1.Adhesion,proliferation,and vascular smooth muscle differentiation of bone marrow mesenchymal stem cells on different electrospinning membranes

		                        			BACKGROUND:Small diameter artificial vessels are urgently needed to treat coronary artery and peripheral artery diseases in clinical practice.At present,vascular tissue engineering has become the main method for preparing small diameter artificial vessels.Selecting suitable biomaterials and cell sources is the key factor for successful construction of small diameter tissue engineered vessels. OBJECTIVE:To observe the effect of four kinds of electrospinning membrane materials on proliferation,adhesion and differentiation of bone marrow mesenchymal stem cells into vascular smooth muscle cells. METHODS:Bone marrow mesenchymal stem cells were isolated and extracted from SD rats.The bone marrow mesenchymal stem cells were inoculated separately on polycaprolactone(PCL),polycaprolactone-hyaluronic acid(PCL-HA),polycaprolactone-silk-filament proteins(PCL-SF),and polycaprolactone-gelatin(PCL-GEL)electrospinning membrane materials.After 1,3,and 7 days of culture,the cell arrangement on the material was observed under scanning electron microscope.The proliferation and adhesion of the material were observed by phalloidin staining.The mRNA expressions of CD90,Meflin,and transforming growth factor β were detected by qRT-PCR.After 7 days of induced differentiation into vascular smooth muscle cells,the mRNA expression ofɑ-smooth muscle actin on the material was detected by qRT-PCR. RESULTS AND CONCLUSION:(1)Bone marrow mesenchymal stem cells were arranged along the fibers of the four kinds of electrospinning membranes under scanning electron microscopy.(2)Phalloidin staining showed the regular distribution of bone marrow mesenchymal stem cells on the four kinds of electrospinning membranes and parallel distribution along the fiber direction.Moreover,PCL-HA,PCL-SF,and PCL-GEL electrospinning membranes were more conducive to the proliferation and adhesion of bone marrow mesenchymal stem cells than PCL electrospinning membranes.Compared with PCL-HA and PCL-GEL electrospinning membranes,PCL-SF electrospinning membranes were more conducive to the proliferation and adhesion of bone marrow mesenchymal stem cells.(3)qRT-PCR showed that the four kinds of electrospun membrane materials could maintain the mRNA expression of CD90 and Meflin in bone marrow mesenchymal stem cells,but there was no significant difference between groups(P>0.05).The mRNA expression of transforming growth factor β in PCL-HA,PCL-SF,and PCL-GEL groups was higher than that in PCL group on days 1 and 7(P<0.05),and the mRNA expression of transforming growth factor β in PCL-SF group was higher than that in the other three groups on days 3 and 7(P<0.05).The mRNA expression of transforming growth factor β in PCL-HA group was higher than that in PCL-GEL group on day 7(P<0.05).(4)qRT-PCR showed that the mRNA expression of ɑ-smooth muscle actin in PCL-SF group was higher than that in the other three groups(P<0.05),and that in PCL-HA group was higher than that in PCL group(P<0.05).(5)The results indicate that compared with PCL,PCL-HA and PCL-GEL electrospinning membranes,PCL-SF electrospinning membranes combined with bone marrow mesenchymal stem cells are more suitable for the preparation of small diameter tissue engineered vessels.
		                        		
		                        		
		                        		
		                        	
2.Research progress on the association between individual psychological factors and adolescent smartphone addiction

		                        			Smartphone addiction adversely effects the physical and mental health of adolescents. Individual psychological factors serve as direct contributors to smartphone addiction in adolescents, and smartphone addiction also affects the individual mental well being. Research suggests that psychological factors such as anxiety and depression, social anxiety and loneliness, self control, sensation seeking, and mindfulness interact with adolescent smartphone addiction. 3.Effects of traditional Chinese medicine on treatment outcomes in severe COVID-19 patients: a single-centre study.

		                        			As the search for effective treatments for COVID-19 continues, the high mortality rate among critically ill patients in Intensive Care Units (ICU) presents a profound challenge. This study explores the potential benefits of traditional Chinese medicine (TCM) as a supplementary treatment for severe COVID-19. A total of 110 critically ill COVID-19 patients at the Intensive Care Unit (ICU) of Vulcan Hill Hospital between Feb., 2020, and April, 2020 (Wuhan, China) participated in this observational study. All patients received standard supportive care protocols, with a subset of 81 also receiving TCM as an adjunct treatment. Clinical characteristics during the treatment period and the clinical outcome of each patient were closely monitored and analysed. Our findings indicated that the TCM group exhibited a significantly lower mortality rate compared with the non-TCM group (16 of 81 vs 24 of 29; 0.3 vs 2.3 person/month). In the adjusted Cox proportional hazards models, TCM treatment was associated with improved survival odds (P < 0.001). Furthermore, the analysis also revealed that TCM treatment could partially mitigate inflammatory responses, as evidenced by the reduced levels of proinflammatory cytokines, and contribute to the recovery of multiple organic functions, thereby potentially increasing the survival rate of critically ill COVID-19 patients.

