1.Relationship Between Physical Activity and Depressive Symptoms Among Middle-Aged and Older Adults in Chengdu,Sichuan,China

Objective To investigate the current status of physical activity and depressive symptoms a-mong middle-aged and older adults in Chengdu,Sichuan and explore the relationship between physical activity and depressive symptoms.Methods Multi-stage proportional stratified random sampling was employed to select middle-aged and older adults aged ≥45 years as the participants,and face-to-face interviews were carried out to collect data.Logistic regression was adopted to explore the relationship between physical activity and depressive symptoms in middle-aged and older adults.The trend test was performed for the relationship between different lev-els of physical activity and depressive symptoms.The subgroup analysis and the test for multiplicative interactions were conducted for the relationship between physical activity and depressive symptoms.Results A total of 4376 middle-aged and older adults were included.Among them,14.58%(638/4376),25.98%(1137/4376),and 27.83%(1218/4376)had depressive symptoms,failed to reach the guideline-recommended standards of physical activity,and were at low levels of physical activity,respectively.There was a negative association be-tween reaching guideline-recommended physical activity standard and depressive symptoms in middle-aged and ol-der adults(OR=0.713,95%CI=0.589-0.861,P<0.001).In addition,moderate levels(OR=0.714,95%CI=0.586-0.871,P=0.001)and high levels of physical activity(OR=0.705,95%CI=0.548-0.906,P=0.006)had negative associations with the presence of depressive symptoms.The trend test revealed that the negative association between physical activity and depressive symptoms in middle-aged and older adults enhanced as the level of physical activity increased(Pfor trend=0.001).The subgroup analysis and the test for multiplicative interactions revealed that neither reaching guideline-recommended physical activity standards or not nor the physi-cal activity level had an interaction with each of the subgroups(all Pfor interaction>0.05).Conclusions The current status of depressive symptoms among middle-aged and older adults in Chengdu,Sichuan needs to be ameliora-ted.A negative association existed between reaching the guideline-recommended physical activity standard and pres-ence of depressive symptoms,and the negative association enhanced as the physical activity level elevated.

Transient receptor potential vanilloid-1(TRPV1) channel is a non-selective ligand-gated cationic channel with multiple activation mechanisms in the transient receptor potential subfamily. In recent years, a large number of studies have found that TRPV1 plays an important role in the field of cardiovascular diseases such as hypertension and atherosclerosis. With the in-depth study of traditional Chinese medicine, it has been found that Chinese medicine monomers and their active components can activate or inhibit TRPV1 channels, which has certain potential in the study of cardiovascular diseases. In this paper, the role of TRPV1 channel in cardiovascular diseases and the research progress of traditional Chinese medicine prevention and treatment of cardiovascular diseases based on TRPV1 channel are reviewed, in order to provide new ideas for prevention and treatment of cardiovascular system diseases.
3.Osteogenic activity of magnesium loaded hydrogel core-shell microspheres by microfluidic chip in vitro

Objective:To explore the osteogenic activity of in vitro cultured preosteoblasts treated by magnesium loaded hydrogel core-shell microspheres.Methods:A microfluidics chip device was designed and made to fabricate magnesium loaded hydrogel core-shell microspheres to precisely control the stable release of magnesium ions in vitro,the mechanism of controllable magnesium ion release in the promotion of osteogenic activity of MC3T3-E1 preosteoblasts was systematically investigated.Results:The magnesium loaded hydro-gel microspheres(PLGA/MgO-alginate microspheres)prepared by the microfluidics chip exhibited to be monodisperse with special core-shell structure and could control the stable release of magnesium ions at a concentration of 50 ppm in vitro.The in vitro study showed that it significantly enhanced the activity and proliferation ability of MC3T3-E1 preosteoblasts,promoted their osteogenic differ-entiation and mineralization ability,and increased ALP activity.Conclusion:Magnesium loaded hydrogel core-shell microspheres can promote the osteogenic activity of MC3T3-E1 osteoblasts in vitro.
4.The current situation and influencing factors of online psychological help seeking behavior among college students in Guangxi

