1.Establishment and verification of a prognostic nomogram for survival of tongue squa-mous cell carcinoma patients who underwent cervical dissection

		                        			Objective:To evaluate the prognostic significance of inflammatory biomarkers,prognostic nutritional index and clinicopathological characteristics in tongue squamous cell carcinoma(TSCC)patients who underwent cervical dissection.Methods:The retrospective cohort study consisted of 297 patients undergoing tumor resection for TSCC between January 2017 and July 2018.The study population was divided into the training set and validation set by 7:3 randomly.The peripheral blood indices of interest were preoperative neutrophil-to-lymphocyte ratio(NLR),lymphocyte-to-monocyte ratio(LMR),platelet-to-lymphocyte ratio(PLR),systemic immune-inflammation index(SII),systemic inflammation score(SIS)and prognostic nutritional index(PNI).Kaplan-Meier survival analysis and multivariable Cox regression analysis were used to evaluate independent prognostic factors for overall survival(OS)and disease-specific survival(DSS).The nomogram's accuracy was internally validated using concordance index,receiver operating characteristic(ROC)curve,area under the curve(AUC),calibration plot and decision curve analysis.Results:According to the univariate Cox regression analysis,clinical TNM stage,clinical T category,clinical N category,differentiation grade,depth of invasion(DOI),tumor size and pre-treatment PNI were the prognostic factors of TSCC.Multivariate Cox regression analysis revealed that pre-treatment PNI,clinical N category,DOI and tumor size were independent prognostic factors for OS or DSS(P<0.05).Positive neck nodal status(N≥1),PNI≤50.65 and DOI>2.4 cm were associated with the poorer 5-year OS,while a positive neck nodal status(N≥1),PNI≤50.65 and tumor size>3.4 cm were associated with poorer 5-year DSS.The concordance index of the nomograms based on independent prognostic factors was 0.708(95%CI,0.625-0.791)for OS and 0.717(95%CI,0.600-0.834)for DSS.The C-indexes for external validation of OS and DSS were 0.659(95%CI,0.550-0.767)and 0.780(95%CI,0.669-0.890),respectively.The 1-,3-and 5-year time-dependent ROC analyses(AUC=0.66,0.71 and 0.72,and AUC=0.68,0.77 and 0.79,respec-tively)of the nomogram for the OS and DSS pronounced robust discriminative ability of the model.The calibration curves showed good agreement between the predicted and actual observations of OS and DSS,while the decision curve confirmed its pronounced application value.Conclusion:Pre-treatment PNI,clinical N category,DOI and tumor size can potentially be used to predict OS and DSS of patients with TSCC.The prognostic nomogram based on these variables exhibited good accurary in predicting OS and DSS in patients with TSCC who underwent cervical dissection.They are effective tools for predicting sur-vival and helps to choose appropriate treatment strategies to improve the prognosis.
		                        		
		                        		
		                        		
		                        	
2.Analysis of medication and glycemic control of patients with type 2 diabetes under chronic disease management in Xi'an pharmacies

		                        			Objective To investigate the medication and blood glucose control of type 2 diabetes patients under chronic diseases management in Xi'an chain pharmacies,and provides reference for improving the management policy of diabetes at grassroots level.Methods A number of chain pharmacies in the sixth district of Xi'an were selected by random sampling method,and on-site interviews were conducted by questionnaire survey to patients with type 2 diabetes under the management of chronic diseases.The basic information of patients,medication status(medication plan,drug adherence,etc.),diabetes-related conditions(blood glucose status,family history,course and complications,etc.)were collected.Multivariate logistic regression was used to analyze the relevant factors of blood glucose control in patients.Results A total of 403 patients were surveyed,the largest number of patients use oral hypoglycemic drugs alone(53.4%),followed by insulin medication(including insulin only and insulin in combination with oral hypoglycemic drugs)(35.7%),and the differences between disease course and glycemic control among patients with different drug regimens were statistically significant(P<0.05).Only 43.7%of patients had good medication compliance.In addition,the patient's fast plasma glucose compliance rate was only 39.2%.The results of multivariate logistic regression analysis showed that good medication compliance(OR=1.744,95%CI 1.104 to 2.754,P=0.017)were independent influencing factors for achieving glycemic control.Conclusion The medication compliance of type 2 diabetes patients with poor blood glucose control in chronic disease management of chain pharmacies in Xi'an needs to be strengthened.Pharmacies should emphasize and give full play to the professional and service advantages of pharmacists to realize the functional role of pharmacies and strengthen diabetes management.
		                        		
