1.Danggui Shaoyaosan Regulates Nrf2/SLC7A11/GPX4 Signaling Pathway to Inhibit Ferroptosis in Rat Model of Non-alcoholic Fatty Liver Disease

ObjectiveTo investigate the effect of Danggui Shaoyaosan on ferroptosis in the rat model of non-alcoholic fatty liver disease (NAFLD) and explore the underlying mechanism based on the nuclear factor E2-related factor 2 (Nrf2)/solute carrier family 7 member 11 (SLC7A11)/glutathione peroxidase 4 (GPX4) signaling pathway. MethodsThe sixty SD rats were randomly grouped as follows: control, model, Yishanfu (0.144 g·kg-1), and low-, medium-, and high-dose (2.44, 4.88, and 9.76 g·kg-1, respectively) Danggui Shaoyaosan. A high-fat diet was used to establish the rat model of NAFLD. After 12 weeks of modeling, rats were treated with corresponding agents for 4 weeks. Then, the body weight and liver weight were measured, and the liver index was calculated. At the same time, serum and liver samples were collected. The levels or activities of total cholesterol (TC), triglycerides (TG), alanine aminotransferase (ALT), aspartate aminotransferase (AST), and Fe2+ in the serum and TC, TG, free fatty acids (FFA), malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GPX), and Fe2+ in the liver were measured. Hematoxylin-eosin staining and oil red O staining were employed to observe the pathological changes in the liver. Immunofluorescence was used to assess the reactive oxygen species (ROS) content in the liver. Mitochondrial morphology was observed by transmission electron microscopy. The protein levels of Nrf2, SLC7A11, GPX4, transferrin receptor 1 (TFR1), and divalent metal transporter 1 (DMT1) in the liver were determined by Western blot. ResultsCompared with the control group, the model group showed increases in the body weight, liver weight, liver index, levels or activities of TC, TG, ALT, AST, and Fe2+ in the serum, levels of TC, TG, FFA, MDA, Fe2+, and ROS in the liver, and protein levels of TFR1 and DMT1 in the liver (P<0.01), and decreases in the activities of SOD, GPX and the protein levels of Nrf2, SLC7A11, and GPX4 in the liver (P<0.05, P<0.01). Meanwhile, the liver tissue in the model group presented steatosis, iron deposition, mitochondrial shrinkage, and blurred or swollen mitochondrial cristae. Compared with the model group, all doses of Danggui Shaoyaosan reduced the body weight, liver weight, liver index, levels or activities of TC, TG, ALT, AST, and Fe2+ in the serum, levels of TC, TG, FFA, MDA, Fe2+, and ROS in the liver, and protein levels of TFR1 and DMT1 in the liver (P<0.01), while increasing the activities of SOD and GPX and the protein levels of Nrf2, SLC7A11, and GPX4 in the liver (P<0.01). Furthermore, Danggui Shaoyaosan alleviated steatosis, iron deposition, and mitochondrial damage in the liver. ConclusionDanggui Shaoyaosan may inhibit lipid peroxidation and ferroptosis by activating the Nrf2/SLC7A11/GPX4 signaling pathway to treat NAFLD.

Osteoporosis is a chronic skeletal disease characterized by low bone mass, destruction of bone tissue microarchitecture, and imbalance of bone homeostasis, leading to increased bone fragility and increased risk of fractures. Oxidative stress caused by the disruption of the balance between excess reactive oxygen species (ROS) and the anti-oxidative system is an important factor in the occurrence and progression of osteoporosis. Nuclear factor E2-related factor 2 (Nrf2)/heme oxygenase-1 (HO-1) is an important anti-oxidative stress pathway. Nrf2 is a primary factor in regulating cellular oxidative stress. Activating Nrf2 can stimulate the expression of HO-1. HO-1 is a key enzyme whose metabolites are bile green Oxygen, carbon monoxide, and free iron. The metabolites can scavenge ROS, thereby exerting an antioxidant effect in cells. At present, domestic and foreign scholars have reported that the Nrf2/HO-1 signaling pathway is closely related to the occurrence and development of osteoporosis and the mechanism of drugs. Chinese medicine can effectively solve the insufficiency of western medicine with multi-target, multi-channel, and multi-level advantages. Chinese medicine can resist oxidative stress, inflammatory response, and apoptosis by regulating the Nrf2/HO-1 signaling pathway, thus treating osteoporosis. This article reviewed the relationship between Nrf2/HO-1 signaling pathway and its key target protein factors and osteoporosis, to clarify the important role of the Nrf2/HO-1 signaling pathway in osteoporosis. At the same time, a systematic summary of Chinese medicines targeting and regulating the Nrf2/HO-1 signaling pathway for the treatment of osteoporosis was conducted, to provide a theoretical basis for further precise treatment of osteoporosis.
3.Study on the effect of relaxation training in the treatment of patients with obsessive-compulsive disorder

