1.Evaluation of Animal Models of Chronic Obstructive Pulmonary Disease Based on Clinical Characteristics in Traditional Chinese and Western Medicine

Chronic obstructive pulmonary disease (COPD) is a common clinical chronic airway disease with high morbidity, high mortality, a heavy disease burden, and complex mechanisms that have not been fully elucidated. Clinical problems promote the continuous progress of basic research. The establishment and evaluation of animal models is an important way to delve into the pathogenesis and treatment strategies of COPD. On the basis of the etiology, pathogenesis, diagnostic criteria, and syndrome differentiation of COPD in traditional Chinese medicine (TCM) and Western medicine, this paper summarizes the establishment methods and characteristics of existing animal models of COPD and analyzes the fitting degree of the models with the disease characteristics in Western medicine and syndrome characteristics in TCM. The results showed that the animal models of COPD in Western medicine were mainly induced by single factors and compound factors, and the model with the highest fitting degree was established by cigarette smoke combined with lipopolysaccharide and hormone. The model showed a fitting degree of 84% with the disease characteristics in Western medicine and 70% with the syndrome characteristics in TCM, being consistent with the clinical characteristics of COPD induced by multiple factors. Most of the animal models of COPD in TCM were established on the basis of disease models and combined with TCM etiology and pathogenesis characteristics, and prepared by syndrome differentiation. Among them, the model of accumulation of cold and water retention in lung had the highest fitting degree of 92% with the TCM diagnostic criteria. The models of phlegm-heat obstructing lung and phlegm-stasis obstructing lung had the fitting degree of 94% with clinical manifestations in Western medicine and the highest fitting degree with the diagnostic criteria in Western medicine. Different animal models of COPD have their own advantages and disadvantages, and most of them simply replicate the manifestations of COPD at a certain stage, failing to reflect the multiple causes and the dynamic changes of TCM etiology and pathogenesis. Moreover, the syndromes of these models fail to match the clinical syndromes in TCM. Therefore, establishing the animal models reflecting clinical characteristics of COPD in TCM and Western medicine and improving the model evaluation criteria are important contents to promote the overall development of integrated traditional Chinese and western medicine for COPD.
2.Problems and Thinking on the Methods for Establishing the Effectiveness Evaluation Index System of Traditional Chinese Medicine

The collection, screening, weighting, and comprehensive evaluation of indicators are key steps in establishing the effectiveness evaluation index system of traditional Chinese medicine (TCM). This article systematically reviewed the research methods in different stages, explored their existing problems, and provide reflections and recommendations. For indicator collection, literature research and expert interviews are generally used, but patient participation is not sufficient; for indicator screening, the Delphi method is the most widely used method, but there is still a lack of criteria and evidence to support indicator screening, and a few studies have applied clinical investigation methods; for indicator weighting, including subjective weighting and objective weighting, but they are mostly the application of a single type of method, and the indicator weights lack reliability; for comprehensive evaluation, analytic hierarchy process, TOPSIS, fuzzy comprehensive evaluation, rank sum ratio, the fuzzy-hierarchy technique for order preference by similarity to an ideal solution, grey relational analysis, and data envelopment analysis have gradually been applied to the study of TCM effectiveness evaluation, however, they often reflect the short-term effectiveness of TCM and can not highlight their long-term effectiveness advantages. Therefore, it is necessary to strengthen the participation of patients and consider their perspectives; in the process of selecting indicators, evidence support for importance judgments should be provided to experts; in indicator weighting, we should integrate subjective and objective data information and actively carry out research on optimizing combination weighting methods; finally, a disease information management platform could be established and a dynamic comprehensive evaluation model could be constructed to provide new ideas and methods for evaluating the effectiveness of TCM.
3.Research and Application Prospect of Organoid Technique in Radiotherapy of Pancreatic Cancer

Pancreatic cancer,as a common malignant tumor of the digestive system,has a very low survival rate.In recent years,pancreatic cancer has made great progress in diagnostic methods,radiation therapy techniques,and systemic chemotherapy,but its therapeutic effect has not been considerably improved.As a new type of tumor research platform,organoids have made research progress in many fields.Constructing pancreatic cancer organoids is of great research value to guide the individualized treatment of pancreatic cancer.This article reviews the research and clinical application prospect of organoid model in radiotherapy of pancreatic cancer.
4.Therapeutic effects of Isaria felina combined with cyclophosphamide in hepatoma H22 tumor-bearing mice

