1.Application of pedicled transverse rectus abdominis muscle flap in reconstructing the breast immediately after breast cancer operation

		                        			Objective:To investigate the clinical effect of the transverse rectus abdominismuscle (TRAM) on reconstruction of the breast.Methods:The clinical data of 23 patients receiving TRAM breast reconstruction in our department from Jan. 2018 to Dec. 2019 were retrospectively analyzed.Results:The operation time of 23 patients ranged from 240 to 360 mins, andthe average time was about 300 mins. Intraoperative bleeding was about 120 to 200 ml, with an average of 170 ml. All the flaps survived successfully, but 2 cases were complicated with local fat necrosis. The postoperative period was between 6 and 12 months. No local tumor recurrence or metastasis was found inall patients during postoperative follow-up, and the breast shape was maintained in good condition.Conclusion:TRAM can make up for the regret of breast loss caused by breast cancer in female patients. 2.Cyclic RNA Molecule circ_0007766 Promotes the Proliferation of Lung Adenocarcinoma Cells by Up-regulating the Expression of Cyclin D1/CyclinE1/CDK4.

		                        			BACKGROUND:
		                        			Cyclic RNA (circRNA) is a new type of non-coding RNA (ncRNA) which is different from traditional linear RNA. More and more studies suggest that circRNA can be used as a biological marker of many malignant tumors and becomes a potential target for treatment. Therefore, searching for new molecular targets of lung adenocarcinoma from the circRNA will help to reveal the new mechanism of the occurrence and development of lung adenocarcinoma, and provide new ideas for clinical diagnosis and treatment. In this study, the biological function of circ_0007766, a highly expressed circRNA found in a screen of lung adenocarcinoma tissue, was verified and analyzed in vitro, so as to preliminarily explore the mechanism of circ_0007766 in promoting the proliferation of lung adenocarcinoma.
		                        		
		                        			METHODS:
		                        			The expression level of circ_0007766 in lung adenocarcinoma cells was detected by qPCR. Then siRNA was used to knock down the expression of circ_0007766. The effects of knockdown of circ_0007766 on proliferation, cell cycle and apoptosis of lung adenocarcinoma cells were detected by CCK8, scratch test, PI staining and Annexin V/PI double staining. In addition, the biological mechanism of circ_0007766 in lung adenocarcinoma was preliminarily studied by qPCR and Western blots.
		                        		
		                        			RESULTS:
		                        			The expression of circ_0007766 in lung adenocarcinoma cell lines was detected by qPCR. The expression of circ_0007766 was interfered in SPCA-1 cells. The proliferation and migration abilities of cells were inhibited. The cell cycle was arrested in G0/G1 phase, but the apoptosis was not affected. The deletion of circ_0007766 did not affect the expression of ERBB2, but influenced the mRNA and protein expression of Cyclin D1/Cyclin E1/CDK4.
		                        		
		                        			CONCLUSIONS
		                        			In vitro functional studies have shown that circ_0007766 may promote the proliferation and migration of lung adenocarcinoma cells. Further molecular mechanism studies have found that circ_0007766 can up-regulate the expression of Cyclin D1/Cyclin E1/CDK4, which are the key proteins of cell cycle, and thus promote the malignant proliferation of lung adenocarcinoma. From the perspective of circRNA, this study will provide new clues for the pathogenesis, development and prognosis of lung adenocarcinoma, and provide new target for clinical treatment.

4.Clinical effect of polymer-free sirolimus-eluting Nano stent on patients with coronary artery stenosis

		                        			Objective To study the safety and effectiveness of polymer-free sirolimus-eluting Nano stent in patients with unstable angina pectoris. Methods Three hundred and twenty-one patients with unstable angina pectoris were divided into Nano stent group(group A,n=157)and Endeavor resolute stent group(group B,n=164). The cardiovascular events were compared postoperative 12 months. The minimal intima cavity area and mini-mum bracket section area and neointimal area were compared postoperative 12 months by intravascular unltrasound (IVUS). Results There were 7 cases of cardiovascular events in group A and 6 in group B postoperative 12 months(P=0.727)and 2 patients in group A and 3 in group B were re-implanted stent because of restenosis post-operative 12 months(P=0.672). The neointimal area were(0.31 ± 0.11 mm2)in group A and(0.29 ± 0.12 mm2) in group B postoperative 12 months(P = 0.985). The minimal intima cavity area(P = 0.921)and the minimum bracket section area(P=0.934)were narrower postoperative 12 months than immediately after the operation in two groups. Conclusion With less cardiovascular events and being safe and reliable,the clinical effect of polymer-free sirolimus-eluting Nano stent implantation is similar to that of Endeavor resolute stent implantation.
		                        		
