1.Data Mining Analysis and Experience Summary of LAO Shaoxian in the Treatment of Gastric Stuffiness(Weipi)

Objective To analyze the medication rules of Professor LAO Shaoxian in the treatment of gastric stuffiness based on syndrome differentiation.Methods The effective prescriptions for patients with gastric stuffiness treated by Professor LAO Shaoxian from March 2017 to March 2022 were collected,and the general information,chief complaints,diagnosis,syndrome differentiation and prescriptions of patients were extracted.Excel software and the ancient and modern medical records cloud platform(V 2.3.7)were used to construct the prescription database.Data mining function was used to carry out analysis of the syndrome type of prescription,analysis of the frequency and property of Chinese herbs,as well as association rule analysis and cluster analysis.Results A total of 272 prescriptions were included,involving 164 kinds of medicinal herbs.The main traditional Chinese medicine(TCM)syndrome types are damp-heat syndrome and qi stagnation syndrome.The frequency of 25 herbs was more than or equal to 30 times.The representative herbs is Pinellinae Rhizoma Praeparatum,Glycyrrhizae Radix et Rhizoma,Citri Reticulatae Pericarpium,Perillae Caulis and Aucklandiae Radix.The medicinal properties are mainly warm and flat.The medicinal flavors are spicy,bitter and sweet.The drug meridians mainly included the spleen,stomach and lung meridians,followed by the liver meridian.There were 23 core drug pairs obtained by association rules,such as"Aucklandiae Radix-Perillae Caulis","Citri Reticulatae Pericarpium-Perillae Caulis",and"Pinellinae Rhizoma Praeparatum-Citri Reticulatae Pericarpium".Clustering analysis of drugs can be divided into three combinations,which have the effects of regulating qi and relieving distension,resolving dampness,and clearing heat and detoxifying.Conclusion The core prescription of Professor LAO Shaoxian in the treatment of gastric stuffiness is Aucklandiae Radix,Perillae Caulis,Citri Reticulatae Pericarpium,Pinellinae Rhizoma Praeparatum,Glycyrrhizae Radix et Rhizoma,Kaki Calyx,Aurantii Fructus Immaturus and Arecae Pericarpium.It focuses on regulating qi movement of middle jiao and treating spleen and stomach simultaneously.The main therapeutic method is regulating qi and relieving distension.At the same time,attention should be paid to the application of dampness-dispelling and stagnation-removing,heat-clearing and detoxifying drugs.The clinical therapy of Professor LAO Shaoxian on gastric stuffiness is significant,which can be used as a reference for diagnosis and treatment.
2.Amplitude of low-frequency fluctuations of behavioral characteristics in different phenotypes of attention deficit hyperactivity disorder

Objective:
To investigate the difference in behavioral characteristics among different phenotypes of attention deficit hyperactivity disorder (ADHD) using amplitude of low-frequency fluctuation (ALFF), so as to provide insights into clinical differentiation of behavioral characteristics among different phenotypes of ADHD.
Methods:
The children with ADHD admitted to The Affiliated Hospital of Hangzhou Normal University were enrolled and classified into the inattentive type (ADHD-I), hyperactive/impulsive type (ADHD-HI) and combined type (ADHD-C). The reaction time (RT) was measured using integrated visual and auditory continuous performance test, and the mean (RT-mean) and standard deviation of RT (RT-SD) were estimated. In addition, the ALFF was calculated at 0.010 to 0.027, 0.027 to 0.073, 0.073 to 0.167 Hz, and the difference of ALFF was compared among children with different types of ADHD.
Results:
A total of 107 children with ADHD were enrolled, including 95 boys and 12 girls, with a mean age of (8.89±1.93) years. There were 69 children with ADHD-I, 8 children with ADHD-HI and 30 children with ADHD-C. The RT-SD was significantly higher among children with ADHD-C than among children with ADHD-I [(126.003±51.619) ms vs. (97.720±45.302) ms; P=0.007]; however, there was no significant difference in RT-mean among children with various ADHD phenotypes (F=1.386, P=0.255). There was an interaction between frequency and ADHD phenotypes (F=2.754, P=0.032), and the ALFF was significantly higher among children with ADHD-C than among children with ADHD-I at 0.010 to 0.027 [(5 590.567±231.595) ms vs. (4 694.001±154.397) ms; P=0.002] and 0.073 to 0.167 Hz [(4 312.609±174.709) ms vs. (3 690.805±116.473) ms; P=0.005].
Conclusions
The ALFF varies in ADHD phenotypes, and there is a frequency-specific difference.
3.Analysis of related factors of postoperative fever in patients with oral and maxillofacial tumor radical treatment and simultaneous repair and reconstruction

