1.Current Research on Modulation of NF-κB Signaling Network by Traditional Chinese Medicine to Intervene in Rheumatoid Arthritis

		                        			Rheumatoid arthritis (RA) is a chronic systemic autoimmune disease with complex pathogenesis, poor cure rate, long course of disease, and high disability rate, which seriously affects patients' quality of life. Western medicine treatment of RA mainly includes non-steroidal anti-inflammatory drugs, rheumatic drugs, glucocorticoids, biologics, and targeted small-molecule drugs, which have large side effects and many adverse reactions. Traditional Chinese medicine (TCM) has systematic, comprehensive, multi-target, and multi-mechanism advantages in the treatment of RA. Through the overall syndrome pattern identification, it is effective in relieving symptoms, delaying onset, and relieving pain in RA patients by dispelling wind and removing dampness, dissipating cold and dredging collaterals, clearing heat and removing toxin, resolving phlegm and removing blood stasis, and relaxing sinew and activating collaterals. In recent years, TCM has made remarkable progress in the intervention of RA by regulating the nuclear transcription factor-κB (NF-κB) signaling network. They include Toll-like receptor 4 (TLR4), phosphoinositol 3 kinase (PI3K)/protein kinase B (Akt), mitogen-activated protein kinase (MAPK), Nod-like receptor protein 3 (NLRP3), nuclear factor E2 associated factor 2 (Nrf2), Janus kinase 2 (JAK2)/signaling and transcriptional activator 3 (STAT3), and Toll-like receptor 2 (TLR2). The regulatory mechanism of NF-κB signaling network is complex: TLR4 is the upstream receptor of NF-κB, PI3K/Akt and MAPK pathways can not only regulate the activity of NF-κB, but also serve as its downstream effect pathway, Nrf2 and NF-κB often have antagonistic effects in the regulation of inflammatory response. Therefore, the research progress of regulating NF-κB signaling network by traditional Chinese medicine in the intervention of RA was summarized to provide a reference for the treatment of RA disease and the development of new drugs. 
		                        		
		                        		
		                        		
		                        	
2.Traditional Chinese Medicine Intervention in Signaling Pathways Related to Benign Prostatic Hyperplasia: A Review

		                        			Benign prostatic hyperplasia (BPH) is a common chronic progressive disease in middle-aged and elderly men, characterized by prostate enlargement and bladder outlet obstruction, leading to symptoms such as frequent urination, urgency, and difficulty urinating. The pathogenesis of BPH involves factors such as aging, hormonal metabolic abnormalities, inflammatory responses, and imbalances in cell proliferation and apoptosis. Currently, the main treatment methods for BPH include medication, physical therapy, and surgical intervention. However, medication may cause side effects like sexual dysfunction and hypotension, physical therapy has limited efficacy, and surgery carries risks and postoperative complications. Therefore, there is an urgent need to find safer and more effective treatment options. Traditional Chinese medicine (TCM), with its focus on treatment based on syndrome differentiation and a holistic approach, offers therapeutic advantages through multiple pathways and mechanisms. Recent studies have shown that TCM regulates pathways such as phosphoinositide-3-kinase/protein kinase B (PI3K/Akt), nuclear factor-κB (NF-κB), mitogen-activated protein kinases (MAPK), nuclear factor E2-related factor 2/antioxidant response element (Nrf2/ARE), androgen receptor (AR), transforming growth factor-β (TGF-β)/Smad, and hypoxia-inducible factor-1α/vascular endothelial growth factor (HIF-1α/VEGF) to inhibit oxidative stress and inflammatory response, reduce prostate cell proliferation, and promote apoptosis, thus exerting therapeutic effects. This article summarizes and analyzes the roles of these signaling pathways in the occurrence and development of BPH and the mechanisms of TCM intervention, aiming to provide scientific evidence for clinical treatment and drug development for BPH.

