Objective:To investigate the correlation between cognitive function and addiction, impulsivity, and anhedonia in adolescent depressive disorder patients with self-injury behavior. Methods:From September 2021 to October 2022, adolescents with depressive disorders who visited the outpatient department of the Qingdao Mental Health Center were enrolled and divided into self-injury group and non self-injury group based on the presence or absence of self-injury behaviors, each with 60 participants.A self-compiled general information questionnaire, the 17 items Hamilton depression rating scale (HAMD-17), the Ottawa self-injury inventory (OSI), the Chinese version of the Barratt impulsiveness scale (BIS-11), and the temporal experience of pleasure scale(TEPS) were used to evaluate both groups. The Chinese brief cognitive test(C-BCT) was used to assess cognitive function in both groups.SPSS 26.0 software was used for statistical analysis, including t-test, χ2 test, Pearson correlation analysis, and multiple linear regression analysis. Results:The self-injury group had higher scores for OSI addiction factors (9.43±8.29) and BIS-11 (67.09±11.48) compared to the non self-injury group (OSI addiction factor scores: 0, BIS-11 scores: 53.70±7.12, t=6.22, 5.91, both P<0.05). TEPS score and C-BCT scores in various dimensions were lower in the self-injury group than those in the non self-injury group ( t=-2.93, -2.01, -2.88, -2.20, -5.35, all P<0.05). Information processing speed was negatively correlated with BIS-11 score ( r=-0.296, P<0.05), and attention score were negatively correlated with OSI addiction factor score and BIS-11 score ( r=-0.303, -0.561, both P<0.05) and positively correlated with TEPS score ( r=0.364, P<0.05), including a positive correlation with the scale of anticipatory anhedonia score ( r=0.318, P<0.05). Working memory score was negatively correlated with OSI addiction factor score and BIS-11 score ( r=-0.312, -0.416, both P<0.05). Comprehensive ability and executive function scores were negatively correlated with OSI addiction factor score and BIS-11 score ( r=-0.308, -0.679, both P<0.05), and positively correlated with TEPS score ( r=0.304, P<0.05). Multiple linear regression analysis showed that BIS-11 scores were influencing factors of C-BCT dimensions ( β=-0.260, -0.592, -0.557, -1.797, t=-2.150, -3.314, -2.285, -5.165, all P<0.05). Conclusion:In adolescent depressive patients with self-injury, cognitive function is correlated with addiction, impulsivity and anhedonia, among which impulsivity is a risk factor for cognitive function.

Objective:After using proximal femoral nail antirotation (PFNA) fixation to treat femoral intertro-chanteric fractures (ITFs), closed negative pressure drainage systems are often used for drainage, but the clinical effect of this method is uncertain. A prospective randomized controlled trial study was conducted to analyze the negative effects of negative pressure drainage systems after PFNA fixation. Methods:Sixty patients with ITFs and underwent PFNA fixation were randomly divided into two groups. Patients in the drainage group were equipped with negative pressure drainage systems after PFNA fixation. At 4, 10, and 90 days after the surgery, the visual analog scale (VAS) score, analgesic dosage, thigh swelling width, wound and infection status, hemoglobin, hematocrit level, total blood loss and transfusion blood volume of the patients were recorded to evaluate the prognosis. Results:The total blood loss of the drainage group [(579.1±236.7) ml] was greater than that of the non-drainage group [(427.8±239.8) ml; P=0.01]. The transfusion blood volume of the drainage group [(443.3±176.3) ml] was greater than that of the non-drainage group [(307.8±155.4) ml; P=0.01]. Compared with the non-drainage group, the drainage group had a higher VAS score at 4 days after the surgery (2.3±0.6, P=0.02). There was no difference between the two groups in analgesic dosage, thigh swelling width, wound infection and hematoma, hospital stay and the total number of complications (all P>0.05). Conclusions:In the patients with ITFs treated with PFNA fixation, the transfusion blood volume and total blood loss of the patients in the drainage group are higher than those in the non-drainage group. In addition, drainage systems may not have short-term benefits for postoperative wound conditions.

The TEA domain (TEAD) family proteins (TEAD1‒4) are essential transcription factors that control cell differentiation and organ size in the Hippo pathway. Although the sequences and structures of TEAD family proteins are highly conserved, each TEAD isoform has unique physiological and pathological functions. Therefore, the development and discovery of subtype selective inhibitors for TEAD protein will provide important chemical probes for the TEAD-related function studies in development and diseases. Here, we identified a novel TEAD1/3 covalent inhibitor (DC-TEADin1072) with biochemical IC

Unlike adult mammalian heart, zebrafish heart has a remarkable capacity to regenerate after injury. Previous study has shown Notch signaling activation in the endocardium is essential for regeneration of the myocardium and this activation is mediated by hemodynamic alteration after injury, however, the molecular mechanism has not been fully explored. In this study we demonstrated that blood flow change could be perceived and transmitted in a primary cilia dependent manner to control the hemodynamic responsive klf2 gene expression and subsequent activation of Notch signaling in the endocardium. First we showed that both homologues of human gene KLF2 in zebrafish, klf2a and klf2b, could respond to hemodynamic alteration and both were required for Notch signaling activation and heart regeneration. Further experiments indicated that the upregulation of klf2 gene expression was mediated by endocardial primary cilia. Overall, our findings reveal a novel aspect of mechanical shear stress signal in activating Notch pathway and regulating cardiac regeneration.

