1.Study on protective effects and mechanism of Melastoma sanguineum Sims fruit extract on chemical liver injury in mice

		                        			Objective:To study the protective effects and mechanism of Melastoma sanguineum Sims fruit extract (MSE) on chronic chemical liver injury induced by ethanol, acetaminophen and carbon tetrachloride in mice; To discuss it mechanism.Methods:Totally 96 mice were divided into normal control group, ethanol model group, ethanol+bifendate control group and ethanol+MSE high-, medium- and low-dosage groups, APAP model group, APAP+bifendate control group and APAP+MSE high-, medium- and low-dosage groups, CCl 4 model group, CCl 4+bifendate control group and CCl 4+MSE high-, medium- and low-dosage groups, with 6 mice in each group. Except for the normal control group, the other groups were respectively prepared for the ethanol model, the APAP model and the CCl 4 model. The mice in the MSE high-, medium- and low-dosage groups were intragastrically administrated with 10, 5 and 2.5 g/kg of MSE, respectively; the bifendate control group was intraperitoneally injected with 15 mg/ml bifendate solution at 75 mg/kg; the normal control group was intraperitoneally injected with equal volume of normal saline/peanut oil solution once a day for 25 consecutive days. The levels of GPT, GOT and total bilirubin (TBIL) in serum were detected; the activities of SOD and GSH-Px and the content of MDA in liver tissue were detected; the mRNA expressions of TNF-α, IL-6, alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) were detected by qRT-PCR; the protein expressions of cytochrome P450 CYP1A1, CYP1A2, and CYP3A in liver tissue were detected by Western blot; the pathological changes of the liver tissue were observed by HE staining. Results:Compared with the corresponding ethanol, APAP and CCl 4 model groups, the serum GPT, GOT and TBIL levels of mice in the ethanol+bifendate control group and ethanol+MSE high- and medium-dosage groups, the APAP+bifendate control group and APAP+MSE high- and medium-dosage groups, and the CCl 4+bifendate control group and CCl 4+MSE high- and medium-dosage groups decreased ( P<0.01 or P<0.05), the activities of SOD and GSH-Px in the liver tissue increased ( P<0.01 or P<0.05), and the MDA level decreased ( P<0.01 or P<0.05), and the mRNA levels of IL-6 and TNF-α decreased ( P<0.01); the mRNA levels of ADH and ALDH in the ethanol+MSE high-, medium-, and low-dosage groups decreased ( P<0.01). Compared with the APAP model group, the expressions of CYP1A1 and CYP1A2 in the APAP+MSE groups increased ( P<0.01), and the expression of CYP3A protein decreased ( P<0.05); compared with the CCl 4 model group, the expressions of CYP1A1, CYP1A2 and CYP3A proteins in the CCl 4+MSE groups decreased ( P<0.05). Conclusion:MSE has a protective effect on chronic chemically-induced liver injury induced by ethanol, APAP, and CCl 4 in mice, and its mechanism may be related to antioxidant stress, inhibition of inflammatory response, and regulation of the expression of cytochrome P450-related enzymes.
		                        		
		                        		
		                        		
		                        	
2.Barriers and facilitators for exercise rehabilitation in patients with atrial fibrillation after radiofrequency ablation: a mixed study

		                        			Objective:To explore the barriers and facilitators of exercise rehabilitation in patients with atrial fibrillation (AF) after radiofrequency ablation.Methods:From March to August 2022, 285 patients with atrial fibrillation who underwent radiofrequency ablation within three months were recruited from the "Cloud Ward" of the Department of Cardiovascular of the First Affiliated Hospital with Nanjing Medical University by convenience sampling. The Cardiac Rehabilitation Barriers Scale (CRBS) and the Self-efficacy for Exercise Scale (SEE) were used to evaluate the barriers and self-efficacy of patients participating in exercise rehabilitation, respectively. Among them, 22 patients were subjected to semi-structured interviews to extract themes.Results:Among 285 patients with atrial fibrillation, 202 (70.88%) participated in exercise rehabilitation programs. Work/time conflicts, lack of cardiac rehabilitation knowledge, and health/physical function limitations affected the participation of patients in exercise rehabilitation ( P<0.05). Qualitative research extracted three themes that affected the patient's exercise rehabilitation, including capability, opportunity, and motivation. Conclusions:The participation of atrial fibrillation patients in exercise rehabilitation is influenced by multiple factors such as family responsibility, capability, motivation, and opportunity. Medical and nursing staff should comprehensively consider the above factors, develop targeted interventions from all aspects and multiple angles, so as to effectively improve the participation of patients in exercise rehabilitation.
		                        		
		                        		
		                        		
		                        	
3.Sedation and analgesia effects of dexmedetomidine on multiple trauma patients with mechanical ventilation

