A virus was successfully isolated from a sick cat exhibiting clinical signs such as oral mu-cosal ulceration, nasal mucosal redness, and increased nasal secretions utilizing F81 cells.Through a comprehensive analysis as such PCR amplication, sequencing, morphology, serology, and animal re-gression tests, the virus was identified as a feline calicivirus and named FCV-BJ,an inactivated vac-cine was developed from this isolated strain its safety and efficacy were assessed. The results re-vealed that the isolated FCV-BJ strain exhibited characteristic serological and morphological fea-tures consistent with caliciviruses.Furthermore, inoculation of cats with the FCV-BJ demonstrated the strain is highly virulent and the cats manifested the clinical signs of feline calicivirus infection. For the vaccination trial, domestic cats were immunized with inactivated fifth-generation virus cell culture at varying dilutions, followed by a booster immunization after 21 days.Fourteen days after the challenge with the virus, cats immunized with 107.0 TCID50/mL or higher remained largely healthy, while all cats in the control group developed clinical signs of FCV.These findings suggest that the inactivated vaccine derived from the FCV-BJ isolate exhibits strong immunogenicity and protective efficacy at a minimum immunization dose of 107.0 TCID50/mL.This strain holds promise as a candidate for vaccine production, providing a valuable reference and foundation for future re-search and development of feline calicivirus vaccines.

OBJECTIVE To establish the method for determination of bile acid active constituents (cholic acid, hyodeoxycholic acid and sodium taurocholate) in Babao jingfeng powder, and to compare the differences of 3 constituents in Babao jingfeng powder from different manufacturers. METHODS UPLC-MS method was adopted to determine the contents of cholic acid, hyodeoxycholic acid and sodium taurocholate in 9 batches of Babao jingfeng powder from 3 manufacturers. The separation was performed on a Phenomenex Luna® C18 column with mobile phase consisted of acetonitrile-water (gradient elution) at a flow rate of 0.5 mL/min. The column temperature was 40 ℃ and the sample size was 2 μL. MS system was operated in a negative ion single quadrupole mode, and the m/z of cholic acid, hyodeoxycholic acid and sodium taurocholate were 407.2, 392.2 and 514.2, respectively. With the contents of three components as the index, cluster analysis and principle component analysis were performed on 9 batches of samples. RESULTS The linear ranges of cholic acid, hyodeoxycholic acid and sodium taurocholate were 5.48- 548.40, 5.38-538.40, 4.74-474.05 ng/mL, respectively (r≥ 0.999 3). RSDs of precision, repeatability and stability tests(24 h) were all lower than or equal to 3.7% (n=6), and the average recoveries were 103.3%, 103.3% and 101.6% with RSDs of 3.3%, 3.4%, 4.2% (n=6), respectively. The contentsof cholic acid ranged 0.702-1.711 mg/g, those of deoxycholic acid ranged 0.599-2.049 mg/g, and those of sodium taurocholate ranged 0.664-1.752 mg/g in 9 batches of samples. The average content of 3 components in samples from manufacturer A was high, and the difference between batches was the smallest; the average content of 3 components from manufacturer C was the lowest, and the difference between batches was large. Cluster analysis could basically distinguish samples from different manufacturers; cluster analysis was conducted by using the comprehensive score of principal components as an indicator, the results of which were basically consistent with those of cluster analysis for content. CONCLUSIONS The method is established successfully for the content determination of the 3 bile acid active constituents of artificial bezoar in Babao jingfeng powder. The contents of 3 components in Babao jingfeng powder from 3 manufacturers are significantly different.

A male patient aged 1 year and 8 months with type 2 spondyloepimetaphyseal dysplasia with joint laxity (SEMDJL2) was reported. The clinical characteristics included short stature, flat middle face, hypotonia, limb joint relaxation, hyperextension of metacarpophalangeal articulation, etc. In addition, the patient had a history of congenital laryngeal stridor. Thus, SEMDJL2 was determined according to the above symptoms and medical history. Sanger sequencing showed that the child carried a c.443C>T missense mutation in the KIF22 gene, which resulted in an amino acid variation namely p.Pro148Leu. This phenotype was preliminarily determined as a pathogenic mutation. Therefore, it is suggested that next-generation sequencing genetic testing could be helpful for genetic diagnosis in children with congenital laryngeal stridor, systemic joint relaxation, and excessive joint extension.

