Objective :
To investigate the effects of milk derived pentapeptide prolin⁃glycine⁃proline⁃isoleucine⁃pro⁃
line ( PGPIP) on chronic alcoholic liver injury in mice and its related molecular mechanism .
Methods :
Forty C57BL/6 mice were randomly divided into control group , model group , glutathione(GSH) group , PGPIPN group .The mice model of chronic alcoholic fatty liver was established by 10 d ad libitum oral feeding with the Lieber⁃DeCarli(LD) ethanol liquid diet plus one binge . Drug intervention was given at the same time . Based on the reported RNA sequencing data in gene expression omnibus (GEO) database , the differential expression of liver endoplasmic reticulum stress⁃related genes was analyzed by cluster heat map . Liver hematoxylin⁃eosin (HE) staining was used to analyze the pathological effects of each treatment group on alcoholic liver injury in mice . Oiled Red O staining was used to analyze the effects of each treatment group on the accumulation of liver lipid droplets in mice . Transtransduction protein expression was detected by Western blot .
Results :
The pathological examination of PGPIP group was similar to that of Control group , and the liver injury of mice was significantly reduced . The accumulation of lipid droplets in the liver of mice in the model group was manifested as mixed lipid droplets of different sizes , and PGPIP treatment significantly reduced the accumulation of liver lipid droplets induced by alcohol . PGPIP had significant effects on the PERK⁃eIF2α⁃ATF4 pathway and the expression of transcriptional activator 6 ( ATF6 ), Cleaved Caspase 3 protein .
Conclusion
Pentapeptide PGPIP can alleviate chronic alcoholic fatty liver and liver injury in mice . The mechanism may be attributed to reducing the accumulation of lipid droplets in hepatocytes , en⁃ doplasmic reticulum stress , and hepatocyte apoptosis .

Objective :
To investing ate the milk-derived hexapeptide PGPIPN and its truncated pentapeptide PGPIP to alleviate chronic alcoholic liver injury in mice and its associated molecular mechanisms.
Methods :
Sixty Kun- ming mice were randomly divided into control group, model group,GSH group,PGPIPN group, and truncated pen- tapeptide PGPIP group. The model of chronic alcoholic liver injury in mice was established by gavage with gradient alcohol. Drug intervention was given at the same time for 12 weeks. Liver HE staining was used to analyze the pathological effects of each treatment group on alcoholic liver injury in mice.Primary mouse hepatocytes and human normal hepatocyte line L-02 were isolated and cultured in vitro. The appropriate PGPIPN induction concentrations of both cells were determined by WST-1 method. L-02 cells were induced at different times. The expression of FoxO3a and phosphorylated FoxO3a protein were detected by Western blot to determine the appropriate induction time. The subcellular localization of FoxO3a in L-02 cells was detected by cellular immunofluorescence. The mR- NA changes of FoxO3a and MnSOD genes in primary hepatocytes and L-02 cells of mice in different treatment groups were detected by qRT-PCR.
Results :
The pathological examination of PGPIPN group and PGPIP group was similar to that of GSH group, and the liver injury of mice was significantly reduced.Medium and high concentra- tions of PGPIPN were respectively selected to induce mouse primary hepatocytes and L-02 cells. At 16 hours, the expression of FoxO3a protein in L-02 cells increased significantly.FoxO3a protein was mainly expressed in the nu- cleus. In addition,mRNA levels in both types of cells increased significantly after induction with the corresponding dose of PGPIPN.
Conclusion
PGPIPN and truncated pentapeptide PGPIP can reduce chronic alcoholic liver injury in mice. The mechanism may be to reduce alcohol-induced oxidative stress through FoxO3a-MnSOD signaling path- way.

Ca2+ leak via ryanodine receptor type 2 (RyR2) can cause potentially fatal arrhythmias, and RyR2
mutations have been shown in the aetiology of catecholaminergic polymorphic ventricular tachycardia.
We report the case of a patient with catecholaminergic polymorphic ventricular tachycardia resulting
from a RYR2 mutation who had not only typical electroencephalogram changes, but also epileptiform
discharges in electroencephalogram. We believe the changes were closely related to the RYR2 mutation.

