1.Research Progress of Traditional Chinese Medicine Compound in Prevention and Treatment of Type 2 Diabetes Based on Cell Signaling Pathway

As a chronic metabolic disease,type 2 diabetes poses a significant threat to human health with increasing incidence.An increasing number of studies confirm that the pathogenesis of diabetes is closely related with alterations in multiple cellular signaling pathways.Although numerous studies have reported that traditional Chinese medicine compounds prevent diabetes by modulating cell signaling pathways,asystematic review of the mechanism of action of traditional Chinese medicine compounds in modulating cell signaling pathways is still lacking.Therefore,this paper summarizes the research of type 2 diabetes prevention and treatment,which was found mainly related to the signaling pathways such as PI3K/AKT,AMPK,MAPK,NF-κB,PPAR,TGF-β.This family of signaling pathways can treat type 2 diabetes by inhibiting pancreatic islet cell apoptosis,protecting pancreatic β-cell function,ameliorating insulin resistance,inhibiting hepatic gluconeogenesis,promoting glycogen synthesis,attenuating inflammation,and resisting oxidative stress.At the same time,we analyze the problems in current research and the future development trend,in order to provide a scientific theoretical basis for the drug development and clinical application of traditional Chinese medicine compound in the prevention and treatment of diabetes.
2.Effect of Lymph Node Metastasis on Prognosis of Small Cell Lung Cancer with M1a Disease: A Study Based on SEER database

Objective To investigate the impact of lymph node metastasis on the survival of SCLC patients with M1a disease. Methods We retrospectively analyzed the medical records of 7027 SCLC patients with M1a disease from 2004 to 2015 in SEER database. The Kaplan-Meier method and log-rank test were used to estimate the OS in all N stage subgroups. Cox proportional hazard model was used to assess whether N stage was an independent risk factor for prognosis. Results The median OS of all patients was 7 months. Among all M1a patients, the patients without lymph node involvement (N0) had the best OS, followed by N1 stage patients; N2 and N3 stage patients had the worst OS (

As a large category of natural products widely present in traditional Chinese medicine, iridoid glycosides have multiple pharmacological activities. Recent researches suggest that iridoid glycosides mainly exist in the forms of original form, aglycone and a series of their Ⅰ and Ⅱ metabolites under the biotransformation effect, and their metabolites have been proved to have multiple pharmacological activities. The research progress on metabolism and metabolite activities of several iridoid glycosides would be reviewed in this article, to provide a theoretical basis for the further development and utilization of iridoid compounds and their metabolites.

4.Roles of 15-lipoxygenases in chronic myeloid leukemia

[ ABSTRACT] Tyrosine kinase inhibitors ( TKIs) are now advocated as the first-line treatment for chronic myeloid leukemia ( CML) , but facing resistance and relapse .Leukemia stem cells ( LSCs ) are leukemia-initiating cells as the source of resistance and relapse .It is therefore important to discover the molecular biomarker of LSCs for developing anti -LSC strategies in leukemic therapy .15-Lipoxygenase (15-LO) is a key enzyme in the pathway of arachidonic acid and plays an important role in the occurrence and development of CML , which is specifically required for chronic myeloid LSCs . This review summarizes the influence of 15-LO on the chronic myeloid LSC characteristics of marked survival , self-renewal, proliferation , differentiation and apoptosis .
5.The effect of Skp2 gene silencing on the proliferation and apoptosis of SPC-A-1 lung cancer cells

Objective To investigate the effect of Skp2 gene silencing on the proliferation and apoptosis of SPC-A-1 lung cancer cells .Methods Specific small hairpin RNA (shRNA ) targeting Skp2 gene was introduced into lung cancer cells by Lipo-fectamine 2000 and the positive clones were screened by G418 .The Skp2 mRNA and protein expression level of lung cancer cells were detected by RT-PCR and Western blot .M TT method and flow cytometry (FCM ) analysis were used to observe the effect of RNAi on proliferation of lung cancer cells .Cell apoptosis was analyzed by FCM .Results Transfected with Skp2 shRNA expression vector significantly reduced the expression of Skp2 protein in lung cancer cells .Inhibition efficiency was respectively (75 .3 ± 5 .1) , (70 .4 ± 3 .2) .P27 protein expression was increased significantly in lung cancer cells .The growth of lung cancer cells transfected with Skp2 shRNA was blocked ,with Gl phase cells increased and S phase cells decreased .The apoptosis rate of cancer cells was higher in Skp2 shRNA groups than in control groups .Apoptosis rates were (17 .5 ± 2 .8)% ,(15 .6 ± 3 .1)% in Skp2 shRNA groups .Conclusion Specific inhibition of the expression of Skp2 can slow down the growth of lung cancer cells ,increase cell apop-tosis .
6.Intercalated Combination of Chemotherapy and EGFR-TKIs versus Chemotherapy Alone in the First-line Treatment of Advanced Non-small Cell Lung Cancer:A Meta-analysis