		                        		
		                        		
		                        	
3.Metagenomic next-generation sequencing-based retrospective investigation of the drug resistance sites of Mycoplasma pneumoniae in children

		                        			Objective:To analyze the drug-resistant gene loci of Mycoplasma pneumoniae (MP) using metagenomic next-generation sequencing (mNGS). Methods:From November 2022 to October 2023, 697 clinical samples (including sputum, alveolar lavage fluid and blood) of 686 children with Mycoplasma pneumoniae positive detected by mNGS were retrospectively analyzed. Samples were divided into intensive care unit (ICU) group and non-ICU group, Chi-square test was used to compare groups, and Mann-Kendall trend test was used to analyze the change trend of the detection rate of drug resistance gene loci over time. Results:Of the 697 samples, 164 were from the ICU group and 533 were from the non-ICU group. The detection rate of Mycoplasma pneumoniae resistance gene was 44.3% (309/697), and all detected drug-resistant gene loci of MP were A2063G. The detection rate of Mycoplasma pneumoniae in ICU group was 50.0% (82/164), and the detection rates of Mycoplasma pneumoniae resistance gene loci in sputum, alveolus lavage fluid and blood samples were 75.0% (18/24) and 48.4% (62/128), respectively. The detection rate in sputum was higher than alveolus lavage fluid samples ( χ2=5.72, P=0.017). The detection rate of Mycoplasma pneumoniae in non-ICU group was 42.6% (227/533), the detection rate of Mycoplasma pneumoniae resistance gene loci in sputum and alveolar lavage fluid was 40.0% (16/40), 44.3% (201/454), and no detection rate in blood samples (0/12). There was no significant difference in the detection rate of alveolar lavage fluid and sputum ( χ2=0.27, P=0.602). From November 2022 to October 2023, the detection rate of submitted samples showed an increasing trend month by month (overall: Z=3.99, ICU inspection group: Z=2.93, non-ICU group: Z=3.01, all P<0.01). Among the bacteria commonly detected with Mycoplasma pneumoniae, Streptococcus pneumoniae accounted for the highest proportion, the detection rate was 15.5% (108/697), and Epstein-Barr virus accounted for the highest proportion of 17.6% (123/697). Conclusions:From November 2022 to October 2023, the detection rate of Mycoplasma pneumoniae drug resistance gene loci showed an increasing trend. The detection rate of drug resistance gene loci in sputum samples of ICU group was higher than alveolus lavage fluid. 4.Relationship between eyebrow and eye aesthetics after correction of aponeurotic ptosis