Objective To investigate the therapeutic effects of Isaria felina derived from Cordyceps sinensis combined with cyclophosphamide(CTX)in hepatoma H22 tumor-bearing mice.Methods An H22 tumor-bearing mouse model was established and mice were divided randomly into a normal control group(NC group,distilled water),model control group(MC group,distilled water),positive control group(CTX group,25 mg/kg),Isaria felina group(IF group,400 mg/kg),and combined administration group(IF+CTX group,IF 400 mg/kg+CTX 25 mg/kg),with 5 mice in each group.Distilled water and IF were administered by gavage,and CTX was administered by intraperitoneal injection.The administration cycle was 10 days.At the end of the experiment,the mean tumor volume and weight,tumor inhibition rate,q value,and immune organ index were calculated,and routine blood indexes and cytokine levels were determined.Histopathological changes in tumor tissues were observed by HE staining.Results The tumor volume and mass were significantly lower in mice in each treatment group compared with those in mice in the MC group(P<0.05).The tumor inhibition rates in the CTX,IF,and IF+CTX groups were 49.3%,34.2%,and 72.8%,respectively,and the q value was 1.09.The numbers of white blood cells,Lymph,and platelets were significantly higher in the IF+CTX group than in the CTX group(P<0.05).The spleen index was significantly higher in the MC group compared with that in the NC group,and significantly lower in the IF+CTX group compared with that in the MC group(P<0.05).Serum interferon-γ levels were significantly lower in the MC group than in the NC group,and were significantly higher in the IF and IF+CTX groups compared with those in the MC and CTX groups(P<0.05).Pathologically,tumor cells in the MC group grew well and were numerous and closely arranged,while cells in the CTX,IF,and IF+CTX groups were arranged loosely,with focal necrosis and nuclear pyknosis of necrotic cells in many places.Conclusions The combination of IF and CTX has an additive anti-tumor effect on H22 tumor-bearing mice,which can alleviate immunosuppression and have an immunomodulatory function.
5.Advances in application of organoids to research on radiotherapy of tumors

Organoids are in vitro-cultured three-dimensional (3D) miniature structures derived from human pluripotent stem cells or adult stem cells from healthy individuals or patients. Compared to traditional two-dimensional (2D) cell lines or animal models, organoids are regarded as more promising high-fidelity models, possessing unique advantages in terms of disease modeling, drug development, the establishment of living biobanks, and the exploration of personalized treatment. Over recent years, the rapid development of organoid technology has brought new hopes for innovating preclinical experimental tumor models and promoting clinical personalized diagnosis and treatment. This review is intended to introduce the development status and latest progress of organoids in the field of radiotherapy for tumors, explore the advantages and limitations of organoid models for cancer, and prospect for its application in the field of radiotherapy.
6.miR-206 inhibits the proliferation and invasion of estrogen-induced ER-α36-positive gastric cancer BGC-823 cells by targeting CDK14

[摘 要] 目的:探讨miR-206对雌激素诱导的ER-α36阳性胃癌(gastric cancer, GC)细胞BGC-823增殖和侵袭的影响及其相关机制。方法: 用不同浓度(1、10和100 pmol/L)的雌二醇(estradiol,E2)刺激ER-α36阳性BGC-823细胞后,用qPCR法检测miR-206表达水平,MTT法和Transwell实验分别检测细胞的增殖和侵袭能力,WB法检测细胞中CDK14的表达。将miR-206 mimic、miR-NC、pcDNA-CDK14、pcDNA-vector等转染ER-α36阳性BGC-823细胞,并给予100 pmol/L的E2处理后,用MTT法和Transwell小室法分别检测细胞的增殖和侵袭能力,WB法检测细胞中CDK14的表达。用双荧光素酶报告基因实验验证miR-206与CDK14之间的靶向关系。结果: E2能显著降低ER-α36阳性BGC-823细胞中miR-206表达水平(P<0.05或P<0.01)、增强细胞的增殖和侵袭能力(P<0.05或P<0.01)、上调细胞中CDK14的表达水平(P<0.01)。过表达miR-206能显著降低E2诱导的ER-α36阳性BGC-823细胞的增殖和侵袭能力(均P<0.01)。miR-206通过直接结合CDK14 mRNA的3'-UTR发挥抑制作用,从而负向调节CDK14的表达,进而抑制ER-α阳性BGC-823细胞的增殖和侵袭能力(均P<0.01)。结论: miR-206通过靶向CDK14从而抑制雌激素诱导的ER-α36阳性GC细胞的增殖和侵袭。
7.Bioinformatics analysis of differentially expressed miRNAs and their target genes in growth hormone-secreting pituitary adenoma

Objective:To explore the biological functions of differentially expressed miRNAs and their target genes, and their regulatory relations in growth hormone (GH)-secreting pituitary adenoma.Methods:Four databases (miRDB, miRwalk, Targetscan7.2 and Starbase) were used to predict the target genes of differentially expressed miRNAs in GH-secreting pituitary adenoma, and GO, KEGG and protein-protein interaction network (PPI) analyses were performed on the target genes. Meaningful hub target genes were screened out and compared with mRNA sequencing results from the database; and then, a visual miRNA-target gene interaction network was constructed.Results:Totally, 113 up-regulated miRNAs target genes and 128 down-regulated miRNAs target genes were obtained with the protein interaction degree≥20. Further intersection was conducted to obtain 13 hub target genes; these genes were mainly involved in ubiquitin mediated proteolysis pathway. The regulatory miRNAs of the hub target genes were miR-15b, miR-365, miR-32-3p, and miR-486-5p.Conclusion:A miRNAs-hub target gene mutual regulation network closely related to GH-secreting pituitary adenoma is constructed with bioinformatics analysis tools, and some potential proteins and pathways related to the pathogenesis and pathological process of GH-secreting pituitary adenoma are discovered.
8.Role of Pellino-1 Overexpression on Ubiquitin of Tumor Necrosis Factor Receptor-associated Factors 3 and MAPK Signaling Pathway in Endotoxin-tolerant Kupffer Cells

