1.Treating stasis-heat based on the ministerial fire axis of "pericardium-sanjiao-gallbladder"

Stasis-heat is a pathological factor associated with numerous exogenous and internal injuries, representing a pivotal mechanism in disease progression. Its primary cause is fire-heat toxicity. Based on the theory of qi and the holistic perspective of traditional Chinese medicine, this concept emphasizes that the biochemistry of all natural phenomena relies on the dynamic movement of qi ascending, descending, exiting, and entering. Within six qi, "fire" includes sovereign and ministerial fires. While physiological ministerial fire is the source power of life, pathological ministerial fire manifests as violent, intense energy that readily interacts with blood, leading to the formation of stasis-heat. Therefore, this article examines the formation and treatment of stasis-heat resulting from ministerial fire dysfunction. From the perspectives of ministerial fire gasification, the shape and quality of meridians, and the elevation of ministerial fire, it elucidates why the "pericardium-sanjiao-gallbladder" axis is regarded as pivotal. Furthermore, when the "pericardium-sanjiao-gallbladder" ministerial fire axis is unfavorable, and the stagnation of ministerial fire elevation and blockage is crucial to stasis-heat formation. Additionally, the depletion of essence and blood in the liver and kidneys, preventing the proper storage of ministerial fire, forms the pathological foundation. Drawing upon The Inner Canon of Yellow Emperor, this article explores therapeutic principles based on the rules of odor treatment: "when fires in the interior, the treatment of salty and cold, accompanied by bitter and pungent, acid to astringe, bitter to disperse." These principles are applied to achieve specific therapeutic goals: tempering the excess of ministerial fire to cool the nutritive level and transform stasis; adjusting the imbalance of elevation and depression to vent heat and unblock stasis; and restoring the misplaced fire by nourishing blood to expel stasis. Through these approaches, the article aims to reestablish the proper circulation of ministerial fire, dissipate blood stasis, and ultimately eliminate stasis-heat, thereby offering an integrated perspective on its pathogenesis and treatment.
2.Effect of ABCB13435C>T Genetic Polymorphism on Plasma Concentration and Adverse Reactions of High-dose Methotrexate in Acute Lymphoblastic Leukemia Children

Objective: To investigate the association between the genetic polymorphism of ABCB1 C3435T and plasma concentration and adverse reactions of high-dose methotrexate(HD-MTX) chemotherapy in children with acute lymphoblastic leukemia(ALL).Methods: A total of 70 peripheral blood samples were obtained from the children with acute lymphoblastic leukemia for extracting genome DNA.The genetic polymorphism of ABCB1 C3435T locus was examined by using PCR technology and direct sequencing;enzyme-amplified immunoassay (EMIT) was employed to determine the plasma concentration of MTX in 48 h;the clinical data of patients were collected during the HD-MTX chemotherapy, the adverse reactions were statistically analyzed.The association between ABCB1 C3435T genotypes and MTX plasma concentration and adverse reactions were investigated.Results: Genetic polymorphism existed at the SNP of ABCB1 C3435T.At the SNP of ABCB1 C3435T, the percentage of CC, CT and TT genotype in the children with ALL was 31.43%, 47.14% and 21.43%, respectively.The order of C/D ratio of MTX from low to high was CC, CT and TT genotypes, there was a significant association between the gene polymorphism and the C/D ratio of MTX (P<0.05);the order of incidence of oral mucosa damage from low to high was CC, CT and TT genotypes, there was a significant association between the gene polymorphism and oral mucosa damage (P<0.05);the order of incidence of hepatotoxicity from low to high were CC, CT and TT genotypes, the gene polymorphism and hepatotoxicity had a significant association (P<0.05).Conclusion: ABCB1 C3435T genetic polymorphism and C/D ratio of MTX, gastrointestinal side effects and hepatotoxicity after HD-MTX chemotherapy in the children with ALL exhibit significant association.
3.Antihypertensive Effect of the Extract from Compound Prunella Vulgaris L. in Spontaneous Hypertension Rats