		                        		
		                        		
		                        	
5.Influence of classification of psychological guidance on anxiety and pain in endovascular interventional treatment of patients with cerebrovascular stenosis disease

		                        			Objective To discuss the effects of the classification of psychological guidance on the emotion and prognosis of patients with cerebrovascular stenosis disease undergoing intravascular interventional therapy and to investigate effective interventions.Methods A total of 101 patients with cerebrovascular stenosis disease who underwent intravascular interventional treatment from June 2014 to September 2016 were selected and divided into experimental group (n=52) and control group (n=49) using random number method. Patients in the control group accepted conventional preoperative education instruction, while patients in the experimental group received additional postoperative classified psychological guidance. The Hamilton Anxiety Scale (HAMA), pain scores, postoperative blood pressure changes and adverse reactions were compared between the two groups before and after surgery.Results The number of preoperative patients who were not anxiety, possible anxiety and anxiety were 5, 20, and 27 respectively in the experimental group, and were 3, 19, and 27 in the control group. There was no statistic difference between two groups (Z=0.444,P=0.657). The number of postoperative patients who were not anxiety, possible anxiety and anxiety were 29, 17, and 6 respectively in the experimental group, and were 14, 24, and 11 in the control group. There was a statistically significant difference between two groups (Z=2.710,P=0.007). There were significant differences in pain score, blood pressure, dysuria and insomnia between the two groups after operation (P<0.05).Conclusions The classification of psychological guidance can effectively stabilize the patients' psychology and emotion who underwent the cerebral vascular interventional operation, relieve the pain, help to control the blood pressure, reduce complications and adverse reactions, and improve the prognosis of patients.
		                        		
		                        		
		                        		
		                        	
6.Target in target radiotherapy for 90 patients with spinal metastases

		                        			Objective To analyze the result of dose-escalated radiotherapy vs conventional radiotherapy for spinal metastases. Methods Ninety patients (118 lesions) underwent radiotherapy..There were 52 patients (63 lesions) in target in target radiotherapy (TIT) receiving PTV 30~40 Gy, GTV 50~60 Gy/15~25 f. 38 patients (55 lesions) received PTV 30~40 Gy/10~20 f in the conventional radiotherapy. Results The overall responses were 90.4% and 81.6%,respectively.but no significant difference was found. There were 31 and 25 improved lesions, 28 and 20 steady lesions, 4 and 10 worsening lesions,in two goups, respectively. 1-year local control rates and survival were 84.7%vs 68.6%and 49.4%vs 32.2%, respectively. The significant differences were found in 1-year local control rates and survival. The main acute adverse reactions were mainly hematologic toxicities , no significant difference was found. Conclusion TIT for spinal metastases is a more safe and effective treatment, with better efficacy and fewer side-reactions.
		                        		
		                        		
		                        		
		                        	
7.Analysis of setup errors in helical tomotherapy for bone metastases

		                        			Objective To analyze the setup errors of bone metastases patients by tomotherapy with megavoltage CT (MVCT) and calculate the CTV-PTV margins.Methods 30 patients with bone metastases were enrolled.All patients received tomotherapy,fixed with body net and received MVCT scanning before radiation.The MVCT images were registered with the kilovoltage CT (kVCT) images,the setup errors of X (lateral),Y (vertical),Z (longitudina) and Roll (transverse profile rotation) were obtained according to the formula M =2.5Σ+0.7σ calculated CTV-PTV margin.Results 30 patients were received 494 MVCT images.The errors of systemic±random were (2.85±0.77) mm,(3.11±0.95) mm,(2.21±0.55) mm,and (0,55±0.24)° on X,Y,Z and Roll directions,respectively.The CTV-PTV margins were 3.64 mm,4.17 mm,and 2.86 mm on X,Y,Z directions,respectively.Conclusion The application of image-guided technology for bone metastases can correct positioning in time,which greatly reduces setup errors of the fractionated treatment,further improves the treat accuracy and has a positive value in guiding clinical radiotherapy.
		                        		