4.Improvement effects and mechanism of Bazheng powder on chronic urinary tract infection in rats

		                        			OBJECTIVE To investigate the improvement effects and its mechanism of Bazheng powder on chronic urinary tract infection (CUTI) induced by Escherichia coli in rats. METHODS The rats were divided into normal control group, model group, levofloxacin group (45 mg/kg) and Bazheng powder group (4.95 g/kg), with 10 rats in each group. Except for the normal control group, other groups were administered an intravesical injection of Escherichia coli suspension (1×10⁸ cfu/mL) via the urethra to establish CUTI model; at the same time, rats in each group were administered the corresponding medicinal solution or water by gavage once a day for 4 consecutive weeks. After the last medication, blood routine tests (white blood cell count and lymphocyte percentage), the levels of serum inflammatory factors [interleukin-1β (IL-1β), IL-6, IL-8, tumor necrosis factor-α (TNF-α)], and immune indicators [CD4, CD8, secretory immunoglobulin A (SIgA)], renal function indicators [cystatin C (Cys-C), α1- microglobulin (α1-MG), urea and creatinine] were all determined; the pathological changes in renal and bladder tissues in rats were observed. The protein expressions of Toll-like receptor 4 (TLR4), nuclear factor-κB (NF-κB), and nucleotide-binding domain leucine-rich repeat and pyrin domain-containing receptor 3 (NLRP3) in rat bladder tissues were detected. RESULTS Compared with the normal control group, the levels of IL-1β, IL-6, IL-8, TNF-α, CD8, Cys-C, α1-MG, urea and creatinine in serum, as well as the protein expressions of TLR4, NF-κB and NLRP3 in bladder tissues, were significantly elevated in the model group (P<0.05). Conversely, the levels of CD4 and SIgA were significantly decreased (P<0.05). Pathological changes, such as extensive infiltration of inflammatory cells, were observed in both renal and bladder tissues. Compared with the model group, the above quantitative indicators in the Bazheng powder group were significantly improved (P<0.05), with no obvious inflammatory lesions observed in either renal or bladder tissues. CONCLUSIONS Bazheng powder can alleviate inflammatory reaction and improve the immune function of CUTI rats, and its mechanism may be related to the inhibition of TLR4/NF-κB/ NLRP3 signaling pathway.
		                        		
		                        		
		                        		
		                        	
5.Optimization of alcohol precipitation process for Quhan Zhufeng Granules by overall desirability and central composite design

		                        			Objective:To optimize the alcohol precipitation process of Quhan Zhufeng Granules.Methods:Taking the volume fraction of ethanol, the relative density of the concentrated solution and the standing time as the investigation factors, the Z value of the total evaluation of gentiopicrin content, oleanolic acid content and dry paste yield of gentiana macrophylla as the evaluation indexes, the star-point design-response surface method was used to optimize the alcohol precipitation process.Results:The optimal the alcohol precipitation process of Quhan Zhufeng Granules: concentration relative density 1.08 g/ml (90-95 ℃), alcohol precipitation at the end of volume fraction of 62% ethanol, standing for 16 h.Conclusion:The alcohol precipitation process using overall desirability and central composite design is stable and feasible, and has good predictability, which can provide experimental basis for further scale production.
		                        		
		                        		
		                        		
		                        	
6.Simultaneous Determination of 9 Components in Shujin Tongluo Black Plaster by HPLC

		                        			Objective To establish an HPLC-based method for the determination of α-obscurine,ferulic acid,hydroxysafflower yellow A,benzoylneaconitine,benzoylaconitine,periplosin,4-methoxysalicylsalicylate,kaempferol and oleanolic acid simultaneously in Shujin Tongluo Black Plaster.Methods HPLC method was used to determine the 9 components in Shujin Tongluo Black Plaster simultaneously.The 70%methanol extracts were analyzed using Waters SunFire C18 chromatography column(4.6 mm×250 mm,5 μm),in mobile phase containing acetonitrile-0.2%phosphoric acid solution for gradient elution with volume flow rate of 1.0 mL/min;column temperature was set at 25℃;detection wavelength was set at 230 nm.Results The 9 components had good linear relationship in their respective ranges(r≥0.999 7).The average recoveries were between 98.06%-100.56%and RSD was between 0.48%-2.56%,respectively.Conclusion The method has the advantage of repeatability,simple and fast,and can be used as the quality control of Shujin Tongluo Black Plaster.
		                        		