Objective:To understand the fever rate, disease distribution, fever degree and fever related factors of the patients in the same period of oral and maxillofacial tumor repair and reconstruction.Method:A retrospective analysis was performed on 153 patients who underwent radical treatment of oral and maxillofacial tumors from January 2018 to December 2018 in the affiliated stomatological hospital of sun yat-sen university. The patients were divided into fever group and non fever group, and their fever influencing factors were analyzed.Result:Among the 153 patients, 97 (63.40%) had fever symptoms.It’s mainly moderate fever. The incidence of fever was the highest in gingival cancer and oropharyngeal cancer.The results of univariate analysis showed that there were statistically significant differences in serum albumin, tracheotomy and infection between the two groups ( χ2= 7.74, 7.48, 8.58, P<0.01 or 0.05).Logistic multivariate regression analysis showed that infection and tracheotomy were independent influencing factors for postoperative fever in patients with oral and maxillofacial tumor repair and reconstruction (or = 4.74, 2.47, P < 0.05). Conclusion:Patients with oral and maxillofacial tumors undergoing radical treatment and simultaneous repair and reconstruction are more likely to have fever after surgery. Tracheotomy patients and patients with infection need to be paid more attention. They need to be strengthened tracheotomy care and actively prevent infection, so as to reduce the postoperative fever rate, and make the patients with oral malignant tumors repaired and reconstructed at the same time go through the perioperative period smoothly.
4.Risk model of breast cancer prognosis based on the expression profile of long non-coding RNA

Objective:To construct a prediction model for the prognosis of breast cancer patients with long non-coding RNA expression characteristics.Methods:To construct a long non-coding RNA(LncRNA) model for predicting the prognosis of breast cancer patients.Methods Analyzing LncRNA expression profiles and clinical characteristics of 1 081 breast cancer patients in the cancer genome atlas (TCGA) database.Performing differential expression analysis and univariate analysis on 112 paired breast cancer and normal breast tissues′ transcriptome sequencing data in the TCGA database, and screened for differentially expressed (DELncRNAs) that significantly correlated with the prognosis of BRCA (To reduce batch effects, sequencing data has been normalized using the DESeq function). One thousand eighty-one breast cancer patients were randomly divided into two groups: training set (541) and validation set (540). Performing Cox proportional hazard regression using DELncRNAs and establishing a multi-LncRNA prognosis model in the training set, followed by proportional hazards assumption test(PH assumption test). Patients were divided into high-risk and low-risk groups based on calculated risk score.Kaplan-Meier method was used for survival analysis, and 540 patients′ data were used for validation.To evaluate the prognostic value of the model in patients with squamous cell carcinoma of the lung and hepatocarcinoma in TCGA database.Gene Set Enrichment Analysis (GSEA) was used to analyze the specific mechanism of lncrna affecting the survival of patients.Results:There were 2815 differentially expressed genes screened by transcriptome sequencing, 91 of which were significantly related to the prognosis of breast cancer patients ( P<0.05). Based on the Cox regression analysis of 91 delncrna expression data from 541 breast cancer patients in training set, a Cox proportional risk regression model was constructed based on 5 LncRNA (training set AUC=0.746, validation set AUC=0.650): AC004551.1, MTOR-AS1, KCNAB1-AS2, FAM230G and LINC01283, and PH assumption test( P=0.388). K-M survival analysis showed that the survival time of high-risk group was significantly worse than that of low-risk group (median survival time: 7.049 and 12.21 years, HR 0.367, 95% CI0.228-0.597, P<0.001), and the survival time of high-risk group was significantly shorter than that of low-risk group (median survival time: 7.57 and 10.85 years, HR 0.412, 95% CI0.214-0.793, P<0.001). Similar prediction results were also obtained in other cancer species of TCGA: lung squamous cell carcinoma ( HR 0.604, 95% CI0.383-0.951, P=0.007) and liver cell carcinoma ( HR 0.551, 95% CI0.307-0.987, P=0.011). GSEA results suggested that the expression patterns of the above five LncRNA were related to the cell cycle regulation of tumor cells. Conclusion:The prognostic model constructed based on expression profile of AC004551.1, MTOR-AS1, KCNAB1-AS2, FAM230G and LINC01283 can be used to predict the prognosis of breast cancer patients, which is helpful to further guide clinical treatment.
5.Ameliorate effect and mechanism of verbascoside on memory impairment in hypoxic mice

