1.Analysis and summary of clinical characteristics of 289 patients with paroxysmal nocturnal hemoglobinuria in Zhejiang Province

		                        			Objective:To further improve the understanding of paroxysmal nocturnal hemoglobinuria (PNH), we retrospectively analyzed and summarized the clinical characteristics, treatment status, and survival status of patients with PNH in Zhejiang Province.Methods:This study included 289 patients with PNH who visited 20 hospitals in Zhejiang Province. Their clinical characteristics, comorbidity, laboratory test results, and medications were analyzed and summarized.Results:Among the 289 patients with PNH, 148 males and 141 females, with a median onset age of 45 (16-87) years and a peak onset age of 20-49 years (57.8% ). The median lactic dehydrogenase (LDH) level was 1 142 (604-1 925) U/L. Classified by type, 70.9% (166/234) were classical, 24.4% (57/234) were PNH/bone marrow failure (BMF), and 4.7% (11/234) were subclinical. The main clinical manifestations included fatigue or weakness (80.8%, 235/289), dizziness (73.4%, 212/289), darkened urine color (66.2%, 179/272), and jaundice (46.2%, 126/270). Common comorbidities were hemoglobinuria (58.7% ), renal dysfunction (17.6% ), and thrombosis (15.0% ). Moreover, 82.3% of the patients received glucocorticoid therapy, 70.9% required blood transfusion, 30.7% used immunosuppressive agents, 13.8% received anticoagulant therapy, and 6.3% received allogeneic hematopoietic stem cell transplantation. The 10-year overall survival (OS) rate was 84.4% (95% CI 78.0% -91.3% ) . Conclusion:Patients with PNH are more common in young and middle-aged people, with a similar incidence rate between men and women. Common clinical manifestations include fatigue, hemoglobinuria, jaundice, renal dysfunction, and recurrent thrombosis. 2.Chinese expert consensus on multidisciplinary minimally invasive diagnosis and treatment of pulmonary nodules

		                        			With the widespread application of high-resolution and low-dose computed tomography (CT), especially the increasing number of people participating in lung cancer screening projects or health examinations, the detection of pulmonary nodules is increasing. At present, the relevant guidelines for pulmonary nodules focus on how to follow up and diagnose, but the treatment is vague. And the guidelines of European and American countries are not suitable for East Asia. In order to standardize the diagnosis and treatment of pulmonary nodules and address the issue of disconnection between existing guidelines and clinical practice, the Lung Cancer Medical Education Committee of the Chinese Medicine Education Association has organized domestic multidisciplinary experts, based on literature published by experts from East Asia, and referring to international guidelines or consensus, the "Chinese expert consensus on multi-disciplinary minimally invasive diagnosis and treatment of plmonary nodules" has been formed through repeated consultations and thorough discussions. 3.Prognostic value and potential mechanism for the role of SNHG4 in gastric cancer

		                        			Objective:To investigate the expression, prognostic value and potential mechanism for the role of SNHG4 in gastric cancer.Methods:The expression of SNHG4 in gastric cancer was analyzed by UALCAN database. The relationship between SNHG4 and prognosis of gastric cancer was analyzed by Kaplan-Meier Plotter. SNHG4-miRNA-mRNA regulatory network was constructed by StarBase, Targetscan, microT-CDS and Cytoscape. The target genes were analysis GO and KEGG pathway enrichment by DAVID database.Results:The expression of SNHG4 in gastric cancer was significantly higher than that in normal tissues ( P=8.882E-16) . The overall survival time of patients with high SNHG4 expression was lower than that of patients with low expression ( P=8.900E-05) . Through the construction of RNA regulatory network, we found that hsa-let-7a-5p ( P=1.02E-03) , hsa-miR-152-3p ( P=4.51E-06) , hsa-miR-204-5p ( P=6.68E-04) and hsa-miR-363-3p ( P=8.06E-03) could be used as the binding sites of SNHG4 in gastric cancer, and these four miRNAs further regulated 250 downstream target genes. Through GO and KEGG enrichment analysis of the target genes, we found that these target genes played roles in the biological process of protein phosphorylation regulation, transcription negative regulation, RNA polymerase II promoter transcription, and participated in the occurrence and development of gastric cancer by blocking or activating Wnt and other signal pathways. Conclusions:SNHG4 can be used as a potential tumor marker for gastric cancer to judge the prognosis of gastric cancer. By constructing a SNHG4-miRNA-mRNA regulatory network, the pathogenesis of gastric cancer can be studied at the molecular level. 5.The creation of the reconstruction of right ventricular outflow tract in rats

