2.Effects of Tangtong Formula on RSC96 Schwann Cells Apoptosis and Proliferation Induced by High Glucose

		                        			Objective To screen the optimal dose of Tangtong Formula in vitro. Methods RSC96 Schwann Cells were cultivate by DMEM mediums which contains different concentrations of glucose (5–125 mmol/L). The prevention effects of Tangtong Formula at different concentrations (0.25, 0.5, 0.75, 1.0, 1.5, 2.0, 3.0, 5.0 mg/mL) on the proliferation of RSC96 Schwann Cells induced by high glucose were detected. After the RSC96 Schwann Cells were cultivated in 100 mmol/L and 125 mmol/L high glucose mediums for 72 h, the apoptosis of RSC96 Schwann Cells was detected by flow cytometry Annexin V/PI, and the apoptosis rate was calculate; the proliferation situation of RSC96 Schwann cells in different times was detected by CCK-8 method. Results RSC96 Schwann cells were in apoptosis after being intervened by 100 mmol/L and 125 mmol/L high glucose mediums. The apoptosis rates were respectively(7.46±0.96)% and(16.53±1.01)%, with statistical significance compared with control group (P<0.01). Different concentrations of Tangtong Formula could alleviate the inhibitory effect of high glucose on the proliferation of RSC96 Schwann cells, and the threshold concentration of Tangtong Formula in 24 h was 0.25 mg/mL. The concentrations of Tangtong Formula in 0.5 mg/mL, 1.0 mg/mL, and 1.5 mg/mL could inhibit the apoptosis of RSC96 Schwann cells induced by high glucos, compared with 125 mmol/L high glucose group, the apoptosis rate of RSC96 Schwann cells decreased significantly (P<0.05, P<0.01). Conclusion Among three different doses, when the dose of Tangtong Formula is in 1.5 mg/mL, the effects on inhibiting apoptosis are the best.
		                        		
		                        		
		                        		
		                        	
3.Mechanism of salvianolate regulating activity of NLR family pyrin domain containing 3 inflammasome in treatment of mice with colitis

		                        			Objective To study the role of salvianolate in the treatment of inflammation of intestinal mucosa by colitis mice model.Methods A total of 60 C57BL/6J mice were divided into acute control group,acute model group,acute interventional group,chronic control group,chronic model group and chronic interventional group with 10 mice in each group.Dextran sodium sulfate (DSS) was used to establish the model.Meanwhile,the mice of interventional group received salvianolate peritoneal injection during modeling.The expression levels of NLR family pyrin domain containing 3 (NLRP3),apoptosis-assoeiated speck-like protein containing CARD (ACS) and cysteinyl aspartate specific proteinase 1 (caspase-1) were detected.And the absorbances of serum superoxide dismutase (SOD) and malonaldehyde (MDA) in mice serum were determined.Kruskal Wallis single factor analysis of variance,single factor analysis of variance,Student-Newman-Keuls (SNK) method and least-significant difference method were performed for statistical analysis.Results The expressions of NLRP3 at protein level of acute control group,acute model group,acute interventional group,chronic control group,chronic model group and chronic interventional group were 9 965.20 (196.81),16 703.38 (291.21),13 423.74 (209.28),10 112.01 (183.55),16 247.90 (505.44) and 12 674.95 (229.32),respectively.The expressions of NLRP3 of acute and chronic interventional group were both lower than those of acute and chronic model group,and the differences were statistically significant (both x2 =15.158,P<0.01).The expressions of ACS at protein level were 9 744.09-±-546.58,16 555.44 ± 407.80,12 568.21±586.49,10066.32±435.63,14 911.73±384.51 and 13 751.37±322.30,respectively.The expressions of caspase-1 at protein level were 5 300.40±843.14,15 478.98± 174.09,10 587.46±821.60,5 517.28±876.16,13 164.01 ±416.47 and 8 856.27 ± 545.01,respectively.The expressions of ACS and caspase-1 of acute and chronic interventional groups were both lower than those of model groups,and the differences were statistically significant (F=260.56,329.36,240.38 and 236.26,all P<0.01).The serum levels of SOD were (282.08±28.24),(221.87±5.81),(231.45±1.84),(267.08±43.63),(237.09±34.09) and (257.78 ± 4.68) U/mL,respectively.There was no significant difference between acute model group and acute interventional group (P>0.05).SOD level of chronic interventional group was higher than that of model group,and the difference was statistically significant (t=-2.65,P =0.021).The serum levels of MDAwere (1.31±0.40),(6.95±1.57),(2.98±1.41),(1.21±0.16),(9.38±3.73)and (3.74±0.81) nmol/L,respectively.MDA levels of acute and chronic interventional groups were lower than those of acute and chronic model groups,and the differences were statistically significant (t=4.88 and 4.02,both P<0.01).Conclusion Salvianolate may alleviate oxidative stress response by adjusting the activity of NLRP3 inflammasome which takes part in reducing intestinal inflammation.
		                        		