Objective:To study the antihypertensive effect of the extract from compound Prunella vulgaris L. in spontaneous hy-pertension(SH)rats. Methods:Forty SH rats were randomly divided into the model group,high,middle and low dose groups of ex-tract from vompound Prunella vulgaris L. ,the compound kendir leaves tablets Ⅰ group with 8 ones in each. Non-invasive blood pres-sure measurement was used to detect the SBP and DBP of the SHR rats. Then the serum NO,AngⅡ ,ET-1 and ANP content were measured after the eight-week treatment. The pathological changes were observed after kidney HE staining in the SH rats. Results:Compared with that in the model group,the blood pressure in high-dose treatment group,middle-dose treatment group and the positive model group was significantly decreased(P < 0. 05). The AngⅡ and ET-1 levels in high-dose treatment group,middle-dose treatment group and the positive model group were decreased(P < 0. 01),and NO and ANP contents in serum were significantly increased when compared with those in the model group(P < 0. 01). The pathological examination showed that the pathological changes in the model group were faster than those in all the drug-treatment groups,the pathological changes included glomerular and renal tubular atrophy, glomerular vascular wall thickening and renal tubular epithelial cell degeneration or necrosis. Conclusion:The extract from compound Prunella vulgaris L. can reduce blood pressure of SH rats. The mechanism may be associated with the level reduction of AngII and ET-1 and content elevation of NO and ANP.
4.Findings in Positron Emission Tomography of Brain in Rats with Syndrome of Stagnation of Liver-Qi

Objective To explore the characteristics of brain function in rats with Stagnation of Liver-qi Syndrome. Methods Model ofStagnation of Liver-qi was made with chronic stress combined with isolated raising in 10 normal Wistar rats. They were scanned with positronemission tomography (PET) before and after modeling. Data analysis was carried out using SPM (statistical parametric mapping 2) inMATLAB software. Results After modeling, the glucose metabolism decreased (P<0.001) in left frontal lobe, parietal lobe, hypothalamus;bilateral posterior cingulate; right anterior cingulate, while the glucose metabolism increased (P<0.001) in left parahippocampal gyrus, insula,temporal lobe, frontal lobe; right basal ganglia, thalamus. Conclusion The function of some cerebral areas can be changed undergoingStagnation of Liver-qi Syndrome.
5.Study on Point-wise Correlation Dimension of Electroencephalogram Nonlinear Analysis in Patients with Stagnation of Liver-Qi Syndrome

Objective To investigate the changes of non-linear dynamics characteristics of electroencephalogram(EEG) in patients with the stagnation of Liver-Qi syndrome.Methods 15 Liver-Qi stagnation syndrome patients and 15 normal persons were paired by sex and age.The data of EEG under three states(eyes closed,eyes opened,and mental arithmetic with eyes opened) were analyzed by the parameter of point-wise correlation dimension(PD2).Results PD2 of the stagnation of Liver-Qi syndrome group increased significantly compared with that of the healthy control group and always maintain high level.The difference of PD2 between two groups was gradually reduced along with the increase of stimulated condition.Conclusion Non-linear dynamic analysis is more appropriate for the study of mental functions changes of stagnation of Liver-Qi syndrome and can be used to investigate the brain mechanism of the stagnation of Liver-Qi syndrome.
6.Characteristics of Minnesota Multiphasic Personality Inventory in Patients with Depression

ObjectiveTo investigate the characteristics of personality in patients with depression. Methods24 patients with depression and 24 normal controls were measured with Minnesota Multiphasic Personality Inventory (MMPI) software. ResultsCompared with the control, patient with depression showed significantly difference in neuroticism, extraversionn-introversion, anxiety, repression, manifest anxiety scale, ego strength, dependency, dominance (P<0.05). the T score of N and repression is higher than the Chinese usual score. ConclusionThe depression patients show their personality as repression, inferiority, melancholy, anxiety, bad tolerance, overstrung and dependent.

Objective To explore the characteristics of patients with stagnation of liver-qi syndrome using brain functional imaging techniques.Methods10 patients with stagnation of liver-qi syndrome and 20 normal persons whom paired by sex and age were scanned with positron emission tomography(PET).Data analysis was carried out using SPM in MATLAB software.ResultsCompared with normal,some cerebral areas of the patients with stagnation of liver-qi syndrome showed glucose metabolism letdown(P<0.001),including right parietal lobe(postcentral gyrus,BA1/2),temporal lobe(middle temporal gyrus,BA37),frontal lobe(middle frontal gyrus,BA9/10)etc.,while some cerebral areas showed glucose metabolism heighten(P<0.001)including right frontal lobe(superior frontal gyrus,inferior fontal gyrus,BA22/28/44),right cingulated gyrus,double midbrain and cerebella,etc.ConclusionThe function of some cerebral areas can be changed undergoing stagnation of liver-qi syndrome.

ObjectiveTo investigate the analgesic effect of TL-Ⅰ prescription.MethodsHealth ICR mouses were randomly divided into 6 groups: negative control group, positive control group, Tongtian group, large doses group,middle doses group and small doses group. The pain threshold of mouses were detected with hot plate and acetic acid writhing.ResultsTL-Ⅰ prescription can raise the threshold of pain induced by hot plate and reduce the numbers of writhing induced by acetic acid in mice, which was more significant in large doses group. ConclusionTL-Ⅰ prescription can be an effective analgesic.
