1.Metabonomics Combined with Prescription Compatibility Theory to Explore the Lipid-lowering Effect of Qige Decoction Compatibility

Objective To explore the lipid-lowering effect of Qige Decoction before and after compatibility through the combination of pharmacodynamics and liver metabolomics,and to provide new research strategies for exploring the scientific notation of traditional Chinese medicine compatibility.Methods According to the pharmacodynamic strategy,three groups of drug administration were set up as Qige Decoction group,Astragali Radix-Puerariae Radix group,and Pericarpium Citri Tangerinae group.Four indices of blood lipids,serum biochemical indicators,and liver morphology and pathology were used to evaluate the intervention effect of Qige Decoction on hyperlipidemic rats.Liver metabolomics technology was used to analyze the effects of Qige Decoction on metabolites before and after compatibility,and multivariate statistical analysis was used to evaluate the differences between groups in terms of differential metabolites and metabolic pathways.Results Compared with the model group,the callback abilities of four indices of blood lipid in the Qige Decoction group were higher than those in Astragali Radix-Puerariae Radix group and Pericarpium Citri Tangerinae group,among which the total cholesterol(TC)and triglyceride(TG)levels in the Qige Decoction group decreased(P<0.05).A total of 86 potential biomarkers were identified by liver metabolomics,with 23,13,and 7 metabolites being significantly different in the Qige Decoction group,Astragali Radix-Puerariae Radix group,and Pericarpium Citri Tangerinae group,respectively(P<0.05).Metabolic pathway analysis of 29 specific biomarkers with significant callback effects showed that they were related to glycerophospholipid metabolism,linoleic acid metabolism,α-linolenic acid metabolism,sphingolipid metabolism,arachidonic acid metabolism,and unsaturated fatty acid biosynthesis.Qige Decoction mainly regulates glycerophospholipid and linoleic acid metabolism,and uniquely acts on sphingolipid metabolism.Conclusion Qige Decoction has more lipid-lowering targets after compatibility,with better lipid-lowering effects than the Astragali Radix-Puerariae Radix group and Pericarpium Citri Tangerinae group.This study provides experimental evidence and research strategies for further revealing the scientific notation of traditional Chinese medicine compatibility.
2.Application of whole-body magnetic resonance diffusion weighted imaging in evaluating the chemotherapy response for lung cancer

Objective To study the clinical value of whole -body magnetic resonance diffusion weighted imaging(WB-DWI) in evaluating the chemotherapy response for lung cancer,thus to provide evidence for optimizing clinical imaging examination. Methods From October 2017 to May 2018,60 patients with lung cancer confirmed by histopathology in Linfen Central Hospital were selected. The patients underwent DWI examinations before chemotherapy and after two cycles of chemotherapy. The change of tumor size,distant metastasis and apparent diffusion coefficient (ADC) value were compared before and after chemotherapy. The correlation between the change rate of ADC value and the shrinkage rate of tumor size in the effective group was analyzed. Results Of 60 cases,1 case had new cerebral metastases after chemotherapy. There were statistically significant differences in ADC value [(1. 12 ± 0.33) ×10 -3mm2/svs.(1.56±0.40) ×10 -3mm2/s]andtumorsize[(4.63±2.75)cmvs.(2.28±1.45)cm] between before and after chemotherapy in the effective group(t= -3. 954,4. 711,all P<0. 01). There was correlation between the change of ADC value and tumor size(r=0. 34,P<0. 05). Conclusion WB-DWI can not only detect the change of tumor size and distant metastasis quickly and effectively,but also can observe the microscopic changes of tumor cells by measuring ADC value. So it can predict the early therapeutic response of the tumor and make effective evaluation for the staging and chemotherapy response of lung cancer.
3.Extraction Technology Optimization for Qige Granule by Orthogonal Design

Objective:To optimize the extraction technology of Qige granule. Methods:An HPLC method was established to de-termine the contents of astragaloside IV and puerarin in the extract of Qige granules. Based on the orthogonal design of L9(34),three factors including the water amount,decocting duration and extracting times were evaluated. Results:The best extraction process was as follows:adding 12-fold amount of water and extracting 3 times with 30 min for each time. Conclusion:The extraction technology opti-mized by the components evaluation is reasonable and practicable.
4.Effect of electroacupuncture on therapeutic effects and onset time of paroxetine for mild or moderate depression patients