		                        		
		                        		
		                        	
4.EBM methodology-based analysis of surgical time influencing factors

		                        			Objective To improve the utilization ratio of operating room in Affiliated Hospital of Central South Uni-versity Xiangya Medical School by providing more comprehensive reference data for the design of surgical time pre-diction and dispatch system. Methods A questionnaire was designed according to the review of literature and consul-tation of experts for investigating the surgical time influencing factors. The surgical time influencing factors were ana-lyzed by stratified sampling. Results The surgeons-related factor was the highest influencing factor while the pa-tients themselves-related factor was the lowest influencing factor in the 2-dimensional factors. The selected 38 1-dimensional factors could affect the operating time with their mean influencing value>2 . 45 . The recognition of sur-gical time influencing factors was different in different operating rooms. Conclusion There are a variety of surgical time influencing factors. However, the surgeons-related factor is the highest influencing factor. 5.Study on inhibition role of SFRP2 on cervical cancer cell line proliferation

		                        			Objective To understand the expression change of SFRP2 in human cervical cancer tissue and to investigate the effect of SFRP2 on cervical cancer cell proliferation.Methods The expression of SFRP2 in cervical cancer tissue was detected by using Western blot and qRT-PCR;the SFRP overexpressed human cervical cancer line was constructed by using lentivirus,the effect of SFRP2 on the proliferation of human cervical cancer cell line was analyzed by CCK-8 and plate cloning.The effect of SFRP2 on the expression of WNT pathway related proteins and genes in human cervical cancer cell was detected by Western Bolt and qRTPCR.Results Compared with paracancerous tissue,SFRP2 was lowly expressed in human cervical cancer tissue;overexpressed SFRP2 cervical cancer cell proliferation was inhibited;SFRP2 inhibiting cellular proliferation was occurred via WNT signal pathway.Conclusion The role of SFRP2 as a candidate gene for cervical cancer remains to be deeply studied.
		                        		
		                        		
		                        		
		                        	
6.Effect of bone marrow mesenchymal stem cells transfected by Zrt/Irt-like protein 1 on adipogenic differentiation via tumor necrosis factor alpha signaling pathway

		                        			BACKGROUND:Previous studies have confirmed that ethanol can promote adipogenic differentiation of bone marrow mesenchymal stem cells (BMSCs), and up-regulate the expression of PPARγ and aP2 in the tumor necrosis factor α (TNF-α) signaling pathway. As a member of the ZIP protein family, Zrt/Irt-like protein 1 (ZIP1) is closely related to bone metabolism and osteogenic differentiation.OBJECTIVE:To study the effect of BMSCs transfected by ZIP1 on TNF-α signaling pathway in the process of adipogenic differentiation.METHODS:The BMSCs from rabbits were isolated and cultured under different concentrations of alcohol (0.03, 0.09,0.15, 0.21 mol/L), followed by transfection by ZIP1 siRNA and ZIP1 expression vector.RESULTS AND CONCLUSION:After culture in alcohol, the expression levels of aP2 and PPARγ proteins were both significantly increased (P < 0.05), and the level of triglyceride was increased in all alcohol groups except for 0.03 mol/L alcohol group (P < 0.05). After siRNA transfection, the expression levels of aP2 and PPARγ as well as the level of triglyceride were increased significantly in all the alcohol groups (P < 0.05); however, ZIP1 transfection decreased the expression levels of aP2 and PPARγ proteins (P < 0.05). To conclude, ZIP1 siRNA could promote the adipogenic differentiation of BMSCs through the activation of TNF-α signaling pathway.
		                        		
