1.Effect of acute ozone exposure on miRNA expression profile of exosomes in alveolar lavage fluid of rats

Objective To clarify the changes in the microRNA(miRNA)expression profile of rat alveolar lavage-fluid-derived exosomes after acute ozone exposure and analyze its potential relationship with lung injury.Methods Adult male Wistar rats were exposed to 0 ppm,0.5 ppm,and 2 ppm ozone for 6 h in a gas exposure chamber.After 24 h,the rats were anesthetized,the alveolar lavage fluid was collected,and exosomes were extracted.Differentially expressed miRNAs in exosomes were detected by miRNA-seq technology.miRDB databases were used to predict the target genes of the top 10 differentially expressed miRNAs among the groups,and Gene Ontology function enrichment and KEGG Pathway analyses were performed on the target genes.The differentially expressed miRNAs with high expression abundance and good uniformity were verified by quantitative reverse-transcription PCR(qRT-PCR).Results Compared with the findings in the 0 ppm group,there were 8 up-regulated and 55 down-regulated miRNAs in the 0.5 ppm group,and 32 up-regulated and 61 down-regulated miRNAs in the 2 ppm group.Compared with the findings in the 0.5 ppm group,90 exosomal miRNAs were up-regulated and 74 were down-regulated in the 2 ppm group.The target genes of top 10 differentially expressed miRNAs were mainly involved in regulating DNA binding,heterocyclic compound binding,and affecting transcription,and were closely related to cancer,MAPK,PIK3-AKT,TNF,ErbB,mitophagy,and FoxO signaling pathways.The qRT-PCR showed that the sequencing result were basically reliable.Conclusions Acute ozone exposure can lead to changes in the expression profile of exosomal miRNA in the alveolar lavage fluid of rats.Differentially expressed miRNAs may be involved in pathological processes,such as lung tissue inflammation and lung injury,caused by ozone exposure.

3.Clinical Features and Current Treatment Status for In-hospital Elderly Patients With Mitral Regurgitation

Objective: To summarize the clinical features and treatment status for elderly in-hospital patients with mitral regurgitation (MR). Methods: A single center retrospective study was conducted in 1 741 patients admitted in our hospital from 2014-05-01 to 2015-04-30 with echocardiography confirmed moderate to severe MR. The patients were divided into 2 groups: Elderly group, n=680(39.06%)patients≥60 years of age and Non-elderly group,n=1 061(60.94%)patients<60 years.Clinical features and treatment status were studied and compared between 2 groups. Results: The mean age in Elderly group was (66.98±5.94) years and the most common type was degenerative MR (41.18%). Compared with Non-elderly group, Elderly group had more patients combining coronary artery disease (37.79% vs 17.43% ), more risk factors of atherosclerosis such as hypertension (45.44% vs 25.17%), diabetes (19.56% vs 8.48%) and hyperglycemia (35.29% vs 19.51%) all P<0.05; Elderly group had the higher EuroSCORE Ⅱ score (5.54±2.42) vs (3.15±1.66), greater left ventricular end diastolic diameter (57.72±12.37) mm vs (57.33±10.19) mm and less patients combining multiple valve disease (35.59% vs 40.81%), less patients received surgical treatment (54.71% vs 63.9%), all P<0.05. The surgery procedures (mitral valve replacement or mitral-plasty) were similar between 2 groups; compared with Non-elderly group, Elderly group had the higher application rate of bio-prosthetic valve (53.88% vs 18.67%), P<0.001. Conclusion: About 40% in-hospital moderate to severe MR patients were the elderly crowd, the most common pathogenesis was degenerative changes which leaded the higher incidences of cardiac complications, worse cardiac function and the higher risk scores for surgical treatment, there were less patients received surgery.

5.Treatment of nonalcoholic steatohepatitis by Jianpi Shugan Recipe: a multi-center, randomized, controlled clinical trial.

