(OE-TIMP-2)group,and miR-184 mimic+OE-TIMP-2 group according to the transfection(n=3).The expression of miR-184,TIMP-2 mRNA and matrix metalloproteinase-14(MMP-14)mRNA was measured by RT-qPCR,and the protein expression of TIMP-2 and MMP-14 was determined by Western blot.The proliferation of the cells was measured by CCK-8 and colony formation assays.(3)C57BL/6J mice were divided into pneumonectomy(PNX)group and PNX+miR-184 mimic group(n=5).The flexiVent system was used to measure the vital capacity and lung compliance of the mice.Lung volume was measured by water dis-placement method,and lung tissue changes were observed by HE staining.RESULTS:The expression of miR-184 was significantly higher in the patients with better recovery after lobectomy(P<0.01).Overexpression of miR-184 promoted the proliferation of human alveolar epithelial cells and the recovery of lung function in mice after PNX.In terms of mecha-nism,miR-184 showed targeted binding with TIMP-2,and overexpression of miR-184 promoted the expression of MMP-14 by inhibiting TIMP-2,thereby promoting the proliferation of human alveolar epithelial cells and the recovery of mouse lung function after PNX.CONCLUSION:miR-184 promotes CLG after PNX through the TIMP-2/MMP-14 axis.
6.Comparison of the effects of Wiltse approach and minimally invasive percutaneous approach in the treatment of thoracolumbar fracture patients without neurological deficit

【Objective】 To compare the clinical efficacy of minimally invasive percutaneous approach with Wiltse approach in the treatment of thoracolumbar fracture without neurological deficit in young and middle-aged patients. 【Methods】 A prospective study was conducted in 108 patients with thoracolumbar fracture without neurological symptoms treated in Department of Orthopedics, The First Affiliated Hospital of Xi’an Jiaotong University from March 2015 to March 2018. We randomly assigned 54 patients to minimally invasive percutaneous approach group (Group A) and 54 ones to Wiltse approach group (Group B). We compared the operation time, intraoperative blood loss, incision length, the number of intraoperative fluoroscopy times, and postoperative hospital stay of Group A and Group B. We also compared the VAS scores of the two groups before and 3 days after surgery and at each review, the vertebral frontal height compression ratio and Cobb Angle before and immediately after surgery, 3 months 1 year and 3 years after surgery, and ODI index before surgery and 3 months, 1 year and 3 years after surgery. 【Results】 All the 108 patients were followed up. There were no significant differences in operation time, intraoperative blood loss or postoperative hospital stay between the two groups (P>0.05). The number of intraoperative fluoroscopy times was significantly smaller in Group B than in Group A (P<0.05), and the incision length was shorter than that in Group A (P<0.05). The two groups did not significantly differ in VAS score or ODI index before operation (P>0.05), after operation, or at follow-up, while the anterior vertebral body height ratio and Cobb angle were significantly lower in Group B than in Group A (P<0.0125). 【Conclusion】 Minimally invasive percutaneous approach and Wiltse approach are both safe and effective in the treatment of thoracolumbar fracture without neurological deficit. However, the number of intraoperative fluoroscopy times of Wiltse approach was significantly reduced, the incision length was smaller than that of the percutaneous pedicle group, and the postoperative anterior vertebral compression rate and Cobb Angle were lower than those of the percutaneous group, indicating better clinical efficacy.
7.Characteristics and Clinical Application of Commonly Used Wound Dressings.

The pathological mechanism of wound healing is complicated and affected by multiple factors. Modern wound dressings are widely used in the clinical management of wound healing and have achieved good therapeutic effects. Clinically, wounds are often caused by different etiologies. However, there are few reviews focus on the selection of reasonable dressings for different types of wounds. This study mainly focuses on the characteristics of commonly used wound dressings and summarizes the characteristics of the most commonly used wound dressings in clinical practice and their effects. The advantages and disadvantages of pathology wounds: diabetic foot ulcers, pressure injuries, burns, and leg ulcers are reviewed. This study aims to provide references for the development and clinical selection of wound dressings for scientific researchers and first-line nursing staff who are engaged in wound dressings.

8.Parameters optimization in Heparin affinity chromatography purification of human coagulation factor Ⅸ

【Objective】 To optimize the purification conditions of heparin affinity chromatography in the purification of human coagulation factor Ⅸ by response surface method and establish the optimal chromatography process parameters. 【Methods】 The effect of sample loading temperature on purification efficiency was analyzed through single factor test. Three-factor three-level response surface method was used to optimize the chromatographic elution conditions. The Folin phenol method and the automatic hemagglutination analyzer were used to determine the total protein content and human coagulation factor Ⅸ titer, respectively. The purification effect was evaluated by activity index and process recovery rate. 【Results】 The optimized optimal chromatographic conditions were loading at 5 ℃, washing 4 CV, eluent formulation of sodium citrate 20 mmol/L, arginine hydrochloride 18.7 mmol/L, NaCl 611.6 mmol/L and pH 7.5; under this optimal setting, the recovery rate of the chromatographic process was (46.6±2.9) %, titer of factor Ⅸ rated to (68.4±4.7) IU/mL and specific activity was (62.8±3.3) IU/mg. 【Conclusion】 The optimized parameters of heparin affinity chromatography process by response surface method can produce better purification effect on human coagulation factor Ⅸ.

10.Progress of liquid biopsy in urinary neoplams

Urinary malignancy is a disease with high morbidity and mortality. Early detection, diagnosis, treatment response and prognosis can significantly improve patients' quality of life. The development of non-invasive liquid biopsy represents a major innovation in the field of precision medicine, opening up a new era for precision medicine and individualized treatment of cancer. Circulating biomarkers have shown a great potential utility in the diagnosis, treatment response, and prognosis of urinary tumors. This paper reviews the recent progress of liquid biopsy in urinary neoplasms.