Background and objective The combination therapy of chemotherapy and epidermal growth factor receptor-tyrosine kinase inhibitors (EGFR-TKIs) has attracted the attention of more and more investigators. The aim of this meta-analysis is to evaluate the clinical effcacy and safety of intercalated combination of chemotherapy and EGFR- TKIs versus chemotherapy alone in the ifrst-line therapy of advanced non-small cell lung cancer (NSCLC).Methods We retrieved the Co-chrane Library, PubMed, EMBASE, CBM, CNKI and Wanfang databases for randomized controlled trials which involved the intercalated combination of chemotherapy and EGFR-TKIs, and chemotherapy alone in the first-line treatment of advanced NSCLC. hTe progression-free survival (PFS), overall survival (OS), objective response rate (ORR), disease control rate (DCR), and adverse events were analyzed. hTe quality evaluation and cross-checked data were independently performed by two in-vestigators according to the Cochrane Systematic Reviews Handbook. hTe Stata 12.0 sotfware was used to conduct themeta-analysis.Results This study included 933 NSCLC patients from 6 RCTs. Themeta-analysis demonstrated that the intercalated combination of chemotherapy and EGFR-TKIs signiifcantly prolonged the PFS (HR=0.72, 95% CI: 0.53-0.98,P=0.037) of advanced NSCLC patients compared with mono-chemotherapy. However, there was no statistical difference in OS (HR=0.85, 95%CI: 0.72-1.01,P=0.060), ORR (OR=1.59, 95%CI: 0.86-2.95,P=0.142) and DCR (OR=1.09, 95% CI: 0.95-1.25,P=0.226) between the two groups. Further, the subgroup analysis showed that the intercalated combination markedly improved the PFS in female, adenocarcinoma, never smoking,EGFR mutant patients. In the aspect of safety, the main side effects of the interca-lated combination therapy were rash (OR=7.81, 95%CI: 3.74-16.34,P<0.001) and diarrhea (OR=2.73, 95% CI: 1.92-3.89, P<0.001).Conclusion hTe intercalated combination of chemotherapy and EGFR-TKIs signiifcantly prolonged the PFS in the first-line therapy of advanced NSCLC patients compared with mono-chemotherapy, and the main adverse events were toler-able rash and diarrhea. Together, the intercalated combination shows promising results, and more large-scale and high-quality RCTs are still needed.
7.The clinical value of lung function and blood gas analysis in early diagnosis and treatment of interstitial lung disease

Objective To evaluate the value of lung function and blood gas analysis in early diagnosis and treatment of inter‐stitial lung disease(ILD) .Methods The pulmonary function and blood gas of 26 cases of ILD patients were tested ,and indicator change was analyzed by paired t test .Results Detection comparison showed that the indicators were significantly improved :VC in‐creased 24 .6% (P<0 .01) ,FVC increased 18 .1% (P<0 .01) ,DLCO increased 25 .6% (P<0 .01);blood gas analysis of PaCO2 decreased 21 .2% (P>0 .05) ,PaO2 increased 8 .2 % (P>0 .05) .Conclusion Lung function and blood gas measurement are impor‐tant in the early diagnosis and treatment of ILD ,which can provide the necessary basis for clinical diagnosis ,treatment and progno‐sis .
8.Transfection of PDCD5 plasmid for promoting cisplatin-induced A549 cells apoptosis

Objective To observe the role of programmed cell death 5(PDCD5) gene combined with cisplatin for inducing the ap-optosis of human lung adenocarcinoma A549 cells and to investigate its possible mechanism .Methods The PDCD5 recombinant plas-mid was transiently transfected into A549 cells by lipofectamine .Its transfection efficiency was detected by RT-PCR .The expressions of trasfected PDCD5 protein ,Bcl-2 and Survivin protein were examined by Western bolt .The apoptosis of A549 cells by single PDCD5 and its combination with cisplatin was measured by the MTT method and the flow cytometry .Results PDCD5 recombinant plasmid was transfect into A549 cells successfully .The Western blot results showed that the expression of PDCD5 protein in the transfection recombinant plasmid group was higher than that in the blank control group and the transfection empty plasmid group ,while the expres-sion of Bcl-2 and Survivin protein was lower than that in the other two groups ,the difference was statistically significant (P<0 .05) . The MTT and flow cytometry results demonstrated that the cell apoptosis rate in the transfection recombinant plasmid group was higher than that in the blank control group and the transfection empty plasmid group (P<0 .05) .Conclusion PDCD5 gene may pro-mote cisplatin induced A549 cells apoptosis .
9.The influence shuxuetong on the membrane viscoelasticity of erythrocyte taken from patients with chronic pulmonary heart disease.

The present paper was aimed to explore the effect of Shuxuetong on the membrane viscoelasticity of erythrocyte taken from the acute phase patients suffering from chronic pulmonary heart disease. The membrane viscoelasticity of erythrocyte was taken from the acute phase patients suffering from chronic pulmonary heart disease. The changes of membrane viscoelasticity of erythrocyte after treated with shuxuetong were detected by micropipette aspiration technique. The results showed that the Shuxuetong of certain concentration could cause the decrease of membrane elastic modulus and viscous coefficients in acute phase patients suffering from chronic pulmonary heart disease. The study offers experimental evidences that the comprehensive treatment of pulmonary heart disease should involve the drug or measure to improve the erythrocyte deformability.

10.Effect of different doses of cyclophosphamide on drug-resistance of implanted human lung cancer in nude mice

Objective To explore the effect of different doses of cyclophosphamide(CTX) on the drug-resistance of implanted human A549 lung cancer in nude mice,and investigate the anti-tumor effect and toxicity.Methods BALB/c nude mice bearing A549 lung carcinoma were randomized into low-dose metronomic(LDM) CTX group,maximum tolerate dose(MTD) CTX group and normal saline group.The changes of tumor volume,weight and toxicity were observed.Expression of glutathione S-transferase-?(GST-?) and lung resistance-related protein(LRP) in the tumor were detected by immunohistochemistry SP method.Results In LDM CTX group,the tumor grew fast at first,then slowed down gradually and grew slowly for a long time,while in MTD CTX group tumor grew slowly at first,then it accelerated.Side effects of chemotherapy:the weight of nude mice was lost persistently in MTD CTX group,while lost insignificantly in LDM CTX group.No obvious toxicity was observed in LDM CTX group.GST-? and LRP expression in LDM CTX group were not significantly different from those in the control,but significance was seen in MTD CTX group compared with those in the control(P
