1.Nutritional Status of Children Aged 3 to 7 Years in Zhag′yab, Tibet

ObjectiveTo understand the physical development level of 3-7 years old children in Zhag′yab, and to provide reference for local health decision-making. MethodsA cross-sectional study was conducted between June 2021 and July 2021, with a sample of 1 247 Tibetan children aged 3-7 years from kindergartens in 13 districts of Zhag′yab. Their height and weight were measured and the hemoglobin was detected by a unified method. Standard statistical method was adopted (Z-score method). Z-scores of length /height-for-age (HAZ), Z-scores of weight-for-age (WAZ), Z-scores of body mass index (BMI)-for-age (BAZ) and Z-scores of weight-for-height (WHZ) were calculated by WHO Anthro v3.2.2 and WHO Anthro Plus. The nutritional status of children was evaluated according to WHO diagnostic criteria for malnutrition and anemia. ResultsThe average WAZ and HAZ of children aged 3 to 7 in Zhag′yab were lower than the WHO standards,except for the WAZ of 4-year old, the differences were statistically significant (P<0.05). The overall detection rate of malnutrition was 25.7%, stunting, underweight, wasting, overweight, obesity and anemia were 11.6%, 11.8%, 10.8%, 3.3%, 1.8% and 29.3%, respectively. The detection rates of all indicators in boys were higher than those in girls, and the differences were not statistically significant (P>0.05). The overweight rate and obesity rate of rural children were lower than those of urban children, and the other detection rates were higher than those of urban children. The differences of underweight rate, obesity rate and anemia rate were statistically significant between urban and rural children (P<0.05). Among the detection rates of all indicators in different age groups, there were statistically significant differences in the overweight rate and the anemia rate (P<0.05). The overweight rate of children aged 4 and the anemia rate of children aged 5 were the highest. ConclusionsThe physical development of children aged 3 to 7 in Zhag′yab is poor, and the prevalence of malnutrition and anemia is high. Underweight and anemia are more serious in rural children, and the overweight and obesity problem of urban children is emerging. More attention should be paid to promote their nutritional status. The prevention and intervention of children’s malnutrition should be strengthened in Zhag′yab.
2. Bioinformatics analysis on structure and function and expression vector construction of SARS-CoV-2 related protein TMPRSS2

Aim Human TMPRSS2 is a transmembrane serine protease.In this paper, the structure and func¬tion of the protein were systematically analyzed by bioinformatics, the codon was optimized and the pro- karvotie expression vector was constructed to explore the molecular mechanism of SARS-CoV-2 infecting host cells.Methods The recombinant expression vector pET-22b-TMPRSS2 was generated by molecular clo¬ning technology.The homology, functional sites, sub¬cellular localization, three-dimensional structure and evolutionary characteristics of TMPRSS2 protein were systematically analyzed by using analytical tools such as Protparam, NetPhos3.1, Blast, Clustal X2 and MEGA7.0.Results The prokarvotic expression plas- mid was constructed correctly; TMPRSS2 belongs to medium molecular weight protein, which is composed of 492 amino acid residues.The theoretical isoelectric point is 8.12, the molecular extinction coefficient is 118 145 L • mol~1 • cm"1 , and the half-life is 30 h; TMPRSS2 has 15 potential glycosylation sites and 49 possible phosphorylation sites.It is a transmembrane hydrophilie protein without signal sequenee.In addi¬tion, the protein has 13 potential B-cell epitopes and 7 T-eell epitopes.Seeondarv structure analysis showed that random coil accounted for the highest proportion of TMPRSS2 protein ( 0.453 3) , followed by extended strand (0.252 0).Sequence comparison and evolu¬tionary analysis showed that the highest sequence con¬sistency and closest genetic relationship with human TMPRSS2 was Pan troglodytes, followed by gorilla.Conclusions Human-derived TMPRSS2 protein is ev- olutionarilv conserved and functionally important.Hie results of this study can help to reveal the structure and mechanism of action of TMPRSS2 protein, provide ide¬as for the diagnosis and treatment of COYID-19, and accelerate the research and development process of new drugs targeting TMPRSS2 protein.
3.Early-Life Nutrition Intervention Influences Pubertal Development in Female Rats

