ObjectiveTo investigate the anti-inflammatory effect and mechanism of Danggui Shaoyaosan （DSS） in ameliorating vascular dementia （VaD）. MethodsSeventy 8-week-old C57BL/6J male mice were randomized into 7 groups： control， model， sham， positive control （donepezil， 10 mg·kg^-1）， and low-， medium-， and high-dose （12， 24， 36 g·kg^-1， respectively） DSS groups （n=10）. After drug administration， behavioral tests and cerebral blood flow detection were carried out. Enzyme-linked immunosorbent assay was used to assess the levels of interferon （IFN）-α， tumor necrosis factor （TNF）-α， interleukin （IL）-6， and CXC motif chemokine ligand 10 （CXCL10） in the brain tissue. Hematoxylin-eosin staining was employed to observe the changes in hippocampal morphology. Transcriptomics was used to predict the potential signaling mechanisms of DSS in ameliorating neuroinflammation， and Western blot and Real-time fluorescence quantitative polymerase chain reaction （Real-time PCR） were conducted to verify the expression changes of related genes. ResultsCompared with the control group and the sham group， the model group showed deceases in recognition index in the new object recognition test， movement distance and time in the target quadrant， and number of crossings in the Morris water maze test （P<0.01）. In addition， the model group showed slow cerebral blood flow and down-regulated protein levels of postsynaptic density protein 95 （PSD95） and occludin （P<0.05， P<0.01）， and there was no significant difference between the control group and the sham group. Compared with the model group， the DSS at each dose increased the new object recognition index， the distance and time in the target quadrant， the number of crossings， and cerebral blood flow （P<0.05， P<0.01）. In terms of brain tissue injury-related protein expression and inflammatory factor content， the medium-dose DSS group had the best effect， with higher protein levels of PSD95 and occludin （P<0.05） and lower levels of IFN-α， TNF-α， and IL-6 （P<0.01） than the model group. The hippocampus of model mice showed pathological manifestations such as cell loss and disarrangement， which were alleviated after administration of DSS at each dose. Transcriptomic results indicated that interferon regulatory factor 7 （IRF7）， signal transducer and activator of transcription 3 （STAT3）， and bone marrow stromal cell antigen 2 （BST2） were differentially expressed genes （down-regulated） in control group and medium-dose DSS group compared with the model group. The KEGG pathway enrichment analysis showed that RIG-Ⅰ-like receptor signaling pathway， Toll-like receptor signaling pathway， cytosolic DNA-sensing pathway， and downstream stimulator of interferon genes （STING） were both upstream signaling pathways affecting IFN expression. Real-time PCR results indicated that the mRNA levels of cyclic good manufacturing practice（GMP）-adenosine monophosphate（AMP） synthase （cGAS）， STING， IRF7， STAT3， and BST2 in the model group were higher than those in the sham group （P<0.05， P<0.01）. The mRNA levels of all the genes in the medium-dose DSS group were down-regulated compared with those in the model group after administration （P<0.05， P<0.01）. Western blot results indicated that the relative expression levels of cGAS， STING， p-IRF7， p-STAT3， and BST2 in the model group were higher than those in the sham group （P<0.05， P<0.01）. The protein levels of cGAS， STING， p-IRF7， p-STAT3， and BST2 in the medium-dose DSS group were decreased compared with those in the model group （P<0.05， P<0.01）. ConclusionDSS ameliorates cognitive impairment in the VaD model mice by inhibiting the cGAS-STING/IRF7/STAT3-mediated neuroinflammation.