Objective:
To understand the current situation and influencing factors of online psychological help seeking behavior among college students in Guangxi, China, so as to provide a scientific basis for improving the mental health of college students.
Methods:
From March to June 2022, a convenient sampling method was employed to select 2 239 university students from eight universities in Guangxi. The survey included the College Student Online Psychological Help Behavior Questionnaire, General Help Seeking Questionnaire (GHSQ), Attitudes Toward Seeking Professional Psychological Help Scale short Form (ATSPPH-SF), and the 10 item Kessler Psychological Distress Scale (Kessler10) to statistically analyze online psychological help seeking behavior and associated influencing factors.
Results:
The score of online psychological help seeking behavior among college students was ( 13.96± 5.20), and the scores of the three dimensions were as follows: online psychological self-help behavior (7.13± 2.95 ), online non-professional psychological help seeking behavior (4.28±2.19), online professional psychological help seeking behavior (2.54± 1.29 ). The factors influencing online psychological help seeking behavior were ranked as follows: mental health ( β = 0.32), CHSQ ( β =0.21), gender ( β =-0.09), ATSPPH-SF ( β =0.09) and registered residence ( β =0.08) ( P <0.05).
Conclusion
The online psychological help seeking behavior of college students in Guangxi is influenced by factors such as mental health. Seeking psychological help online has become a new form of support aimed at meeting the psychological health needs of college students, and constitutes an important supplement to offline psychological assistance. While there are similarities and differences between both modes, universities should pay attention to them.

Currently, in precision cardiac surgery, there are still some pressing issues that need to be addressed. For example, cardiopulmonary bypass remains a critical factor in precise surgical treatment, and many core aspects still rely on the experience and subjective judgment of cardiopulmonary bypass specialists and surgeons, lacking precise data feedback. With the increasing elderly population and rising surgical complexity, precise feedback during cardiopulmonary bypass becomes crucial for improving surgical success rates and facilitating high-complexity procedures. Overcoming these key challenges requires not only a solid medical background but also close collaboration among multiple interdisciplinary fields. Establishing a multidisciplinary team encompassing professionals from the medical, information, software, and related industries can provide high-quality solutions to these challenges. This article shows several patents from a collaborative medical and electronic information team, illustrating how to identify unresolved technical issues and find corresponding solutions in the field of precision cardiac surgery while sharing experiences in applying for invention patents.

Empathy is one of the basic qualities that modern clinicians need to possess.Although traditional Chinese medicine has no definition for empathy,it does have practical applications.TCM clinical empathy,nurtured by Chinese excellent traditional culture,can be seen in TCM classics and clinical practice of famous doctors of previous generations,and has been inherited in the spiritual thinking of modern colleges and universities of Chinese medicine.However,at present,there is a relative lack of special training courses for"empathy"in Chinese medicine higher education.This paper first briefly describes the connotation,history,and neuroscience mechanism of the concept of"empathy".Secondly,the manifestations and characteristics of"empathy"in the clinical practice of traditional Chinese medicine were sorted out.Finally,we use modern medicine and psychology for reference,and combine the characteristics of traditional Chinese medicine to make suggestions for the cultivation of the ability of"empathy"in clinical Chinese medicine.
7.Research progress in mRNA drug modification and delivery systems.

Messenger RNA (mRNA) has shown tremendous potential in disease prevention and therapy. The clinical application requires mRNA with enhanced stability and high translation efficiency, ensuring it not to be degraded by nucleases and targeting to specific tissues and cells. mRNA immunogenicity can be reduced by nucleotide modification, and translation efficiency can be enhanced by codon optimization. The 5´ capping structure and 3´ poly A increase mRNA stability, and the addition of 5' and 3' non-translational regions regulate mRNA translation initiation and protein production. Nanoparticle delivery system protects mRNA from degradation by ubiquitous nucleases, enhances mRNA concentration in circulation and assists it cytoplasmic entrance for the purpose of treatment and prevention. Here, we review the recent advances of mRNA technology, discuss the methods and principles to enhance mRNA stability and translation efficiency; summarize the requirements involved in designing mRNA delivery systems with the potential for industrial translation and biomedical application. Furthermore, we provide insights into future directions of mRNA therapeutics to meet the needs for personalized precision medicine.

8.Xiaoai Jiedu Prescription-containing Serum Enhances Lethal Effect of NK Cells on Colon Cancer Cells