Objective To analyse the risk factors associated with spontaneous intestinal perforation (SIP) in extremely premature infants/extremely low birth weight infants. Method From January 2015 to December 2018, infants with gestational age (GA)<28 weeks or birth weight (BW)<1000 g admitted to our neonatal intensive care unit were enrolled to the retrospective nested case-control study. The clinical data of SIP infants (SIP group) and infants with the same GA but without SIP (control group) were randomly selected and compared. Multivariable Logistic regression was used to analyse the risk factors of SIP. Result A total of 409 extremely premature infants/extremely low birth weight infants were born during the study period. Among them, 25 SIP infants and 55 controls were enrolled. The incidence of SIP in infants with GA 22~25 weeks was 11.8%(16/136), which is higher than infants with GA 26~27 weeks (2.0%, 5/247) (χ2=16.057, P<0.001). The incidence of SIP in infants with BW 400~749 g was 13.0%(14/108), which is higher than infants with BW 750~999 g (3.4%, 8/236) (χ2=11.343, P=0.001). Multivariate Logistic regression analysis showed that twins (OR=4.153, 95%CI 1.392~12.384, P=0.011), umbilical veins catheterization (OR=15.942, 95%CI 1.026~247.789, P=0.048) and ibuprofen use within 3 days after birth (OR=15.387, 95%CI 1.519~155.883, P=0.021) were independent risk factors of SIP. Conclusion The smaller the GA and BW, the higher the incidence of SIP. Twins, umbilical veins catheterization and ibuprofen use early after birth may be independent risk factors of SIP.

Objective: To explore the mediating effects of self-focused attention between social anxiety and interpersonal needs. Methods: Totally 467 college students [159 males and 221 females, mean age (21 ±2)] were assessed with the Social Anxiety Scale (SAS), Self-Focused Attention Scale (SFAS) and Interpersonal Needs Questionnaire (INQ ). The bootstrap method was used to test the mediating effect Results: The SFAS and INQ scores were higher in female students than in males (Ps <0.01). The scores of SAS, SFAS, and INQ were positively correlated each other (r = 0.20 - 0.90, Ps < 0.01). Self-focused attention has a full mediating effect between social anxiety and interpersonal needs, and the 95% confidence interval was (0.34, 0.59). Conclusion: It suggests that the effect of interpersonal needs on social anxiety may be achieved through self-focused attention in college students. Reducing the level of self-focused attention may be an important way to alleviate social anxiety among college students.

Aberrant expression of annexin A2-S100A10 heterotetramer (AIIt) associated with PML/RARα fusion protein causes lethal hyperfibrinolysis in acute promyelocytic leukemia (APL),but the mechanism is unclear. To facilitate the investigation of regulatory association between ANXA2 and promyelocytic leukemia/retinoic acid receptor α (PML/RARα) fusion protein, this work was performed to determine the transcription start site of ANXA2 promoter with rapid amplification of S'-cDNA ends analysis.Zinc-induced U937/PR9 cells expressed PML/RARα fusion protein, and resultant increases in ANXA2 transcripts and translational expressions of both ANXA2 and S100A10,while S100A10 transcripts remained constitutive. The transactivation of ANXA2 promoter by PML/RARα fusion protein was 3.29 ± 0.13 fold higher than that by control pSG5 vector or wild-type RARα.The overexpression of ANXA2 in U937 transfected with full-length ANXA2 eDNA was associated with increased S100A10 subunit, although S100A10 transcripts remained constitutive. The tPA-dependent initial rate of plasmin generation (IRPG) in zinc-treated U937/PR9 increased by 2.13-fold,and cell invasiveness increased by 27.6%.Antibodies against ANXA2,S100A10,or combination of both all remarkably inhibited the IRPG and invasiveness in U937/PR9 and NB4.Treatment of zinc-induced U937/PR9 or circulating APL blasts with all-trans retinoic acid (ATRA) significantly reduced cell surface ANXA2 and S100A10 and associated reductions in IRPG and invasiveness.Thus,PML/RARα fusion protein transactivated the ANXA2 promoter to upregulate ANXA2 and accumulate S100A10.Increased AIIt promoted IRPG and invasiveness, both of which were partly abolished by antibodies against ANXA2 and S100A10 or by ATRA.
