1.Effect of chromosome-instability-associated gene GALNT7 on proliferation and apoptosis of HCT116 colon cancer cells

Objective :
To investigate the effect of chromosome instability(CIN) associated gene polypeptide N-acetylgalactosaminyltransferase 7(GALNT7) on proliferation and apoptosis of HCT116 colon cancer cells.
Methods :
The HCT116 cell line withGALNT7knockdown was constructed by lentiviral infection. The correlation betweenGALNT7and CIN was verified by chromosome spread assay. The effect ofGALNT7on cell proliferation was detected by live cell counting, and the effect ofGALNT7on cell cycle distribution was detected by flow cytometry and Western blot. Caspase-3 activity and Western blot assays were used to detect the effect ofGALNT7on apoptosis.
Results :
HCT116 cells showed a slower proliferation rate upon knocking down ofGALNT7, and exhibited a more scattered karyotype distribution and a phenotype of increased degree of CIN. Inhibition ofGALNT7in HCT116 cells resulted in cell cycle arrest, upregulation of P21 and downregulation of CDK6 protein levels, as well as increased levels of Caspase-3 activity, cleaved PARP1 and PUMA protein expression, and decreased levels of BCL-2 protein expression.
Conclusion
TheGALNT7gene may promote proliferation and inhibit apoptosis of HCT116 colon cancer cells through the suppression of CIN generation.
2.Clinical characteristics of patients with puerperal schizophrenia

BackgroundWomen may develop severe symptoms of stress disorder following childbirth, which may be exposed to a risk of developing mental health problems, and even lead to the recurrence of the illness in female patients with schizophrenia, while comparatively limited research has been undertaken concerning the clinical characteristics and treatment of puerperal schizophrenia in China. ObjectiveTo explore the clinical characteristics of puerperal schizophrenia, so as to provide references for the clinical treatment. MethodsA total of 24 patients with puerperal schizophrenia who were hospitalized in the female ward of adult psychiatry department of the Affiliated Brain Hospital of Guangzhou Medical University from 2012 to 2020 and met the Diagnostic and Statistical Manual of Mental Disorders, fourth edition (DSM-IV) diagnostic criteria for schizophrenia were included as puerperal group. Another 48 non-puerperal women with schizophrenia were concurrently enrolled as control group. Then the basic data, scores on Positive and Negative Symptom Scale (PANSS) and the discharge medication were recorded. ResultsThe percentages of newly onset and positive family history of psychosis in puerperal group were larger than those in control group, with statistical significance (χ2=9.321, 5.240, P<0.05 or 0.01). Puerperal group scored higher on PANSS excitement factor (t=-2.220, P<0.05) and lower on negative factor (t=3.377, P<0.01) compared with control group. In terms of discharge medication, puerperal group reported a higher dosage of antipsychotic drugs (t=-2.095, P<0.05), and a larger proportion of combined use of benzodiazepines or antidepressants (χ²=21.316, 5.114, P<0.05 or 0.01) compared with control group, with statistical significance. ConclusionPatients with puerperal schizophrenia display increased ratings of excitement symptoms and decreased ratings of negative symptoms, which necessitates the use of high doses of antipsychotic drugs, and combined use of benzodiazepines and antidepressants.

In recent years,the field of nucleic acid therapeutics has been flourishing,progressively establishing itself as the third generation of drug modalities following small molecules and antibody-based drugs.Artificial intelligence technology based on machine learning is advancing rapidly,which can significantly accelerate the development process of nucleic acid therapeutics.This review provides an overview of the foundational aspects of artificial intelligence algorithms,databases,and characterizations in the field of nucleic acid drug development.It elucidates the advances in the application of artificial intelligence in nucleic acid structural prediction,small nucleic acid drug design,and other research and development phases of nucleic acid therapeutics,aiming to offer some reference for the interdisciplinary development of artificial intelligence and nucleic acid drugs.
4.CT-guided hook-wire localization of ≤10 mm pulmonary ground-glass nodules via different path ways before video-assisted thoracoscopic surgery:a comparative study