ObjectiveTo explore the influence of Xiaoai Jiedu prescription (XJP)-containing serum on natural killer (NK) cells′ lethal effect on colon cancer cells and the molecular mechanism. MethodXJP-containing serum (0.1%, 0.5%, 1%, 5%, 10%) was used to treat HCT-116 cells and NK-92MI cells respectively for 24 h, and methyl thiazolyl tetrazolium (MTT) assay was employed to detect cell proliferation. Then, low-concentration (0.1%, 0.5%, 1%) XJP-containing serum was selected to treat co-cultured HCT-116 cells and NK-92MI cells for 24 h and calcein acetoxymethyl ester/propidium iodide (Calcein-AM/PI) was applied to detect the killing effect of NK cells on colon cancer cells. Flow cytometry was used to detect apoptosis of colon cancer cells, Western blot the expression of apoptosis-related proteins and signal transducer and activator of transcription 4 (STAT4) pathway-related proteins, and enzyme-linked immunosorbent assay (ELISA) the secretion of interferon (IFN)-γ. ResultHigh-concentration (5%, 10%) XJP-containing serum inhibited the proliferation of HCT-116 and NK-92MI cells (P<0.01), while low-concentration (0.1%, 0.5%, 1%) XJP-containing serum had no obvious influence on cell proliferation compared with the blank group. As compared with the blank group, low-concentration XJP-containing serum enhanced the killing activity of NK cells against colon cancer cells in a concentration-dependent manner (P<0.01), and induced apoptosis of colon cancer cells (P<0.01). Moreover, XJP-containing serum (0.1%, 0.5%, 1%) down-regulated the expression of B-cell lymphoma 2 (Bcl-2) and B-cell lymphoma-extra large (Bcl-xl), and up-regulated the expression of Bcl-2-associated X (Bax) compared with the blank group (P<0.05, P<0.01). Compared with the co-culture group, XJP-containing serum (0.1%, 0.5%, 1%) increased the expression of p-STAT4 and IFN-γ (P<0.05). ELISA result showed that XJP-containing serum (0.1%, 0.5%, 1%) raised IFN-γ secretion (P<0.01). ConclusionXJP-containing serum can enhance the activity of NK cells to kill colon cancer cells. The mechanism is the likelihood that it activates STAT4 pathway, increases IFN-γ secretion by NK cells, down-regulates the expression of Bcl-xl and Bcl-2, and up-regulates the expression of Bax, thereby promoting the apoptosis of colon cancer cells.
9.Prediction and risk factors of recurrence of atrial fibrillation in patients with valvular diseases after radiofrequency ablation based on machine learning

bjective To use machine learning technology to predict the recurrence of atrial fibrillation (AF) after radiofrequency ablation, and try to find the risk factors affecting postoperative recurrence. Methods A total of 300 patients with valvular AF who underwent radiofrequency ablation in West China Hospital and its branch (Shangjin Hospital) from January 2017 to January 2021 were enrolled, including 129 males and 171 females with a mean age of 52.56 years. We built 5 machine learning models to predict AF recurrence, combined the 3 best performing models into a voting classifier, and made prediction again. Finally, risk factor analysis was performed using the SHApley Additive exPlanations method. Results The voting classifier yielded a prediction accuracy rate of 75.0%, a recall rate of 61.0%, and an area under the receiver operating characteristic curve of 0.79. In addition, factors such as left atrial diameter, ejection fraction, and right atrial diameter were found to have an influence on postoperative recurrence. Conclusion Machine learning-based prediction of recurrence of valvular AF after radiofrequency ablation can provide a certain reference for the clinical diagnosis of AF, and reduce the risk to patients due to ineffective ablation. According to the risk factors found in the study, it can provide patients with more personalized treatment.
10.Machine learning models for analyzing valvular heart disease combined with atrial fibrillation using electronic health records

Objective To establish a machine learning based framework to rapidly screen out high-risk patients who may develop atrial fibrillation (AF) from patients with valvular heart disease and provide the information related to risk prediction to clinicians as clinical guidance for timely treatment decisions. Methods Clinical data were retrospectively collected from 1 740 patients with valvular heart disease at West China Hospital of Sichuan University and its branches, including 831 (47.76%) males and 909 (52.24%) females at an average age of 54 years. Based on these data, we built classical logistic regression, three standard machine learning models, and three integrated machine learning models for risk prediction and characterization analysis of AF. We compared the performance of machine learning models with classical logistic regression and selected the best two models, and applied the SHAP algorithm to provide interpretability at the population and single-unit levels. In addition, we provided visualization of feature analysis results. Results The Stack model performed best among all models (AF detection rate 85.6%, F1 score 0.753), while XGBoost outperformed the standard machine learning models (AF detection rate 71.9%, F1 score 0.732), and both models performed significantly better than the logistic regression model (AF detection rate 65.2%, F1 score 0.689). SHAP algorithm showed that left atrial internal diameter, mitral E peak flow velocity (Emv), right atrial internal diameter output per beat, and cardiac function class were the most important features affecting AF prediction. Both the Stack model and XGBoost had excellent predictive ability and interpretability. Conclusion The Stack model has the highest AF detection performance and comprehensive performance. The Stack model loaded with the SHAP algorithm can be used to screen high-risk patients for AF and reveal the corresponding risk characteristics. Our framework can be used to guide clinical intervention and monitoring of AF.
