1.Study on the liver function protective effect and mechanism of ethanol extract from Atractylodes lancea

		                        			Objective Based on the network pharmacology and animal experiments,to investigate the protective effect and possible molecular mechanism of ethanol extract from Atractylodes lancea on liver function in mice with liver fibrosis induced by bile duct ligation.Methods The main active ingredients atractylodin,atractylenolide Ⅰ,Ⅱ and Ⅱ from Atractylodes lancea were selected,which had been verified by literature and experiments,and the targets of these active ingredients were obtained through the SwissTargetPrediction database.The liver fibrosis disease targets were obtained through On-line Mendelian Inheritance in Man (OMIM),DisGeNET and GeneCards databases.The targets were added to the Wei Sheng Xin platform to find the intersection target for Atractylodes lancea in treating liver fibrosis.Cytoscape 3.10.1 was used to construct the "drug-component-target-disease" network diagram and protein-protein interaction core target network diagram.GO functional enrichment analysis and KEGG pathway analysis were performed,and molecular docking was performed between active components and core targets.Liver fibrosis was induced in mice by bile duct ligation,and liver function markers were measured.Results A total of 91 corresponding targets of atractylodin,atractylenolide Ⅰ,Ⅱ and Ⅲ and 9296 liver fibrosis disease targets were obtained,including 74 intersecting targets and 31 core targets.KEGG enrichment analysis showed that the main signaling pathways involved included inflammatory pathways such as epidermal growth factor receptor (EGFR) and phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt).Molecular docking results showed that the active ingredients had strong binding activity with the core target protein.The results of animal experiments showed that,compared with the sham surgery group,the model group displayed notable,the liver index,spleen index,activity of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST),degree of liver fibrosis,mRNA and protein expression of α-smooth muscle actin (α-SMA) and recombinant collagen type Ⅰ alpha 1 (COL1A1),and mRNA of recombinant collagen type Ⅳ alpha 2 (COL4A2) were significantly increased,and the thymus index was sigficantly decreased (P<0.05);compared with the model group,the liver injury of mice in the Atractylodes lancea administration group reduced liver injury,its liver index,spleen index,activity of serum ALT and AST,degree of liver fibrosis,mRNA and protein expression of α-SMA and COL1A1,and mRNA of COL4A2 were significantly decreased,and the thymus index was sigficantly increased (P<0.05).Conclusion Atractylodes lancea can improve liver function and alleviate tissue pathological damage in mice with liver fibrosis,which may be related to activating pathways such as PI3K/Akt,inhibiting oxidative stress and inflammatory reactions,and intervening in liver fibrosis.

		                        			According to the pathological characteristics of symptomatic chronic thoracic and lumbar osteoporotic vertebral fracture (SCOVF), the different clinical treatment methods are selected, including vertebral augmentation, anterior-posterior fixation and fusion, posterior decompression fixation and fusion, and posterior correction osteotomy. However, there is still a lack of a unified understanding on how to choose appropriate treatment method for SCOVF. In order to reflect the new treatment concept and the evidence-based medicine progress of SCOVF in a timely manner and standardize its treatment, the clinical guideline for surgical treatment of SCOVF is formulated in compliance with the principle of scientificity, practicability and advancement and based on the level of evidence-based medicine.
		                        		
		                        		
		                        		
		                        	
3.Research on the learning curve of contrast-enhanced ultrasonography in sentinel lymph node of breast cancer

		                        			Objective To investigate the learning curve of contrast-enhanced ultrasonography ( CEUS) in sentinel lymph node( SLN ) of breast cancer and provide a theoretical basis for leaners to learn SLN CEUS . Methods The multi-center study of SLN CEUS in breast cancer" was planned by Sichuan Cancer Hospital . According to the uniform inclusion and exclusion criteria , 511 patients with complete clinical data and follow-up results from 9 hospitals in Multi-center were included in this study . According to the inspection time ,the patients were divided into 3 groups named as group A ( 170 patients) ,group B ( 170 patients) and group C ( 171 patients ) ,respectively . The basic clinical data ,ultrasound imaging data , intraoperative and postoperative pathological findings of all patients were recorded . With the accumulation of cases examined ,analysis was performed to find the learning curve of the SLN CEUS examination time , SLN CEUS detection rate ,SLN CEUS surface marking accuracy rate and SLN CEUS diagnosis rate ,the learning curve was analyzed . Results ① There was no statistical significant difference in patients ages , tumors sizes ,tumors locations ,SLNs numbers and LCs numbers among the three groups( all P > 0 .05) . ②As the number of cases examined increases ,the examination time was reduced gradually ,but SLN detection rate ,surface marking accuracy and SLN diagnostic coincidence rate were increased gradually( F = 151 .75 , 1 .96 ,7 .49 ,5 .50 ; P = 0 .000 ,0 .143 ,0 .001 ,0 .005 ) . Conclusions The skill of the doctor is improved gradually when learning SLN CEUS . With the number of the cases increase ,the operating time of SLN CEUS is shorted ,and the SLN detection rate ,surface marking accuracy and SLN diagnostic coincidence rate of SLN-CEUS are gradually increased . 		                        			Objective To evaluate the diagnostic value of contrast-enhanced ultrasound in breast precancerous lesions . Methods Retrospectively analyzed the contrast-enhanced ultrasound model and angiographic predictive model of 465 cases of the A prospective multicenter study of breast nodules contrast-enhanced ultrasound" that led the Sichuan Provincial People′s Hospital from January 2016 to April 2017 ,which included 69 cases of breast precancerous lesions and 396 other types benign lesions ,and the sensitivity ,specificity and accuracy of the diagnosis of breast precancerous lesions were calculated . Results The sensitivity of ultrasound predictive model for the diagnosis of precancerous lesions was 60 .9% and AUC was 0 .681 . Precancerous lesions mainly showed non-concentricity , increased homogeneity , and increased lesions;other types of benign lesions mainly showed non-centripetal ,high uniformity enhancement and lesion size unchanged . Conclusions Contrast-enhanced ultrasound shows a potential value in the differential diagnosis of precancerous lesions and other types of benign lesions ,that can help clinicians to take early intervention measures for breast precancerous lesions ,but there are still many problems to be solved .