Objective To compare the clinical safety and efficacy of CT-guided hook-wire localization of≤10mm pulmonary ground-glass nodule(GGN)via different path ways before video-assisted thoracoscopic surgery(VATS).Methods The clinical data of a total of 128 patients with 10 mm pulmonary GGN,who received CT-guided hook wire localization before VATS at The Third Hospital of Mianyang of China between July 2018 and March 2023,were retrospectively analyzed.According to the puncturing localization path way mode,the patients were divided into vertical puncturing group(n=88)and non-vertical puncturing group(n=40).The number of puncturing times,the time spent for puncturing localization,the success rate of puncture,the operation time of VATS,and puncture-related complications of the two groups were recorded.Results No statistically significant differences in the gender,age,smoking history,GGN location,puncture position,nodule size,density characteristics of GGN,emphysema,and nodules-pleura distance existed between the two groups(all P>0.05).Compared with non-vertical puncturing group,in vertical puncturing group the number of puncturing times was smaller,the time spent for localization was shorter,the incidence of pneumothorax was lower,and the operation time of VATS was shorter,the differences in all the above indexes between the two groups were statistically significant(all P<0.05);and the subgroup analysis of patients whose GGN was overlapped with rib shadow obtained the same results.Binary logistic regression analysis revealed that non-vertical puncturing and the number of puncturing times were the independent risk factors for the occurrence of pneumothorax.Conclusion CT-guided hook-wire localization of≤10mm pulmonary GGN before VATS is clinically safe and effective.Under the condition when the lesion can be localized within the range of 2.0cm and the shadow overlapping of GGN with the rib and blood vessel can be effectively avoided,vertical puncturing path way mode should be preferred,which can effectively reduce the incidence of pneumothorax and shorten the operation time of VATS.
5.Towards understandings of serine/arginine-rich splicing factors.

Serine/arginine-rich splicing factors (SRSFs) refer to twelve RNA-binding proteins which regulate splice site recognition and spliceosome assembly during precursor messenger RNA splicing. SRSFs also participate in other RNA metabolic events, such as transcription, translation and nonsense-mediated decay, during their shuttling between nucleus and cytoplasm, making them indispensable for genome diversity and cellular activity. Of note, aberrant SRSF expression and/or mutations elicit fallacies in gene splicing, leading to the generation of pathogenic gene and protein isoforms, which highlights the therapeutic potential of targeting SRSF to treat diseases. In this review, we updated current understanding of SRSF structures and functions in RNA metabolism. Next, we analyzed SRSF-induced aberrant gene expression and their pathogenic outcomes in cancers and non-tumor diseases. The development of some well-characterized SRSF inhibitors was discussed in detail. We hope this review will contribute to future studies of SRSF functions and drug development targeting SRSFs.
6.Mediating effect of rumination between affiliate stigma and post-traumatic growth in parents of autistic children

Objective:To explore the relationship among ruminating,affiliate stigma and post-traumatic growth in parents of autistic children.Methods:Totally 339 parents of autistic children were selected.The Affiliate Stigma Scale(ASS),Post-traumatic Growth Inventory(PTGI),ERRI Intrusive Subscale(ERRI-I)and ERRI De-liberate Subscale(ERRI-D)of Event Related Rumination Inventory(ERRI)were used to measure affiliate stigma,post-traumatic growth,intrusive ruminating and deliberate ruminating.SPSS macro program PROCESS was used to test the mediating role.Results:There were positive correlations among the scores of ASS,ERRI-I and ERRI-D(r=0.39-0.72,Ps<0.01).The PTGl scores were negatively correlated with ASS scores(r=0.26,Ps<0.01)and positively correlated with ERRI-D scores(r=0.10,Ps<0.05).The ERRI-D scores played a partial mediating role in the relationship between ASS and PTGl scores(95%CI:0.01-0.07).The ERRI-I scores and ERRI-D scores played a serial mediating role in the relationship between ASS and PTGI scores(95%CI:0.03-0.14).Both me-diation paths had suppression effect.Conclusion:In the negative correlation between affiliate stigma and post-trau-matic growth in parents of autistic children,the deliberate ruminating plays a partial mediating role,while intrusive ruminating and deliberate ruminating play a serial mediating role.
7.Expression and functional mechanism of KCTD7 gene in hepatocellular carcinoma