		                        		
		                        		
		                        	
8.Exploration on new assessment methods for cell biology among postgraduates

		                        			This paper summarized the assessment model reform practiced in the postgraduate cell biology course for several years and successfully obtain a new testing method based on research papers re-porting and questioning. Before the assessment, each student searched and selected one required research paper to read and make a PowerPoint reporting document. During assessment , some students were ex-tracted to speak and comprehensive score, which was regarded as student's final examination results was made by teaching group. The rest of the students listened to speeches , questioned and filled the questions on the record sheet.Teaching group reported a score , which was regarded as part of student's final exami-nation result based on questioning record sheets and printed PowerPoint documents.This method can well evaluate students' scientific thinking and strengthen the training of scientific thinking of postgraduates.
		                        		
		                        		
		                        		
		                        	
9.Anticancer activities of curcumin on human hepatocarcinoma cell line Sk-hep-1.

		                        			To study the anticancer activities of curcumin on human hepatocarcinoma cell line Sk-hep-1 and its related molecular mechanism which has not been elucidated. In the present study,we showed that curcumin inhibited proliferation of Sk-hep-1 cells in a dose-dependent manner through MTF assay. The effect of curcumin on apoptosis in Sk-hep-1 cells was investigated by DAPI staining and the various apoptosis was observed in hepatocarcinoma cell lines Sk-hep-1, HepG2 and Hep3B, but not in normal liver cell line Chang's liver with curcumin treatment. Cell cycle analysis results showed that curcumin treatment resulted in dramatic accumulation of Sk-hep-1 cells at the G0/G1 or G2/M phase. The effect of curcumin on the expression of anti-apoptosis genes (Survivin and BCl-xL) and drug resistance genes (DRG2 and MDR1) was studied by reverse transcription-polymerase chain reaction (RT-PCR). The expression of MDR1 mRNA was significantly decreased in Sk-hep-1 cells treated with curcumin, while no alterations in the amount of DRG2 and anti-apoptosis genes' mRNA levels were found. These results indicate that curcumin is able to inhibit proliferation and induce apoptosis in Sk-hep-1 cells and it may cause by down-regulating the expression of MDR1 mRNA.

10.The value of quantitative parameters of dynamic-enhanced MRI and the significance of the maximum linearity slope ratio in the differential diagnosis of benign and malignant breast lesions

		                        			Objective To find the effective quantitative parameters for the differentiation of the breast lesions using the post-processing of time.signal curve of 3D dynamic-enhanced MRI.Methods Thirty patients with 35 lesions underwent 3D dynamic-enhanced MRI and the time-signal cHIve was deduced.The four quantitative parameters including SImax,PH,Slope and SlopeR were analyzed in benign andmalignant lesions of the breast.Independent samples t test and rank sum test were used for the statistics.Results Seyenteen benign lesions and 18 malignant lesions were included in this study.The SImax(M)of benign and malignant lesions were 375.2 and 158.1,the 95% confidence intervals of SImax were 278.2-506.0 and 160.5-374.8.The PH(M)of benign and malignant lesions were 114.4 and 87.8,the 95% confidence intervals of PH were 73.7-196.5 and 71.3-162.9.The Slope(M) of benign and malignant lesions were 22.3×10-3 and 44.0×10-3,the 95% confidence intervals of Slope were 13.7×10-3-41.1×10-3 and 46.1×10-3-81.8×10-3.The Slope"(M) of benign and malignant lesions were 2.6 and11.4,the 95% confidence intervals of SlopeR were 1.9-3.4 and 9.8-14.5.There were no significant differences on SImax and PH between benign and malignant lesions(P>0.05).The significant differences existed on Slope(P<0.01)and SlopeR(P<0.01)between benign and malignant lesions of the breast.Conclusion SlopeR is a very effective parameter in t}le differential diagnosis of breast lesions.
