1.Design and development of centralized management and sharing system of medical equipment

Objective To solve the problems in medical equipment utilization,supervision,management,clinical safety and etc.Methods A centralized management and sharing system of medical equipment was developed with functional requirements analysis,C/S architecture and programming.Results The system eliminated the needs for manual accounting and allocation,enhanced medical equipment usage rate and improved equipment borrowing procedure.Conclusion The system contributes to dynamic management and efficacy evaluation of medical equipment,so as the working efficiency can be increased while the costs can be decreased for manpower and materials.
2.Construction strategy of nerve conduits bridging long peripheral nerve defects

The repair, regeneration and functional recovery of peripheral nerve defects are the clinical problems needed to be solved urgently.Researches showed that the hollow conduits of bridging nerve defects only had the functions of support and isolation, but the composite conduits could simulate nerve regeneration microenvironment and possess biological activity, which could effectively guide the migration of Schwann cells and the regeneration of axons to achieve satisfactory effects of restoration and functional recovery.With the development of tissue engineering, regenerative medicine, biomaterial and the techniques of cells and molecular biology, the construction of biomimetic and functional nerve conduits to bridge long nerve defects has attracted more and more attention. This review will provide a brief overview of the construction strategy of nerve conduits repairing long nerve defects of peripheral nerves.
3.Proteomic study of myocardial mitochondria with ischemia/reperfusion injury and pinacidil postconditioning in isolated rat hearts

AIM:To investigate the protective effect of pinacidil postconditioning on rat myocardium suffering ischemia/reperfusion injury by mitochondrial proteomics .METHODS: Langendorff apparatus was used to establish the model of myocardial ischemia/reperfusion injury .Sprague-Dawley rats were randomly divided into 2 groups:pinacidil post-conditioning group (Pina group) and ischemia/reperfusion injury group (I/R group).After 20 min of perfusion with K-H solution, the perfusion was suspended for 40-min (global ischemia) follow by 60 min of reperfusion in I/R group.In Pina group at the end of 40 min global ischemia , the isolated hearts were perfused with K-H solution containing pinacidil ( 50μmol/L) for 2 min followed 58-min perfusion with regular K-H solution.Total proteins extracted from the mitochondria were applied to the two-dimensional gel electrophoresis (2-DE).The differentially expressed protein spots over 2 times were evaluated by a software .Then they were subjected to in-gel digestion , and analyzed by spectrometry .RESULTS:The expression levels of NDUFA10, NDUFS2 and NDUFV2 were elevated but those of IDHA and ECH 1 were decreased in Pina group compared with I/R group.Interestingly, 2 spots in the 2-DE map were identified as ATPase subunit δ.The ex-pression levels of one spot was elevated , while the other was decreased .CONCLUSION:Pinacidil postconditioning may decrease the degree of increased expression levels of NDUFA 10, NDUFS2 and NDUFV2, promote the expression of IDHA and ECH1, and induce the phosphorylation of ATPase subunit δ, which may be related to the protective mechanism of pinacidil postconditioning .
4.The expression of estrogen sulfotransferase and steroid sulfatase in formalin-fixed paraffin-embedded endometrioid adenocarcinoma samples

Objective To explore the expression of estrogen sulfotransferase(EST) and steroid sulfatase (STS) in formalin-fixed paraffin-embedded endometrioid adenocarcinoma samples. Methods The RNA of EST and STS in 30 formalin-fixed paraffin-embedded endometrium samples were extracted using Roche products. Results The RNA expression of EST and STS were 0.25±0.03 and 0.08±0.02 respectively and the STS/EST was 0.11±0.08 in normal endometrium. While in endometrioid adenocarcinoma the RNA expression of EST and STS were 0.06±0.02 and 0.24±0.92 respectively and the STS/EST was 4.40±0.64. There were significant differences between these two groups. Conclusions (1) Formalin-fixed paraffin-embedded tissues could be used to study the endometrioid adenocarcinoma. (2) EST is decreased, STS and STS/EST are increased in human endometrioid adenocarcinoma. STS/EST may be related with the prognosis of the endometrioid adenocarcinoma.
5.Treatment of proximal ureteral stones: a comparison study among extracorporeal shock wave lithotripsy, retrograde ureterolithotripsy and percutaneous ureterolithotripsy