Objective To investigate the effects and mechanism of verbascoside on hypoxia-induced memory impairment.Methods The eight-arm maze was used to train mice′s spatial memory ability.After successful training, mice were randomly divided into five groups:a normoxic control group (distilled water, 0.1ml/10g), hypoxic model group (distilled water, 0.1ml/10g), the verbascoside low dose group (50 mg/kg), medium dose group (150 mg/kg), and high dose group (300mg/kg) were administered orally once a day for a total of 7days.After administration on the fourth day, except for the normoxic control group was placed in the animal room (1 500m), the remaining four groups were placed in a large-scale hypobaric chamber to simulate the hypoxic environment of the plateau (7 500m, 3days).Eight-armed maze test (4 000m) was used and the plasma and brain tissues were dissected out and measured for reactive oxygen species (ROS) in the brain, malondialdehyde (MDA), reduced glutathione (GSH) and total superoxide dismutase (T-SOD) activity in plasma and brain.Results Compared with the normoxic control group, the indexes of the eight-armed maze, ROS and MDA in the brain, MDA in the plasma of the hypoxia model group were significantly increased, and the GSH and T-SOD enzyme activities in the brain and plasma were notably decreased.Compared with the hypoxic model group, the indexes of the eight-armed maze, ROS and MDA in the brain, MDA in the plasma in the various groups of verbascoside were reduced more or less, the GSH and T-SOD enzyme activities in the brain and plasma slightly were increased.Conclusion Verbascoside could ameliorate the hypoxic memory impairment at high altitude, which might be related to the stabilization of the body′s antioxidant enzyme system balance and
6.The study on the incidence and risk factors of lactase deficiency in newborns

Objective To study the incidence of lactase deficiency and the risk factors affecting intestinal lactase secretion in newborns with lactase deficiency.Method From February to December 2016,newborns admitted to the neonatal ward of the Affiliated Hospital of Hangzhou Normal University were enrolled in this prospective study.Urine samples were taken within one to two hours after feeding for galactose qualitative tests,and the related clinical data were recorded.The newborns were assigned into lactase deficient group and non-lactase deficient group according to the test results.Then the risk factors of lactase deficiency were analyzed comparing the clinical data between the two groups.Result A total of 1 022 newborns were hospitalized during the research period,of whom 213 were enrolled in this study according to the inclusion criteria.154 cases had positive results in the urine galactose qualitative tests,yielding the incidence of lactase deficiency of 72.3 %.42 cases had lactose intolerance symptoms,and the incidence of lactose intolerance was 27.3 % (42/154).Age and positive family history in lactase deficient group were higher than non-lactase deficient group (10.3 ±6.4 d vs.8.1 ±5.8 d and 23.4% vs.10.2%),while the gestational age of lactase deficient group was lower than non-lactase deficient group (37.8 ±2.9 weeks vs.39.0 ± 1.7 weeks),and the differences between the two groups were statistically significant (P < 0.05).No significant differences existed in gender,birth weight,antibiotics use and feeding volumes between the two groups (P > 0.05).Multivariate Logistic regression analysis showed that age (OR =1.065,95%CI 1.007 ~ 1.127) and positive family history (OR =2.912,95% CI 1.053 ~ 8.056) were the risk factors of lactase deficiency.Gestational age (OR =0.747,95% CI 0.617 ~ 0.904) was the protective factor of lactase deficiency in newborns.Conclusion The incidence of lactase deficiency in newborns is high,but not all the newborns manifest lactose intolerance symptoms.Age and positive family history were the risk factors while gestational age was the protective factor for lactase deficiency in newborns.

This study was aimed to analyze and summarize the intervention rule on traditional Chinese medicine (TCM) external treatment of primary dysmenorrhea,in order to provide theoretical evidences for clinical treatment.Articles on TCM external treatment of primary dysmenorrhea published both at home and abroad were search in the CNKI from January Ft,2014 to January Ft,2017.After standard screening,the frequency analysis,cluster analysis and association analysis were conducted.The results showed that acupuncture was the mostly used TCM external treatment of primary dysmenorrhea,which was far more than other types of TCM external treatments.Theoretical studies on single usie of acupuncture or single use of moxibustion were far more than clinical trials.The acupoint used with the highest frequency in primary dysmenorrhea treatment was SP-6 Sanyinjiao.RN-4 Guanyuan was the second and SP-8 Diji was the third.A total of 22 classic acupoint combinations were identified.It was concluded that the main external intervention method of primary dysmenorrhea was acupuncture with SP-6,RN-4 and SP-8 as its main acupoints.There are a total of 22 classic acupoint combinations
8.Correlation between preoperative CRP/Alb ratio and lymph node metastasis in pa-tients with gastric cancer