Objective :
Using RNA sequencing data from The Cancer Genome Atlas (TCGA) to explore the expres⁃
sion , mechanism , and clinical significance of KCTD7 in hepatocellular carcinoma ( HCC) .
Methods :
The RNA sequencing data and clinical information of HCC patients were downloaded from the TCGA database . Univariate and multivariate Cox regression analyses were used to explore the correlation between the expression level of the KCTD7 gene in HCC and the clinical information and prognosis of patients . Gene Set Enrichment Analysis ( GSEA) was used to predict possible pathways regulated by the KCTD7 gene in HCC . Single⁃sample GSEA (ssGSEA) was used to compare immune infiltration between KCTD7 high expression group and low expression group . KCTD7 knockdown HCC cell lines were established to explore its function and possible mechanism in HCC regulation .
Results:
KCTD7 was highly expressed in HCC tissues compared to normal tissues (P < 0. 01) . The overall survival rate of HCC patients with high expression of KCTD7 gene was worse (P < 0. 05) . The expression level of KCTD7 was an independent risk factor affecting the overall survival of HCC patients . GSEA results showed that the KCTD7 gene was related to cell cycle signaling pathways . In the tumor microenvironment , high expression of the KCTD7 gene was positively correlated with activated CD4 + T cells , central memory CD4 + T cells , and natural killer cells . Knocking down KCTD7might inhibit HCC cell proliferation , impair cell cycle distribution , and promote apoptosis .
Conclusion
KCTD7 gene is highly expressed in HCC and affects the prognostic survival of HCC patients . Knocking down KCTD7 can inhibit the proliferation of HCC cells and promote apoptosis .
8.Expression of Tim-3 in the peripheral blood of pancreatic cancer patients and the diagnostic value in combination with serum glucose chain antigen 19-9

Objective:To investigate the expression of soluble T cell immunoglobulin and mucin domain-3 (Tim-3) in peripheral blood of patients with pancreatic cancer and its diagnostic value in combination with serum Carbohydrate antigen 19-9 (CA19-9) .Methods:106 newly diagnosed pancreatic cancer patients and 65 age and sex matched healthy individuals were enrolled. Tim-3 concentration was quantitatively determined by enzyme-linked immunosorbent assay (ELISA). According to the expression levels of soluble Tim-3 and serum CA19-9, a binary logistic regression model of receiver operating characteristic (ROC) curve was established to compare the diagnostic effects of serum CA19-9 and soluble Tim-3 alone or combined with the two tests.Results:The levels of soluble Tim-3 in the pancreatic cancer group were significantly higher than those in the healthy control group ( P<0.001). The expression level of soluble Tim-3 was significantly higher in patients with stage III-IV pancreatic cancer than in patients with stage I-II ( P=0.003). The AUC of soluble Tim-3 diagnosis for stage I-II pancreatic cancer was 0.856 (95%CI: 0.765 to 0.992 P<0.001), Serum CA19-9 The AUC used for the stage I-II pancreatic cancer diagnosis was 0.862 (95%CI: 0.772 to 0.926 P<0.001), The AUC for the combined diagnosis was 0.949 (95%CI: 0.880 - 0.985 P<0.001) ; In a healthy population and in patients with stage III-IV pancreatic cancer, the AUC of soluble T I I-IV pancreatic cancer in stage III was 0.927 (95%CI: 0.873 to 0.963 P<0.001), the AUC of serum CA19-9 used for the diagnosis of stage III-IV pancreatic cancer was 0.933 (95%CI: 0.881 to 0.968 P<0.001), the AUC for the combined diagnosis was 0.989 (95%CI: 0.956 to 0.999 P<0.001) . Conclusions:The combination of soluble Tim-3 and serum CA19-9 can improve the diagnostic rate of pancreatic cancer patients.
9.Application of WeChat-based PBL teaching method combined with micro-classroom in the teaching of contrast-enhanced gastrointestinal ultrasound imaging