		                        		
		                        		
		                        	
7.Optimization of Phenylphthalein and Phenolic Acid Components Extracts from Angelicae Sinensis Radix-Chuanxiaong Rhizoma Decoction Pieces by Central Combination Design Response Surface Methodology

		                        			Objective To optimize the optimal extraction process for phthalein and phenolic acid components in Angelicae Sinensis Radix-Chuanxiaong Rhizoma decoction pieces.Methods On the basis of a single factor experiment,central combination design-response surface methodology was adopted,and the extraction time,ethanol concentration,and ethanol dosage were used as influencing factors,and the total normalized values of the content of Senkyunolide I,Senkyunolide A and ligustilide,and extract yield were used as evaluation indicators to optimize the extraction process of phthalein components in Angelicae Sinensis Radix-Chuanxiaong Rhizoma decoction pieces;the total normalized values of chlorogenic acid,caffeic acid,ferulic acid,and extract yield were used as evaluation indicators to optimize the extraction process of phenolic acids components in Angelicae Sinensis Radix-Chuanxiaong Rhizoma decoction pieces.Results The optimal extraction process was to add 7 times the amount of 90%ethanol to the phthalein components,extract for 130 minutes each time,and extract twice;phenolic acid components were extracted twice with 7.5 times the amount of 65%ethanol for 120 minutes each time.Conclusion The optimized extraction process for phthalein and phenolic acids in Angelicae Sinensis Radix-Chuanxiaong Rhizoma decoction pieces is stable and feasible,which can provide a basis for subsequent research.
		                        		
		                        		
		                        		
		                        	
8.Extraction process optimization for preparation of Ziyin Qinggu Pills by overall desirability and response surface methodology

		                        			Objective:To optimize the extraction process of Ziyin Qinggu Pills.Methods:The solvent amount, extraction time and soaking time were taken as the influencing factors, and the overall desirability (OD) of the contents of timosaponin BⅡ, paeoniflorin, calycosin 7-O-glucoside, liquiritin and the dry extract yield were taken as the evaluation indexes, so that the optimal extraction process of Ziyin Qinggu Pills was optimally selected by the centralized composite design-response surface method.Results:The optimal extraction process of Ziyin Qinggu Pills was as follows: extraction time of 115 min, soaking time of 26 min, solvent volume of 10 times, and extraction twice.Conclusion:The optimal extraction process is reasonable, stable and reliable, which can provide the basis and foundation for the extraction process and further molding process of Ziyin Qinggu Pills.
		                        		
		                        		
		                        		
		                        	
9.Ultrasonic characteristics and histopathologic comparison of vascular remodeling after carotid balloon injury in rats

		                        			Objective:To detect the characteristics of vascular remodeling after carotid balloon injury model in rats using ultrasound biomicroscopy(UBM), and to discuss the application value of UBM technique by comparing ultrasonic characteristics with histopathological results.Methods:Carotid balloon injury was performed in 10-week-old SD rats(11 female and 11 male) by 2F Fogarty balloon catheter. The left common carotid artery(CCA) was injured and the right side in the same animal was used as an uninjured control. Arterial structures and hemodynamics were evaluated pre-procedure and post-procedure at 7, 14 days.The intima-media thickness(IMT) inner diameter, outer diameter, lumen area, vessel area, peak systolic velocity, end diastolic velocity of CCA were measured by UBM, and the vascular resistance index, shear stress and blood flow were calculated to evaluate the vascular hemodynamics. The histological data were obtained by H&E staining in cross-sections at 14 days after balloon injury. The characteristics of arterial structure and hemodynamic changes at various time points were compared, the structural changes of CCA between injured and control side after injury were compared. The Spearman correlation and linear regression were used to test the correlation between ultrasonic and histological measurements 14 days after balloon injury.Results:①Compared with pre-procedure, the IMT at 14 days after balloon injury was increased, the inner diameter was decreased, the shear stress in ultrasound was increased(all P<0.05). H&E staining histological test showed that IMT and neointima area in male rats were larger than those of female rats (all P<0.001). ②After carotid balloon injury, the lumen area decreased, but the CCA underwent compensatory positive remodeling and the vessel area increased. ③Significant correlations were demonstrated between UBM and histology in IMT, inner diameter, outer diameter and vessel area of CCA( rs=0.819, 0.965, 0.896, 0.955; all P<0.001). The vessel area value measured by UBM was larger than that of histology( P=0.006). Conclusions:The CCA of rats can be showed clearly by UBM in males and females. The arterial structure cab be measured by UBM accurately with good correlation with histology, as did arterial hemodynamic parameters, which may be benefit for the study in carotid balloon injury model of rats.