		                        			Objective To investigate the feasibility of animal model of the reconstruction of right ventricular outflow tract in rats. Methods A total of 15 female Sprague-Dawley (SD) rats underwent right ventricular outflow tract reconstruction surgery. Before the operation, the collagen scaffolds were treated with g 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride chemistry (EDC), and seeded with human bone marrow stem cells (h-MSCs). Three days after the surgery, 3 rats were randomly sacrificed to evaluate the transmural resection of right ventricular outflow tract. One or 3 months later, other 3 rats at each timepoint were sacrificed, stained with Masson’s Trichrome to observe the degradation of scaffold. Furthermore, 4 weeks after the surgery, 4 rats were sacrificed and the hearts were sliced. Anti-human mitochondria staining was used to identify the survival of seeding cells. Results The transmural resection of right ventricular outflow tract was feasible in rats at an acceptable mortality (13.3%). After EDC treatment, the degradation rate of collagen scaffold was extended greatly. The seeding cells were detected by anti-mitochandria immunofluorescent staining in all patches 4 weeks after the operation. Conclusion Rat model of right ventricular outflow tract reconstruction could be a stable, reliable and economical screening model for engineered heart tissue research.

7.Accelerated and enhanced osteointegration of MAO-treated implants: histological and histomorphometric evaluation in a rabbit model.

		                        			Microarc oxidation (MAO) has become a promising technique for the surface modification of implants. Therefore, the aims of this study were to further quantitatively and qualitatively evaluate the osteointegration abilities of MAO-treated and smooth surface (SF) implants in vivo and to investigate the areas in which the superiority of MAO-treated implants are displayed. In a rabbit model, a comprehensive histomorphological, osteogenic, mineralizational, and integrative assessment was performed using light microscopy, fluorescence microscopy, confocal laser scanning microscopy, and radiographic analyses. Compared with the SF groups, the MAO-treated groups exhibited more active contact osteogenesis, as well as distant osteogenesis, under fluorescence examination, the mineral apposition rate was found to be greater for all of the MAO-treated implants, and the osteointegration index (OI) value was greater in the MAO-treated groups at different times. In conclusion, the calcium-rich amorphous layer created by MAO provided a better environment for osteointegration, with more active contact osteogenesis, a more rapid mineral apposition rate and greater OI values.

8.Repair of left ventricular aneurysm secondary to myocardial infarction with cytokine-conjugated collagen scaffolds in rat models

		                        			Objective To compare the vascularization of collagen scaffolds with or without growth factors and their efficacy on cardiac function in postinfarcted rats underwent surgical ventricular restoration.Methods Collagen scaffolds were activated with 1-ethyl-3-[3-dimethylaminopropyl]carbodiimide hydrochloride chemistry (EDC) as control or continually covalently immobilized with vascular endothelial growth factor (VEGF) and VEGF + basic fibrohlast growth factor (bFGF) as experimental groups.Adult SD male rats underwent left anterior descending artery (LAD) ligation to generate transmural myocardial infarction(MI).Four weeks later,by echocardiography,rats with moderate scar size(25%-35% akinetic area of freedom wall of left ventricle) were screened out,assigned into 3 groups randomly and received the surgical ventricular restocation (SVR).Then,cardiac function was measured by echocardiography at 1w,2w and 4w after patch implantation.At endpoint of study (4w after patch implantation),the rats were sacrificed and the hearts were harvested.Vascularization of patch were determined by capillary density (evidenced by vWFⅧ staining) or mature vessel density (evidenced by SMA staining) respectively.Results The general mortality of the animal model is 15% (6/40).A significant improvement of cardiac function was observed in all animals at 1 w after patch implantation but that was better preserved in both cytokine-conjugated groups 4w later (control group vs.VEGF group,P < 0.05,control group vs.VEGF + bFGF group,P < 0.01).More capillaries were present in patch with growth factors (P <0.05),while significant functional vessel formation was observed only in VEGF + bFGF group (P <0.01 vs.control or VEGF group).Additionally,we identified a positive correlation between heart function and mature vessel density (P =0.0297,r2 =0.998).Conclusion The mechanical property of collagen scaffold can be effectively improved by EDC,the growth factors immobilized in scaffold were in favor of vascularization of patch,which may facilitate the preservation of cardiac function posterior to SVR.
		                        		