Objective:To investigate the relationship between Creactive protein(CRP)/albumin ratio (CAR) and lymph node metastasis of gastric cancer. Methods:A total of 96 cases of gastric cancer were included in the study. The clinical pathological stage and lymph node detection in patients with gastric cancer were analyzed, with the preoperative CAR as the dependent variable. Results:1) The pa-tients with preoperative CAR>0.04 has higher transfer rate and metastasis than the patients with preoperative CAR≤0.04, and the dif-ference was significant (P<0.05). The lymph node stage of patients with preoperative CAR>0.04 was significantly higher than that of the patients with preoperative CAR≤0.04, and the difference was significant (P<0.05). 2) The mean CAR of patients with Borrmann typesⅠ,Ⅱ,Ⅲ, andⅣgradually increased;with the progression of the pathological stage of gastric cancer, the mean value of CAR in-creased. 3) During the operation, the total number of lymph node dissections was high and the mean value of CAR before the opera-tion was high. Conclusion:The correlation between preoperative CAR and lymph node metastasis in patients with gastric cancer may reflect the degree of lymph node metastasis to a certain extent.
9.Study on the Extraction Technology for Active Constituents of Tibetan Medicine Pedicularis kansuensis

OBJECTIVE:To optimize the extraction technology of Tibetan medicine Pedicularis kansuensis and compare con-tent of verbascoside and isoverbascoside differences in P. kansuensis from various habitats. METHODS:Using verbascoside and iso-verbascoside and dry paste yield as comprehensive evaluation indexes,single factor test and orthogonal test were used to investigate the extraction solvent,solvent dosage,extraction time and times to optimize extraction technology,and the verification test was conducted. Contents of the 2 constituents verbascoside and isoverbascoside in P. kansuensis from Gansu,Qinghai and Sichuan were compared. RESULTS:The optimal extraction technology was as follows as 8-fold 50% ethanol,extraction for 3 times,90 min each time. The verification results showed that the average contents of verbascoside and isoverbascoside were 3.49%(RSD=1.28%,n=3),1.26%(RSD=1.32%,n=3),and average dry paste yields were 37.99%(RSD=1.97%,n=3). The contents of verbascoside and isoverbascoside in P. kansuensis from Qinghai were relatively higher. CONCLUSIONS:Optimized extraction tech-nology is reasonable,stable,feasible;the contents of index constituents in P. kansuensis from different habitats have certain differ-ences. The study can provide scientific evidence for the development and utilization of extraction,and the in-depth study of quality evaluation for medicinal material.
10.Phenylethanoid glycosides of Pedicularis muscicola ameliorate high altitude-induced memory impairment by activating mTOR signal pathway

Objective To investigate the mechanism of phenylethanoid glycosides of pedicularis muscicola Maxim ameliorating high altitude memory impairmentby activating mTOR signal pathway.Methods 60 clean male Wistar rats were randomly divided into normoxic control group, hypoxia group, PhGs low, medium and high dose groups(50, 200, 400 mg/kg by oral administration).Normoxic control and hypoxia groups were administered with sterile injection water for 7 days.On the fourth day of drug treatment, hypoxia and PhGs groups were exposed to a specially designed animal decompression chamber, which simulated 7 500 m high altitude environment.The expression levels of mTOR, P70S6K and 4E-BP1 mRNA in hippocampus were detected by SYBR Green real-time PCR.The expression levels of p-mTOR, p-P70S6k and p-4E-BP1 protein in hippocampus were detected by Western blot.Results For hypoxia group rats, mTORand P70S6k mRNA repression, p-mTOR and p-P70S6K protein repression were respectively decreased by 22.50%, 26.00%, 42.28% and 11.70%(P<0.05, P<0.01), 4E-BP1 mRNA repression and p-4E-BP1 protein repression were respectively increased by 41.28%, 111.86%(P<0.01) in comparison tonormoxic control group.Compared with hypoxia group,for PhGs low dose group rats, 4E-BP1 mRNA repression and p-4E-BP1 protein repression were respectively decreased by 77.33% and 82.4%(P<0.01), p-P70S6K protein repression was increased by 32.53%(P<0.01).For PhGs medium, high dose groups, mTOR and P70S6k mRNA repression,p-mTOR and p-P70S6K protein repression were respectively increased by 64.56%, 60.76%;14.86%, 20.27%;65.12%, 94.17% and 56.63%, 78.31%(P<0.01), 4E-BP1 mRNA repression and p-4E-BP1 protein repression were respectively decreased by 72.67%, 71.57% and 57.6%, 40%(P<0.01).Conclusion Phenylethanoid glycosides of Pedicularis muscicola Maxim can ameliorate high altitude-induced memory impairment.This protective mechanism may due to the activation of mTOR signal pathway.