Objective To investigate the expression level of serum ubiquitin-conjugating enzyme 2C(UBE2C)and tripartite motif-containing protein 27(TRIM27)in patients with endometrial cancer(EC)and their correlation with pathological parameters.Methods A total of 96 EC patients from March 2020 to March 2023 were selected as EC group;65 patients with endometrial atypical hyperplasia(EAH)as EAH group,and 80 healthy subjects as the control group.Enzyme linked immunosorbent assay was applied to analyze the expression level of serum UBE2C and TRIM27.The relationship between serum UBE2C,TRIM27,and pathological data was analyzed;receiver operating characteristic was applied to evaluate the predictive value of serum UBE2C and TRIM27 level for EC.Results The level of serum UBE2C and TRIM27 of EC patients was obviously higher than that in healthy subjects(P<0.05),and was correlated with tumor diameter,tumor differentiation,lymph node metastasis,FIGO stage,muscle layer invasion depth,cervical involvement,estrogen receptor expression,and progestogen receptor expression(P<0.05).There was positive correlation between serum UBE2C and TRIM27(r=0.475,P<0.001);and the level of serum UBE2C and TRIM27 was positively correlated with tumor diameter,lymph node metastasis,FIGO stage,and depth of musclular invasion,but negatively with tumor differentiation,estrogen receptor expression,and progestogen receptor expression(P<0.05).The combination of UBE2C and TRIM27 had obviously higher AUC in evaluating EC than single detection(ZUBE2C-combination=3.406,P<0.001,ZTRIM27-combination=3.285,P=0.001).Conclusion The expression level of UBE2C and TRIM27 in serum of EC patients is up-regulated,which is closely related to pathological parameters.The level of serum UBE2C and TRIM27 can provide reference for early diagnosis of EC.

Objective:To investigate the effects of Longmu Anshen prescription on microglia(MG)polarization in the medial prefrontal cortex(mPFC)and anxiety behavior of chronic restraint stress(CRS)mice.Methods:Forty-eight healthy male KM mice were randomly divided into normal group,paroxetine hydrochloride group and Longmu Anshen prescription high,middle and low dose groups.From the 15th day,after 30 minutes of CRS every day,the normal group and the model were given distilled water by gavage,the paroxetine hydrochloride group was given paroxetine hydrochloride(2.6 mg/kg)by gavage,and the traditional Chinese medicine group was given Longmu Anshen prescription at high,medium and low doses(4.16,2.08,1.04 g/kg)by gavage for 21 days.The changes of anxiety behavior in mice were evaluated by light-dark boxes experiment and elevated plus maze;ELISA was used to detect IL-1β,IL-6,TNF-α and IL-10 levels;protein expression of inducible nitric oxide synthase(iNOS)and arginase-1(Arg-1)in mice mPFC tissue were detected by Western blot;mRNA expression of ionized calcium binding adaptor molecule-1(Iba-1),iNOS and Arg-1 in mice mPFC tissue were detected by RT-PCR;the protein expression of Iba-1 in mice hippocampal tissue was detected by immuno-histochemistry(IHC).Results:Compared with normal group,the residence time in the light boxes,the number of times of shuttle between the light-dark boxes,the percentage of times of entering the open arm(OE%)and the percentage of time spent in the open arm(OT%)of mice in model group were significantly reduced(P<0.01);moreover,in the mPFC tissue of the model group mice,the protein and mRNA expressions of Iba-1 and iNOS were significantly increased(P<0.01),and the levels of IL-1β,IL-6 and TNF-α were significantly increased(P<0.01),while the protein and mRNA expressions of Arg-1 were significantly decreased(P<0.01),and the level of IL-10 was significantly decreased(P<0.01).Compared with model group,the residence time in the light boxes,the number of shuttles between the light-dark boxes,the OE%and OT%of mice in the paroxetine hydrochloride group and the high and medium dose groups of Longmu Anshen prescription were significantly increased(P<0.01);the expression levels of Iba-1 and iNOS protein and mRNA in mPFC tissue were significantly decreased(P<0.05,P<0.01),and the levels of IL-1β,IL-6 and TNF-α were significantly decreased(P<0.01),while the expression levels of Arg-1 protein and mRNA were significantly increased(P<0.01).IL-10 level was significantly increased(P<0.01).Conclusion:Longmu Anshen prescription can inhibit the activation of MG in mPFC tissues of CRS mice and may reduce nerve inflammation and increase the level of anti-inflammatory cytokines by regulating the polarization phenotype of M1/M2,thereby ameliorate CRS-induced anxiety behavior.

