Objective To analyze the structural and phylogenetic characteristics of the mitochondrial genome from Bulinus globosus, so as to provide a theoretical basis for classification and identification of species within the Bulinus genus, and to provide insights into understanding of Bulinus-schistosomes interactions and the mechanisms of parasite transmission. Methods B. globosus samples were collected from the Ruya River basin in Zimbabwe. Mitochondrial DNA was extracted from B. globosus samples and the corresponding libraries were constructed for high-throughput sequencing on the Illumina NovaSeq 6000 platform. After raw sequencing data were subjected to quality control using the fastp software, genome assembly was performed using the A5-miseq and SPAdes tools, and genome annotation was conducted using the MITOS online server. Circular maps and sequence plots of the mitochondrial genome were generated using the CGView and OGDRAW software, and the protein conservation motifs and structures were analyzed using the TBtools software. Base composition and codon usage bias were analyzed and visualized using the software MEGA X and the ggplot2 package in the R software. In addition, a phylogenetic tree was created in the software MEGA X after sequence alignment with the software MAFFT 7, and visualized using the software iTOL. Results The mitochondrial genome of B. globosus was a 13 730 bp double-stranded circular molecule, containing 2 ribosomal RNA (rRNA) genes, 22 transfer RNA (tRNA) genes, and 13 protein-coding genes, with a marked AT preference. The mitochondrial genome composition of B. globosus was similar to that of other species within the Bulinus genus. Phylogenetic analysis revealed that the complete mitochondrial genome sequence of B. globosus was clustered with B. truncatus, B. nasutus, and B. ugandae into the same evolutionary clade, and gene superfamily analysis showed that the metabolism-related proteins of B. globosus were highly conserved, notably the cytochrome c oxidase family, which showed a significant consistency. Conclusions This is the first whole mitochondrial genome sequencing to decode the compositional features of the mitochondrial genome of B. globosus from Zimbabwe and its evolutionary relationship within the Bulinus genus, which provides important insights for further understanding of the phylogeny and mitochondrial genome characteristics of the Bulinus genus.

Objective To study the changes and mechanism of skin stem cells in microgravity.Methods The skin stem cells of SD rats were used to establish a suspension culture system and compare the proliferation of skin stem cells with 1G gravity.Results The simulated microgravity significantly affected the velocity of skin stem cell sphere proliferation in suspension culture,which was about 12%higher than the 1G gravity group.Transcriptome sequencing showed that 1673 genes were up-regulated and 1409 genes were downregulated;Calcium signaling;cytokine-cytokine receptor interaction and PPAR pathway were different in the two environments.Conclusion Simulation of microgravity can affect the proliferation behavior of skin stem cells in suspension culture by regulating the expression of key signaling pathways,which provides an experimental basis for further research in spatial microgravity environ ment.
9.Action and mechanism of GATA2 overexpression regulating the transcriptome expression of chicken preadipocytes

Objective To clarify the impact of GATA2 overexpression on transcriptional expression in chicken preadipocytes.Methods Chicken preadipocytes ICP1 were cultivated,pCMV-myc-GATA2 or pCMV-myc plasmid was transfected,Western blot was used to verify the overexpression of GATA2 in cells,RNA-seq was used to study the changes in cell transcriptome expression caused by overexpression of GATA2,ChIP-seq was used to study the binding of GATA2 in the genome,Real time PCR was used to verify some differentially expressed genes,ChIP-PCR was used to verify the collection of GATA2 to TAF3 genes.Results Transfection of pCMV-myc-GATA2 allowed overexpression of GATA2 protein in ICP1 cells;overexpression of GATA2 resulted in up-regulation of 942 genes and down-regulation of 840 genes in ICP1 cells(P<0.05),and enrichment analysis showed that differentially ex-pressed genes were associated with ribosome and mitochondrial structure and function(P<0.01);GESA analysis showed that the expression of GATA2-overexpressing ICP1 cells showed up-regulation of RNA polymerase Ⅱ tran-scription initiation complex,ribosome biogenesis,propionate metabolism,and oxidative phosphorylation-related gene sets,and down-regulation of histone lysine N-methyltransferase,nuclear transcription repression complex formation,adipokines,and calcium ions signaling pathway-related gene sets.ChIP-seq in ICP1 cells identified 2833 GATA2-binding genomic peaks involving 2018 genes.motif analysis revealed GATA2-binding(T/A)GATA motifs;enrich-ment analysis showed that these genes are involved in embryonic development,signaling,cellular metabolism,and cell-cell interactions.Differently expressed genes and GATA2-binding genes were taken to intersect to obtain 105 genes,and enrichment analysis showed that these genes were associated with transcription,post-transcriptional regu-lation,cell-cell interactions,signaling,and cell cycle(P<0.001).Conclusion GATA2 can at least bind to the genome of chicken preadipocytes to regulate their transcriptome expression patterns.
10.Establishment and evaluation of a streptozotocin-induced diabetic encephalopathy rat model

BACKGROUND:Animal models of diabetic encephalopathy that have been studied mainly include streptozotocin-induced model,high-sugar and high-fat diet-induced model and spontaneous animal model.Establishing a simple,easy,short-cycle,safe and effective model of diabetic encephalopathy can help to explore the subsequent pathogenesis and screen therapeutic drugs. OBJECTIVE:To further explore and evaluate the method of building diabetic encephalopathy rat models. METHODS:Twenty Sprague-Dawley rats were randomly divided into control(n=10)and model(n=10)groups.Rats in the model group were given a single injection of 45 mg/kg streptozotocin in the left lower abdominal cavity,and those in the control group were given the same amount of citrate buffer.During the experiment,the body mass,feed intake,water intake and blood glucose were measured.After 8 weeks,the glucose tolerance and oxidative stress levels were measured,and the pathological changes of brain tissue and the expression of apoptotic proteins were compared between groups. RESULTS AND CONCLUSION:Compared with the control group,the food intake,water intake,encephalization quotient,blood glucose and area under the blood glucose curve were significantly increased in the model group,while the body mass decreased significantly(P<0.01).Histopathological examination of the brain showed that compared with the control group,the number of surviving nerve cells was significantly reduced in the model group(P<0.01),with more significant pathological damage of nerve cells.Compared with the control group,the activities of serum superoxide dismutase,catalase and glutathione in the model group were significantly decreased(P<0.01),and the content of oxidative malondialdehyde was significantly increased(P<0.05).The expression levels of apoptosis-related proteins Bax and Caspase-3 in brain tissue increased in the model group compared with the control group,while the expression of Bcl-2 decreased(P<0.01).In conclusion,an 8-week injection of 45 mg/kg streptozotocin can cause obvious pathological damage to the brain tissue of diabetic rats,to successfully establish the rat model of diabetic encephalopathy.
