1.A cohort study of lipid levels and recurrence risk of ischemic stroke in a community-based natural population in Songjiang District, Shanghai

		                        			ObjectiveTo investigate the recurrence of ischemic stroke (IS) and to analyze the association between four indices of total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), and triglycerides (TG) and the risk of IS recurrence by analyzing the follow-up data related to IS in the community-based natural population of Songjiang District, Shanghai, so as to provide a scientific basis for improving the prognosis of stroke patients in the community and controlling IS recurrence. MethodsA prospective follow-up study was conducted among the IS patients in the community-based cohort population, collecting data about patient’s age, gender, disease history, biochemical indicators, and etc. Cox regression model and restricted cubic spline model were used to analyze the relationship between different levels of plasma lipids and the recurrence of IS in these patients. ResultsA total of 1 368 patients with IS were included. The total follow-up duration was 7 171.46 person-years, with a median follow-up time of 6.24 years. There were 420 cases of IS recurrence, resulting in a cumulative recurrence rate of 30.70%. The results of multivariate Cox regression analysis showed that the recurrence risk of IS was reduced when the baseline TC and LDL-C levels of IS patients were in the ranges of 4.65‒5.67 mmol·L-1 and 2.52‒3.46 mmol·L-1, respectively. The results of restricted cubic spline analysis showed a U-shaped relationship between baseline TC and LDL-C levels and the recurrence risk in IS patients. ConclusionThe cumulative recurrence rate of patients with IS in the community of Songjiang District in Shanghai is high, and the levels of TC and LDL-C at baseline survey are correlated with the recurrence of IS in these patients. It is suggested to pay more attention to the levels of LDL-C and TC in patients with IS, so as to improve the prognosis.

		                        			Objective To study the application effect of quality control circle(QCC)in reducing the dissatisfaction rate of physical examination clients in health management center.Methods To establish QCC,selected the health check-up popula-tion in our hospital in September-2019 and March-2020,through the questionnaire investigation and analysis,compare the dis-satisfaction of the clients before and after the quality control circle.Results After carrying out QCC activities,the dissatisfaction of physical examination clients was significantly lower than that before QCC,and the difference was statistically significant(P<0.05).Conclusion The activities of QCC in the health management center can effectively improve the quality of the physical examination work and reduce the dissatisfaction of the customers in the physical examination.It is of great significance to the health management.

		                        			Diabetes mellitus(DM)is a disease syndrome characterized by chronic hyperglycaemia.A long-term high-glucose environment leads to reactive oxygen species(ROS)production and nuclear DNA damage.Human umbilical cord mesenchymal stem cell(HUcMSC)infusion induces significant antidiabetic effects in type 2 diabetes mellitus(T2DM)rats.Insulin-like growth factor 1(IGF1)receptor(IGF1R)is important in promoting glucose metabolism in diabetes;however,the mechanism by which HUcMSC can treat diabetes through IGF1R and DNA damage repair remains unclear.In this study,a DM rat model was induced with high-fat diet feeding and streptozotocin(STZ)administration and rats were infused four times with HUcMSC.Blood glucose,interleukin-6(IL-6),IL-10,glomerular basement membrane,and renal function were examined.Proteins that interacted with IGF1R were determined through coimmunoprecipitation assays.The expression of IGF1R,phosphorylated checkpoint kinase 2(p-CHK2),and phosphorylated protein 53(p-p53)was examined using immunohistochemistry(IHC)and western blot analysis.Enzyme-linked immunosorbent assay(ELISA)was used to determine the serum levels of 8-hydroxydeoxyguanosine(8-OHdG).Flow cytometry experiments were used to detect the surface markers of HUcMSC.The identification of the morphology and phenotype of HUcMSC was performed by way of oil red"O"staining and Alizarin red staining.DM rats exhibited abnormal blood glucose and IL-6/10 levels and renal function changes in the glomerular basement membrane,increased the expression of IGF1 and IGF1R.IGF1R interacted with CHK2,and the expression of p-CHK2 was significantly decreased in IGF1R-knockdown cells.When cisplatin was used to induce DNA damage,the expression of p-CHK2 was higher than that in the IGF1R-knockdown group without cisplatin treatment.HUcMSC infusion ameliorated abnormalities and preserved kidney structure and function in DM rats.The expression of IGF1,IGF1R,p-CHK2,and p-p53,and the level of 8-OHdG in the DM group increased significantly compared with those in the control group,and decreased after HUcMSC treatment.Our results suggested that IGF1R could interact with CHK2 and mediate DNA damage.HUcMSC infusion protected against kidney injury in DM rats.The underlying mechanisms may include HUcMSC-mediated enhancement of diabetes treatment via the IGF1R-CHK2-p53 signalling pathway.