Objective: To investigate the effect of astragalus polysaccharides (APS) on proliferation, invasion, apoptosis and drugresistance of cisplatin-resistant colorectal cancer (CRC) HT-29/DDP cells through regulating miR-20a/TGFBR2 axis, and to explore the possible mechanism. Methods: Human CRC HT-29 cells and HT-29/DDP cells were used as non-drug resistant and resistant cell models, respectively; HT-29/DDP cells were randomly divided into four groups, including untreated (HT-29/DDP) group, APS treatment group, miR-20a mimics + APS group, and si-TGFBR2 + APS group. qPCR and Western blotting were applied to detect the expressions of miR-20a and TGFBR2 in HT-29/DDP cells treated with different concentrations ofAPS (0, 0.5, 1.0, 1.5 and 2.0 mg/ml). Subsequently, dual luciferase reporter gene assay was used to verify whether TGFBR2 was a target gene of miR-20a. In addition, CCK-8, Transwell andAnnexin V-FITC/PI double staining were applied to examine the effect ofAPS on proliferation, invasion and apoptosis of HT29/DDP cells. Furthermore, subcutaneous HT-29/DDP cell xenograft model was established on nude mice, and the effect ofAPS on the growth of transplanted tumor was observed. Results: APS significantly inhibited the proliferation of HT-29/DDP cells in a dose-dependent manner (P<0.01). Meanwhile, the expression of miR-20a was down-regulated in HT-29/DDP cells treated with APS, while the expression of TGFBR2 was significantly up-regulated (all P<0.01). Additionally, dual luciferase reporter gene assay result showed that TGFBR2 was a direct target of miR-20a in HT-29/DDP cells and its expression was suppressed. Furthermore, APS could enhance the drug sensitivity of HT-29/DDP cells through downregulating the inhibitory effect of miR-20a on TGFBR2 expression, thereby suppressed proliferation and invasion, and induced apoptosis of HT-29/DDP cells in vitro and in vivo. It was also found that this effect was related with the suppression of PCNA and Bcl-2 proteins and promotion of Bax and Caspase-3 proteins. Conclusion: APS reverses the resistance of HT-29/DDPcells to cisplatin by down-regulating the inhibitory effect of miR-20a on TGFBR2 expression.

Objective To learn the effect of breast removal of polyacrylamide hydrogel by areola incision;and to provide an effective method for removal of polyacrylamide hydrogel.Methods According to the accepted operation,45 cases with breast polyacrylamide hydrogel were divided into 25 cases of observation group and 20 cases of control group.The observation group was treated with areola medial semicircular incision breast removal of polyac-rylamide hydrogel,and the control group was treated with areola small incision suction to remove polyacrylamide hydrogel.The operative time,intraoperative blood loss,postoperative drainage tube extraction time,postoperative 7d breast shape and perioperative complications of the two groups were observed;postoperative follow -up was performed to evaluate the effect of surgery.Results The operation time of the observation group (190.2 ±36.1)min was longer than (119.5 ±44.0)min of the control group,the difference was statistically significant between the two groups (t =5.924,P <0.05);the intraoperative blood loss of the observation group (250.4 ±23.9)mL was higher than (89.7 ± 30.4)mL of the control group,the difference was statistically significant (t =19.865,P <0.05 ).The difference between postoperative 7d breast normal morphology rates of the two groups was not statistically significant (χ2 =2.571,P >0.05);the postoperative drainage tube extraction time of the observation group (3.7 ±0.5)d was more than (2.3 ±0.3)d of the control group,the difference was statistically significant (t =11.021,P <0.05 ).The complication rate of the observation group was 12.0%(3 /25),which of the control group was 10.0%(2 /20),the difference was not statistically significant (χ2 =0.045,P >0.05).All cases of the two groups were followed up;the mean follow -up time was (6.7 ±2.1)months.The good rate of surgery of the observation group was 92.0 % (23 /25),which of the control group was 70.0%(14 /20),the observation group was higher than the control group (χ2 =5.718,P <0.05).Conclusion For patients with complications after polyacrylamide hydrogel injection for augmentation mammoplasty;the breast polyacrylamide hydrogel removal surgery by areola medial semicircular incision is recommen-ded as the preferred.