Objective:To investigate the application value of WeChat-based problem-based learning teaching method combined with micro-classroom in the teaching of contrast-enhanced gastrointestinal ultrasound imaging for residents in standardized residency training.Methods:A total of 40 trainees who underwent the standardized residency training in the Department of Ultrasound, The First Affiliated Hospital of Chongqing Medical University from January 2019 to June 2021 were selected as the research objects. They were randomly divided into the experimental group (WeChat-based PBL teaching method combined with micro-classroom) and the control group (traditional teaching method). The teaching content was the diagnosis of gastrointestinal ultrasound imaging. After the theoretical learning and practice, assessments of contrast-enhanced gastrointestinal ultrasound imaging including theory and operating practice were performed to all students. SPSS 22.0 was used for t test and rank sum test.Results:After training, the theoretical test scores of the experimental group were higher than those of the control group [(93.09±2.31) vs. (90.63±2.26)], and the difference was statistically significant ( P<0.05). However, there was no significant difference between the two groups in the number distribution of students at all levels of operational practice ( P>0.05). Conclusion:The WeChat-based PBL teaching method combined with micro-classroom is beneficial to improve students' knowledge of contrast-enhanced gastrointestinal ultrasound imaging, and is worth being popularized and applied in clinical teaching.
10.Effect of Danggui Shaoyaosan on Improvement of Cognitive Ability of SAMP8 Mice and Its Mechanism via Regulating Ubiquitin Proteasome Pathway

ObjectiveTo investigate the mechanism of Danggui Shaoyaosan (DSS) in the improvement of the cognitive ability of SAMP8 mice with Alzheimer's disease (AD) via regulating the ubiquitin-proteasome pathway (UPP). MethodFifteen SAMR1 mice were used as a normal group, and 60 SAMP8 mice were randomly divided into a model group and DSS high, medium, and low-dose groups (57.6, 28.8, and 14.4 g·kg-1·d-1), with 15 mice in each group. Intragastric administration was conducted for eight continuous weeks. Place navigation and spatial capacity were evaluated by Morris water maze. Pathological structure changes in neurons in the hippocampal CA1 area was detected by hematoxylin-eosin (HE) staining. The protein expression levels of hippocampal β-amyloid protein(Aβ) and phosphorylation(p)-Tau were determined by immunohistochemical staining (IHC) and enzyme-linked immunosorbent assay (ELISA), and the mRNA and protein expression levels of hippocampal ubiquitin (Ub), ubiquitin ligase E3 (E3), 26S proteasome, ubiquitin carboxyl terminal hydrolase-1 (UCHL1), and UCHL3 were determined by real-time fluorescent quantitative polymerase chain reaction (Real-time PCR) and Western blot, respectively. ResultAs compared with the normal group, the escape latency was prolonged in the model group (P<0.05) with the reduced number of crossing platform quadrants and time ratio in the platform quadrant (P<0.05). The model group decreased neurons and condensed cell bodies in the CA1 area, and increased β-amyloid precursor protein (β-APP) and p-Tau positive cells (P<0.05). In the model group, the protein expression levels of Aβ and p-Tau were increased (P<0.05), the mRNA and protein expression levels of Ub were increased (P<0.05), and the mRNA and protein expression levels of E3, 26S proteasome, UCHL1, and UCHL3 were decreased (P<0.05). As compared with the model group, the escape latency was shortened in the DSS high and medium-dose groups (P<0.05) with an increased number of crossing platform quadrants and residence time ratio (P<0.05). The pathological changes in CA1 of each DSS group were significantly improved, and the number of β-APP positive staining cells decreased (P<0.05). The number of p-Tau positive staining cells decreased in the DDS medium and low-dose groups (P<0.05). The protein expression levels of Aβ and p-Tau in each DDS group decreased (P<0.05), and the mRNA expression level of Ub in each group decreased (P <0.05). The mRNA expression levels of 26S, E3, and UCHL3 in the DDS high and medium-dose groups increased (P<0.05), and the mRNA expression level of UCHL1 in the DDS medium-dose group increased (P<0.05). The protein expression level of Ub in each DDS group decreased, and the protein expression levels of 26S, E3, UCHL1+3 in the DDS high and medium-dose groups increased (P<0.05). ConclusionDSS can improve the cognitive ability of SAMP8 mice, and its mechanism may be related to the reduction of the abnormal deposition of Aβ and p-Tau via decreasing the expression of Ub and increasing that of E3, 26S, UCHL1, and UCHL3 in the UPP.