		                        			Objective To explore the mechanism of dendrobine in the treatment of diabetic kidney disease(DKD)by intervening immunity using network pharmacology,bioinformatics and Western blot.Methods The targets of dendrobine were screened by Pharmmaper,Superpred and other databases.The disease targets of DKD were screened from GeneCards,OMIM and other databases.The immune-related targets were searched in Immport,In-nateDB and other databases.The intersection of the above three was obtained via Venn diagram.The PPI map of intersection targets was constructed by STRING software combined with Cytoscape software,and the core targets in the interaction network were mined by CytoHubba plug-in.The data set GSE142025 was obtained from GEO,and immune infiltration and WGCNA analysis were performed using the R language CIBERSORT package.GSE1009,GSE30122 and GSE142025 were used for gene differential expression analysis and ROC verification.The correla-tion between core targets and immune cell phenotype was analyzed.Molecular docking and molecular dynamics sim-ulation analysis were used to test the binding force of dendrobine to key targets.Western blot was used to detect the expression of HSP90AA1 in HK-2 cells.Results A total of 128 intersecting genes,including HSP90AA1,LCK and EGFR,were obtained,and the core targets involved HSP90AA1,LCK and STAT3;molecular docking and molecular dynamics simulation analyses showed that the protein of HSP90AA1 could bind well with dendrobine.Western blot experiments showed that compared with the control group,HSP90AA1 was significantly down-regula-ted in the high glucose group.Compared with the high glucose group,HSP90AA1 was significantly up-regulated in the dendrobine group.Conclusion Dendrobine can intervene in diabetic kidney disease by regulating the expres-sion of HSP90AA1.

		                        			"Biochemical Engineering Experiment" is a compulsory curriculum for the concentrated practical teaching of biotechnology majors in Hunan University of Science and Engineering. It is also an experimental curriculum for improving the overall quality of bioengineering students under the context of "Emerging Engineering Education". The course includes comprehensive experiments and designable experiments, and the contents of which are designed by combining the local characteristic resources of Yongzhou, the research platform and the characteristics of the talents with engineering background. In the teaching practice, methods such as heuristic teaching, research cases-embedded teaching and interactive teaching are comprehensively used to boost students' interest in learning and stimulate their innovative thinking and application capability. Through curriculum examination and post-class investigation, it was found that the students' abilities of knowledge transfer and application were significantly improved, and they achieved excellent performances in discipline competitions and approved project proposals. 7.Application of training methods of clinical skills competition for college students in standardized training of anesthesiologists

		                        			This study summarizes the skills training work of the National College Students' Clinical Skills Competition for 9 consecutive years, and explores a set of teaching methods suitable for the standardized training of residents of anesthesiology. This method is applied to the clinical operation, professional theory, clinical thinking, humanistic care, doctor-patient communication and other aspects of the standardized training of residents. 8.Effect of Danggui Shaoyaosan on Improvement of Cognitive Ability of SAMP8 Mice and Its Mechanism via Regulating Ubiquitin Proteasome Pathway