		                        		
		                        		
		                        	
7.Expression changes of DNA repair enzymes and DNA damage in diabetes rat hearts

		                        			Objective To identify if there was increased DNA damage in cardiomyocytes of rats with diabetic cardiomyopathy and also to evaluate the change in gene expression of DNA repair enzymes 8-oxoguanine DNA glycosylase 1(OGG1) and AP endonuclease 1(APE1).Methods Total DNA,RNA and proteins of hearts were isolated in diabetes rat hearts.DNA damage was examined with quantitative polymerase chain reaction(Q-PCR) assay.mRNA and protein expressions of OGG1 and APE1 were detected with real time-polymerase chain reaction(RT-qPCR) and Western blot analysis.The levels of 8-hydroxy-2′-deoxy-guanosine(8-OHdG) in DNA were studied with ELISA.ResultsThe damage of mtDNA was increased in rat hearts(P<0.05) and there was not significant damage in nDNA.The amount of 8-OHdG in DNA was significantly increased(P<0.05),mRNA and protein expressions of OGG1 and APE1 were increased in diabetic rat hearts(P<0.01).Conclusions The damage of mtDNA increased in diabetic rat hearts.Although the expression of OGG1 and APE1 increased in diabetic rat hearts,which fails to repair the damage of the mtDNA.The increased mtDNA damage may contributes to myocardium damage.
		                        		
		                        		
		                        		
		                        	
8.Analysis of the effect that nursing students acting as social volunteers to participate in gerontological nursing practice teaching

		                        			Objective To compare the effect of nursing students acting as social volunteers to participate in gerontological nursing practice teaching with traditional practice teaching mode, and promote the reform of teaching mode in nursing practice. Methods Students of two classes from department of nursing, medical college of Wuhan University of Science and Technology were selected as research objects. In class one nursing students acted as social volunteers to participate in gerontological nursing practice teaching (the experimental group), In class two nursing students took practice in geriatric wards according to traditional practice mode(the control group). The practice effect was compared between two groups. Results The average final grade of the experimental group was 86.50, and 78.51 in the control group. Ridit analysis revealed that, setting class one as the experimental group, R=0.796 8,95% confidence interval was 0.715 1-0.878 4, in the control group, R =0.500 0. The investigation showed that the experimental group and the control group were different in moral elevation aspect, teaching level, capability improving aspect and employment promotion aspect, the difference was significant (P<0.01). Conclusions The mode of nursing students acting as social volunteers to participate in gerontological nursing practice teaching has advantage over traditional practice teaching, and it can gain satisfying teaching effect.
		                        		
		                        		
		                        		
		                        	
9.Clinical analysis on juvenile dermatomyositis combined with soft-tissue calcification

		                        			Objective To analyze the clinical features of juvenile dermatomyositis (JDM) combined with soft-tissue calcification. Methods Forty-seven patients with JDM combined with soft-tissue calcification (soft-tissue calcification group) were retrospectively analyzed, and they were contrasted with 89 patients with non-calcification (non-calcification group). Results The rates of Gotton signe, muscle contracture and joint dysfunction in soft-tissue calcification group were significantly higher than those in non-calcification group:87.23% (41/47) vs. 43.82% (39/89) and 68.09% (32/47) vs. 21.35% (19/89), and there were statistical differences (P<0.05). The dosage of glucocorticoid (conversion of prednisone measuring more than 1.5 mg/kg), rate of using immunodepressant, level of creatine kinase in soft-tissue calcification group were significantly lower than those in non-calcification group:17.02%(8/47) vs. 68.54%(61/89), 25.53%(12/47) vs. 88.76%(79/89), (566.45±240.41) U/L vs. (1 680.12±656.50) U/L, and there were statistical differences (P<0.05). Conclusions The patients with JDM combined with Gotton signe are more prone to soft-tissue calcification. The rate of muscle contracture and joint dysfunction in soft-tissue calcification patients is significantly higher than that in non-calcification patients. For the patients whose creatine kinase are not obviously elevated, they are more prone to soft-tissue calcification. Early active application of glucocorticoid and immunodepressant therapy can reduce or prevent the occurrence or development of late calcium deposition.
		                        		
		                        		
		                        		
		                        	
10.Analysis and identification of B cell epitopes of the OMP18 from Campy lobacter jejuni

		                        			In this study ,we aimed to understand the sequence characteristics ,transmembrane structures ,line B cell epitopes present in the OMP18 from Campylobacter jejuni ,and provide candidate antigens for the antibody detection and vac-cine development .NCBI/Blast ,TMHMM Server V2 and DNA Star softwares were used for the OMP18 sequence analysis . Based on the ELISA ,the whole bacterial antibody IgG of Campylobacter jejuni was used for the identification of the predicted line B cell epitopes .The OMP18 gene was found conserved in different Campylobacter jejuni strains .The OMP18 was predic-ted to be located on the outer surface of the bacteria .And three line B cell epitopes were determined to be present in the OMP18 protein .As a conclusion ,the OMP18 protein was confirmed to be an important outer membrane protein ;three line B cell epitopes were identified in the OMP18 ,which could be further used for Campylobacter jejuni antibody detection and vaccine development .
