1.Babaodan Alleviates APAP-induced Acute Liver Injury in Mice by Inhibiting NLRP3/Caspase-1 Pathway

		                        			ObjectiveTo explore the effect of Babaodan (BBD) on the NOD-like receptor pyrin domain containing 3/cysteine aspartate-specific protease-3 (NLRP3/Caspase-1) pathway proteins in mice with acetaminophen (APAP)-induced acute liver injury. MethodC57BL/6 mice were randomly grouped, and BBD (75, 150, 300 mg·kg-1, ig) was administered twice a day for three days. After 2 hours of the last administration, the mice were treated with APAP (400 mg·kg-1, ip), and the eyeballs were removed to collect blood after 14 hours. Then they were sacrificed by cervical dislocation for sample collection. Hematoxylin-eosin (HE) staining was used to observe the morphological changes of liver tissue cells, and biochemical methods were used to detect the activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST), superoxide dismutase (SOD), malondialdehyde (MDA) and myeloperoxidase (MPO) in serum of mice in each group. Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) was performed to determine the mRNA expression of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and IL-6, and Western blot was performed to determine the protein expression of cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS), NLRP3, Caspase-1 and IL-18 in the liver of mice. ResultCompared with the conditions in normal group, the hepatic lobule structure of mice in the model group was partially destroyed, and the hepatic sinusoids were dilated. And the expression levels of ALT and AST in serum, the protein levels of NLRP3, Caspase-1, iNOS, IL-18 and COX-2 and the mRNA levels of IL-1β, IL-6 and TNF-α were increased (P<0.05, P<0.01). Compared with the model group, the administration groups had improvement in liver cell rupture and hepatic sinusoidal compression, and a dose-dependent decrease in the levels of ALT and AST in serum as well as the protein levels of NLRP3, Caspase-1, iNOS, IL-18 and COX-2 and the the mRNA levels of IL-1β, IL-6 and TNF-α in liver tissue (P<0.05, P<0.01). ConclusionBBD can reduce APAP-induced acute liver injury in mice. The mechanism may be related to anti-oxidative stress, inhibition of NLRP3/Caspase-1 pathway, and decreased expression levels of IL-1β, IL-18, TNF-α and IL-6. 
		                        		
		                        		
		                        		
		                        	
2.Clinical analysis of the treatment of maxillary odontogenic cyst by nasal endoscope fenestration through nasal base.

		                        			Objective:To investigate the feasibility and clinical effect of the surgical approach and method of transnasal fenestration under nasal endoscope for the treatment of maxillary odontogenic cyst. Methods:The clinical data of 23 cases with maxillary odontogenic cysts treated by nasal endoscopy through nasal fenestration were retrospectively analyzed. All cases underwent nasal endoscopy and CT examination before the operation. The mucosal membrane of the parietal wall of the cyst was excised through fenestration of the nasal base. The cyst fluid was removed by decompression, and the bony opening of the nasal base was trimmed and enlarged to the edge of the cyst. The intraoperative and postoperative effects were observed. Results:All cases were well exposed under the direct vision of nasal endoscope. The top wall of the cyst was removed to maximize the communication between the cyst cavity and the nasal floor. There were no complications such as nasolacrimal duct injury, turbinate atrophy, necrosis, and facial numbness. All patients were followed up for 6-12 months, and the clinical symptoms gradually disappeared after surgery. The inferior turbinate was in good shape, the cyst cavity was smooth, the cyst wall was determined, and no cyst recurrence was observed. Conclusion:The treatment of odontogenic cyst of maxilla under nasal endoscope through nasal fenestration is convenient. It has less trauma, fewer complications and a satisfactory curative effect, which is worthy of clinical promotion.

3.Multimorbidity developmental trajectory among middle-aged and older adults and its impact on new-onset disability in China