Objective:To explore the effect of relaxation training in the treatment of patients with obsessive-compulsive disorder.Methods:From January 2017 to June 2018, 90 patients with obsessive-compulsive disorder were admitted to the fourth ward of the Mental Health Center of Shanghai Jiaotong University School of Medicine. According to the random number table method, they were divided into the control group and the intervention group with 45 cases each. The control group was given medication, and the intervention group used relaxation training on the basis of the control group. The obsessive-compulsive disorder scores, quality of life, depression, and anxiety of the two groups were compared before and after treatment.Results:The obsessive thinking and compulsive behavior scores of the control group after treatment were (14.02±3.12) and (15.01±2.45) points; the intervention group was (11.91±3.01) and (12.23±2.34) points after treatment, and lower than the control group ( t values were 3.265, 5.504, P<0.05 or 0.01). The quality of life score of the control group after treatment was (283.64±51.28) points; the intervention group was (305.67±49.28) points after treatment, and the intervention group was higher than the control group after treatment (t value was -2.078, P<0.05). After treatment, the scores of the depression scale and anxiety scale of the control group were (12.35±2.89) and (15.35±4.03) points; the intervention group was (8.05±1.72) and (10.35±3.65) points after treatment, and the intervention group after treatment all lower than the control group ( t values were 8.577, 6.169, P<0.01). Conclusion:Using relaxation training combined with conventional drugs to treat patients with obsessive-compulsive disorder can improve obsessive-compulsive symptoms, improve quality of life, and relieve negative emotions.
4.Impacts of carvedilol combined with pravastatin on NT-proNBP cTnI and cardiac function in coronary heart disease patients with chronic heart failure

Objective To investigate the impacts of carvedilol combined with pravastatin,on aminoterminal pro-brain natriuretic peptide(NT-proNBP),cardiac troponin Ⅰ(cTnI)and cardiac function in coronary heart disease patients with chronic heart failure.Methods One hundred and tewnty five cases of coronary heart disease patients with chronic heart failure were randomly divided into the carvedilol group(63 cases)and carvedilol combined with pravastatin group(62 cases).In addition to using certain dosage of the above-mentioned drugs respectively,both groups underwent routine anti heart failure treatment with the course of 12 weeks.The class of heart function and changes of heart rate(HR) were observed before and after treatment.Left ventricular end diastolic diameter(LVEDD),left ventricular end systolic diameter(LVESD) and left ventricular ejection fraction(LVEF) were determined by using ultrasound heartbeat graph.Six min walk test(6MWT)was also observed before and after treatment and the level of NT-proBNP and cTnI level were determined by using an enzyme immunoassay method.And patients readmission rates and the incidence of cardiovascular events were also observed.Results The condition of carvedilol and pravastatin group were improved after treatment compared with before treatment[HR:(78±12) CICCS/min vs(100±112) CICCS/min,t =13.682,P < 0.05 ;LVEDD:(43±5)mmvs(53±8)mm,t=5.284,P<0.01;LVESD:(42±6)mmvs(56±7)mm,t=6.454,P<0.01;LVEF:(50±5)% and(35±8)%,t=-6.091,P<0.01);NT-proBNp:(986±713)ng/Lvs (3328±1109) ng/L,t =17.626,P < 0.05) ; CInI:(0.85±0.16) μg/L vs(2.03±0.63) μg,/L,t =5.879,P < 0.01 ;6MWT:(355.6±92.5)m vs(238.8±101.4) m,t =-8.255,P < 0.01].After 3 months follow-up,the condition of carvedilol combined with pravastatin group were better than carvedilol group;LVEDD:(43±5)mm vs(57±6)mm,t =5.892,P <0.05 ;LVESD:(42±6)mm vs(49±7) mm.t =3.243,P <0.01 ;LVEF:(50±5) % vs(42±8) %,t =-12.036,P < 0.01 ; NT-proBNP:(986±713) ng/L vs(1626±968) ng/L,t =3.603,P <0.01 ;cTnI:(0.85±0.16) μg/L vs(1.15±0.36) μg/L,t =3.200,P < 0.01 ;6MWT:(355.6±92.5) mvs(296.2±99.5) m,t =-10.119,P < 0.01].The rehospitalization rate(3.3 % vs 12.7%,x2 =6.224,P < 0.05) and the incidence of cardiovascularevents(3.3% vs 15.9%,x2 =5.974,P < 0.05) were both decreased Significantly after treated with carvedilol combined with pravastatin.Conclusion Carvedilol combined with pravastatin can reduce the level of NT-proBNP and cTnI,improve heart function in coronary heart disease patients with chronic heart failure.
5.Effects of Fosinopril vs Amlodipine on brachial ankle pulse wave velocity and pulse pressure in patients of primary hypertension