		                        			Objective:To evaluate the specific changes and importance of the proportions of the eyebrows and eyes after surgery by analyzing and measuring the changes of the values of the eyebrows and eyes in elderly patients with aponeurotic ptosis before.Methods:A total of 54 cases (108 eyes) of elderly patients with aponeurotic ptosis who were treated in our department from 2019.01 to 2020.12 were collected preoperatively and 6 months postoperatively, and they were set as the study group. At the same time, the data of 48 cases (96 eyes) of healthy people with good face were collected and set as the young control group, and the data of 50 cases (100 eyes) of elderly healthy people without ptosis were collected and set as the elderly control group. The corresponding objective data were analyzed, and the specific changes and proportional relationship of eyebrows and eyes after surgery were evaluated.Results:Compared with the preoperative data, the marginal reflex distance 1 (MRD1), the height of the palpebral fissure (HPF) in the study group increased at 6 months after the operation, and the distance between the eyebrow and eye at each site increased or shortened ( P<0.05). Six months after the operation, the brow height from medial canthus (MBH), the central brow and eye distance (brow height from upper eyelid margin on midpupillary level, CBH), and the lateral brow and eye distance (brow height from lateral, LBH) had been changed. The change rates of canthus compared with preoperatively were that (88.29±11.69)%, (85.04±14.96)%, and (93.61±13.56)%, respectively, among which CBH shortened most significantly, and the shortened distance was (3.76±2.43) mm. The MRD1 and HPF in the group increased significantly at 6 months after operation, which were significantly greater than those in the elderly control group ( P<0.05). CBH was significantly shortened ( P<0.05); compared with preoperative, the ratio of HPF/CBH, HPF/length of the palpebral fissure (LPF) in the study group increased at 6 months after operation, and the lateral canthus brow tail line [ratio of lateral canthus to lateral end of eyebrow (LLE)]/LPF decreased ( P<0.05); HPF/CBH in the study group was lower than that in the young control group at 6 months after operation, but greater than that in the elderly control group ( P<0.05). Monthly HPF/LPF was significantly higher than that of the elderly control group ( P<0.05), and the LLE/LPF of the study group was significantly higher than that of the young control group at 6 months after operation ( P<0.05); intraoperative levator muscle shortening (LMS) was negatively correlated with postoperative MBH, CBH and LBH ( P<0.05), among which LMS had the greatest correlation with CBH ( r=-0.587). Conclusions:After ptosis correction, the palpebral fissures are larger, the distance between the eyebrows and the eyes is shortened, and there is a certain proportional relationship. After surgery, the patient's eyebrow-eye ratio is better than that of the elderly in the same age group, and it is closer to the young people, and the intraoperative LMS has a correlation with the eyebrow-eye distance, which can be used as a factor to predict the shortened distance of the eyebrow-eye distance.
		                        		
		                        		
		                        		
		                        	
5.Assessment of demand for rehabilitation services and analysis of causes of the gap between supply and demand in China

		                        			ObjectiveTo evaluate the overall demand for rehabilitation services from the perspective of the groups with disabilities, diseases, injuries and older adults, and explore the causes of the supply and demand gap of rehabilitation services. MethodsThe literatures in the field of rehabilitation were collected, summarized and analyzed, including policy documents issued by World Health Organization and relevant departments in China, surveys, and researches. ResultsThe demand for rehabilitation services caused by disabilities, chronic diseases, injuries and population ageing were vast in China, but many were still unmet. The main reasons leading to the gap included those population trend and health situation, the insufficient reserve of rehabilitation resources coming from incomplete development of rehabilitation service system early, and inadequate construction of disability reporting system, which could not efficiently meet rehabilitation demand with supply. 6.Association of maternal gestational diabetes mellitus with neonatal metabolites

		                        			Objective:To investigate the effects of gestational diabetes mellitus (GDM) on neonatal metabolites.Methods:This retrospective cohort study recruited 580 singleton newborns who were born to women with GDM from January 2018 to December 2018 in Foshan Maternal and Child Health Care Hospital as the GDM group. Another 580 counterparts born to non-GDM singleton mothers with matching age were selected as the non-GDM group with an allocation ratio of 1 to 1. Neonatal genetic metabolic disease screening was performed within 3-7 days after birth. Two independent sample t-test, and multiple linear regression model were used for statistical analysis. Results:There were significant differences in seven amino acids and 10 fatty acids levels between the GDM and non-GDM group. The serum levels of six amino acids and eight fatty acids were increased in the GDM group, while the levels of piperamide [(140.79±32.60) vs (150.26±35.46) μmol/L, t=-4.733, P<0.001], palmitoyl carnitine [(2.59±0.81) vs (2.73±0.82) μmol/L, t=-2.940, P=0.003], and carbamate [(0.066±0.022) vs (0.069±0.022) μmol/L, t=-1.937, P=0.042] were decreased compared with the non-GDM group. After adjusting for maternal gravidity, parity, neonatal birth weight, and gender, multivariate linear regression analysis showed that GDM was positively correlated with three amino acids levels, which were cysteine ( ?=0.012), homocysteine ( ?=0.263) and leucine ( ?=4.225); and was negatively correlated with glycine ( ?=-6.271) and piperamide level ( ?=-9.885). With regard to the fatty acids, GDM was positively correlated with the neonatal propionyl carnitine ( ?=0.214), butyryl carnitine ( ?=0.014), 3-hydroxybutyryl carnitine ( ?=0.006), isovaleryl carnitine ( ?=0.009), 3-hydroxyisovaleryl carnitine ( ?=0.024), hexadecanoyl carnitine ( ?=0.001), decadienoyl carnitine ( ?=0.045), octadecadienyl carnitine level ( ?=0.128), but was negatively correlated with palmitoyl carnitine ( ?=-0.119), and carbamate ( ?=-0.002) (all P<0.05). Conclusions:Correlations between maternal GDM and the levels of amino acids and fatty acids in neonates was noted in this study, suggesting that maternal GDM may affect the metabolism of amino acids and fatty acids in offspring at early stage of life.
		                        		
