1.Expression of PHB2 in breast cancer and its effect on proliferation, migration, and invasion

Objective :
To explore the expression of prohibitin2(PHB2) in breast cancer and its effect on the biological behaviors of tumor cells.
Methods :
Immunohistochemistry was used to detect the expression of PHB2 protein in breast cancer tissues and its relationship with clinicopathologic features. Breast cancer stable transient cell lines were constructed with knockdown and overexpression ofPHB2, respectively. The effects of PHB2 on cell proliferation, migration and invasion ability were detected by clone formation assay, scratch assay and Transwell assay. Western blot(WB) was used to detect the effects of PHB2 on the expression of epithelial-mesenchymal transition(EMT)-related markers, including E-cadherin, N-cadherin, Snail family transcriptional repressor 1(Snail) protein, Vimentin, and Claudin-1. The effect of PHB2 on tumorigenicityin vivowas detected by subcutaneous tumor formation assay in nude mice.
Results:
The result of immunohistochemical showed that PHB2 was highly expressed in breast cancer and the expression of PHB2 was significantly positive correlated with tumor size, human epidermal growth factor receptor-2(HER-2) status and proliferation index Ki-67 levels(P<0.05). Clone formation assay, scratch assay and Transwell assay revealed that knockdown ofPBH2significantly inhibited the proliferation, migration and invasion ability of breast cancer cells(P<0.01), while the overexpression ofPHB2significantly promoted cell proliferation, migration and invasion(P<0.01). The result of subcutaneous tumor formation experiment in nude mice revealed a significant decrease in tumor volume and weight in knockdownPHB2mice(P<0.000 1), whilePHB2overexpression tumors significantly increased in volume and weight(P<0.001).WB assay showed that the protein expression of epithelial marker E-cadherin increased, while the expressions of mesenchymal markers N-cadherin, Snail and Vimentin decreased significantly afterPHB2knockdown with them in control cells(P<0.01). The expression of Claudin-1 decreased, while the expressions of N-cadherin, Snail and Vimentin increased significantly inPHB2overexpression cells(P<0.05).
Conclusion
PHB2 is highly expressed in breast cancer and promotes multiple malignant biological behaviors in tumor cells, suggesting PHB2 may be a potential target for breast cancer diagnosis and treatment.
2.Study of the Application of Mobile Medical Technology in Construction of Grading Diagnosis and Treatment System.

The combination of mobile medical technology and the grading diagnosis and treatment system (GDTS) can stimulate the allocation of medical resources, reduce medical cost and improve public health significantly. Firstly we summarize development features of mobile medical technology in foreign and domestic market, then we study the application model of mobile medical application in GDTS with field research data and analyzes its advantage and shortage. Finally, we propose four measures for further developing mobile medical application in the GDTS:the government departments should formulate policies and industry standards of products as soon as possible to meet requirement of market; service providers should take the hospitals as core role to achieve mutual benefit and win-win situation; take the daily monitoring of chronic diseases as an entry point to build profitable business model; enhance publicity to promote public health awareness.

3.Group social skills training on the effects of improving psychiatric symptoms and disability and the family relationship for patients with schizophrenia

Objective To discuss the effect of group social skills training on improving psychiatric symptoms and disability and the family relationship in patients with schizophrenia.Methods 101 followup patients of rehabilitation with schizophrenia in mental health center of a top three hospital and in a community were chosen from June 2011 to August 2012.Based on the principle of voluntary,45 and 56 patients were grouped into the control group and the intervention group,respectively.The scores of psychiatric symptoms,mental disability and family relationship were compared between two groups by independent sample test.Results At the end of six months,the score of positive and negative symptoms and mental disability were lower in the intervention group,and there was significant difference in different measuring time,the interaction existed between the intervention and the measuring time.Meanwhile,the total score of family concern in the intervention group was higher than that in the control group,and there was significant difference in different measuring time,no interaction existed between the intervention and the measuring time.Conclusions Group training of social skills could improve psychiatric symptoms and the family relationship of patients with schizophrenia,reduce mental disability and promote recovery of patients.
4.Clinical significance of CT perfusion image on post-operative follow-up of glioma

Objective To assess the clinical value of multi-slice helical CT perfusion on the post-operative follow-up of glioma.Methods CT perfusion image was applied to 32 patients with glioma after operation.Various perfusion parameters,include cerebral blood volume(CBV),permeability surface(PS),relative cerebral blood volume(rCBV)and relative permeability surface(rPS)were measured on recurrent area,nonrecurrent area and normal cerebral area respectively.SPSS 12.0 statistical software was used.Independent t test was used to compare the differences.Results In the recurrent group(16 cases),the value of CBV in creased in 12 cases and was normal in the other 4 cases.The value of PS increased in all the cases of recurrent group.The value of rCBV exceeded 2.6 in 10 cases,among which the biggest value of rCBV was 4.01.The mean value of rCBV was 2.67 and the mean value of rPS was 6.32;In the non-recurrent group(16 cases),the value of CBV was nearly normal in 7 cases,increased in 2 cases and decreased in the remaining 7 cases.The value of PS was nearly normal in 11 case,decreased in 3 cases and increased in the remaining 2 cases.The values of rCBV were all less than 2.6 and the biggest value of rCBV was 2.14.The mean value of rCBV was 0.99 and the mean value of rPS was 1.42.The statistic significance was obtained for all the parameters when compared the recurrent group with the non-recurrent group(P<0.01)and with the normal cerebral area(P<0.01).When compared the non-recurrent group with the normal cerebral area,none of the parameters had statistic significance(P>0.05);The statistic significance was obtained for rCBV and rPS when compared the recurrent group with the non-recurrent group(P<0.01).Conclusion CT perfusion image is valuable in determining glioma recurrence after operation.
5.CT diagnosis of primary mediastinal seminoma

Objective To characterize the CT features of primary mediastinal seninoma. Methods CT scans of 10 cases with pathologically proved primary mediastinal seminoma were retrospectively reviewed.Results 9 of the 10 cases with the maximal dimension of 6. 5 ~ 18.2 cm( mean = 12. 3 cm) were located in the anterior mediastinum and 1 tumor was located in the middle mediastinum. All of the 10 tumors were solid masses and none of cavity, loculus, fat, calcification or fluid was detected. Areas of low density in the center of tumors were seen in 9 cases. Tumors encircling macrovascular or pericardium were seen in 7 cases. Chest wall invasion was detected in 2 cases. Enlarged lymph nodes in supraclavicular or mediastinum were identified in 4 cases. Lung diseases were found in 5 cases. Bilateral pulmonary metastasis was seen in 1 case. Pleural effusions were detected in 4 cases. Pericardial effusions were evident in 5 cases. Conclusion Most of the primary mediastinal seminomas are solid and large mass without calcification or fat. The tumors are usually located in the anterior mediastinum and areas of low density are usually seen in the center of tumors. Some tumors involve in adjacent structures. Seminoma has some features on CT.