Objective To evaluate the effectiveness of extracorporeal shock wave lithotripsy (ESWL), retrograde ureterolithotripsy(URSL) and percutaneous ureterolithotripsy(PCL) in the treatment of proximal ureteral calculi. Methods A total of 397 patients with proximal ureteral calculi treated by ESWL,URSL or PCL ftom September 2001 to December 2005 were retrospectively analyzed. Results Among 397 patients,83 patients with a mean stone size of 1.23 cm were treated by ESW L.Of then.13 patients transferred to URSL or ureterolithotomy and the stone-free rate of ESWL 1 month later was 65.7%(46/70).TWO hundred and thirteen patients with a mean stone size of 1.21 cm were treated by URSL and 101 patients with a mean stone size of 1.50 cm were treated by PCL.The stone-free rate of URSL and PCL 1 month after the treatment was 88.2%(172/195)and 96.9%(95/98),respectively.Eighteen patients in URSL group and 3 patients in PCL group trans-ferred to ureterolithotomy.ESWL had a statistically lower stone-free rate than that of URSL and PCL (P<0.001),both in patients with stone size≤1 cm and>1 cm.For patients with stone size>1 cm,PCL achieved a higher stone-free rate than URSL(P=0.005).PCL also had a higher stone-free rate than URSL in treating patients with stone size≤1 cm but there was no statistical difference between them. Conclusions ESWL can still be used as first-line treatment choice for proximal ureteral stones less than 1cm.For patients with proximal ureteral stones larger than 1cm.URSL and PCL are more proper treatment modalities since they can achieve higher stone-free rate and have acceptable low complications.
6.Osteogenic differentiation of adipose-derived stem cells from rabbits in vitro

[Objective]To observe the differentiation of adipose derived stem cells(ADSCs)into osteogenic cells in rabbit in vitro.[Method]ADSCs were prepared by collagenase Ⅰ digestion of subcutaneous fat from the nape of Japanese white rabbit after being excised and finely minced.Cells were identified by stro-1 immunocy to chemistry,and collagen Ⅰ immunocytochemistry,alkaline phosphatase assay and Von Kossa stain after osteogenic differentiation was performed.[Result](1)ADSCs were stro-1 positive.(2)The expression of collagen Ⅰ,alkaline phosphatase and calcium deposit of ADSCs were all positive under the influence of osteogenic differentiation medium.[Conclusion]ADSCs have similar characteristics to bone marrow stromal cells,but are much more easier to have high number upon harvest and bring less pain will occur during taking ADSCs than taking BMDCs.
7.Effect of concentrations of serum on differentiation of adipose-derived adult stem cells into chondrocytes

[Objective]To study the differentiation of adipose-derived adult stem cells(ADASCs)into chondrocytes induced by TGF-?1 in the presence of different concentrations of serum in vitro.[Method]Adipose tissue-derived adult stem cells were isolated from rabbits subcutaneous adipose tissue using enzymatic digestion.The adipose tissue was minced and digested with collagenase type I,and the released cells were collected by density centrifugation and then placed in culture.After being passaged three times,ADASCs were induced to differentiate into chondrocytes by chondrogenic culture media containing 1% or 10% of FBS for 2 weeks.MTT,alkaline phosphatase toluidine blue staining and type Ⅱ collagen immunohistochemistry staining were then tested in both groups.Gray value was analyzed by Leica patho-image analysis system to observe if there was any difference in differentiative status between the two groups.[Result]MTT showed that the cells of 10% FBS group had higher proliferation activity than those of 1% FBS group(P
8.Effect of reconstituted bone xenografts on tendon-to-bone healing after anterior cruciate ligament reconstruction