		                        			Objective To explore the sedation and analgesia effect of dexmedetomidine (DEX) in patients with multiple trauma during mechanical ventilation.Methods Eighty cases of multiple trauma patients under mechanical ventilation treated from September 2016 to March 2017 were analyzed by retrospective case-control study.There were 58 males and 22 females with an age range of 18-60 years (mean,41.87 years).The injury severity score (ISS) was (18.45 ±4.53)points.The patients were treated with sedation and analgesia,and they were divided into two groups according to the sedative drugs.DEX composite tartaric acid butorphanol were used in 40 patients as DEX group.Midazolam composite tartaric acid butorphanol were used in 40 patients as Midazolam group.The degree of sedation score (Ramsay score) were compared between groups.The time from initial drug use to effective sedation achievement,daily wake-up time,mechanical ventilation duration,emergency intensive care unit (EICU) time,dosage of tartaric acid butorphanol,heart rate,blood pressure changes,and incidence of delirium were compared.Results (1) Two groups could both reach the target of sedation.DEX group had shorter daily wake-up time,shorter mechanical ventilation time and shorter length of EICU stay compared with Midazolam group (P < 0.05).(2) The time to achieve satisfied sedation after initial usage in Midazolam group was shorter than that in DEX group (P < 0.05).(3) DEX group had smaller tartaric dosage of acid butorphanol,and lower incidence of delirium compared those in Midazolam group (P < 0.05).(4) The comparison of systolic blood pressure,diastolic blood pressure,heart rate within either group showed significant difference before and after sedation (P < 0.05),but had no significant difference between two groups (P > 0.05).Conclusions For multiple trauma patients with mechanical ventilation,DEX can attain sedation and analgesia and shorten daily wake-up time,mechanical ventilation time,and length of hospital stay.DEX can reduce the dosage of analgesic (butorphanol) and the incidence of delirium.Blood pressure and heart rate are associated with small variations before and after sedation.
		                        		
		                        		
		                        		
		                        	
4.The Microtox-Based Fast Test of Comprehensive Toxicity of Safflower Injection

		                        			This study aimed at exploring a new method--microtox technology to evaluate the comprehensive toxicity of safflower injections from different drug manufacturers.In the experiment,we investigated the characteristic parameters of vibrio fischeri (CS234) under different conditions,and then locked the optimum parameters for safflower injection assay:recovery liquid volume 0.9 mL in freeze-dried vial,50 μL bacteria suspension in each sample,and the detection time of 10 mins after adding the bacteria suspension.Under this optimum detection condition,we found that the EC50 values were 3.36%,5.58% and 4.33%,being significantly different between 3 representative drug manufacturers (P < 0.05).In conclusion,it was implied that the biological detection standards of safflower injections need improving,while the microtox-based fast test of the comprehensive toxicity of safflower injection showed a favorable and prospective application to controlling the product qualities of different manufacturers.
		                        		
		                        		
		                        		
		                        	
5.Recognizing research fronts and trends of synthetic biology

		                        			Objective To identify research fronts of synthetic biology by mapping knowledge domains .Methods The research articles were clustered by co-cited analysis and bibliographic coupling while research domains of synthetic biology were mapped using software VOSviewer .Results There were seven research clusters by co-cited analysis and 32 clusters by bibliographic coupling .The articles published in the last two years have changed the structure of research domains .Con-clusion Bibliographic coupling is better at recognizing research fronts .Synthetic bioengineering , RNA expression , biology bone, biology library,and biology parts are research fronts of synthetic biology .
		                        		
		                        		
		                        		
		                        	
6.Metrology and visualized analysis of H5N1 virus:a research of literature

		                        			Objective To analyze domestic and foreign papers on H5N1 virus research using literature metrology and visualized knowledge mapping.Methods Pajek, Bibexcel,and VoSviewer software were used based on Web of Knowledge platform.Results Study on H5N1 virus has been a research focus wide since 2005 and a series of institutions and investi-gators have appeared, with US being the leader in this area and followed by China.The most popular topics of studies on H5N1 included its virology, epidemiology, basic research, anti-virus drugs and vaccines.Conclusion Studies on H5N1 have received more attention, which suggests that more efforts have to be made in China in terms of communication and co-operation with key institutions and famous researchers so as to achieve more success in key areas.

		                        			Objective To explore the development of the anthrax research field in order to provide information and strategies for anti-bioterrorism in China.Methods Pajek, Vosviewer, Bibexcel running on Web of Knowledge platform, and Excel running on CSCD platform were used.Results According to the results of anthrax literature metrology,the USA is by far the leader in research while the Academy of Military Medical Sciences, Chinese Academy of Sciences and other in-stitutions are our main anthrax research institutions in China, but there is a gap between China and international research institutions.Conclusion We should strengthen anthrax research, improve the overall research strength, and provide the necessary protection to respond to potential bioterrorism incidents.
		                        		
		                        		
		                        		
		                        	
8.Organization and implementation of aeromedical evacuation of US patients with Ebola virus infection from West Africa

		                        			In 2014,serious Ebola fever epidemic broke out in West Africa .Afterwards, the US transported two Ebola patients from Liberia by air, which aroused world wide concern .During the aeromedical evacuation , there were lots of tech-nical problems and management procedures involved .First, the US government toke emergency measures for the patients in a local hospital ,which made evacuation possible .Secondly , the US CDC and other government departments coordinated this matter during and after the evacuation .Finally,upon arrival at home the two patients received careful isolation and treat-ment.Thus, the successful aeromedical evacuation is instructive for us .

		                        			The outbreak of Ebola hemorrhagic fever in West Africa in February 2014 is the worst one in the history of the disease.Medicines and vaccines for Ebola have attracted special attention .This study utilizes Cortellis database of Thom-son Reuters and knowledge about Ebola virus to analyze current R&D of medicines and vaccines for Ebola in order to offer references for research and development .Study findings shows that there are mainly 5 important categories:vaccines, neu-tralizing antibodies , small-molecule anti-Ebola drugs , RNAi drugs and nucleotide drugs .In each category , there are prom-ising products under development that are worthy of attention .
