1.Correlation of Metabolic Syndrome and Low-level High-density Lipoprotein Cholesterol with Adverse Pathological Features of Prostate Cancer After Radical Prostatectomy

Objective To assess the association of metabolic syndrome (MetS) and low-level high density lipoprotein-cholesterol (HDL-C) with the invasion of prostate cancer (PCa) in the patients after radical prostatectomy (RP). Methods We collected and analyzed the data of 232 PCa patients treated with RP for prospective multicenter clinical study.The MetS was assessed according to NCEP-ATP Ⅲ.Patients were divided into MetS group and non-MetS group.Locally advanced PCa was defined as a pT-stage≥3.ISUP Gleason grading system (GS) were used for pathological grading.Logistic regression analyses assessed the association of MetS and its components with pathological data. Results MetS was an independent risk factor for GS≥8 after RP.Low HDL-C level was associated with locally advanced PCa.The risks of adverse pathological features increased with the number of MetS components. Conclusion Low HDL-C level and 4 or more MetS components are associated with higher risk of adverse pathological features of PCa.
2.Effect of silencing monocarboxylate transporter 4 on the malignant biological behaviors of prostate cancer PC3 cells

[Abstract] Objective: To investigate the effects of silencing monocarboxylate transporter 4 (MCT4) on the proliferation, migration
and invasion of prostate cancer PC3 cells and its possible molecular mechanism. Methods: RNA interference technology was used to
transfect siRNA-MCT4 (si-MCT4) and negative control plasmid (si-NC) into PC3 cells, respectively. The content of lactic acid in the
cell culture medium of transfected PC3 cells was detected by lactic acid assay after culturing for 96 h. The proliferation, migration and
invasion ability of PC3 cells were detected by CCK-8 and Transwell assay, respectively. Western blotting was used to detect the silencing
effect and the expressions of integrin β 4-FAK-SRC-MEK-ERK signaling pathway associated proteins (integrin β4, p-FAK, p-SRC,
p-ERK1/2, p-MEK1/2) and EMT associated proteins (E-cadherin and N-cadherin). Results: PC3 cell line with silenced MCT4 was
successfully constructed. Compared with the control group, the content of extracellular lactic acid in the PC3 cell culture medium of the
si-MCT4 group was significantly decreased (P<0.01), and the proliferation, migration and invasion of cells were significantly decreased
(P<0.05 or P<0.01). Compared with the control group, the protein expressions of integrin β4, p-FAK, p-SRC, p-MEK1/2, p-ERK1/2
and N-cadherin were significantly decreased (all P<0.01), while the protein expression of E-cadherin was significantly increased
(P<0.01). Conclusion: Silencing MCT4 can significantly inhibit the proliferation, migration and invasion of PC3 cells, the mechanism
of which may be related to the inhibition of lactic acid level in cell culture medium and suppression of integrin β4-FAK-SRC-MEKERK signaling pathway associated proteins as well as EMT associated proteins.
3.Application of shuffling fat in the treatment of eyelid cicatricial depression and atrophy due to stick-bone scar

Objective:To investigate the effect and reasonable strategy of shuffling fat in the treatment of eyelid stick-bone scar cicatricial depression and atrophy.Methods:A retrospective analysis consisted of patients admitted in Plastic Surgery Hospital, CAMS from March 2017 to May 2019, and the inclusion criteria was depressive and atrophic deformities caused by eyelid stick-bone scar. By performing subcutaneous liposuction on the abdomen, the harvested fat was further mechanically refined into shuffling fat. Single serialized shuffling fat injection with a 3-month interval was performed in the orbital region. Approximately 3 months after fat transplantation, symptomatic treatment was performed due to complications of upper eyelid ptosis or ectopic, and fat injection combined with the orbicularis oculi muscle flap was carried out to repair the lower eyelid atrophic deformity. Follow up regularly after the operation to observe the occurrence of complications. Six months after the end of treatment, the patient and observer scar assessment scale (POSAS) was used to assess scars, and the doctor-patient comprehensive score was used to assess patient satisfaction.Results:A total of 6 patients with stick-bone scar in the eyelid area were enrolled including 4 males and 2 females, aged 24-53 years old, with an average of 37 years old. The causes of defects included 4 cases of trauma, 2 cases after maxillary osteomyelitis treatment. Duration of disease ranged 1.5-50.0 years, average 14.2 years. Scars were located in the regions of upper eyelid in 4 cases and lower eyelid in 2 cases. Area of depressed region ranged 0.5 cm×2.0 cm-1.5 cm×3.0 cm. After mechanical refinement, the fat texture was more delicate and even. Injection of shuffling fat with the single volume of 0.6 to 1.3 ml was performed 3 to 4 times (average 3.25 times) on the upper eyelid and average 2 times on the lower eyelid, respectively. During the post-operative 6 months, the appearance of all patients was significantly improved, the volume of soft tissues of the eyelid was significantly increased, the pliability and elasticity of the scar tissue was ameliorated, and no complications occurred. The scores of patient scar assessment scale (PSAS) and observer scar assessment scale (OSAS) were (7.0±1.0) and (6.7±1.1), respectively, and the doctor-patient comprehensive score was 4-5 (average 4.5).Conclusions:For upper eyelid, shuffling fat injection has a good therapeutic effect on depressive and atrophic deformity caused by the stick-bone scar by increasing the tissue volume; for lower eyelid, it is necessary to combine the orbicularis oculi myocutaneous flap on the basis of shuffling fat transplantation to further increase the support structure for better appearance.