Objective To study the incidence of positive t(14;18) and myc gene rearrangement, and the clinical features and prognosis of concurrent positive t ( 14;18 ) and myc gene rearrangement“ double-hit lymphoma” (DHL) in diffuse large B cell lymphoma.Me thods The positive t(14;18) and myc gene rearrangement in 106 cases of DLBCL were analyzed using interphase fluorescent in situ hybridization ( FISH ) technique. The expression of myc and bcl-2 proteins was determined by immunohistochemistry.The relationship of positive t ( 14;18) and myc gene rearrangement with clinical features, pathogenesis and prognosis for the patients was analyzed.SPSS 16.0 software was used for statistical analysis.Results Among the 106 cases, there were 27 (25.5%) cases with positive t(14;18) and 13 (12.3%) cases with myc gene rearrangement, and 7 cases (6.6%) of DLBCL with concurrent t(14; 18)-positive and myc gene rearrangement.A relationship was observed between positive t ( 14;18 ) and myc gene rearrangement ( P=0.019) .The follow-up data showed that the 7 DHL patients were in age of 528-4 years, the International Prognostic Index (IPI) scores were 3 in two cases, 4 in four cases and 5 in one case, and the ECOG scores were 3 in all the7 cases .Four patients had bone marrow involvement and were combined with leukemia.The survival time ranged from 0.5 to 6 months, with a median survival of 4 months.The univariate analysis showed that B symptom, Ann Arbor stage, ECOG score, LDH level, IPI score, immunophenotype, bcl-2 protein expression, myc protein expression,and myc gene rearrangement were all associated with poor prognosis ( P<0.05 for all) .The multivariate analysis using a COX proportional hazard model confirmed that ECOG score, bcl-2 protein expression, myc protein expression , myc gene rearrangement, and immunophenotype were independent prognostic factors affecting survival ( P<0.05 for all) , among them, the myc gene rearrangement was the strongest prognostic factor ( OR=4.337,P<0.001) . Conclusions “Double-hit” DLBCL is rare and can be mainly identified only by molecular detection.Perhaps positive t(14;18) and myc gene rearrangement play concurrent role in its “double-hit” pathogenesis.DHL are highly invasive, and most of DHL patients have poor prognosis.Further studies of larger case number are required to determine the pathologic features and the therapeutic strategy of this subgroup.

Based on quantum mechanics, rotational spectroscopy is a branch of fundamental science to study the rotational spectra of molecules, free radicals and ions, and has great applications on radio astronomy and many analytical fields such as atmospheric remote sensing. In this paper we reviewed the basic theory of rotational spectroscopy, two different types of Fourier transform microwave spectrometers, some typical spectroscopic analysis, and future prospect of the microwave spectroscopic techniques as well.

Objective To carry out a taxonomic identification of a strain of claviform bacteria iso-lated from prostatic fluid of a patient who suffered from chronic prostatitis, and to approach its phylogenic and biologic position. Methods We undertaked an initial identification by phenotypic characters such as morphologecal, physiological and biochemical characteristics to ascertain its phylogeny by chemical composi-tion analysis of cell wall and 16S rRNA gene sequencing and alignment. Results A club-shaped gram posi-tive rod bacillus was isolated in pure culture state. Its biochemical reactions were not active. The diamino-acid of cell wall was meso-diaminopimelic acid (meso-DAP) and it had wall chemotype Ⅳ ( contained arabi-nose, galactose and maltose ). Sequence searches of the GenBank database revealed that this strain had a highest level of 16S rDNA sequence similarity (99.4%) to C. tuberculostearicurn strain ATCC35692 with only 8 nucleotides difference. Conclusion On the basis of phenotypic and phylngenetie analysis, it is rea-sonable to assign this strain to the species C. tuberculostearicum, and this is the first isolation of C. tubercu-lostearicum from prostatic fluid home and abroad.