ObjectiveTo explore the influence of early-life nutrition intervention on pubertal development in female rats. MethodsFifty-eight neonatal female rats born small for gestational age (SGA) by using maternal food restriction were randomly divided into postnatal over-nutrition group (Group IS, n=28) and postnatal normal diet group (Group IC, n=30). Another 58 neonatal female rats born appropriate for gestational age (AGA) were divided into postnatal over-nutrition group (Group CS, n=28) and postnatal normal diet group (Group CC, n=30). Then we observed and measured the vaginal opening (VO), sex hormone levels, organ coefficients of uterus and ovary, and numbers of corpora lutea at various stages of each group. ResultsThe VO in Group IS and Group CS was accelerated than that in Group IC and Group CC. The levels of luteinizing hormone (LH) on postnatal day 35 in Group IS and Group CS were higher than that of Group CC. The levels of estradiol and LH on postnatal day 35 were higher than those on postnatal day 21 in Group IS, Group CS and Group CC. The organ coefficients of uterus and ovary on postnatal day 35 were higher than those on postnatal day 21 in each group. The numbers of corpora lutea on postnatal day 35 and day 45 were higher in Group IS and Group CS than those in Group CC. ConclusionsPostnatal normal diet may help most SGA female rats catch up their growth and normal pubertal development, while postnatal over-nutrition promotes precocious puberty in either SGA or AGA female rats.
4. Inhibition of T lymphocytes secreting EphrinAl-Caspase-3 on proliferation of tumor tissue in nude mice with breast cancer

Objective To study the inhibitory effect of T lymphocytes secreting EphrinAl-Caspase-3 in vivo and on the growth of cancer cells in nude mice with breast cancer. Methods Nude mice (n = 35) were inoculated with breast cancer cells to construct a nude mouse model of breast cancer. When the tumor volume reached 0. 1 cm3, 30 nude mice with average size tumor tissue were randomly divided into PBS group, uninfected adenovirus group, T lymphocyte infected with Ad-EphrinAl-Caspase-3 group, and intratumoral transplantation. Tumor size was measured every day 2 to 3. Three groups of tumor-bearing nude mice were selected. After the above-mentioned cell transplantation, the subcutaneous tumor tissue homogenate was obtained every day 2 to 3, and the content of EphrinAl-Caspase-3 was detected by ELISA. At the end of the experiment, the animals in each group were sacrificed by cervical dissection and sliced. The presence of T lymphocytes expressing green fluorescent protein was observed under a fluorescence microscope, and Caspase-3 and Ki-67 were detected by immunofluorescence. Results After one week of inoculation of breast cancer cells into nude mice, the presence of subcutaneous tumors could be touched by hand, which proved that the tumor-bearing animals of breast cancer cells were successfully modeled. On the 8th day after inoculation, the tumor volume of the nude mice in each group became larger, and the difference between the treatment group and the PBS group/T lymphocyte group was extremely significant ( P<0.05). Although the tumor volume of the T lymphocyte transplantation group was slower than that of the PBS control group, there was no statistically significant difference between the two. The expression of EphrinAl-Caspase-3 was detected in the EphrinAl-Caspase-3 treatment group on the 2nd day, reached the peak on the 8th day, and then the secretion decreased gradually. No expression of EphrinAl-Caspase-3 was detected in the PBS control group and the T lymphocyte group. The presence of dispersed green fluorescent protein-labeled EphrinAl-Caspase-3-T lymphocytes was observed in the tumor tissues of the treatment group, while the presence of green fluorescent protein was not detected in the PBS group and the T lymphocyte groups. In the infected cells of the treatment group, the proportion of Caspase-3 positive cells was up- regulated, and the proportion of Ki-67 positive cells was down-regulated. No expression of EphrinAl-Caspase-3 was detected in the PBS group and the T lymphocyte group. Conclusion EphrinAl-Caspase-3 can significantly inhibit the growth of breast cancer cells, thereby exerting an anti-tumor effect.
5.Effects of Early Life Nutritional Status on Weight Catch-up Growth,Puberty Initiation and Obesity in Female Rats with Intrauterine Growth Retardation