		                        			Objective:To explore the developmental trajectory of multimorbidity and its impact on new-onset disability to identify homogeneous groups with similar multimorbidity developmental courses and to provide evidence for interventions for disability risk among middle-aged and older adults in China.Methods:Data was retrospectively collected from China Health and Retirement Longitudinal Study with four consecutive surveys (2011-2018). Group-based trajectory modeling was used to fit multimorbidity developmental trajectories, and the impact of multimorbidity trajectories on new-onset disability was analyzed using the time-dependent Cox regression model.Results:A total of 8 580 participants were included in current analysis, and four multimorbidity trajectories were identified: no multimorbidity ( n=2 136, 24.90%), newly-developing ( n=3 758, 43.80%), moderate-developing ( n=2 270, 26.45%) and severe-developing ( n=416, 4.85%). Participants who belong to moderate-developing and severe-developing tended to be female, single, overweight or obese, live in rural areas, have poorer self-rated health and high levels of annual per capita household expenditure, and developed a new-onset disability. After adjusting for demographic and behavioral covariates, compared to the newly-developing, the severe-developing( HR=3.132, 95% CI:1.884-5.207) had the highest risk of disability, followed by the moderate- developing ( HR=1.400, 95% CI:1.026-1.909) and the risk for the no multimorbidity ( HR=0.631, 95% CI:0.424-0.938) was the lowest. Conclusions:There was great heterogeneity in the developmental trajectory of multimorbidity among middle-aged and older adults in China. Data showed that the risk of disability in the developmental trajectory of multimorbidity increases with increasing levels. 4.HPV E6 and E7 mRNA combined with HPV 16 and 18 or 45 genotyping testing as a means of cervical cancer opportunistic screening

		                        			Objective To evaluate Aptima HPV E6 and E7 mRNA assay (Aptima HPV) combined with Aptima HPV 16 and 18 or 45 (18/45) genotype assay (Aptima HPV-GT) as a means of cervical cancer opportunistic screening. Methods From October 2016 to October 2017, a total of 23 258 women aged 25-65 years were enrolled in the physical examination center and gynecological clinic of Huzhou Maternity and Child Health Care Hospital. All the women had Aptima HPV tested, further Aptima HPV-GT testing for positive women and liquid-based thin layer cytology Thinprep cytologic test (TCT). Women with Aptima HPV (+) or ≥low-grade squamous intraepithelial lesion (LSIL) or obvious clinical symptoms (including vaginal bleeding after intercourse and watery, bloody vaginal discharge) were referred for colposcopy and further biopsy with or without endocervical curettage (ECC) if indicated. Expression of Aptima HPV, HPV 16 and HPV 18/45 with different cytological diagnostic groups and histological diagnosis groups were compared respectively. Sensitivity, specificity, positive predictive value and negative predictive value of Aptima HPV detection and TCT in identifying histological diagnosis of high-grade squamous intraepithelial lesion (HSIL) or worse (HSIL+) were compared. Results (1) The positive rates of Aptima HPV, HPV 16 and HPV 18/45 were 14.00% (3 257/23 258), 1.85% (430/23 258) and 0.86% (199/23 258) respectively.The positive rates of Aptima HPV, HPV 16 and HPV 18/45 increased with cytology grading in squamous epithelium [negative for intraepithelial lesion or malignancy (NILM), atypical squamous cells of undetermined significance (ASCUS), LSIL, atypical squamous cell cannot exclude HSIL (ASC-H), HSIL and squamous cell carcinoma (SCC), all P=0.000)]. According to histology results, the positive rates of Aptima HPV, HPV 16 and HPV 18/45 increased with histology grading in squamous epithelium (normal cervical tissue, LSIL, HSIL and SCC, all P=0.000). The positive rate of Aptima HPV was significantly higher in HSIL+group than that in the LSIL or better (LSIL-) group [98.11% (311/317) vs 12.84% (2 946/22 941), P=0.000]. The positive rate of Aptima HPV-GT was significantly higher in HSIL+group than that in LSIL-group [58.36% (185/317) vs 1.91% (439/22 941), P=0.000]. (2) Compared with cytology, Aptima HPV resulted in significant higher sensitivity (98.11% vs 59.62%, P=0.000) and negative predictive value (99.97% vs 99.42%, P=0.000), significant lower specificity (87.16% vs 95.37%, P=0.000) and positive predictive value (9.55% vs 15.10%, P=0.000) when identified HSIL+. Conclusions Women with Aptima HPV positive, especially those with Aptima HPV-GT positive, are more likely to have histological diagnosis of HSIL+. 5.Expression of PD-1 and PD-L1 in laryngeal carcinoma and its clinical significance