[Objective]To investigate the effect of pellino-1 gene overexpression by lentivirus vector on the ubiquitin of tumor necrosis factor receptor-associated factors 3 (TRAF3) and the activation of mitogen-activated protein kinases (MAPK) signaling pathway in endotoxin-tolerant kupffer cells (KCs).[Methods]Isolated KCs of C57BL/6 mouse were randomly divided into two groups:control group,which transfected with control lentivirus vector for 48 h,then pretreated with 10 ng/mL lipopolysaccharide (LPS)for 24 h,and next treated with 300 ng/mL LPS;overexpression group,which transfected with pellino-1 gene overexpression lentivirus vector for 48 h,then pretreated with 10 ng/mL lipopolysaccharide(LPS)for 24 h,and next treated with 300 ng/mL LPS. The protein expressions of pellino-1,K48-Ub,TRAF3,p38,p-p38,c-Jun N-terminal kinase(JNK)and p-JNK were analyzed by Western blot. The level of interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α)and interleukin-10(IL-10)in superna-tants were measured by enzyme linked immunosorbent assay(ELISA).[Results]Compared with control group,the protein expres-sions of pellino-1,K48-Ub,p-JNK and p-p38 and the levels of IL-1β(P < 0.05),TNF-α(P < 0.05)in supernatants was in-creased in overexpression group,while the protein levels of TRAF3 and the levels of IL-10(P < 0.05)in supernatants were de-creased.[Conclusion]Overexpression of pellino-1 can promote TRAF3 K48 ubiquitination degradation,decrease the protein expres-sion of TRAF3,activate the downstream MAPK signaling pathway,and thus impair the formulation of endotoxin tolerance.
9.Study on the effect of acupoint massage on vital signs in patients underwent percutaneous coronary intervention surgery

Objective To investigate the effect of acupoint massage on vital signs in patients undergoing percutaneous coronary intervention surgery.Methods128 patients underwent percutaneous coronary intervention surgery from January 2014 to June 2016 were divided into the observation group and the control group according to the random number table, 64 cases in each group.The control group were given traditional surgery,and received routine treatment at postoperative rehabilitation period.At the basis of this, the observation group were given acupoint massage.The liver and kidney function indexes, coagulation function, blood routine, heart rate and blood pressure in the two groups were compared.ResultsThe systolic blood pressure was (142.98±18.35mmHg, the diastolic blood pressure was (92.83±17.28) mmHg and the heart rate was (79.4±11.7) times/min in the observation group;the systolic blood pressure was (143.26±17.26) mmHg, the diastolic blood pressure was (82.65 ± 13.17) mmHg and the heart rate was (71.9±12.1) times/min in the control group, the differences in the two groups was statistically significant(P<0.05).The TBIL of the control group was (9.36±3.71)umol·L-1, DBIL was (4.01±1.14)umol·L-1, TP was (53.23±7.29) g·L-1, and the difference was statistically significant (P<0.05, ALB was (32.10±3.17) g·L-1 and BUN was (4.23 ± 1.32) mmol·L-1, which was lower than that of TBIL, DBIL, TP and ALB in the observation group (P<0.05) The levels of PTA and FIB in the observation group were higher than those in the control group.The levels of PT, APTT and TT were lower than those in the control group, the difference was statistically significant (P<0.05).ConclusionAcupoint massage can effectively enhance the liver and kidney function and coagulation function, reduce the blood pressure and heart rate of patients, relieve anxiety and reduce the recovery time of patients.
10.Investigation and analysis of clinical application of TCM nursing therapies in TCM hospitals in China

Objective To evaluate the clinical application of TCM nursing therapies and identify the problems during the therapy implementation, in order to explore the countermeasures to promote its clinical improvement. Methods A descriptive cross-sectional study was conducted,and a self-designed questionnaire was employed to investigate 218 nurses from 148 grade A class three TCM hospitals in China. Results 25 TCM nursing therapies were implemented in these hospitals. Ear acupressure therapy was the most frequently implemented,accounting for 83.5%(182/218);while wax therapy was the least implemented, accounting for 7.8%(17/218). The implementation frequency of these technologies varied significantly, 0~860 times a week. The top three of them were acupoint application therapy, hot compression of Chinese medicine and ear acupoints therapy.TCM nursing therapies were considered to be the most frequently used in insomnia, headache, edema, arthralgia, cold, epigastric pain, hypochondriac pain and fever.Storage of TCM nurses,lacking of TCM knowledge and inapposite therapy implementation were the main problems in the application. Conclusions Scientific and rational allocation of human resources, cultivation of the TCM-specific nursing professional, enhancement of nurses'awareness of Chinese medicine, establishment of a reasonable charging standard, and strengthening the risk management in implementation will promote its clinical development.