【Objective】To investigate the effects of different nutritional status in early life on weight catch-up,puberty initiation and obesity in rats with intrauterine growth retardation(IUGR).【Methods】IUGR model of newborn rats was established by limiting diet during pregnancy in SPF rats. Female newborn rats were used in the experiment. IUGR rats were divided into two groups:small litter IUGR group(SL-IUGR)and common feeding IUGR group(CF-IUGR). Rats of normal birth weight served as normal control group(NC). The SL-IUGR group was used to simulate overfeeding in lactation period. After weaning,the rats in the three groups were fed with basic diet. The weights were measured at postnatal 1,7,14,21,35,42 and 75 days. The time of vaginal orifice opening(VO)was recorded. The levels of estradiol(E2),luteinizing hormone(LH)and follicle estrogen(FSH)were examined on the 21 d and the 35 d.【Results】The birth weight of IUGR rats was(4.92±0.18)g,which was significantly lower than that in the control group(6.00±0.29 g,P < 0.001). IUGR rats showed weight growth catch- up on the 14th day. The body weight of SL- IUGR group was higher than that of control group from 14 to 75 d. The body weight of CF-IUGR group lagged behind that of control group from 1 to 75 d,the difference was statistically significant(P < 0.05). The vaginal opening time of female IUGR rats in SL- IUGR group was (29.88 ± 1.81)d,which was significantly earlier than that of CF- IUGR group(32.03 ± 2.11)days(P = 0.044). There was a correlation between body weight of the 21 d and vaginal opening time in IUGR rats,Rs = -0.174,P = 0.039. The besity rates of puberty and adulthood of IUGR rats in SL-IUGR group were 28.33% and 21.67%,which were significantly higher than those in CF-IUGR group(7.5%,6.25%;P = 0.001,0.007). The weight growth rate of adolescent obese IUGR rats at 7 and 21 d was higher than that of non-adolescent obese rats,and the weight growth rate of adult obese IUGR rats at 7 and 35 d was higher than that of non-adolescent obese rats. The difference was statistically significant(P < 0.05). Binary Logistic regression analysis of vaginal opening time and adult obesity in IUGR rats′ group OR = 0.419,P = 0.24. The time of vaginal orifice opening was not an independent factor of adult obesity. 【Conclusions】The improvement of nutrition level in the early stage of life is beneficial to weight catch-up of IUGR,and the overweight catch-up during lactation can lead to puberty and adulthood obesity of IUGR rats,as well as the advance of vaginal opening time.
6.Effect of Over-nutrition in Postnatal Life on Growth of Female SD Rats

【Objective】 To guide the nutritional intervention of children in the clinical practice ,by exploring the effect of over-nutrition in postnatal life on growth of the female SD rats.【Methods】We established the rat model of intra? uterine growth retardation(IUGR)through diet restriction in pregnant rats and selected female newborn rats that met the criteria as the experimental group(group I). Group I was randomly divided into control group(group IC)and small group(group IS). Fifty-eight female newborn rats with normal diet in pregnant period(group C)were also divided into control group(group CC)and small group(group CS). After 21-days-lactation,group IC and group CC were randomly subdivided into 4 rats per cage,and then fed with normal diet,while group IS and group CS were fed with high fat diet. We measured the body weight and the body length of each rat every week. Serum fasting insulin,fasting blood glucose,IGF-1,IGF- BP3 levels,and rat insulin sensitivity index(ISI)were measured at different growth and development stages.【Results】 The body weight and body length of group IS were higher than those of group IC(P<0.05);the same was true for group CS and group CC. In the different growth and development stages,the FINS,FBG,IGF- 1 and IGF- BP3 of group IS were higher than those of group IC(P < 0.05),the ISI was lower than that of group IC(P < 0.05). Besides,the FINS,FBG of group CS were higher than those of group CC and the ISI of group CS was lower than that of group CC in the 75 days after birth(P < 0.05).【Conclusions】We found that those small for gestational age infants can be added nutrition appropriately for rapid catch-up,but it is necessary to evaluate the growth index closely and adjust the nutrition on time. Children with normal intrauterine development also need to prevent over-nutrition.
7.Research on the Diagnosis of Smith-Magenis Syndrome

[Objective] We explore the diagnosis of Smith-Magenis syndrome and its clinical features of children,to raise the domestic awareness of this disease.[Methods] In this study,the child received peripheral blood chromosome microarray analysis,blood routine and urine routine,growth hormone provocation test,insulin-like growth factor Ⅰ and insulin-like growth factor binding protein Ⅲ test,cortisol (8a) test,prolactin test,adrenocorticotropic hormone test,thyroid function test,liver and kidney function test,blood biochemistry test,fasting insulin test,2-hour plasma glucose test,the antibodies and antigens test of hepatitis B.The bone age measurement and the pituitary gland MRI were also performed.We use the above figures to diagnose Smith-Magenis syndrome,assess and observe the condition of the child in Smith-Magenis syndrome.[Results] In this case,the chromosomal microarray analysis revealed a deletion of about 3.6Mb fragments in the chr17p11.2 region,including main functional gene RAI1,which was associated with Smith-Magenis syndrome.According to the clinical manifestations and the result of chromosome microarray analysis,the diagnosis of children with Smith-Magenis syndrome was made clear.[Conclusion] Genetic tests are the standard for diagnosing Smith-Magenis syndrome.When children have special facial features combined with multiple system disorders,early genetic examination is conducive to early diagnosis,and can reduce the time and economic cost.
8.Predictive value of c-met for long-term mortality in patients with esophageal squamous cell carcinoma.