OBJECTIVETo evaluate the efficacy and safety of Chinese medicine (CM) intervention in the treatment of nonalcoholic steatohepatitis (NASH) from liver enzyme (ALT), imaging (the liver/spleen CT ratio) and syndrome scores, and to establish standard methods for diagnosis and therapeutic efficacy evaluation with characteristics of CM.
METHODSA multi-center, stratified randomized, parallel controlled, blindness-method evaluated, superiority trial was performed. Totally 204 patients were randomly allocated into two groups, 102 patients in the experimental group (treated with CM) and 102 patients in the control group [treated with Western medicine (WM)]. The alanine aminotransferase (ALT), liver/spleen CT ratio, and clinical symptoms were observed in both groups.
RESULTSOf the randomly allocated 204 cases from 4 hospitals, 3 patients were rejected, and 25 were lost. Totally 176 cases con- formed to the plan with complete follow-ups. After 3 months of treatment, syndrome scores and the improvement of partial clinical symptoms (fatigue and sallow complexion) were superior in the experimental group to those in the control group (P < 0.05). After 3 months of follow-up, the syndrome scores and improvement of partial clinical symptoms (fatigue and sallow complexion) were superior in the experimental group to those in the control group (P < 0.05). There was no statistical difference in improving liver enzymes or the liver/spleen CT ratio between the two groups (P > 0.05). There were 4 adverse reactions/adverse events in the two groups in the process of treatment, mainly covering drug-induced liver injury, diarrhea, and epigastric distension. Adverse reactions had nothing to do with CM treatment.
CONCLUSIONSJianpi Shugan Recipe had obvious efficacy in treatment of NASH. It could remove the liver fat and play a role in anti-inflammation and liver protection. It also could improve the indices of liver enzymes and the liver/spleen CT ratio effectively, which was superior to Polyene Phosphatidylcholine Capsule (PPC) in improving clinical symptoms, especially for such symptoms as fatigue and sallow complexion.

6.Oxidative Damage on Testicles of Male Rats Induced by Micro-nano-scale SiO_2

Objective To study the oxidative damage on testis of male rats induced by micro-nano-scale SiO2. Methods Male Wistar rats were exposed to nanometer SiO2(20-40 nm) and micro-meter SiO2(1-10 ?m)by intratracheal injection once two days. The rats were killed after 5 weeks of exposure. Some indicators related to the oxidative damage in the serum and the testis were determined. Results The activity of GSH-Px and SOD in the high dose groups decreased significantly and the MDA levels increased compared to the control group;The MDA levels of nano-silicon dioxide high dose group increased in the serum. Conclusion Nano-silicon dioxide may cause lipid peroxidation in the testis with a tendency of higher toxicity than that of micro-SiO2 at the same exposed level,the detail mechanism needs further discussion.
7.Rat Aortic Endothelium Damage Caused by Carbon Nanotubes Exposure

Objective To investigate the effect of nanotubes on rat aortic endothelium damage.Methods Wistar rats were intratracheally instilled with carbon nanotubes(CNTs) at the doses of 0,3.5 and 17.5 mg/kg,and with a carbon black negative control,or a quartz positive control.The rats were euthanized after 7 or 30 days of the single treatment,the oxidative stress(GSH,O_2~-?) was determined by using biochemical kits,the expression of sICAM-1 and sVCAM-1 in serum was determined by using ELISA kits and ICAM-1 and VCAM-1 on aortic endothelium was determined with immunohischemistry.Results The content of GSH and O_2~1?in serum increased with a dose-dependent and time-dependent manner.A up-regulated expression of sICAM-1 and sVCAM-1 in serum and ICAM-1 and VCAM-1 in the aortic endothelium was seen.No vascular damage was seen in the rat treated with carbon black,in those treated with high-dose CNTs obvious inflammation was revealed.Conclusion Carbon nanotubes exposure can induce oxidative stress which may be followed by the up-regulated expression of sICAM-1 and sVCAM-1 in serum and ICAM-1 and VCAM-1 in the aortic endothelium,and then the endothelium disfunction may occur.
8.Progress in Research on Antibacterial Mechanism and Biological Safety of Silver Nanoparticles

The antibacterial property of silver nanoparticles has resulted in their widespread application in many fields,so the chance of silver nanoparticles exposure for human increased greatly.Thus,there is urgent need to assess the safety of such particle.So far,most toxicological studies of silver nanoparticles mainly focus on the cytotoxicity using different examination endpoint such as morphology,mitochondrial function,cell proliferation,enzyme activity,and so on.In addition,the in vitro studies on the toxicity of silver nanopoarticles are also reported,few of the study on molecule mechanism of toxicity was reported.This review provided a summary of antibacterial mechanism of silver nanoparticles and the current research situation of the safety.The future research direction of toxicological study of silver nanoparticles is also prospected based on the current knowledge.
9.Oxidative Damage of Mouse Liver and Lung Protein Induced by Multi-walled Carbon Nanotube Exposure