		                        		
		                        		
		                        	
7.Effects of low frequency magnetic stimulation on myelin and inflammation in demyelinated mice

		                        			Objective:To explore the effect of low frequency magnetic stimulation on myelin and inflammation in the callosum of demyelinated mice.Methods:Thirty-six 6 to 8-week-old male C57BL/6J mice were randomly divided into a control group, a cuprizone (CPZ) group and a magnetic therapy group. The CPZ group and the magnetic therapy group had demyelination induced by feeding a mixed diet containing 0.3% CPZ for 6 weeks, while the control group was given conventional food. The magnetic therapy group was given 50Hz 10mT magnetic stimulation during the 6 weeks for 20min daily, 5 days a week. The body mass of each mouse was observed every 7 days. At the end of the 6th week elevated cross maze experiments were conducted to observe any anxiety state. The myelin sheath in the corpus callosum was observed using Luxol fast blue staining and myelin basic protein (MBP) immunohistochemistry Tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) in the corpus callosum were detected using enzyme-linked immunosorbent assays.Results:After the 6 weeks of treatment, the average body mass of the mice in the magnetic therapy group had improved significantly compared with the CPZ group. The CPZ group′s times in the elevated cross maze experiments were significantly shorter than those of the control group and also shorter than those of the magnetic therapy group. The Luxol staining showed significant myelin loss in the corpus callosum of the CPZ group, but compared with the CPZ group the average loss of myelin in the magnetic therapy group was significantly less. This was further confirmed by the MBP immunohistochemistry. Compared with the control group, the average expression of MBP in the CPZ group was significantly reduced, while in the magnetic therapy group it was significantly increased. Compared with the control group, the average TNF-α and IL-1β levels in the corpus callosum of the CPZ group increased significantly, but compared with the CPZ group the average levels in the magnetic therapy group had decreased significantly.Conclusions:Low frequency magnetic stimulation improves the body weight and anxiety state of mice. 8.Regulatory Effects of Stilbene Glucoside on JNK and PP 2B in APP/PS1/Tau Transgenic Dementia Mice

		                        			OBJECTIVE:To study the regulatory effects of stilbene glucosid e(TSG)on c-Jun N-terminal kinase (JNK)and protein phosphortase 2B(PP2B)in APP/PS1/Tau transgenic dementia (3×Tg-AD)mice,and to explore its potential mechanism of anti-Alzheimer’s disease (AD). METHODS :Totally 45 male 3×Tg-AD mice were randomly divided into model group ,positive control group (huperzine A ,0.15 mg/kg),TSG low-dose ,medium-dose and high-dose groups (0.033,0.1,0.3 g/kg),with 9 mice in each group. Another 9 normal male C 57BL/6J mice were included into normal control group. Administration groups were given relevant medicine intragastrically ,once a day ,for consecutive 60 d. Normal control group and model group were given constant volume of normal saline intragastrically. After medication ,Morris water maze experiment was used to test the spatial learning and memory ability of mice in each group ;Nissl staining was used to observe the changes of Nissl bodies in cerebral cortex and hippocampus ;mRNA and protein expressions of JNK and PP 2B were detected by qRT-PCR and Western blotting assay. RESULTS:Compared with normal control group ,the escape latency was significantly prolonged (P<0.01),the retention time of the original platform quadrant was significantly shortened (P< and the times of crossing the platform was significantly reduced in model group (P<0.01);the number of Nissl bodies in cerebral cortex and hippocampus was significantly 729011126@qq.com reduced,the staining was slight ;the relative expressions of JNK mRNA and protein were significantly increased (P<  0.01),and the relative expressi ons of PP 2B mRNA and protein were significantly decreased (P<0.01). CONCLUSIONS :TSG can improve the learning and memory ability and neuronal damage of 3 × Tg-AD mice. 9.Effects of Stilbene Glycoside on the Phosphorylation of Thr 205 and Ser 404 Sites of Tau Protein in AD Model Mice