4.Hemostatic effect of oxidized regenerated cellulose hemostatic material on minipig liver hemorrhage models

		                        			BACKGROUND:At present,plant-derived absorbable hemostats are mainly imported,so it is necessary to develop domestic alternatives that are not inferior to similar imported products. OBJECTIVE:To investigate the hemostatic effect of oxidized regenerated cellulose hemostatic material on a model of minipig liver hemorrhage. METHODS:A total of 24 Bama minipigs were selected and randomly divided into three groups.In the sham operation group(n=6),only an open operation was performed,and the damaged side of the liver was removed and put back in situ.In the experimental group(n=12),the liver hemorrhage model was established,and the oxidized regenerated cellulose hemostatic material was applied to the wound.In the control group(n=6),commercially available absorbable hemostatic gauze was used on the wound after establishing liver hemorrhage models.Hemostatic time and blood loss were recorded.The venous blood of the minipig anterior cavity was collected at different time points before and after modeling,and the blood routine and liver and kidney functions were analyzed.Hematoxylin-eosin staining and Masson staining were performed at 2,6,and 14 weeks after modeling.The main organs were observed by histopathology at 14 weeks after modeling.A liver ultrasound examination was performed at different time points after modeling to observe the degradation and absorption of materials. RESULTS AND CONCLUSION:(1)There was no significant difference in intraoperative hemostatic time and blood loss between the experimental group and the control group(P>0.05).(2)The monitoring results of blood biochemical indexes demonstrated that the levels of aspartate transaminase and glutamic pyruvic transaminase in the three groups were higher 24 hours after modeling than before modeling,and the indexes basically recovered to the normal levels 72 hours after modeling.There were no significant differences in blood biochemical indexes such as liver and kidney functions,blood glucose and inflammatory factors among the three groups(P>0.05).(3)Hematoxylin-eosin staining and Masson staining of histopathology revealed that 2 weeks after surgery,granulation tissue formation and a large amount of collagen fiber deposition were observed on the liver wounds of the two groups,and there were obvious fibrous hyperplasia zones and inflammatory cell infiltration,and the wound healed well.14 weeks after modeling,the liver wounds of two groups of minipigs exhibited mild fibrous hyperplasia zone,collagen fiber deposition and a small amount of inflammatory cell infiltration,complete material degradation,and the healed wound.Hematoxylin-eosin staining showed no significant pathological changes in major organs.(4)Ultrasonic examination demonstrated that the materials of both groups were degraded gradually with the extension of implantation time,and most of them were degraded and absorbed by 56 days after molding.(5)The results confirm that the oxidized regenerated cellulose hemostatic material can effectively prevent liver wound hemorrhage in minipigs,and the hemostats are safe and reliable.

		                        			ObjectiveTo study the bioactive secondary metabolites of Talaromyces sp. TP21 and their bioactivities. MethodThe secondary metabolites of Talaromyces sp. TP21 were isolated by high performance liquid chromatography (HPLC), normal phase and reversed phase column chromatography combined with molecular networking and bioassay-guided fractionation, and their structures were determined by nuclear magnetic resonance (NMR) and high resolution mass spectrometry (HR MS). The inhibitory effects of the compounds on the growth of the lung cancer cell line A549 and the liver cancer cell line Hep G2 were measured by themethyl thiazolyl tetrazolium (MTT) method. The antimicrobial activities of the compounds were measured with Staphylococcus aureus and human oral cavity-derived Saccharomyces cerevisiae as the indicator microorganisms. 6.Development and reliability and validity test of the Psychological Birth Trauma Assessment Scale