		                        			ObjectiveTo investigate the mechanism of Danggui Shaoyaosan (DSS) in the improvement of the cognitive ability of SAMP8 mice with Alzheimer's disease (AD) via regulating the ubiquitin-proteasome pathway (UPP). MethodFifteen SAMR1 mice were used as a normal group, and 60 SAMP8 mice were randomly divided into a model group and DSS high, medium, and low-dose groups (57.6, 28.8, and 14.4 g·kg-1·d-1), with 15 mice in each group. Intragastric administration was conducted for eight continuous weeks. Place navigation and spatial capacity were evaluated by Morris water maze. Pathological structure changes in neurons in the hippocampal CA1 area was detected by hematoxylin-eosin (HE) staining. The protein expression levels of hippocampal β-amyloid protein(Aβ) and phosphorylation(p)-Tau were determined by immunohistochemical staining (IHC) and enzyme-linked immunosorbent assay (ELISA), and the mRNA and protein expression levels of hippocampal ubiquitin (Ub), ubiquitin ligase E3 (E3), 26S proteasome, ubiquitin carboxyl terminal hydrolase-1 (UCHL1), and UCHL3 were determined by real-time fluorescent quantitative polymerase chain reaction (Real-time PCR) and Western blot, respectively. ResultAs compared with the normal group, the escape latency was prolonged in the model group (P<0.05) with the reduced number of crossing platform quadrants and time ratio in the platform quadrant (P<0.05). The model group decreased neurons and condensed cell bodies in the CA1 area, and increased β-amyloid precursor protein (β-APP) and p-Tau positive cells (P<0.05). In the model group, the protein expression levels of Aβ and p-Tau were increased (P<0.05), the mRNA and protein expression levels of Ub were increased (P<0.05), and the mRNA and protein expression levels of E3, 26S proteasome, UCHL1, and UCHL3 were decreased (P<0.05). As compared with the model group, the escape latency was shortened in the DSS high and medium-dose groups (P<0.05) with an increased number of crossing platform quadrants and residence time ratio (P<0.05). The pathological changes in CA1 of each DSS group were significantly improved, and the number of β-APP positive staining cells decreased (P<0.05). The number of p-Tau positive staining cells decreased in the DDS medium and low-dose groups (P<0.05). The protein expression levels of Aβ and p-Tau in each DDS group decreased (P<0.05), and the mRNA expression level of Ub in each group decreased (P <0.05). The mRNA expression levels of 26S, E3, and UCHL3 in the DDS high and medium-dose groups increased (P<0.05), and the mRNA expression level of UCHL1 in the DDS medium-dose group increased (P<0.05). The protein expression level of Ub in each DDS group decreased, and the protein expression levels of 26S, E3, UCHL1+3 in the DDS high and medium-dose groups increased (P<0.05). ConclusionDSS can improve the cognitive ability of SAMP8 mice, and its mechanism may be related to the reduction of the abnormal deposition of Aβ and p-Tau via decreasing the expression of Ub and increasing that of E3, 26S, UCHL1, and UCHL3 in the UPP. 
		                        		
		                        		
		                        		
		                        	
9.Protective Effect of Danggui Shaoyaosan-contained Serum on Aβ1-40-injured PC12 Cells via Regulating UPP and Its Mechanism