		                        			Objective To explore labeling efficiency and appropriate conditions of Superpara magnetic iron exide nanopaticles (SPIO) nanoparticles for Bone marrow stromal cells(BMSCs). Methods BMSCs were aquired from skeletally mature dogs via iliac crest aspiration and separated by adherent cell cytopheresis.BMSCs were cultured and incubated with SPIO at different concentrations in vitro. The labeling efficiency of BMSCs with different labeling concentrations SPIO nanoparticles as well as detection of characteristics and signal attenuation rules were evaluated by MRI at 1.5T in vitro. Results BMSCs were efficiently labeled by SPIOin vitro and has no alterations to viability and proliferation profiles at this labeling concentration. BMSCs loaded with SPIO can be detected by MRI at certainly cell quantity in vitro(5 × 104). The quantity of SPIO in cells gradually reduced as cell culture time prolonged, with no statistically significant changes in cell death(P> 0.05). 7.Five-times Sit-to-Stand Test on Physical Performance for Older People

		                        			ObjectiveTo determine the value of the five-times sit-to-stand test (FTSST) on assessment of physical performance for older people.Methods65 volunteers participated in this study. 21 of them were evaluated the FTSST twice by the same raters within one week. Test-retest reliability assessed using intraclass correlation coefficients (ICC). 65 volunteers were tested for the FTSST, the Timed "up & go" test(TUGT) and the strength of lower limbs. The Pearson's correlation (r) and stepwise regression analysis was used to examine the relationship between these variables and the FTSST time.ResultsThe FTSST showed good test-retest reliability(ICC=0-91, 95%CI:0-79~0-96). The TUGT, the strength of lower limbs and age were significantly related to the FTSST time (P<0-05). The strength of lower limbs was important clinical factor to consider in determining the FTSST time.ConclusionThe FTSST showed good test-retest reliability and can reflect the physical functional status of older people.
		                        		
		                        		
		                        		
		                        	
8.Test-retest Reliability and Measurement Error in Using A Jamar Dynamometer to Determine Grip Strength for Elderly Men

		                        			 ObjectiveTo examine the test-retest reliability and measurement error in using a Jamar dynamometer to determine grip strength for elder men. Methods40 elder men with a mean age of 82 years old volunteered to participate in this study. The grip strength was administered twice using a Jamar dynamometer by specially trained testers; subjects were re-tested at the same time of day a week later by the same testers. ResultsThe infraclass correlation coefficient(ICC) was 0.99. The standard error of measurement (SEM and SEM%) were 1.18~1.69 and 4.11%~6.41%. The smallest real difference (SRD and SRD%) were 3.27~4.68 and 11.39%~17.75%. The Bland-Altman analysis revealed no systematic errors between test and retest. 9.Cross-sectional Study on Age-related Changes in Grip Strength of Elderly Men: Compared with Norms of Overseas

		                        			ObjectiveTo investigate the age-related changes in grip strength of the elderly men and compare with the norms of overseas. Methods112 subjects were divided into six groups: 65~69 years old, 70~74 years old, 75~79 years old, 80~84 years old, 85~89 years old, and 90~95 years old. The grip strength was measured with Jamar dynamometer. ResultsThe grip strength of both sides were significantly moderate negative correlated to age (r=-0.681 and -0.68, P<0.001) and stepwise regression analysis showed the age was important clinical factor to consider in determining grip strength in the elderly men (R2=0.468, F=47.951, P<0.001). The grip strength was declined significantly during the 75~79 and 85~89 years old. ConclusionGrip strength had declined in the elderly men and these losses were significantly related to age. The highest loss in men was in the age over 75. 10.Test-retest Reliability of Isometric Muscle Strength Measurement for Lower Limb with Functional Squat System in Elderly

		                        			ObjectiveTo determine the test-retest reliability of isometric muscle strength measurement for the lower limb with functional squat system (FSS) in elderly people. Methods20 elderly participants were measured the isometric muscle strength in 5 s of each lower limb with FSS, and retested a week later. The maximal voluntary contraction and the average strength of each lower limb were recorded. ResultsThe intraclass correlation coefficient were 0.77~0.88 (P<0.01).