Chinese Journal of Medical Imaging Technology 2020;36(3):448-452
Objective: To explore the feasibility of evaluation on the differences of wrist triangular fibrocartilage complex (TFCC) with T2 mapping in healthy adults. Methods: Totally 81 healthy volunteers underwent T2 mapping of wrist joint. Among them 30 subjects underwent bilateral wrist scanning, and the images were divided into handedness group and non-handedness group according to the handedness questionnaire. All images were postprocessed to obtain T2 mapping pseudo-color images. T2 values of radial cartilage, triangular fibrocartilage disc proper (TFC), fibrous vascular tissue, meniscus and TFC's ulnar attachment were measured. The differences of T2 values in different areas of wrist cartilage were compared between different genders and handedness group and non-handedness group. Results: There were statistically differences of T2 values in different regions of wrist cartilage (F=32.235, P<0.01). There was no statistically difference of T2 values of fibrous vascular tissue ([34.22±6.85]ms) and TFC's ulnar attachment ([32.57±7.23]ms), nor T2 values of radial cartilage ([41.37±7.04]ms) and meniscus ([39.26±8.88]ms) (all P>0.05), while there were statistically differences of T2 values of the remaining areas of carpal cartilage (all P<0.05). There was no significant difference of T2 values of wrist TFCC between different genders nor handedness group and non-handedness group (all P<0.05). Conclusion: T2 mapping can visually display the morphology of wrist TFCC and quantitatively evaluate changes of T2 values in different regions of TFCC, which are not affected by gender and handedness.

Journal of Practical Radiology 2019;35(6):970-973
Objective To investigate the reproducibility of magnetic resonance intravoxel incoherent motion in body diffusionG weighted imaging (IVIMGDWI)for normal lumbar disc scanning. Methods 50 healthy volunteers were enrolled with informed consent,30 males and 20 females,2 5.20±2.04 years old.Using 3.0T MR on the lumbar spine, the sagittal T1 WI,the sagittal, axial T2 WI and sagittal IVIMGDWI sequences were scanned once, then the second sagittal IVIMGDWI sequence was scanned after 4 hours. The IVIMGDWI sequence used 10 b values (0,10,20,40,60,80,100,200,400,600 s/mm2 ).The discs were graded according to the Pfirrmann grading standard. The ADCfast ,ADCslow ,and f values of each intervertebral disc were measured by two doctors at the postGprocessing workstation.Paired tGtest was used to analyze whether there was a difference between the two scans. The intraGgroup correlation coefficient (ICC)was used to analyze the consistency of the ADCfast ,ADCslow ,and f values of the two acquisitions (P<0.05)and the consistency of the ADCfast ,ADCslow, and f values between the different doctors (P<0.05).The IVIMGDWI imaging was evaluated to measure the repeatability of normal lumbar discs. Results Of the 50 healthy volunteers,230 intervertebral discs matched the criteria(Pfirrmann gradeⅠandⅡ).The ADCslowvalue between the two scans was significantly different (t=2.460,P<0.05),and the differences in ADCfast and f values were not significant (t=-0.418,1.273,P>0.05). The consistency of ADCfast ,ADCslow ,and f values for the two scans were generally (ICC=0.478,0.306,0.316,P<0.05 ).Different observers had good consistency in the measurement of interverG tebral disc ADCfast ,ADCslow, and f values (ICC=0.929,0.909, 0.9 1 1 ,P<0.05).Conclusion The IVIMGDWI imaging has good consistency in the measurement of normal lumbar disc between different observers. The consistency of IVIMGDWI in two scans of normal lumbar intervertebral discs is general, which may be due to the time interval between the two scans.Because ADCslow represents the diffusion of water molecules in tissues, the microenvironment in the lumbar intervertebral disc has changed, resulting in the difference of ADCslow value. As for the problem that different scanning time may lead to the change of IVIMGDWI data,we will study it further.