A total of 560 patients with primary hypertension were divided into two groups.Of them,260 were treated with Fosinopril(10~20 mg once daily),and the other 300 were treated with Amlodipine (5~10 mg once daily).The treatment lasted for 12 months in both groups.The levels of blood pressure (BP)、braehial ankle pulse wave velocity(baPWV)and pulse pressure(PP)were examined at baseline,6 month and 12 month with automatic analysis system of arteriosclerosis.BP in Amlodipine group(132/76 mm Hg)decreased more significantly than Fosinopril group(136/85 mm Hg),but Fosinopril group showed more remarkable changes in baPWV(1310 cm/s)and PP(52 mm Hg)than Amlodipine group(1400 cm/s and 56 mm Hg).
6.Research on Developing Law of Pathogeny and Mechanism of Chronic Hepatitis Type B

Objective To discuss the developing law of pathogeny and mechanism of chronic hepatitis type B.Methods Patients of chronic hepatitis type B,early hepatocirrhosis and ascites caused by hepatocirrhosis was chosen as objects and clinical information was collected face to face.Patterns of Chinese medicine were confirmed and counted seperately according to efficiency,deficiency and efficiency-deficiency sydrome,harmed Zang-Fu organs and syndrome factors.Finally,the developing law of pathogeny and mechanism was analyzed.Results As the state of chronic hepatitis type Bevolving,simple efficiency and deficiency sydrome decrease with efficiency-deficiency sydrome increasing.Zang-Fu organs damaged by chronic hepatitis type B including mostly liver,gallblader,spleen,stomach,kidney,Trijiao and so on,among which spleen and liver are mainly damaged.As the state of chronic hepatitis type B evolving,the frequencies of damage to gallblader,spleen,stomach,kidney,Trijiao and so on increase.Most simple chronic hepatitis type B cases are due to Shi,Shi-Re,efficiency and stasis of Qi,while most of early hepatocirrhosis stem from Shi-Re,extravasated blood,efficiency and stasis of Qi,with that of ascites caused by hepatocirrhosis originating from Shi-Re,extravasated blood,Shui,efficiency of Qi and Yin. Conclusion There is developing law in the character,location and syndrome factors of chronic hepatitis type B.
7.Clinical significance of plasma tissue factor pathway and urokinase-type plasminogen activator system in cancer patients.

OBJECTIVETo evaluate variations in the plasma tissue factor (TF) and urokinase type plasminogen activator (u-PA) system and their relationship with clinical cancer type, pathological classification and metastatic status in cancer patients.
METHODSPlasma levels of TF and its inhibitor (TFPI), as well as u-PA and its receptor (u-PAR) were measured using ELISA in 76 patients with malignant tumors and 24 patients with benign tumors.
RESULTSPlasma levels of TF and u-PAR in the malignant tumor group were significantly higher than those of the benign tumor group and the normal control. U-PA and u-PAR increased significantly in patients with esophageal and gastric cancer. However, most of these parameters except TFPI did not vary according to pathological classification. A significant elevation was evident in patients with local infiltration, lymph node involvement and distal metastasis, while u-PAR only increased in the latter two categories.
CONCLUSIONSBoth the TF and u-PA systems are activated in cancer patients. U-PA and its receptor might prove to be a clinically useful marker for disease progression.

8.Quantitative assessment of coronary flow reserve by the variables of time-intensity curve with myocardial contrast echocardiography.

The reliability and reliable indexes of quantitative assessment of coronary flow reserve (CFR) by using time-intensity curve (TIC) via myocardial contrast echocardiography were investigated. The TIC variables were obtained by employing acoustic densitometry (AD) technique before and after acetylcholine (Ach) injection in 12 dogs. Meanwhile, the correlation between these variables and CFR was analyzed. Among the variables derived from TIC, peak intensity (PI), area under the curve (AUC) and descending slope (DS) were increased significantly (P < 0.05) with the increase of coronary blood flow after Ach injection. Conversely, time-to-peak (TP), half-time of descent (HT), and mean-transit-time (MTT) were decreased remarkably (P < 0.0001). The PI and AUC ratios from post- to pre-Ach injection were strongly associated with CFR with the correlation coefficient (r) being 0.8366 and 0.8824, respectively. It is reliable by using the variables derived from TIC with myocardial contrast echocardiography to quantitatively evaluate regional myocardial CFR. The PI and AUC ratios from post- to pre-Ach injection are the reliable indexes for quantitative assessment of CFR.