		                        			Objective To investigate the relationship between programmed death 1 (PD-1) and programmed death ligand 1(PD-L1) with clinicopathological characteristics in the patients with laryngeal carcinoma.Methods The expression levels of PD-1 and PD-L1 in 45 samples of laryngeal carcinoma tissue(laryngeal carcinoma group) and 37 samples of vocal cord polyp tissues(polyp group) were detected by using the immunohistochemical method.Then the relationship between PD-1 and PD-L1 with clinicopathological characteristics in the patients with laryngeal carcinoma was analyzed.Results The positive expression rates of PD-1 and PD-L1 in the laryngeal carcinoma group were 57.78% and 66.67 % respectively.PD-1,PD-L1 mainly located in cytoplasm or cell membrane of laryngeal carcinoma cells or interstitial lymphocyte.The positive expression rates of PD-1 and PD-L1 in the polyp group were 8.11% and 13.51% respectively.The positive expression rates of PD-1 and PD-L1 in the laryngeal carcinoma group were significantly higher than those in the polyp group(P<0.01).The expression of PD-1 and PD-L1 in laryngeal carcinoma tissue had a correlation.The positive expression rates of PD-1 and PD-L1 in the laryngeal carcinoma tissue of stage Ⅲ and Ⅳ were significantly higher than those of stage Ⅰ and Ⅱ (P<0.05).The positive expression rates of PD-1 and PD-L1 in highly differentiated laryngeal carcinoma tissue were significantly higher than those in moderately and lowly differentiated carcinoma tissue(P<0.05).Conclusion PD-1 and PD-L1 play an important role in the occurrence and development of laryngeal carcinoma.The expression of PD-1 and PD-L1 is closely correlated to the clinical stage and differentiation degree of laryngeal carcinoma.

7.Separation, Identification and Inhibitory Effect on HepG2 Cells of Flavonoids from Humulus Scandens

		                        			Objective:To extract, separate and identify the flavonoids in Humulus Scandens and detect their cytotoxic effect on cells HepG2. Methods: After Humulus scandens bing was extracted by alcohol, luteolin-7-O-β-D-glucoside ( LGL) and cosmoiin ( AGL) were obtained by using column chromatography and identified by spectral analysis. The inhibitory effects of LGL and AGL on HepG2 cells were detected by a typan blue dye method. Results:The main flavonoids in Humulus Scandens were LGL and AGL. When the concentration of LGL and AGL was increased from 6. 25 μmol·L-1 to 100. 0 μmol·L-1 , the inhibition rate of LGL was increased from 28. 8% to 95. 2% and that of AGL was increased from 3. 6% to 21. 3%. 8.Local recurrence after complete resection of primary retroperitoneal liposarcoma

		                        			Objective To investigate factors impacting on local recurrence in cases of primary retroperitoneal liposarcoma (RPLS) after complete enbloc resection.Methods The clinical data of 47 cases of primary retroperitoneal liposarcoma were retrospectively analyzed.Results 3-year local recurrence rate was 62%.Single and multiple regression analysis showed preoperative needle biopsy,histological subtype,tumor size and capsular invasion were all risk factors related to postoperative local recurrence in primary retroperitoneal liposarcoma undergoing complete resection (P < 0.05).Conclusion Preoperative aspiration biopsy,tumor poor differentiation,large tumor size tumor extracapsular spread all lead to high local recurrence rate after complete resection of primary retroperitoneal liposarcoma.
		                        		
		                        		
		                        		
		                        	
9.The Experimental Study on the Changes of Hearing Function and Cochlear Morphology in Rat Diffuse Brain Injury Model

		                        			Objective To study the changes of hearing function and cochlear morphology on diffuse brain in-jury model in rat .Methods One hundred and fifty SD rats with normal hearing were randomly divided into five groups ,each group consisted of 30 SD rats ,including a control group and four experimental groups which endured diffuse brain injury(DBI) from one to four weeks .Diffuse brain injury model of rats were established ,then ABR , 40 Hz AERP and ASSR examination ,light microscopy ,electron microscopy were used to evaluate the change of hearing function and morphology .Results The difference of ABR ,40 Hz AERP and ASSR thresholds between the experimental and the normal control group were significant (P<0 .05) .The thresholds of ABR ,40HzAERP and AS-SR were increased in the first week of DBI ,then the threshold continuously increased in the second and third week , at last the threshold decreased in the fourth week .The results under scaning electron microscope demonstrated that the ciliums of the majority of outer hair cells lodged in the first week of DBI .The results under transmission electron microscope showed that in the first week of DBI ,there were edema and denuration of mitochondrial ,mitochondrial cristaes were obscured or disappeared .The changes were deteriorate in the second and third week ,whereas the changes were mitigatal in the fourth week .Conclusion Cochlear morphology and hearing damage were observed in diffuse brain injury model of rats .