		                        			OBJECTIVE:To study the e ffects of stilbene glycoside c(TSG)on phosphorylation of Thr 205,Ser404 sites of Tau protein in Aizheimer ’s disease (AD)model mice ,and to investigate the potential anti-AD mechanism of TSG. METHODS :APP/ PS1/Tau three transgenes (3×Tg-AD)mice were randomly divided into model group ,positive control group (huperzine,0.15 mg/kg),TSG low-dose ,medium-dose and high-dose groups (0.033,0.1,0.3 g/kg),with 6 mice in each group. In addition ,6 C57BL/6J mice were chosen as normal control group. Administration groups were given relevant medicine intragastrically. Model group and normal control group were given equal volume of normal saline intragastrically ,once a day ,for consecutive 60 days. After last medication ,immunofluorescence staining was used to detect Tau protein and phosphorylated Tau protein (Thr205, Ser404 sites) distribution and expression in brain tissue of mice in each group. Western blotting assay was used to detect phosphorylated Tau protein (Thr205,Ser404 sites)expression  level in brain tissue of mice in each group. RESULTS :Compared with normal control group ,the expression of Tau  protein,phosphorylated Tau protein (Thr205,Ser404 sites)in 729011126@qq.com the brain tissue of mice were increased in model group ,which  were easy to aggregate and distributed more widely ;theirrelative expression were increased significantly (P<0.01). Results of Western blotting assay showed that the expression levels of phosphorylat ed Tau protein (Thr205,Ser404 sites)were increased significantly (P<0.01). Compared with model group ,the expression of Tau protein ,phosphorylated Tau protein (Thr205,Ser404 sites) in the brain tissue of mice were decreased in positive control group and TSG groups ;aggregation decreased,distribution narrowed and their relative expression were decreased significantly (P<0.01). Results of Western blotting assay showed that the expression levels of phosphorylated Tau protein (Thr205,Ser404 sites)were decreased significantly (P< 0.01). Compared with positive control group ,There was no significant difference in the distribution of Tau protein ,phosphorylated Tau protein (Thr205,Ser404 sites)in the brain tissue of mice in TSG groups ;the relative expression were not statistically significant(P>0.05);but Western blotting assay showed the expression levels of phosphorylated Tau protein (Thr205 site)in TSG medium-dose and high-dose groups as well as the expression levels of phosphorylated Tau protein (Ser404 site)in TSG groups were decreased significantly (P<0.05 or P<0.01). 10. The expression and clinical significance of DDR2 and IFITM1 in nasopharyngeal carcinoma

		                        			 Objective:
		                        			To investigate the expression of discoid domain receptor 2 (DDR2) and interferon-induced transmembrane protein 1 (IFITM1) in nasopharyngeal carcinoma (NPC) patients and their clinical significance.
		                        		
		                        			Methods:
		                        			From November 2014 to November 2015, 65 patients with nasopharyngeal carcinoma were selected. All patients underwent nasopharyngeal biopsy. All specimens were embedded in paraffin after biopsy. All specimens were confirmed by pathology. Another 65 rhinitis mucosa specimens with chronic inflammation confirmed by pathology in the same period were selected as control group. The clinical and pathological data of the two groups were collected. The mRNA and protein expression levels of DDR2 and IFITM1 in nasopharyngeal carcinoma tissues were detected by quantitative real-time polymerase chain reaction (qRT-PCR) and immunohistochemistry. The correlation between them was analyzed by Pearson. All patients were followed up for 3 years, and the 3-year survival period of the patients was analyzed by Kaplan-Meier method.