5. The changing trend of capacity on policy implementation related to the prevention and control of chronic non-communicable disease at the provincial level, from 2011 to 2017

Objective:
To assess the follow-up situstion and changes of policies related to the prevention and control on chronic non-communicable diseases (NCDs), in various provinces from 2011 to 2017.
Methods:
Three national assessment programs on the prevention and control capacity of NCDs were carried out from September 2012 to March 2013, September 2014 to March 2015, and July to November 2018 respectively. Data related to the capacity on policy implementation among the 31 provinces, autonomous regions, municipality directly under the central government and Xinjiang Production and Construction Corps, were collected through online surveys.
Results:
The rate of data collection in all the provinces reached 100%, for all three surveys. In 2011, 2013, and 2017, the capacity for policy development special funding was distributed for prevention and control NCDs under the provincial fiscal revenue by 27 provinces (84.4%), 26 provinces (81.3%) and 25 provinces (78.1%), and the numbers of provincial governments leaders attended the local activities related to prevention and control NCDs was 15 (46.9%), 13 (40.6%) and 19(59.4%), respectively. From 2009 to 2011, 14 (43.8%) proposals related to the topics on prevention and control of NCDs, were raised at the provincial People’s Congress and Political Consultative Conference, while from 2011 to 2013 and2014 to 2017, 13 (40.6%) and 12 (37.5%) were respectively raised. In terms of capacity for policy development, numbers of provincial comprehensive plan which targeting prevention and control of NCDs reached 6 (18.8%), 20 (62.5%) and 27 (84.4%) in 2011, 2013, and 2017 respectively. In 2011, 2013 and 2017, numbers of provincial special plans that targeting on NCDs or the risk factors of NCDs prevention and control were 0, 1, and 3, respectively.
Conclusions
Under the continuous introduction of international and national policies related to prevention and control on NCDs, capacity for policy formulation in various provinces has been greatly improved. However, a slight progress has been made in the capacity for policy making. The increase of capacity building on policy making regarding prevention and control of NCDs, at the provincial government level, has become a key issue.
6. Application of EH composite artificial bone prosthesis fabricated by 3D digital technology in treating structural defect of orbital region

Objective:
To explore the application of the 3D precise digital technology in restoring structural defects of the orbital region.
Methods:
Structural defects of the orbital region concerning on osseous structure and soft tissue were restored in one stage or stages using EH composite artificial bone prosthesis with complex three-dimensional structure. The fabrication of EH composite artificial bone prosthesis was based on CT scanning and 3D reconstruction of the skull with computer aided data analysis and design. The appearance, degree of satisfaction and the complications were evaluated in postoperative regular follow-up.
Results:
Five cases of structural defects in the orbital regions presenting bone defect and soft tissue abnormality, received treatments in the department from January 2016 to September 2017. The cases consist of one patient with dysplasia following surgical treatment, three with post-traumatic and one with Treacher-Collins syndrome. With the application of individualized EH composite artificial bone fabricated by aforementioned method, all the repair materials presented the ideal three-dimensional structure and coincided well with the defects, and soft tissue restoration of 2 cases was performed in one stage or by stages. Appearance and symmetry of the 5 cases was significantly improved, without complications of infection, rejection, exposure or graft tissue necrosis. All the patients were satisfied with the results.
Conclusions
In consideration of the capacity to fabricate accurate individualized repair materials, the three-dimensional digital technology plays an important role in the treatment of structural defects in the orbital region, especially the reconstruction of the bony contour. The simple orbital deformities can be treated with the repair materials and correction of soft tissue. For special orbital deformities, attention should be paid to bone structure repair, eye socket reconstruction and filling of orbital contents sequentially.
7.N-myc downstream regulated gene 2 promotes apoptosis of bladder cancer cells through signal transducer and activator of transcription 3 signaling pathway