		                        			ObjectiveTo investigate the protective effect of Danggui Shaoyaosan (DSS)-contained serum on β-amyloid (Aβ)1-40-injured rat adrenal pheochromocytoma PC12 cells and its mechanism in regulating ubiquitin-proteasome pathway (UPP). MethodAβ1-40 was used to intervene PC12 cells to prepare the cell models of Alzheimer's disease (AD), and the experiment was divided into the blank, model, and DSS-contained serum high, medium, and low-dose groups (10%, 5%, and 2.5%). Cell viability and apoptosis were detected using cell counting kit-8 (CCK-8) method and flow cytometry, respectively. The content of Aβ and p-Tau protein was determined by enzyme-linked immunosorbent assay (ELISA). The ubiquitin (Ub), ubiquitin ligase E3 (E3), 26S proteasome, ubiquitin carboxyl terminal hydrolase1 (UCHL1), and UCHL3 protein expressions of UPP were displayed using immunofluorescence cytochemistry (ICC), and the mRNA and protein expression levels of Ub, E3-parkin, 26S, UCHL1, and UCHL3 were determined by real-time fluorescent quantitative polymerase chain reaction (Real-time PCR) and Western blot, respectively. ResultThe data of the CCK8 experiment verified that 5 μmol·L-1 and 48 hours were the optimal conditions for modeling Aβ1-40-injured PC12 cells. As compared with the blank group, the cell viability rate in the model group decreased (P<0.05) with an increased apoptosis rate (P<0.05), the content of Aβ and p-Tau contents was elevated (P<0.05), the mRNA and protein expression levels of Ub increased, and the mRNA and protein expression levels of 26S, E3, and UCHL1+3 decreased (P<0.05). As compared with the model group, the cell viability rate in the DSS-contained medium-dose group increased (P<0.05), whereas the apoptosis rate in each DSS-contained group decreased (P<0.05). The content of Aβ in each DDS-contained group decreased (P<0.05), and the content of p-Tau in the DDS-contained high and medium-dose groups decreased (P<0.05). The mRNA expression level of Ub decreased, and that of 26S increased in each DDS-contained group (P<0.05). The mRNA expression level of UCHL1 in the DDS-contained medium-dose group increased (P<0.05), and the mRNA expression levels of E3 and UCHL 3 in the DDS-contained high and medium-dose groups increased (P<0.05). The protein expression level of Ub in each DDS-contained group decreased, and the protein expression levels of 26S, E3, and UCHL1+3 in the DDS high and medium-dose groups increased. The DSS-contained serum medium-dose group exerted the optimal effect. ConclusionDSS-contained serum can increase cell viability rate, reduce cell apoptosis rate, eliminate Aβ and p-Tau protein deposits, and exert protective effects on Aβ1-40-injured PC12 cells. 10.Effectiveness research of medicatedγintrauterine device and medicated genefix intrauterine device inserted immediately after abortion

		                        			Objective To compare the effectiveness of medicated γintrauterine device (IUD) and medicated genefix IUD inserted immediately after abortion. Methods A multicenter clinical trail was performed for the study from Mar. 2012 to Jan. 2013. Totally 840 women who volunteered to participate were randomly allocated to γ-group (medicated γ IUD) or genefix-group (medicated genefix IUD) immediately after abortion. While 464 abortion women who had not used IUD or steroids contraceptive methods were chosen as control group. The effectiveness of the IUD were followed up for 1 year. All women were required to record the number of vaginal bleeding days and blood volume of vaginal bleeding within 3 months after abortion. Results At the 12th month, the expulsion was the most common reason for termination. The expulsion rates of genefix-group and γ-group were 2.48/100 women years and 3.12/100 women years, respectively (P>0.05). For the expulsion reasons, IUD moving down could account for more than seventy percent. The removal rate for IUD usage of two IUD groups were almost equal (3.91/100 women years verus 4.35/100 women years), the differences were not statistically significant (P>0.05). At the 90th day after abortion, comparing with control group, the bleeding and (or) spotting days of genefix-group and γ-group extended by 3.9 and 2.6 days respectively, the differences had statistical significance between the three groups (P<0.05). Among the bleeding and (or) spotting days, spotting days prolonged significantly. At the 12th month, spotting days of genefix-group andγ-group were (9.2±5.9) days and (8.5±4.6) days respectively, more than (5.2 ± 4.0) days of control group. The differences had statistical significance between IUD with control group (P<0.05), and had not between the two kinds of IUD (P>0.05). Conclusion The insertion of medicated genefix IUD and medicated γ IUD immediately after abortion is safe, feasible, has slight side effects and could be effective contraception.