		                        			Objective To develop a Psychological Birth Trauma Assessment Scale and to test its reliability and validity.Methods Through literature review,semi-structured interview,expert conference,Delphi expert enquiry and preliminary investigation,a pre-test scale was formed.From March to May 2023,postpartum women from 38 hospitals of different levels in 15 provinces(Jiangsu,Anhui,Zhejiang,Fujian,Tibet,and Shanghai,etc.)were conveniently selected for investigation.The sample size of the first-round survey was 547,which was used for reliability and validity test.The sample size of the second-round survey was 550,which was used for confirmatory factor analysis.Results The psychological birth trauma assessment scale consisted of 2 parts,with a total of 29 items in 6 dimensions.The intrapartum feeling part included 4 dimensions,with a cumulative variance contribution rate of 63.992%,and the postpartum effect part included 2 dimensions,with a cumulative variance contribution rate of 68.208%.The content validity index of the scale was 0.947,and the content validity index of each item was 0.809~1.000.The total score of the scale and the scores of each dimension were significantly negatively correlated with the total score of the calibration scale(r=-0.784~-0.533,P<0.001).The total Cronbach's α coefficient was 0.941,and the split-half reliability coefficient was 0.883.Confirmatory factor analysis showed that the main evaluation indicators were all within the acceptable range.Conclusion The Psychological Birth Trauma Assessment Scale has good reliability and validity,and it can be used as an assessment tool for the degree of maternal psychological birth trauma.
		                        		
		                        		
		                        		
		                        	
7.Correlation among Serum LncRNA NEAT1,miR-424-5p Levels,Clinical Stage and Infertility Diagnosis in Patients with Endometriosis

		                        			Objective To explore the diagnostic value of long non-coding RNA nuclear-enriched abundant transcript 1(LncRNA NEATl)and microRNA(miR)-424-5p in the diagnosis of endometriosis(EM)combined with infertility and its application value in disease staging.Methods A total of 232 EM patients who visited Shijiazhuang Maternal and Child Health Hospital from October 2021 to October 2023 were selected as the study subjects,120 patients with EM combined with infertility were used as the infertility group,and 112 patients with pure EM were regarded as the control group.The infertility group was separated into r-AFS stages Ⅰ~Ⅱ(n=27),r-AFS stage Ⅲ(n=45),and r-AFS stage Ⅳ(n=48)according to the American r-AFS staging standards,and qRT-PCR method was applied to detect the expression levels of serum LncRNA NEAT1 and miR-424-5p.Receiver operating characteristic(ROC)curve was applied to analyze the value of serum LncRNA NEAT1 and miR-424-5p in diagnosing EM combined with infertility,and multivariate Logistic regression was applied to analyze the influencing factors of EM combined with infertility.Bioinformatics analysis of LncRNA NEAT1 targeting miR-424-5p was performed on the TargetScanHuman website.Pearson method was applied to analyze the correlation between serum LncRNA NEAT1 and miR-424-5p expression levels in patients with EM combined with infertility.Results Differences of serum LncRNA NEAT1(1.16±0.15 vs 1.02±0.13),pelvic surgery history ratio(17.50%vs 1.79%),miR-424-5p(0.92±0.11 vs 1.04±0.12)expression levels between infertility group and control group were significant,and the differences were significant(t=7.753,16.018,7.974,all P<0.05).Serum LncRNA NEAT1(1.05±0.10,1.14±0.14,1.24±0.19)increased and miR-424-5p(0.99±0.12,0.93±0.11,0.87±0.10)decreased in r-AFS stage Ⅰ~Ⅱ,Ⅲ,Ⅳ patients,and the differences were significant(F=13.528,10.910,all P<0.05).There was a targeting relationship between LncRNA NEAT1 and miR-424-5p,and there was a negative correlation between serum LncRNA NEAT1 and miR-424-5p expression levels in EM patients with infertility(r=-0.431,P<0.05).The AUCs of serum LncRNANEAT1,miR-424-5p,and combined diagnosis for EM combined with infertility were 0.782,0.719 and 0.835,respectively,and the AUC of combined diagnosis was better than that of individual detection(Z=2.625,3.861,P=0.009,0.001).LncRNA NEAT1 was a risk factor for EM combined with infertility,while miR-424-5p was a protective factor(all P<0.05).Conclusion The expression level of serum LncRNA NEAT1 is increased and miR-424-5p is decreased in EM patients with infertility,both of which are related to the disease stage and the occurrence of infertility.
		                        		