Objective To investigate the effect of N-myc downstream regulated gene 2 (NDRG2) on the apoptosis of bladder cancer cells by regulating the signal transducer and activator of transcription 3(STAT3) signaling pathway.Methods The expression level of NDRG2 in human bladder cancer cell BIU-87 and immortalized cell SV-HUC-1 was detected by Western blot.NDRG2 over expression vector and empty vector control (pcDNA3.1),siRNA-NDRG2,siRNA control were transfected into BIU-87 cells.After transfected 48 h,the expression level of NDRG2,Cleaved caspase 3,STAT3,p-STAT3,JAK2,p-JAK2 were detected by Western blot,the cell proliferation and apoptosis were measured by MTT and flow cytometry.After adding inhibitor AG490 of 45 μmol/L in cultured BIU-87 cells,MTT assay was used to detect cell proliferation and flow cytometry was used to detect the cell apoptosis,Western blot to detect the expression level of Cleaved caspase 3,STAT3,p-STAT3,JAK2,p-JAK2.Results The expression level of NDRG2 in bladder cancer cells was higher than that in bladder epithelial cells.The cell survival rate of pcDNA3.1/NDRG2 group was lower than that of pcDNA3.1 group,the difference was statistically significant (P < 0.01).The cell survival rate of siRNA-NDRG2 group was higher than that of siRNA control group (P< 0.01).The apoptosis rate of pcDNA3.1/NDRG2 group was higher than that of pcDNA3.1 group (P < 0.01).The apoptosis rate of NDRG2 siRNA group was lower than that of siRNA control group (P < 0.01).The level of p-STAT3 and p-JAK2 in pcDNA3.1/NDRG2 group was lower than that in pcDNA3.1 group (P< 0.01).The survival rate and apoptosis rate of BIU-87 cells cultured with AG490 were in agreement with the trend of pcDNA3.1/NDRG2 after transfection.Conclusions NDRG2 could promote the apoptosis of bladder cancer cells,and its mechanism may be related to STAT3 signaling pathway.

Objective:To prepare for mAb of progesterone receptor. It would provide support for the immunohistochemistry behind. Methods:Target gene connected together with a carrier by seamless cloning method. The target protein that expression by inducing was collected. And with cell fusion method , the monoclonal antibodies were preparation. Then the mAb were detected by IHC. Results: The mAb ( clone 7C7 ) was detected and it found positive for the breast, uterine fibroid tissue, showed negative in colorectal cancer tissue, smooth muscle tissue, the goal of the claim were achieve. Conclusion: Finally, we found the method that prepare for mAb was far beyond our imagination. The result of IHC on different samples about mAb(7C7)obtained compliance with an-ticipation. Study on the difference between the PR-A and PR-B had significance.
9.Relationship of the location of nerve compression and different type of trigeminal neuralgia

Objective To investigate the relationship of the location of nerve compression and different type of trigeminal neuralgia.Methods Twenty-one patients with typical trigeminal neuralgia and 16 patients with atypical trigeminal neuralgia were treated by micro-vascular decompression.The position of the responsible vessels was probed during operation.Results Compared with atypical trigeminal neuralgia group, the compression rate of trigeminal root entry/exit zone (REZ) in typical trigeminal neuralgia group was significantly higher(P<0.01).The compression rate of trigeminal REZ which oppressed by artery in typical trigeminal neuralgia group was significantly higher than that in atypical trigeminal neuralgia group ( P<0.01 ) .Conclusions The responsible vessels in typical trigeminal neuralgia patients is mostly located in the trigeminal REZ, and the responsible vessels is mostly not located in other location in the trigeminal REZ.The responsible vessels oppress REZ is closely related to types of trigeminal neuralgia.
10.Role of Fas/FasL signaling pathway in ulinastatin postconditioning-induced attenuation of apoptosis in myocardial cells of patients undergoing cardiac valve replacement with cardiopulmonary bypass

Objective To evaluate the role of Fas/FasL signaling pathway in ulinastatin postconditioning-induced attenuation of apoptosis in the myocardial cells of patients undergoing cardiac valve replacement with cardiopulmonary bypass (CPB).Methods Forty patients of both sexes,aged 21-59 yr,of ASA physical status Ⅱ or Ⅲ (NYHA class Ⅱ or Ⅲ),scheduled for elective cardiac valve replacement with CPB,were randomly divided into 2 groups (n =20 each):control group (group C),and ulinastatin postconditioning group (group U).In group U,ulinastatin 10 000 U/kg was perfused via the aortic root at 4 000-5 000 U·kg-1 ·min-1 starting from 5 min before aortic unclamping.In group C,the equal volume of normal saline was infused instead of ulinastatin.Myocardial specimens were taken from the right auricle at 45 min after aortic unclamping for determination of Fas,Fas ligand (FasL),caspase-8,Bcl-2 and Bax expression and cell apoptosis.The ratio of Bcl-2 expression to/Bax expression (Bcl-2/Bax) and apoptotic index were calculated.Results Fas,FasL,caspase-8 and Bax expression and apoptotic index were significantly lower,and Bcl-2 expression and Bcl-2/Bax were higher in group U than in group C.Conclusion Ulinastatin postconditioning attenuates apoptosis in the myocardial cells through inhibiting Fas/FasL signaling pathway in the patients undergoing cardiac valve replacement with CPB.