		                        		
		                        		
		                        	
9.The value of CT venography in the surgical treatment of falcotentorial junction meningioma

		                        			Objective To explore the value of CT venography in the surgical treatment of falcotentorial junction meningiomas. Methods CT venography was carried out in 25 patients with falcotentorial junction meningiomas. 2-D and 3-D images were reformatted at the workstation. The classification and relationship between the tumors and veins were determined, and the degree of venous stenosis and collateral were assessed. The safe surgical pathway was chosen to avoid the injury of main draining vein. Results Falcotentorial junction meningiomas were divided into 5 types according to the relationship between the tumors and veins and the direction of the tumors. There were 5 cases of forward type, 4 cases of backward type, 3 cases of inferior type, 6 cases of superior type and 7 cases of lateral type in this group. The relationship between tumors and veins was accordant with the findings in surgery. Of all 25 cases, 19 underwent total removal, 6 underwent subtotal removal and all of the main driving veins were reserved. There were 3 cases of quadrantanopsia or hemianopsia after the operation, and all of them recovered after 3 months of follow-up. Conclusion CTV technique can classify the falcotentorial junction meningioma and is helpful for choosing the appropriate surgical approaches.
		                        		
		                        		
		                        		
		                        	
10.Assessment on physical development of Han children in rural area of Huludao city with Heath-Carter somatotype method

		                        			BACKGROUND: There are many methods for investigating the physical development and the somatotype of child. Among them, Heath-Carter so matotype method is a comprehensive evaluation method on somatotype. In this method, 10 items of anthropometric indicators are selected from several dozens of items as assessment indicators for development and somatotype.OBJECTIVE: To analyze the rules and characteristics of soma totype development of Hah children in rural area of Huludao city, so as to supplement the essential data for physical anthropology.DESIGN: Normal children were chosen as the subjects, and cross-sectional investigation was carried out.SETTING: Department of Anatomy, Jinzhou Medical College. PARTICIPANTS: With cluster sampling, the Han children between 7 to 10 years old were selected from central primary school of Mingshuixiang Village of Suizhong County of Huludao city from July 2001 to September 2003. All the selected children should be verified healthy by physical examination at school. The children were divided into two groups according to gender, and each group was divided into 4 subgroups according to age,with 46 to 63 members in each group. There were 8 groups altogether.Complete data of 408 children were collected (male 213, female 195).METHODS: Each indicator was measured twice with Heath-Carter soma to type method and selected the average value. Specialists were responsible for the 10 indicators, and a database was established in computer according to age and gender. Calculated the following indicators in turn: endomorphic factor, mesomorphic factor and ectomorphic factor, coordinate values of X and Y on somatotype chart, Height/Weight1/3, mean of dimensional distances from the average somatotype to all somatotypes in the sample,difference between the two somatotypes in three-dimensional space, percentage of body fat and frequency distribution of each somatotype.MAIN OUTCOME MEASURES: Height, body mass, upper arm circum ference, calf circumference, intracondylar diameters of humerus and femur,skinfold of brachial triceps, subscapular skinfold, skinfold of anterosuperior iliac spine, skinfold of gastrocnemius muscle.RESULTS: All the 408 children entered results analysis. ①Height and body mass of 7 to 10 year old children increased as age went up, indicators of Height/Weight1/3 of 7,8,10 year old girls were higher than boys, that of 9 years old boys was higber than girls, which were related to that the growth of height of 8 & 9 year old boys (5.61 cm) was more than girls (3.88 cm).The content of body fat of girls was more than boys. The 3 somatotype values were 3.2-3.9, 2.3-2.6 and 3.0-3.4 in boys and 3.5-4.6, 1.9-2.6 and 3.3-3.5 in girls, so endomorphic factor and ectomorphic factor were the main factors both in girls and boys, and endomorphic gene in girls was higher than boys, which indicated that the content of body fat of rural children in Hulutao city was relatively high, their bodies were finely high and thin, their skeletal muscle was not so muscular and the hypodermic fat in girls was more abundant. Seen from the comparison of somatotype between boys and girls in each age group, the differences between two somatotypes in the three dimensional space of 7-, 8-, 9-, 10-11 year old groups were 0.46, 0.68, 1.03 and 0.61 respectively, except 7-year old group, there were significant differences in somatotype of boys and girls between each age groups (P < 0.01). ②The average value of somatotype of Han boys in rural area was 3.4-2.5-3.2, which belonged to medial type; that of girls was 4.0-2.3-3.4, which beglonged to ectomorphic endomorphic type. Endomorphic factor and ectomorphic factor were the dominant factors both in boys and girls, whereas mesomorphic factor was relatively low; the frequency of somatotype kept changing as the increase of age. ③Compared with the data at home and abroad, the endomorphic factor of the Han children in rural area in Huludao City was relatively high, while the mesomorphic factor was relatively low, and the ectomorphic factor was almost the same.CONCLUSION: The somatotype of Han children in rural area in Huludao City develops better. Their heights are tall, whereas their skeletal muscles are not so muscular.