Chinese Journal of Biotechnology 2016;32(8):1133-1144
The expression of dentin sialophosphoprotein (DSPP) is the marker for cells differentiated into odontoblasts. This study attempted to analyze the DSPP promoter and build the reporter LacZ expression system driven by this promoter, which allows convenient and quick detection of odontoblast cells. First, we separated the human dental mesenchymal cells in which the expression of DSPP can be effectively induced by dexamethasone. Second, four 5' flanking regions of human DSPP gene (- 4 000-+54, -2 500-+54, -1 447-+54 and -1 027-+54) were analyzed, the results showed that the highest promoter activity lied in the -2 500-+54 region. The promoter activity is reduced when the 5' flanking region was extended from -2 500 to -4 000 bp upstream from the transcription start site; The promoter activity are also decreased when the 5' flanking regions were shorted from -2 500 to -1 447 bp and from -1 447 to -1 027 bp, indicating that potential suppresser elements are lied in the region between -4 000 and -2 500 bp and potential activator elements are lied in the region between -2 500 and -1 027 bp. Then we constructed the lentiviral report vector phDSPP-LacZ containing the -2 500-+ 54 promoter region in front of the LacZ gene. The expression of LacZ was detected using X-Gal staining in both human dental mesenchymal cells and immortalized human dental mesenchymal cells infected with phDSPP-LacZ. The phDSPP-LacZ lentiviral vector may provide a more convenient method to detect the expression of DSPP in human odontogenic differentiation, tooth development and tooth regeneration studies.

OBJECTIVETo analyze the influence factors of body image dissatisfaction among children and adolescents with normal weight.
METHODSThe primary and middle school students who were selected from Changping district of Bejing city using the stratified cluster sampling method were measured body height, weight, and waist circumference. Body image cognitive attitude of students or their parents was surveyed using 'Ma figural shape'. The cognitive attitude of obesity risk factors was surveyed by self-designed questionnaires. The students with normal weight were selected according to 'reference norm for screening overweight and obesity in Chinese children and adolescents (WGOC) in 2005' and 'reference norm for screening underweight in Chinese children and adolescents aged 6-19 years'. The association between body image dissatisfaction and cognitive attitude of obesity risk factors was analyzed for each gender.
RESULTSThe study validly surveyed and measured 680 students, and there were 36.6% (249/680) students with overweight or obesity, 4.0% (27/680) students with underweight, and 59.4% (404/680) students with normal weight. Prevalence of body image satisfaction, expecting to be thinner, and to be fatter in students with normal weight was 32.7% (132/404), 35.1% (142/404), and 32.2% (130/404), respectively. The prevalence of expecting to be thinner in females and middle students was higher than that in males and primary students (46.4% (102/220) vs 21.8% (40/184)), (39.8% (88/221) vs 29.5% (54/183)), respectively; χ² values were 26.65 and 4.67 respectively (P < 0.05). The prevalence of expecting to be fatter in males was higher than that in females ((42.9% (79/184) vs 23.2% (51/220)) (χ² = 17.91, P < 0.001). The concordance ratio on body image cognitive attitude between students and their parent was 60.4% (244/404), and the consistency coefficient was 0.41 (P < 0.001). The factors including parents expecting their children to be thinner, the negative attitude on less drinking sugary beverages and having breakfast per day might increase the risks of children expecting to be thinner in males, the OR (95% CI) value were 5.20 (1.51-17.89), 3.43 (1.11-10.59), and 6.53(1.14-37.58), respectively. The parents expecting their children to be fatter might increase the risk of children expecting to be fatter in males, the OR (95% CI) value were 7.57 (3.47-16.52). The factors including parents expecting their children to be thinner, and the positive attitude on less eating high calorie snacks might increase the risk of children expecting to be thinner in females, the OR (95% CI) value were 65.74 (8.45-511.21) and 3.03 (1.01-9.09), respectively. The parents expecting their children to be thinner or fatter might increase the risk of children expecting to be fatter in females, the OR (95% CI) value were 17.38 (1.53-197.74) and 9.64 (3.98-23.35), respectively.
CONCLUSIONThe prevalence of body image dissatisfaction among students with normal weight was high, and the concordance ratio on body image cognitive attitude between themselves and their parents was low. Guiding students to have the correct cognitive attitude on less drinking sugary beverages, having breakfast per day, and less eating high calorie snacks may be conducive to prevent body image dissatisfaction.