[Objective]To investigate the effect of reconstituted bone xenografts(RBX) on tendon-to-bone healing by means of imaging,histological and biomechanical studies.[Method]Anterior cruciate ligament(ACL) reconstruction was performed bilaterally in 25 skeletally mature rabbits using long digital extensor tendon grafts.RBX were implanted into the treated knee of each rabbit,with the contralateral knees as controls.Every three rabbits were killed at 2,6 and 12 weeks postoperatively for routine histological studies.The samples were processed through Micro CT and subsequent HE and toludine blue staining.The remaining 16 rabbits were sacrificed at 6 and 12 weeks.Their femur-tendon graft-tibia complexes were harvested for subsequent mechanical testing.[Result]At 6 and 12 weeks after operation,the values of BMD in the RBX group were higher than those in the control group(P
9.Effects of Bushen Chinese Medicine on Expression of Cbf?1 in Aging Osteoblast of Rat

Objective To observe the expression of core-binding factor ?1 (Cbf?1) in aging osteoblast, and try to provide new mechanisms of Chinese medicine on bone formation. Methods Rat osteoblast from different age donors (6 months and 16 months) were isolated and cultured by the tissue-fragment-migrating-growth method. The biological characteristic was observed by phase-contrast microscope and identified with mineralizing nodule staining. The expression of Cbf?1 was examined by RT-PCR and immunohistochemistry. Results RT-PCR and immunohistochemistry showed that the expression of Cbf?1 was upregulated by Gubenzhuanggu and Jinkuishenqi, and downregulated by Zhibaidihuang in aging osteoblast, but the effect was different on the Cbf?1 expression in Bushenyijing. Conclusion Buyang Chinese medicine among Bushen Chinese medicine promoted bone formation.
10.Assessment of cerebral arteriostenosis with regional cerebral blood flow loss index formula

BACKGROUND: Transcranial Doppler(TCD) spectrum analysis techniques cannot measure the diameters of blood vessels and therefore and not be used to calculate the changes of brain blood flow. A formula was designed for regional cerebral blood flow loss index(rCBFLI) following arteriostenosis when arteriostenosis index formula was deduced; the former may provide a reference to assess cerebral function.OBJECTIVE: To introduce the formula of cerebral blood flow loss ratio obtained basing on TCD results.DESIGN: Retrospective controlled study based on patients with cerebral middle arteriostenosis diagnosed with TCD and healthy people.SETTING: At the departments of neurology in a university hospital and in a military medical university affiliated hospital of Chinese PLA.PARTICIPANTS: Between October 1994 and December 2000,43 patients who were confirmed of arteriostenosis with TCD at outpatient or neurological ward of the First Affiliated Hospital of Fujian Medical University were enrolled into this study,and their TCD results were compared with 908 normal controls between March 1992 and September 2000.METHODS: Stenosis index (STI) formula was deduced from TCD results of 31unilateral and 12 bilateral stenoses at middle cerebral artery(MCA) . STI = 1- [ normal average blood velocity (Vm0) ÷ average blood velocity at stenosisSince(PI1 ÷PI0) may be equal to [post-stenosis blood flow(Q1) ÷ normal blood flow(Q0)],regional cerebral blood flow loss index is concluded as:MAIN OUTCOME MEASURES: rCBFLI,relationship between STI and Vm,as well as between rCBFLI and STI.RESULTS: rCBFLI formula was used to calculate the rCBFLI of 31 patients following unilateral MCA stenosis. rCBFLI of these 31 patients was obtained by Pearson statistical analysis. STI was found to be related to the average blood velocity at stenosis with relative coefficient(Vm) of(r) =0.76( P < 0. 001 )and r = 0. 83 ( P < 0. 001 ) . rCBFLI was proved to be correlated with Vm and STI with r = 0. 76( P < 0. 01 ) and r = 0. 81 in 55 side MCA stenoses,and the difference was of statistical significance( P < 0. 001 ).CONCLUSION: rCBFLI can be used to assess the decrement of regional brain blood flow due to the stenosis of supplying artery; also,it can be used to observe the changes of brain hemodynamics if combined with STI.