		                        		
		                        		
		                        	
8.Effect of capsaicin on LPS-induced microglial inflammatory response by modulating SIRT1/HMGB1/NF-κB signaling pathway

		                        			Objective:To investigate the effect of capsaicin on lipopolysaccharide(LPS)-induced microglial inflammatory response by modulating silent mating type information regulation 2 homolog 1(SIRT1)/high mobility group box-1 protein(HMGB1)/nuclear factor κB(NF-κB)signaling pathway.Methods:BV2 mouse microglia were cultured in vitro,and pretreated with capsaicin(10 μmol/L),SIRT1 inhibitor EX527(100 μmol/L),capsaicin+EX527(10 μmol/L capsaicin+100 μmol/L EX527)for 3 hours,and then treated with LPS(1 μg/ml)for 24 hours,the corresponding groups were LPS group,LPS+capsaicin group,LPS+EX527 group,and LPS+capsaicin+EX527 group,a normal cultured control group(Control)was also set up.Morphological changes of BV2 cells in each group were observed under a microscope;CCK-8 method was used to detect the viability of BV2 cells in each group;immunofluo-rescence staining was applied to detect the positive expressions of ionic calcium adaptor protein(Iba-1)and the polarization of M1(positive for CD16/32)/M2(positive for CD206)subtypes in BV2 cells in each group;ELISA was applied to detect levels of inflamma-tory factors in BV2 cells in each group;Western blot and co-immunoprecipitation were applied to detect expression of SIRT1/HMGB1/NF-κB signaling pathway-related proteins in each group.Results:Compared with Control group,BV2 cells in LPS group were atro-phied,with shortened processes,the cell viability,and levels of IL-1β,TNF-α and IL-6 were increased,Iba-1 positive expression of BV2 cells and M1 type BV2 cells were increased,while expression of SIRT1 protein was decreased,expressions of acetylated(ace)-HMGB1 protein,cytoplasmic HMGB1 protein and nuclear NF-κB protein were increased(P<0.05);after capsaicin pretreatment,the above conditions were improved,and the M2 type BV2 cells were increased;adding EX527 pretreatment on the basis of capsaicin pre-treatment,the above conditions were all aggravated.Conclusion:Capsaicin can inhibit the inflammatory response induced by LPS-in-duced BV2 cell activation,which may be achieved by modulating the SIRT1/HMGB1/NF-κB signaling pathway.
		                        		
		                        		
		                        		
		                        	
9.Epidemiological characteristics of human respiratory syncytial virus among acute respiratory infection cases in 16 provinces of China from 2009 to 2023

		                        			Objective:To understand the epidemiological characteristics of human respiratory syncytial virus (HRSV) among acute respiratory infection (ARI) cases in 16 provinces of China from 2009 to 2023.Methods:The data of this study were collected from the ARI surveillance data from 16 provinces in China from 2009 to 2023, with a total of 28 278 ARI cases included in the study. The clinical specimens from ARI cases were screened for HRSV nucleic acid from 2009 to 2023, and differences in virus detection rates among cases of different age groups, regions, and months were analyzed.Results:A total of 28 278 ARI cases were enrolled from January 2009 to September 2023. The age of the cases ranged from<1 month to 112 years, and the age M ( Q1, Q3) was 3 years (1 year, 9 years). Among them, 3 062 cases were positive for HRSV nucleic acid, with a total detection rate of 10.83%. From 2009 to 2019, the detection rate of HRSV was 9.33%, and the virus was mainly prevalent in winter and spring. During the Corona Virus Disease 2019 (COVID-19) pandemic, the detection rate of HRSV fluctuated between 6.32% and 18.67%. There was no traditional winter epidemic peak of HRSV from the end of 2022 to the beginning of 2023, and an anti-seasonal epidemic of HRSV occurred from April to May 2023. About 87.95% (2 693/3 062) of positive cases were children under 5 years old, and the difference in the detection rate of HRSV among different age groups was statistically significant ( P<0.001), showing a decreasing trend of HRSV detection rate with the increase of age ( P<0.001). Among them, the HRSV detection rate (25.69%) was highest in children under 6 months. Compared with 2009-2019, the ranking of HRSV detection rates in different age groups changed from high to low between 2020 and 2023, with the age M (Q1, Q3) of HRSV positive cases increasing from 1 year (6 months, 3 years) to 2 years (11 months, 3 years). Conclusion:Through 15 years of continuous HRSV surveillance analysis, children under 5 years old, especially infants under 6 months old, are the main high-risk population for HRSV infection. 		                        			The leucine-rich repeat (LRR) containing 15 (LRRC15) protein is one of the members of the LRR superfamily. In the medical field, LRRC15 protein not only participates in parasitic, viral and other pathogen infections, but also plays a role in anti-tumor and rheumatoid arthritis treatment. It also regulates cell autophagy, stem cell ossification, and changes in hair follicle dermal papilla cell characteristics.