OBJECTIVETo explore the correlation of c-met protein with the clinical staging and cell differentiation of esophageal squamous cell carcinoma (ESCC).
METHODSA total of 100 patients with ESCC were enrolled were examined for expression of c-met protein using immunohistochemistry, and the patients in negative and positive c-met expression groups were compared for clinicopathological characteristics and overall survival.
RESULTSs The 100 ESCC patients included 67 male and 33 female patients with a median age of 59 years; 49 of the patients were negative and 51 were positive for c-met expression. Positive c-met expression was significantly correlated with advanced TMN stages and lower tumor differentiation. Kaplan-Meier survival curve showed that the median survival time of c-met-positive patients was significantly reduced compared with that of c-met-negative patients (30.9 vs 48.2 months, P<0.05). COX regression analysis showed that c-met was a independent risk factor for the overall survival of the patients (HR: 2.34, 95% CI: 1.63-4.54, P<0.05).
CONCLUSIONA positive expression of c-met protein is significantly correlated with an advanced TMN stage, lower tumor differentiation and a poor prognosis, and may serve as a indicator for predicting the prognosis of ESCC.

9.Application of the real-time three-dimensional transesophageal echocardiography in the quantification of aortic anuulus diameter in patients with aortic stenosis

Objective To investigate the application of real-time three-dimensional transesophageal echocardiography (RT-3D TEE) in quantification of the aortic anuulus diameter in patients with severe aortic stenosis.Methods RT-3D TEE image of aortic root was get perioperatively in 65 patients with sever aortic valve stenosis during the surgery procedure.The aortic annulus was reconstructed in the quantitative analysis software to generate the parameters of 3-dimensional area based aortic anuulus diameter(3D-AAAD) and 3-dimensional circumference based aortic anuulus diameter(3D-C-AAD).The parameters was compared with intraoperative aortic anuulus diameter(IO-AAD),which was get by the standard cylindrical valve sizer.Results The three dimensional form of the aortic annulus was related to the number of the valves.The value of 3D-C AAD and 3D-A-AAD were (22.4 ± 2.1) mm,(23.4 ± 1.9) mm,no significant difference could be detected between them and the IO-AAD (P < 0.05).The 3D-C-AAD had a better agreements with IO-AAD than 3D-A-AAD.Conclusions RT-3D TEE can reconstruct the three dimensional sharp of aortic anuulus,and can be used as quantitative tools to calculate its diameter.
10.Study of the location of coronary ostial in the artic valve stenosis by the real-time three-dimensional transesophageal echocardiography

Objective To study the the feasibility of real-time 3D transesophageal echocardiography (RT-3D TEE) to measure the distance between the coronary ostial and the junction of left ventricle and artery.Methods The RT-3D TEE image of the aortic root was collected intraoperative in 65 cases who were refered to our center for valve replacement.The reconstruction of the aortic annulus was done in the QLAB quantitative analysis software and the height parameter of the annulus could be generated.Meanwhile the coronary artery position was observed and the distance between the coronary ostial and the junction of left ventricle and artery was measured respectively.Results In the 65 cases,49 cases clearly showed bilateral coronary ostial,three cases only showed the left coronary ostial,six cases only showed the right coronary ostial,seven cases of both sides of the coronary ostial were unclear.The shows rate of right coronary ostial was 84.61% (55/65),with the left coronary ostial 81.54% (52/65).The measured value of the L-AA in systole and diastole were (15.5 ± 1.9)mm,(12.8 ± 1.5)mm,the R-AA systole and diastole measured values were (13.7 ± 1.8)mm,(12.4 ± 1.7)mm,respectively,there was significant difference between systole and diastole respectively.The aortic annulus height (H) got by 3D reconstruction of the aortic annulus was lower than ventricular-arterial junction to the coronary artery distance in both systole and diastole.Conclusions Not only can RT-3D TEE reconstruct the annulus,but also can show and measure the distance between the coronary ostial and the junction of left ventricle and artery respectively.