Objective To observe the effect of electroacupuncture on onset time and effectiveness of Paroxetine for mild or moderate depression patients and on the scores of Hamilton depression rating scale for depression(HAMD) and self-rating depression scale(SDS). Methods Fifty-five patients with mild or moderate depression were randomly divided into A group( Paroxetine group,29 patients) and B group (Electroacupuncture combining with Paroxetine group,26 patients). Therapeutic periods for the two groups were 6 weeks. HAMD and SDS were used to evaluate the efficacy before the research and after treatment on 1 week,2 weeks,4 weeks,6 weeks.Results HAMD scores and SDS scores for A group were significant difference after 2 weeks treatment compared with the beginning of the research (P< 0.05 ) ;HAMD scores and SDS scores for B group were significant difference after 1 week treatment compared with the beginning of the research(P<0. 05 ). Total effective rate of HAMD for A group was 71.4% ,total effective rate of SDS for A group was 75% ,total effective rate of HAMD for B group was 88% ,total effective rate of SDS for B group was 84%. The total effective rate between the two groups ordered by the individual chi-square test was significant difference( X2 = 4. 1790, P= 0. 0408). Conclusion Electroacupuncture can shorten the onset time and increase the effectiveness of paroxetine for mild or moderate depression patients.
5.Determination of Human Serum Strychnine Concentration by Reversed-Phase HPLC with Solid Phase Extraction

[ Objective ] To establish a method for the determination of serum strychnine concentration in rheumatoid arthritis patients treated with Tongbiling prescription and to monitor the serum strychnine concentration. [ Methods ] Serum strychnine was extracted with solid phase extraction column. Reversed-phase HPLC conditions were: Hypersil Division ODS C18 column (150 mm ?4.6 mm, 5?m), mobile phrase being methanol: water (70 : 30) with 0.6 g potassium dihydrogen phosphate and 1.0 g dodecyl sodium sulfonate added in 1000 mL mixture (adjust pH to 4.5 with acetic acid), detecting wavelength at 254nm and a flow rate of 1.0 mL/min at room temperature. [Results] Strychnine has a good linearity in the range of 0.031- 2.000?g?mL-1. The minimum detecting concentration was 0.01?g?mL-1 and the average recovery was over 90%. Intra-day RSD and inter-day RSD ranged from 1.53% to 5.32%. [Conclusion] This method is simple and efficient, and can be used for the determination of strychnine.
6.Study on Quality Standards for Shenchang Injection

OBJECTIVE:To study the quality standards for Shenchang injection.METHODS:TLC was applied to identify the properties of Borneol and volatile oil of Rhizoma Acori Tatarinowii in the formula.HPLC was adopted to determine the content of ?-asarone in Shenchang injection.RESULTS:?-asarone showed good linear relationship in the range of 0.632 3~ 1.264 6? g.The average recovery of ammonium ?-asarone was 99.92%(RSD=0.91%).CONCLUSION:The standard can be used for the quality control of Shenchang injection.
7.Determination of ?-Asarone and ?-Asarone in Xing Nao Nasal Drops by HPLC

Objective: To determine the contents of ?-Asarone and ?-Asarone in Xing Nao nasal drops. Methods: HPLC was used with ODS C18 column (150mm?4.6mm, 5?m). Methanol mixture : water being 6 : 4 and Patassium biphoepate 1.4 and Dodecyl Sulphonic acid sodium salt 1.2g in 1000mL mixture served as mobile phase, detection wavelength at 257nm and flow rate being 1.0mL/min. Results: The mean recovery was 99.38 %(RSD=2.25 %) for ?-Asarone and 96.59 %(RSD=2.16) for ?-Asarone. Conclusion: this method is simple,rapid and accurate.
8.Content Determination of Brucine and Strychnine in Tongbiling Tablet by Reversed-phase HPLC

[Objective] To establish a method for the determination of brucine and strychnine contents in Tongbiling Tablet. [Methods] Reversed-phase HPLC conditions were: Nucleodur C18 Gravity column (125 mm ? 4.6 mm, 5?m), mobile phrase being methanol-water (60:40) with 0.5 g potassium dihydrogen phosphate and 1.0 g dodecyl sodium sulfouate and 0.5 mL triethylamine in 1 000 mL mixture (adjusting pH to 4.5 with acetic acid) , detecting wavelength at 254 run and a flow rate of 1.0 mL/min at room temperature. [ Results ] The average recovery of brucine was 97.69 % ( RSD = 2.03%) and that of strychnine 97.93% ( RSD = 2.61%). [Conclusion] This method is efficient and can be used for the quality control of Tongbiling Tablet.
9.Study on HPLC Fingerprint of Amino Acids Composition in Buzhong Yiqi Pill

Objective To establish the HPLC fingerprint of amino acids compounds from Buzhong Yiqi Pill.Methods A pre-column derivatization HPLC method was adopted with praline as a reference.Separation was performed on a AccQ? TagTM C18(3.9? 150 mm)analytical column with mobile phase consisting of 0.15 mol/L sodium acetate buffer solution and 60 % acetonitrile,gradient elution with the flow rate of 1.0 mL/min.The column temperature was at 37 ℃.The Fluorescence detecting wavelength was set at 250 nm(excitation wavelength)and 395 nm(emission wavelength),the analysis time was 35 min.Results Twelve common peaks on the HPLC fingerprints of Buzhong Yiqi Pill were presented.The similarity was determined by the correlation coefficient,included angle cosine coefficient and evaluation software of chromatogram fingerprint similarity.The results showed the similarity for 10 bathes of samples was in 0.97~ 1.00.Conclusion Good fingerprints were obtained,which can be used for the quality control of Buzhong Yiqi Pill.
